Histopathologic Features in Animal Model of Atopic Dermatitis Induced by Topical Application of Oxazolone

  • Animal models of atopic dermatitis (AD) are widely used to investigate therapeutic effects of candidates for AD. However, the characteristics of each model are not fully understood. This study was designed to compare the animal models of dermatitis induced by dinitrofluorobenzene (DNFB) and oxazolone (Ox). We investigated the effects of DNFB and Ox on skin thicknesses and weights as well as skin lesions associated with AD such as scale, crust and erythematous eruption, and histopathological changes such as hyperkeratosis, dermal and epidermal hyperplasia and immune cell infiltration in inflamed tissues. Multiple application of 0.5% Ox onto the skin increased skin thickness and weight compared to those of DNFB treated mice, as well as those of normal mice. In addition, topical application of DNFB induced marked scale, crust and erythematous eruption, while Ox induced erythematous eruption and mild scale and crust. Histopathological examination revealed that 0.5% Ox induced marked hyperplasia in the dermis and epidermis, large vesicles, spongiotic changes, mild hyperkeratosis and immune cell infiltration in balb/c mice. These data suggest that multiple applications of Ox can induce chronic AD like dermatitis in balb/c mice.

마우스, 랫트 및 토끼에서 paraquat 중독의 병리학적 관찰 (Pathological findings of paraquat poisoning in mice, rats and rabbits)

  • This study was carried out to investigate the pathological changes with paraquat(1.1'-dimethyl-4.4'-dipyrildiylium dichloride) administered by intraperitoneally, orally, skin applied in mice, rats and rabbits. Results were obtained as follows; In 2 days after paraquat administration clinical signs anorexia, depression, tachypnea, and tachycardia, respiratory failure, coma and death were observed in mice, rats and rabbits. Compared toxicity of paraquat with mouse were observed toward to paraquat that resistance was strong than rats and rabbits. Also, mortality of skin application were found the low than intraperitoneal and high than oral administration. In the case of gross observation were appear lips moisture in orally administered rats and rabbits by skin application. Lung of all laboratory animals were observed congestion and haemorrhage, swelling or atrophy. In the case of microscopic findings were severe congestion and haemorrhage, interstitial pneumonia of lung. Congestion and haemorrhage of liver, congestion and haemorrhage, renal tubule epithelium necrosis of kidney were observed in mice, rats and rabbits. Skin application group of mice, rats and rabbits showed infiltration of inflammatory cells and folliculitis of epidermis and dermis. Also, in oral administration group showed congestion and haemorrhage, tachment, necrosis of alimentary tract mucosa.

  • Skin color reappearance problem in color processing system is necessary to transform a specific color sand improve color reappearance quality as a reference color. The skin color has been situated as an important memory color not only in our lives but also in color application systems such as TV. Thus, skin color reappearance problem is more important than other color processing problem. In this paper, we propose a skin color reappearance algorithm for color enhancement in TV which use phase detector to detect the skin colors at real-time from 3.58 MHz color burst signal and color signal, comparators to discriminate the types of skin color, and micom to reprodece standard skin colors for races.

  • Preventive effect of red ginseng saponin on experimentally-induced hyperkeratosis was investigated by ultrastructural observation, skin weight and epidermal lipid analysis. Hexadecane increased skin weight per unit area and epidermal lipids, free fatty acids, cholesterol and triglyceride in guinea pig skin. Topical application of ginseng saponin reduced these hyperkeratotic responses regradless of the concentration and the purity of ginseng saponin. Ultrastructurally, lipids and empty space-containing multiple horny cells were piled and nuclear remnants, desmosome, desmosomal bodies, tight junction were shown in the stratum corneum of hexadecane-treated skin.

  • There have been various inflammatory skin disorders in humans including atopic dermatitis, eczema and psoriasis. Although some drugs have been used for these disorders, there is an urgent need for safer and more effective topical anti-inflammatory agents. Plant flavonoids possess anti-inflammatory activity and some of them have multiple pharmacological mechanisms, inhibition of eicosanoid metabolizing enzymes, histamine release and/or down-regulation of pro inflammatory gene expression. These properties of flavonoids may be suitable for treating chronic skin inflammatory disorders. Especially, wogonin, some prenylated flavonoids and biflavonoids have a strong potential as new anti-inflammatory agents by topical application.

  • The skin color model is a very important concept in face detection, face recognition and face tracking. Usually, this model is obtained by estimating a probability density function of skin color distribution. In many cases, it is assumed that the underlying density function follows a Gaussian distribution. In this paper, a new method for non-parametric estimation of the probability density function, by using feed-forward neural network, is used to estimate the underlying skin color model.   • A new capsaicin analog modified with 4-hydroxyl and alkyl chain of capsaicin was a very potent antiinflammatory analgesic drug and may be clinically useful for those who have rheumatoid arthritis, diabetic neuropathy and cancer. The purpose of this study was to investigate histopathology after short and long term application of poloxamer-based gels, and percutaneous absorption of various topical formulations. Poloxamer-based gel was prepared by cold method using poloxamer 407. The poloxamer gels was applied to dorsal sites of hairless mouse skin during one week or one month for the evaluation of skin irritation. The applied site was then sectioned for histopathologic examination. The topical formulations were also prepared using CMC, HPMC, MC, carbopol and glycerylmono stearate. Skin variation of poloxamer gels was studied using excised hairless mouse, rat, hamster and human penis skin. Franz-type diffusion cells were used far skin penetration of drug against receptor phase filled with about 10$m\ell$ of 0.9% saline solution kept at 32$^{\circ}C$. The concentration of drug was determined by the reverse phased C18, Symmetry HPLC with fluorometeric detector. No skin erythema was observed after dorsal application of poloxamer-based gels for one week or one month. No histopathologic changes was also examined, suggesting no skin toxicity of poloxamer-based gels. The order of flux rate was HPMC > MC ( CMC > poloxamer >> glycerylmono stearate ( carbopol. There was a skin variation of poloxamer gels. The flux rate of poloxamer gels was highest in case of hairless mouse followed by rat, human and hamster skin. 가미대황고(加味大黃膏)의 멜라닌세포 활성억제가 자외선 조사로 인한 피부 손상 완화에 미치는 영향 (The effect of Daehwanggogasangbakpi(大黃膏加桑白皮) on the skin damage induced by ultraviolet irradiation)

