Combination of multiplex reverse transcription recombinase polymerase amplification assay and capillary electrophoresis provides high sensitive and high-throughput simultaneous detection of avian influenza virus subtypes

  • The pandemic of avian influenza viruses (AIVs) in Asia has caused enormous economic loss in poultry industry and human health threat, especially clade 2.3.4.4 H5 and H7 subtypes in recent years. The endemic chicken H6 virus in Taiwan has also brought about human and dog infections. Since wild waterfowls is the major AIV reservoir, it is important to monitor the diversified subtypes in wildfowl flocks in early stage to prevent viral reassortment and transmission. To develop a more efficient and sensitive approach is a key issue in epidemic control. In this study, we integrate multiplex reverse transcription recombinase polymerase amplification (RT-RPA) and capillary electrophoresis (CE) for high-throughput detection and differentiation of AIVs in wild waterfowls in Taiwan. Four viral genes were detected simultaneously, including nucleoprotein (NP) gene of all AIVs, hemagglutinin (HA) gene of clade 2.3.4.4 H5, H6 and H7 subtypes. The detection limit of the developed detection system could achieve as low as one copy number for each of the four viral gene targets. Sixty wild waterfowl field samples were tested and all of the four gene signals were unambiguously identified within 6 h, including the initial sample processing and the final CE data analysis. The results indicated that multiplex RT-RPA combined with CE was an excellent alternative for instant simultaneous AIV detection and subtype differentiation. The high efficiency and sensitivity of the proposed method could greatly assist in wild bird monitoring and epidemic control of poultry.

  • The large-area Compton camera (LACC), featuring significantly high detection sensitivity, was developed for high-speed localization of gamma-ray sources. Due to the high gamma-ray interaction event rate induced by the high sensitivity, however, the multiplexer-based data acquisition system (DAQ) rapidly saturated, leading to deteriorated energy and imaging resolution at event rates higher than 4.7 × 103 s-1. In the present study, a new simultaneous multi-channel DAQ was developed to improve the energy and imaging resolution of the LACC even under high event rate conditions (104-106 s-1). The performance of the DAQ was evaluated with several point sources under different event rate conditions. The results indicated that the new DAQ offers significantly better performance than the existing DAQ over the entire energy and event rate ranges. Especially, the new DAQ showed high energy resolution under very high event rate conditions, i.e., 6.9% and 8.6% (for 662 keV) at 1.3 × 105 and 1.2 × 106 s-1, respectively. Furthermore, the new DAQ successfully acquired Compton images under those event rates, i.e., imaging resolutions of 13.8° and 19.3° at 8.7 × 104 and 106 s-1, which correspond to 1.8 and 73 μSv/hr or about 18 and 730 times the background level, respectively.

PIV measurement of step cavity with driven flow (구동류를 갖는 계단 캐비티의 PIV계측)

  • An experimental study was carried out in a three-dimensional cubic cavity driven by 2-dimensional plane Poiseuille flow for three kinds of Reynolds number, $10^4$, 3 $\times$ $10^4$ and 5 $\times$ $10^4$ based on the cavity width and cavity inlet mean flow velcoity. Instant simultaneous velocity vectors at whole field were measured by 2-D PIV system. Laser based illumination and two-frame grey-level cross correlation algorithm are adopted. Severe unsteady flow fluctuation within the cavity are remarkable at above Re = 3 $\times$ $10^4$ Reynolds numbers and sheared mixing layer phenomena are also found at the region where inlet driving Poiseuille flow is collided with the clock-wise rotating main primary vortex at upper center area. Instant velocity profiles reveal that deformed forced vortex formation is observed throughout the separate two areas.

  • A simultaneous visualization of the behavior of bubbles and dry spots has been carried out for pool boiling of R-1l3 on a horizontal heater. From video imaging and image processing analysis, the formation of bubbles and dry spots occurs simultaneously, and therefore they should be considered as a synchronized concept rather than independent identities. The dry spot density is equivalent to the active site density in the region before CHF. At CHF point, large dry areas due to the coalescence of neighboring dry areas cover the heater surface.

