Antigenotoxicity of Quercetin and Its Glycosides Against Benzo(a)pyrene-induced Genotoxicity (퀘르세틴 및 퀘르세틴 배당체들의 벤조피렌에 대한 유전독성억제효과)

  • In order to compare the suppressive effect of quercetin and its glycosides, such as quercitrin (quercetin-3-rhamnoside), isoquercitrin (quercetin-3-glucoside), hyperin (querceti n-3-galactoside)and rutin (quercetin-3-rhamnosyl glucoside), on the genotocicity by benzo(a)pyrene(B(a)P), in vitro sister chromatid exchange(SCE) test using mouse spleen lymphocytes and in vivo micronucleus test using mouse peripheral blood were performed. B(a)P-induced SCEs in vitro were slightly decreased by the simultaneous treatment of quercetin and its glycosides, although there was no significant decrease. On the other hand, MNU induced micronucleated reticulocytes(MNRL7s) in vivo were significantly decreased with a dose-dependent manner in all compounds tested. However, there were no differences between quercetin aglycone and glycosides in the suppressive effects under experimental condition of this study. To elucidate, the action mechanism of quercetin aglycone and its glycosides against B(a)P-induced genotoxicity, the assay of DNA binding with B(a)P was studied. Quercetin aglycone and its glycosides inhibited B(a)P metabolism in the presence of S-9 mix and decreased the B(a)P/DNA binding in the calf thymus DNA with S-9 mix. These results suggest that antigenotoxicity of quercetin antiglycosides on B(a)P-induced genotoxicity is due to decrease of DNA binding with B(a)P through the inhibition of metabolism with B(a)P in the calf thymus DNA. Therefore, quercetin and its glycosides may act as an antigenotoxicity agent and may be useful as a chemopreventive agent of polycyclic aromaic hydrocarbons like B(a)P.

Hop2 and Sae3 Are Required for Dmc1-Mediated Double-Strand Break Repair via Homolog Bias during Meiosis

  • During meiosis, exchange of DNA segments occurs between paired homologous chromosomes in order to produce recombinant chromosomes, helping to increase genetic diversity within a species. This genetic exchange process is tightly controlled by the eukaryotic RecA homologs Rad51 and Dmc1, which are involved in strand exchange of meiotic recombination, with Rad51 participating specifically in mitotic recombination. Meiotic recombination requires an interaction between homologous chromosomes to repair programmed double-strand breaks (DSBs). In this study, we investigated the budding yeast meiosis-specific proteins Hop2 and Sae3, which function in the Dmc1-dependent pathway. This pathway mediates the homology searching and strand invasion processes. Mek1 kinase participates in switching meiotic recombination from sister bias to homolog bias after DSB formation. In the absence of Hop2 and Sae3, DSBs were produced normally, but showed defects in the DSB-to-single-end invasion transition mediated by Dmc1 and auxiliary factors, and mutant strains failed to complete proper chromosome segregation. However, in the absence of Mek1 kinase activity, Rad51-dependent recombination progressed via sister bias in the $hop2{\Delta}$ or $sae3{\Delta}$ mutants, even in the presence of Dmc1. Thus, Hop2 and Sae3 actively modulate Dmc1-dependent recombination, effectively progressing homolog bias, a process requiring Mek1 kinase activation.

A Multilevel Approach to Sibling Warmth and Conflict among Korean Young Adults by Gender: Roles of Sibling and Parent-Child Characteristics (청년의 형제자매 온정 및 갈등 관련 요인 성별 다층모형 분석: 형제자매 및 부모자녀 특성을 중심으로)

  • The purpose of this study was to examine which sibling and parent-child characteristics were associated with siblisng warmth and conflict among Korean young adults. The data came from an online survey of Koreans aged 19-34 years who had at least one sibling and two living parents. We conducted a multilevel regression analysis using data from 193 men with 305 siblings and 203 women with 286 siblings. The results showed that, for men, the levels of warmth were higher when the sibling was younger, when they received more frequent financial and instrumental support from the sibling, and when maternal favoritism was relatively lower. Women reported higher levels of warmth when the sibling was a sister, when they provided more frequent support for the sibling, when they had a more affectionate relationship with the mother, and when paternal favoritism was lower. As for sibling conflict, the number of siblings was positively associated with the levels of conflict among both men and women. Maternal affection was negatively related with conflict for men, and paternal favoritism was positively related to conflict for women. Interestingly, women's sibling warmth and conflict were both higher when the sibling was a sister, which meant that relationships between sisters tended to be ambivalent. This study contributes to our understandings of Koreans' sibling relationships in young adulthood.

