Manila clam, Ruditapes philippinarum Cathepsin D: Molecular analysis and immune response against brown ring disease causing Vibrio tapetis challenge

  • Cathepsins are lysosomal/cysteine proteases belong to papain family (C1 family) that is involved in intracellular protein degradation, antigen processing, hormone maturation, and immune responses. In this study, member of cathepsin family was identified from Manila clam (Mc-Cathepsin D) and investigated the immune response against brown ring disease (BRD) causing Vibrio tapetis challenge. The identified Mc-Cathepsin D gene encodes characteristic features typical for the cathepsin family including eukaryotic and viral aspartyl protease signature domain and two highly conserved active sites ($^{84}VVFDTGSSNLWV^{95}$ and $^{270}IADTGTSLLAG^{281}$). Moreover, MC-Cathepsin D shows higher identity values (-50-70%) and conserved amino acids with known cathepsin D members. Transcriptional results (by quantitative real-time RT-PCR) showed that Mc-Cathepsin D was expressed at higher levels in gills and hemocytes than mantle, adductor muscle, foot, and siphon. After the V. tapetis challenge under laboratory conditions, Mc-Cathepsin D mRNA was up-regulated in gills and hemocytes. Present study indicates that Mc-Cathepsin D is constitutively expressed in different tissues and potentially inducible when infecting BRD by V. tapetis. It is further suggesting that Mc-Cathepsin D may be involved in multiple role including immune response reactions against BRD.

Effects of Temperature and Salinity on Development of Sea Peach Halocynthia aurantium (붉은멍게 Halocynthia aurantium 발생에 관한 수온 및 염분의 영향)

  • The solitary ascidian, Halocynthia aurantium, which is commonly called the sea peach because of its coloration and general shape, is a valuable organism of benthic marine population in the northern region of the East Sea, Korea. It is seldom found at a depth of less than 10 meters and the sea peach is frequently observed in large populations between 20 and 100 meters. It appears to prefer attachment to vertical rocks faces and artificial cement blocks exposed to the currents. Mass mortality and reduction of resources in sea peach, H. aurantium, were occurred in the benthic area of the northern region of the East Sea because of the rapid fluctuation of environmental factors such as temperature and salinity due to mass rainfall in summer and going up north of a strong warm current in winter. Therefore, we examined the effects of temperature and salinity on embryonic development of fertilized eggs, tadpole larva to metamorphosis, and attachment to siphon development. Laboratory-raised larvae were studied using a two-factorial experimental design with four levels of temperature(8, 12, 16 and $20^{\circ}C$) and four levels of salinity(20, 25, 30 and 34 psu). The ascidian larvae of H. aurantium survived environmental conditions between temperature of $8{\sim}20^{\circ}C$ and salinity of 25~34 psu and exhibited positive growth at $8{\sim}16^{\circ}C$ and 30~34 psu. Fertilized eggs have not developed at lower salinity of 20 psu irrespective of temperature range tested and have showed an abnormal development at the salinity of 25 psu between higher temperatures of 20 and $24^{\circ}C$. This result suggests that temperature increase and salinity reduction depending on environmental fluctuation may have significant impacts on population variation of H. aurantium in the northern region of the East Sea.

  • A Li recovery technology has been developed and related experimental verification efforts were carried out to improve the economical viability and environmental friendliness of the 'Advanced Spent Fuel Conditioning Process' being developed at KAERI. This technology is characterized by the combination of 1) the electrolysis of $Li_2O$ in a molten salt by using a porous non-conducting magnesia container at the cathode, 2) the separation of the Li in the container from the molten salt by elevating the container above the level of a molten salt, 3) the transport of the Li in the container by using a vacuum siphon to a separated reservoir. Li was semi-continuously recovered from a LiCl-$Li_2O$ molten salt with a more than 95% yield by using the developed technology.

The Experimental Research for the Collecting Characteristics of the Passive and Active type Domestic Solar Hot Water Systems (자연형 및 설비형 태양열 온수기의 집열특성에 대한 실험적 연구)

  • Domestic solar hot water system can be divided into a passive type and an active type. In a passive type the storage tank is horizontally mounted immediately above the solar collectors. No pumping is required as the hot water naturally rises into the storage tank from the collectors through thermo-siphon flow. While, in an active type the storage tank is ground- or floor-mounted and is below the level of the collectors; a circulating pump moves water or heat transfer fluid between the storage tank and the collectors. We installed two types solar hot water systems consisting of the same storage tank and collectors at the same place, and were measured and compared typical operating characteristics under the same external conditions. In particular, the daily system performance was presented through the stirring test after the sunset. The results show that the amount of solar radiation obtained for an active type were less than a passive type on a cloudy day, because the operation of the circulation pump stops frequently took place on that day. However, on a sunny day, depending on the stable operation of the circulation pump, the amount of solar radiation obtained for an active type were increased than a passive type.