  • A sol-gel processing of $YBa_2Cu_3O_{7-x}$ superconductor using metal nitrate salts and sodium hydroxide as the starting materials has been investigated because of the need to produce pure, hompgeneous superconducting materials. Since the precipitation of barium hydroxides can be obtained only at high basicities, the process has to be carried out Ca. pH 13 to get the simultaneous coprecipitation with the other metal hydroxides. The involved reaction mechanisms were investigated and intermediate and final products were characterized by means of thermogravimetric analysis(TGA), infra-red(IR) spectroscopy, X-ray diffraction(XRD) analysis, scanning electron microscopy(SEM), thermal mechanical analysis(TMA) and electrical measurement.

  • Study on the use of enzymes for textile wet processing has been very active. The exploratory research conducted herein is related to the bioscouring process for cotton fabric. The optimum concentration of alkaline pectinase(BioPrep) was in the range of 0.05~0.2 g/l, the proper treatment time was 30~60 minutes, the appropriate treatment temperature was $60^{\circ}C$ for both the batch method and the padding method. The simultaneous desizing/bioscouring by padding method did not give water absorbency as good as the bioscouring after desizing. Color of fabrics which were bioscoured and dyed with direct dyes and a reactive dye was just a little darker than that of NaOH scoured one. K/S and Lab values of the bioscoured fabrics, regardless of the degree of water absorbency, were quite similar to each other.

  • Fire Sequencing Problem (FSP) is to find a sequence of targets, where there exist a number of targets with different time units required to fire. Because of the weapon's specifications and the size of the targets, several weapons may fire on the same targets, and the time units required on firing for each weapon may be different from each other. The objective is to minimize the completion time of firing for given number of targets. Mathematical formulation is given, and the heuristic algorithm based on the paring of targets in advance is suggested. Performance of the heuristic is evaluated by comparison of heuristic appeared in the literature through the computational experiments.

  • The success of cell manufacturing applications in FMS rests on the effective cell formation to maintain the independent relations both between machine cells and between part families. This paper presents an integrated method for concurrent formation of cells and families with no E.E (Exceptional Element) in FMS with alternative routings. To determine the maximum number of cell and family with no E.E, mathematical conditions and properties are derived. New concept of nonsimilarity is introduced for each machine and part based on machine-operation incidence matrix and part-operation incidence matrix. To concurrently form the cells and families, integer programming based mathematical models are developed. For the predetermined number of cell or family, model I is used to identify whether E.E exists or not. Model II forms cells and families considering only nonsimilarity. But model III can consider nonsimilarity and processing times. The proposed method is tested and proved by using numerical examples.

  • Pulsed ultrasonic Doppler system is a useful diagnostic instrument to measure blood-flow-velocity, velocity profile, and volume-blood-flow. This system is more powerful compare with 2-dimensional B-scan tissue image. A system has been deve- loped and ii being evaluated using TMS 32010 DSP. We use this DSP for the purpose of real-time spectrum analyzer to obtain spectrogram in singlegate pulsed Doppler system and for the serial comb filter to cancel clutter and zero crossing counter to estimate Doppler mean frequency in multigate pulsed Doppler system. The Doppler shift of the backscattered signals is sensed in a phase detector. This Doppler signal corresponds to the mean velocity over a some region in space defined by the ultrasonic beam dimensions, transmitted pulse duration, and transducer ban(iwidth. Multi- gate pulsed Doppler system enable the transcutaneous and simultaneous assessment of the velocities in a number of adjacent sample volumes as a continuous function of time. A multigate pulsed Doppler system processing the information originating from presented.

  • Although the use of apodization functions in connection with postprocessing of a 2D NMR spectrum proves improved spectral quality, there is usually a trade-off between resolution enhancement and noise suppression due to a classical "uncertainty principle." In this study, therefore, a mathematical deconvolution technique called "Maximum Likelihood Deconvolution (MLD)" was adopted to achieve the spectral resolution and sensitivity enhancement simultaneously. The MLD technique greatly facilitates visualization and restoration of the genuine spectral information from complex 2D NMR spectra that would be problematic with the conventional apodization/FT processing. In particular, application of the MLD to the 2D-NOE spectrum would be very useful to derive the important proton connectivities, which are essential to achieve elucidating the 3D molecular structure.