  • Objective : As a result of increasing amount of ultraviolet ray, skin problems including sunburn, rapid skin aging, melanoma, and even skin cancer continue to rise. In the present study, the effect of oriental herbal extract, Daehwanggo(大黃膏,DH) and Daehwanggogasnagbakpi(大黃膏加桑白皮,DS), as external application, on the skin damage, was investigated. Methods : 30 mice were equally distributed into 3 groups : control, UVB-control and UVB-irradiated and DS-treated group. Also mouse melanoma cell lines were cultured. Tyrosinase inhibition was measured to analyze the UN-protection effect. Melanogenesis in the UV-irradiated melanoma cell lines was compared in DS-treated cell line and control cell line. Sample skin from the ear tissue of the 3 groups were analyzed to observe the inflammatory response, T cell differentiation, apoptosis of keratinocytes. Results : The tyrosinase was more significantly inhibited in the DS group compared to DH group. Antioxidative effects was more prominent in DS group when superoxide dismutase was measured. Both the DS- and DH-treated cell lines showed significantly reduced melanogenesis. The reduction of external skin damage including erythematous papule, eczema, keratinocyte, pyopoiesis was observed in the DS- and DH-treated sample cells. In terms of the effect on the skin damage, sunburn cell, activated skin mast cells, secretion of IL-12, manifestation of HSP70, hyperplasia of epithelial cells, MMP-9 and destruction of the collagen were all significantly improved in the DS-treated sample cells. Melanin cells and the apoptosis in the melanoma cell line were decreased. Conclusion : DH and DS were traditionally applied externally for the scald in the oriental medicine. The present study elucidated the possibility of herbal extracts to be used as ultraviolet protectives. Further investigations are needed to assure the clinical application.

동종유래각질세포(Cultured Allogenic Keratinocytes, Kaloderm®)를 이용한 부분층 피부 결손의 치료 (Treatment of Partial Thickness Skin Defect with Cultured Allogenic Keratinocytes (Kaloderm®))

  • Purpose: Grafting with autograft skin remains the most effective method for treating skin defects. When insufficient donor sites are present or patients are afraid of the operation, a skin graft is impossible. Cultured allogenic keratinocytes speed wound healing by providing cover and by producing growth factors and extracellular matrix protein. We report an application of cultured allogenic keratinocytes ($Kaloderm^{(R)}$, Tegoscience, Seoul, Korea) in the treatment of an acute partial thickness skin defect. Methods: From March 2005 to January 2006, 20 patients with a partial thickness skin defect were treated with cultured allogenic keratinocytes. The wound was covered with a sheet of cultured allogenic keratinocytes and ointment with $Bactigras^{(R)}$ gauze. The wound was inspected every two or three days. We regarded completion of epithelialization as wound healing. Results: The mean period between time of injury and time of $Kaloderm^{(R)}$ application was 7.5 days. The time taken from application of $Kaloderm^{(R)}$ to complete closure of the wounds was 7.2 days. Conclusion: In view of the favorable outcome, cultured allogenic keratinocytes are safe and effective biologic dressing materials for use in the treatment of open wounds.

질려자 추출물의 피부 볼륨 증진 및 주름개선 효과 (Skin Volume Augmentation and Anti-wrinkle Effects of Tribulus terrestris Fruit Extract)

  • Aging of the face is mainly related to the features that are sagging or loss of elasticity of the skin by reducing the volume around the eyes or cheek. Intrinsic aging can be seen to cause thinner dermis, reduction of extracellular matrix and subcutaneous fat. This study was carried out to investigate the skin volume augmentation and anti-wrinkle effects of Tribulus terrestris fruit extract. Skin anti-aging effect of Tribulus terrestris fruit extract was evaluated by using lipid accumulation, expressin of type I procollagen and elastin in preadipocytes and human dermal fibroblasts. Tribulus terrestris fruit extract augmented preadipocytes differentiation about 56% at 100 µg/mL. The type I procollagen and elastin were increased about 35% and 25% by treatment 20% Tribulus terrestris fruit extract, respectively. The clinical study also showed that skin sagging, skin elasticity, and dermal density improved without adverse effect following 4 week application of cream containing 2% Tribulus terrestris fruit extract. We suggest that Tribulus terrestris fruit extract can have the good possibility as skin volume augmenting, skin elasticity and wrinkle improving agent.