  • Some hydromedusae were collected from the coasts of Seogwipo(Jejudo Island), Hoenggando Island and Ilsanhaesuyokjang(Ulsan), Korea on July 11, 1985; July 23, 1990 and July 16, 1994. They were identified into Aequorea coerulescens (Brandt, 1838) of the order Leptomedusae, and Physalia physalis utriculus La Martiniere, 1829 of the Siphonophora, respectively. The unique morphological characteristics of A. coerulescens are smooth even surface of exumbrella, large mouth with 60 highly fringed oral lobes, shallow stomach, 120 simple radial canals and flat beret-shaped bell. In P. physalis utriculus its morphological characteristics are a triangular large pneumatophore with very low or rudimentary crest, a ribbon like long slender main tentacle, siphon-shaped gastrozooid with mouth, finger-shaped dactylozooid and branched gonozooid with gonophores. P. physalia. utriculus is the Pacific form and distinguished from the Atlantic form, P. p. physalis which has a much larger pneumatophore with high crest, numerous large main tentacles, and compact arrangement of basal and ventral cormidia. As a result of this work the Korean hydromedusan fauna consists of 15 species of five orders.

  • 물넘이 형식에는 수문과 밸브로 조절할 수 있는 조절형식, 개수로 및 관수로 형식의 자연적으로 수위조절이 되는 비조절형식, 레버린스(Labyrinth), 피아노건반(Piano Key Weir), 사이펀(Siphon) 등의 특수형식으로 구분할 수 있다. 이 중에서 피아노건반 물넘이는 레버린스 물넘이를 피아노건반 모양으로 개량한 것으로 2003년 알제리 Biska 대학의 Quamane 교수에 의해 제안되었다. 제한된 물넘이 폭에 물넘이의 길이를 연장하여 방류능력을 증가시키는 효과를 나타내는 물넘이 형식으로 여수로의 월류수심을 낮게 유지하면서 방류량을 증가시킬 수 있다. 피아노건반 물넘이는 2006년에 프랑스에서 최초로 적용되었으며, 현재까지 약 30여 지역(프랑스, 베트남, 호주 등)의 댐에 적용되었다. 본 연구에서는 국내 저수지를 대상으로 여수로에 피아노건반 물넘이를 적용하여 방류능력과 수리적 안정성 검토를 위해 농어촌연구원 국제융합수리시험센터에서 수리모형실험을 수행하였다. 농어촌연구원은 2018년도에 기존 수리모형실험 시설을 확장하여 가로 250m, 세로 102m의 국내 최대규모의 실내 수리모형실험 시설을 준공하였고, 대형유사순환수로, 고정식 급경사수로, PIV 전용실험수로 등 기능별 7종의 실험수로를 갖추고 있다. 수리실험은 Froude 상사에 의한 축척을 적용하여 모형을 제작하였고, 기존의 수리시설과 하류하천까지 포함하여 70m(L)×20(B)×2m(H) 규모이다. 실험은 유량을 다양하게 변화시키면서 수위-방류량을 검토하였고, PMF 홍수위에서 방류량이 배제 가능한지 검토하였다. 또한, 물넘이의 안정성 검토를 위한 접근유속을 게측하였고, 물넘이를 통해 월류하는 흐름의 유황을 분석하여 수리학적으로 안정한 흐름이 발생하는지 검토하였다.

An experimental study on increased pressure in Shinwol rainwater storage and drainage system by undular bore (불규칙 단파에 의한 신월 빗물저류배수시설 내 압력상승에 관한 실험 연구)

  • An underground deep tunnel system is a facility in form of a reverse siphon for an under flood defense structure. In this study, the 'Shinwol rainwater storage and drainage system', which is under construction for the first time in South Korea, in order to confirm the effects of undular bore and pressurized air on the hydraulic stability of the facility in various flood scenarios a hydraulic model experiment was performed. As a result of this study, it was analyzed that the undular bore generated downstream pushed the pressurized air collected in the facility while moving upstream, and the pressure inside the pipe increased at this time. It was analyzed that the pressure during the passage of the undular bore was greater than the sum of the static pressure and dynamic pressure at the time and overflow occurred when the cross-sectional size of the pressurized air was more than 40% of the cross sectional area of the tunnel. It is determined that this is correlated with the volume of pressurized air collected in the facility, and it is determined that it is necessary to study the relationship between velocity of undular bore and the volume of pressurized air in the future.