A Case of Adult-onset Type II Citrullinemia Confirmed by Mutation of SLC25A13 (SLC25A13 유전자 돌연변이로 확진된 성인형 제 2형 시트룰린혈증 1례)

  • Adult-onset type II citrullinemia (CTLN2) is characterized by episodes of neurologic symptoms associated with hyperammonemia leading to disorientation, irritability, seizures, and coma. CTLN2 is distinct from classical citrullinemia, which is caused by a mutation of the argininosuccinic acid synthetase (ASS) gene. The serum citrulline level is elevated, while the activity of ASS in liver tissue is decreased. CTLN2 is known to have a poor prognosis if the proper treatment is not taken. We reported a female aged 37 years who developed recurrent attacks of altered consciousness, aberrant behavior, and vomiting. We initially suspected the patient had CTLN2 because of the signs of hyperammonemic encephalopathy, such as altered mentality, memory disturbance, and aberrant behaviors provoked by exercise-induced stress and excessive intravenous amino acid administration. Through her peculiar diet preferences and laboratory findings that included hyperammonemia and citrullinemia, we diagnosed the patient as CTLN2, and SLC25A13 sequencing revealed known compound heterozygous mutations (IVS11+1G>A, c.674C> A). Her parents were heterozygous carriers, and we identified that her older sister had the same mutations. The older sister had not experienced any episodes of hyperammonemia, but she had peculiar diet preferences. The patient and her sister have been well with conservative management. When considering the clinical course of CTLN2, it was meaningful that the older sister could be diagnosed early in an asymptomatic period and that preemptive treatment was employed. Through this case, CTLN2 should be considered in adults who present symptoms of hyperammonemic encephalopathy without a definite etiology. Because of its rare incidence and similar clinical features, CTLN2 is frequently misdiagnosed as hepatic encephalopathy, and it shows a poor prognosis due to the lack of early diagnosis and proper treatment. A high-carbohydrate diet, which is usually used to treat other urea cycle defects, can also exaggerate the clinical course of CTLN2, so proper metabolic screening tests and genetic studies should be performed.

A Study on the North Korean's Modern Adaptation of the Classic Folktale (설화 <해와 달이 된 오누이>에 대한 북한의 현대적 수용 방식 고찰)

  • The North Korean animation is a puppet movie that is adapted The Brother and Sister Who Became the Sun and the Moon, a traditional Korean lore. The quality of this animation is acknowledged because of not only North Korea's considerably advanced animation technology but also the animation's retention of the folklore's traditional essence rather than intention to disseminate ideological propaganda. Nevertheless, the animation reveals the trasformation of its original purpose from general educative intentions for children to the educative concept of salvation by heaven is replaced by salvation by people and cultural education insteadof salvation by heaven. The appearance of the hero Jangsoe is the key adaptation of this animation, and it suggests the main principal of salvation lies in man rather than in heaven. Such adaptation complies with the requirements of children's literature suggested by the North Korea's literary history office. Furthemore the hero Jangsoe as the examplary figure of revolutionary self-reliance ideology and as a leader. Theory of self-reliance literature stipulates that children's literature is used for ideological education that develops people to be successors of revolutionary feats and become active workers for the construction of socialism and communism, therefore it is possible to understand the purpose of the adaptation to reflect the educational aims. This study investigates the change in meaning form the original folktale through such adaptation, and highlights problems related to limiting the meaning implied in "heaven's salvation" in the original story only to the vague meaning of religious hope. This vague implied meaning is considered as "an awareness activity to examine their own existence in the universe". With regard to this, the concept of heaven's salvation that is prevalent in the classic stories can be interpreted as a positive self-belief that enables the use of rationality in any helpless situation that cannot be understood with existing empirical knowledge. It considers that heaven expresses the power that exists in the human mind through self-viability and self-belief. This creates the power of reason in the character to fight against the evil disguised as the mother, in the absence of the real mother.