MR Imaging of Slow-flow Using a Flow Phantom (유동모형을 이용한 저속유동의 자기공명영상)

  • Purpose : To find sensitivity of MRI imaging methods to slow flow phantom study was performed with conventional Spin-Echo, gradient echo based Phase Contrast, fast GRASS, and heavily T2-weighted Fast Spin Echo pulse sequences. Materials and Methods : A siphon driven flow phantom was constructed with a ventriculo-peritoneal shunt catheter and a GE phantom to achieve continuous variable flow. Four different pulse sequences including Spin-Echo, Phase Contrast, GRASS and Heavily T2-weighted Fast Spin Echo were evaluated to depict slow flow in the range from 0.08 ml/min to 1.7 ml/min and to compare signal intensities between static fluid and flowing fluid. Results : In the slow flow above 0.17 ml/min conventional Spin-Echo showed superior apparent contrast between static and flowing fluid while GRASS was more sensitive to the very slow flow below 0.17 ml/mim. It was not accurate to calculate flow and velocity below 0.1 ml/min with a modified PC imaging. Conclusion : Four different MR pulse sequences demonstrated different sensitivity to the range of slow flow from 0.08 ml/min to 1.7 ml/min. This finding may be clinically useful to measure CSF shunt flow or detecting CSF collection and thrombosis.

Induction of Soft Tunic Syndrome by Water Temperature and Physiological and Histological Responses of the Sea Squirt, Halocynthia roretzi (수온에 의한 멍게(Halocynthia roretzi)의 물렁증 유도와 생리 및 조직학적 반응)

  • In this study, we investigated the changes in the physiological and histological traits of a sea squirt (Halocynthia roretzi) with the emergence of the soft tunic syndrome induced by the water temperature control (6, 9, 12, 15, 18, 21, 24 and $27^{\circ}C$). It was observed that the induction rate of the soft tunic syndrome was highest at $15^{\circ}C$, but lowest at $24^{\circ}C$. Based on the tunic color condition and contraction strength, the whole process were classified into 4 stages as S0, S1, S2 and S3. Interestingly, there were significant differences in oxygen consumption and filtration rate were observed during S0-S3. The most distinctive aspects were change of blood cell composition at stage S3, whereas multi-vacuole cell ratio was decreased by 1/2 and morula cell ratio expanded about 10 times during S0-S3. Further, change of organ structure started following the syndrome such as degeneration of epithelial cells, microfilaments, increment in hemocytes and damage in muscle fiber have been detected in tunic, siphon, branchial sac, body wall musculature and pyloric gland. Briefly, our study results indicated that the normal physiological functions of the sea squirt can be affected due to the soft tunic syndrome induced by water temperature.

Studies on the carotenoids in the viscera of abalone (Haliotis discus hannai) (전복(Haliotis discus hannai) 내장(內臟)의 Carotenoid에 관(關)한 연구(硏究))

  • The studies on the carotenoids in the viscera of abalone (Haliotis discus hannai) have been-carried out. The pigments were extracted with acetone-methanol mixture (4 : 1) from the viscera of abalones which were caught around the coastal water of Korea from March to August. The individual carotenoid in the extracts was separated and purified by the silica gel TLC, $Mg(OH)_2$ impregnated paper chromatography and $Mg_2(OH)_2CO_3$ TLC. The isolated eleven carotenoids were investigated and identified by epoxide test, partition test, reduction with sodium borohydride, alkaline hydrolysis, co-chromatography and comparative test with reference carotenoids and electronic and IR absorption spectrophotometry. ${\alpha}$-Carotene, ${\beta}$-carotene. lutein, zeaxanthin, siponaxanthin, siponein, fucoxanthin, loroxan-thin-like and fucoxanthinol-like have been identified among the eleven carotenoids isolated. It has been found that fucoxanthin, on alkaline treatment, was transformed to the product of which chromophore was the same one as fucochrome and semifucoxanthol. Among the identified nein carotenoids siphonaxanthin, siponein, fucoxanthin, loroxanthin-like and fucoxanthinol-like have not been reported previously to be contained in the shellfish.