New record of the red algae, Halarachnion parvum (Gigartinales) and Champia lubrica (Rhodymeniales), from Korea

  • We report the first finding of Halarachnion parvum and Champia lubrica from Korea based on morphology and the plastid rbcL sequence analyses. H. parvum occurs in the subtidal zone of Munseom, the southern part of Jeju. Thalli have short stipe, and elliptical to ovate fronds with marginal proliferations of up to 3 cm in height. H. parvum has zonately divided tetrasporangia and cystocarp immersed under the cortical layer. Champia lubrica appears in Namhae, Gyeongnam and Seopseom, Jeju. Thalli are erect, irregularly branched, terete, obtuse apex, up to 3-5 cm high, and have tetrahedrally divided tetrasporangia. Molecular analyses of the plastid rbcL gene reveal that two species are clearly separated from other species of their respective genera. H. parvum is sister with Halarachnion latissimum in 3.1-3.2% sequence divergence, and C. lubrica is closely related to the sample from Japan with 0.2% sequence divergence.

  • A new species of gobiid fish Acentrogobius pellidebilis is described from specimens collected in the southern and western coasts of Korea. The new species is distinguished from its apparant sister species, A. pflaumi, by 7~8 predorsal scales and the 5 darker blotches arranged in a lateral line on body.

First Record of Ulva torta (Ulvales, Chlorophyta) in Korea

  • A marine ulvalean species (Chlorophyta) was collected from Imgok, which is located on the eastern coast of Korea. This species is morphologically characterized by distromatic, filiform to strap compressed or tubular thallus. Many branches were found near the base, but lacked proliferations. Cells were longitudinally aligned in the younger part of the branch and were disordered in the older part of the branch. A cap-like parietal chloroplast with one or two pyrenoids was contained in each cell. In a phylogenetic tree based on ITS and rbcL sequences, this species was nested in the same clade as Ulva torta and U. clathratioides from Australia, but formed a sister clade to U. torta from Japan. However, the genetic divergence between them was included in an intraspecific variation range within Ulva. This finding suggests that U. clathratioides should be reduced to a synonym of U. torta. Accordingly, the Korean alga was identified as U. torta based on the morphological and molecular data. This investigation is the first record of U. torta in the Korean marine algal flora.

  • Subjects included Hyomin Kim and Eunha Sohn (fictitious names) of Purum Class (a class for 5-year-olds) at 'H' Kindergarten. Subjects were selected after interviews with teachers and observation by the researcher. This study examined communication with classmates focusing on spontaneous dialog situations and functions. Results showed that Hyomin and Eunha exercised power in the art group and the math groups, respectively. The power exercise processes comprised a variety of behaviors including : to become similar with each other; to assign an exclusive seat; to act as an elder sister; to allow to do something; to keep a friend ata distance; to defend oneself; to threaten or persuade with a diversity of linguistic strategies; to teach like a teacher.

Monostroma alittorale, a marine green algal species newly recorded in Korea

  • A marine green algal species (Chlorophyta) was collected from the eastern coast of Korea. It is morphologically characterized by monostromatic thallus, usually undulate and entire margins, cap-like chloroplast and several pyrenoids per cell. In a phylogenetic tree based on molecular data, the Korean alga nests in the same clade as Monostroma alittorale originally described from Japan, as a sister clade of M. grevillei from France. The genetic distance for ITS(Internal Transcribed Spacer) sequences among Monostroma species ranges from 2.3% to 38.2%. The value between the Korean entity and M. alittorale was calculated as 0.01%, considered to be intraspecific divergence. This Korean entity is identified as Monostroma alittorale based on morphological and molecular analyses. This is the first record of M. alittorale in Korea.
