Causal Pathogenesis on the Silkworm, Bombyx mori, Associated with Entomopathogenic Nematoda (곤충 병원성 선충에 의한 집누에 감염증과 병인론적 발병생리)

  • Entomopathogenic nematodes, Heterorhabditidae and Steinernematidae, were isolated from the soil of mulberry field, and the high infectivity and invesiveness were confirmed in the silkworm, Bombyx mori. The cause of non-microbial and acute flacherie was found as an disease by infection with soil-born nematodes through the mulberry leaves contaminated with soil and rainwater. The causal nematodes were isolated by silkworm trap from all of the 5 soil samples collected on the 5 mulberry fields, and identified as 3 strains of Heterorhabditis sp. and 2 of Steinernema sp. Rainwater itself, however, wasn't engaged in the silkworm disease, mulberry leaves with rainwater was rather profitable for cocoon production when the leaf quality was too hard to feed silkworm. Feeding of wet mulberry leaves with rain might not so harm to silkworm when the condition of rearing room to be kept at suitable temperature and ventilated well. Nematode infection of silkworm could be occurred by harvesting and feeding of contaminated mulberry leaves on the weather condition of rainy and wind. For the prevention of nematode infection, silkworms should be fed the leaves harvested from the higher portion of the mulberry tree in rainy days. For an oppositional application of this susceptibility of silkworms to nematode, might be useful on the collection and amplification of nematode agents for biotic control of pest insects.

The Detection and Multiplicity of Entomopathogenic Nematodes Using Silkworms (Bombyx mori) (누에를 이용한 곤충병원성 선충의 검색 및 배양)

  • Entomopathogenic nematodes were isolated through the investigation in soils collected from cultivated and non-cultivated fields using silkworms (Bombyx mori) and Galleria mellonella trap. The detectable rate of entomopathogenic nematodes of silkworms trap was higher than the G. mellonella trap. This study indicates the detection of entomopathogenic nematodes from soils that silkworms are sensitive superior to the G. mellonella to entomopathogenic nematodes. The steinernema, rhabditidae, and diplogatroidae strains successfully cultured on the silkworms host as well as on artificial media. Reproductivity in the living silkworm larva and pupa was 1.5 to 3.5${\times}$ 10$\^$5/ nematodes per host However, G. mellonella could be multiplied less than 5${\times}$l0$\^$5/ nematodes. The dried pupa of the silkworm following mositurize was cultured 0.5 to 2${\times}$10$\^$5/) nematodes per host. The culture methods of the steinernema, rhabditidae, and diplogatroidae strains, using silkworm powder, extracted chicken intestine, and food waste fertilizer could be applicative, but rate of reproduction was low.

Use of Enthomopathogenic Nematode to Biological Control of the Pets of Korean Ginseng (곤충병원성 선충을 이용한 인삼해충방제)

  • In round numbers the 100 strains of entomopathogenic nematodes were isolated through the investigation of cultivated including the ginseng forming cultivated and forest soil samples by silkworm trap. The 28 strains of nematodes were selected among the isolated entomopathogenic nematodes that were confirmed the pathogenicity against Holotrichia morosa, Hoiotrichia diomphalia and Ectinus sericeus, the pest of korean ginseng and silkworm. Pathogenicity of the 2025, 2027, 2028, 2034, 2039 and 2057 strains was excellent. Selected entomopathogenic nematodes are classified of two species by morphologiacl and molecular studies, which were Sterinernma carpocapsae sp. and Diplogaster lethier sp.. Diagnostic characters include body length, lateral field pattern, tail shape and so on. The DNA sequences of the ITS region of rDNA shows similar to S. carpocapsae and .D. lethier. Isolated entomopathogenic nematodes were demonstrates that are quite within the realms of possibility for biological control agents of the pests of Korean Ginseng.

Isolating Entomopathogenic Nematode in South Korea (남한 토양에서 곤충병원성 선충의 분리)

  • Entomopathogenic nematodes were isolated through the investigation of soil samples from various biotopes in south Korea, the efficiency of isolation for highly pathogenic nematodes to silkworms (Bombyx mori) was as high as 28 %. Twenty-eight strains of nematodes, selected among 100 samples by silkworms were confirmed the pathogenicity, multiplicity, and tolerance against various condition of preservation. Pathogenicity of the nematode isolates to agricultural and environmental pests such as Calliphora vomitoria, Pseufaletia separata, Palomena angulosa, and Melolontha incana were high. Mortality was varied from 20 to 100% by the pest insects and nematode strains. The high detectablity of entomopathogenic nematodes resulted from the methods of collection for soil samples within 10 cm depth after eliminating dried soil surface and the use of silkworm trap. High population of entomopathogenic nematodes represented the strong activity and broad action radius in the environment. Most of the nematode isolates were successfully cultured on the silkworm host as well as on artificial media, and proved their potential for the use of biological control agent.

A Study of Entomopathogenic Nematode at Heavy Metal Contents in the Polluted Soil of Kyungsangbuk-do Area (경상북도지역의 오염된 토양에서의 중금속 함량에 관한 곤충병원성 선충에 관한 연구)

  • The variety and density of entomopathogenic nematodes from the polluted soils of heavy metals were examined. In order to investigate the pollution of heavy metals in soil, 300 sites in kyungsangbuk-do were collected from March to October in 2001. We measured the contents of seven heavy metal elements (Cd, Cu, As, Hg, Pb, Cr^{6+}$, CN) and than cheesed soil of 25 sites with high concentration of heavy metals. The seven strains of nematodes were isolated from seven samples by silkworm host (Bombyx mori mori) and white trap. Isolated nematodes are composed of two families, one order. The members of Rhabditida were isolated in the soil with mean Cd content of 0.870 ppm. And they were isolated in the soil samples with As content less than 0.745 ppm. However they were isolated regardless of concentration of Cu and Pb. The members of Cylindrocorpidae were isolated in the soil samples with Cr^{6+}$ content less than 0.05 ppm. Any entomopathogenic nematode was not detected in the CN polluted soil. Isolated nematodes successfully cultured on the silkworm host and were confirmed the pathogenicity, multiplicity, and tolerance against various condition of preservation. Which proved its potential usefulness as biological agent.

Isolation and Identifieation of Entomopathogenic Nematodes from Soil and Insect (토양과 곤충 사체로부터 곤충병원성 선충의 분리 및 동정)

  • Nematodes were isolated using silkwom trap through the investigation of 100 soil samples from various biotopes in Korea. The 30 nematode strains from soil and dead insects by the pathogenicity aganinst silkworms (Bombyx mori mori) and insect pests of Calliphora vomitoria, Pseufazetia separata, Palomena angulosa, and Melolontha incana. Mortailty of the silkworm larvae and pupae were as high as 100% by nematode infection, those of insect of pests were varied from 20 to 100%. The 30 strains of entemopathogenic nematodes were classified into five groups of Rhabditidae, Diplogatroidae, Heterorhabitidae, Steinernematidae, and Tylenchida by morphological criteria. The genetic relationships among the 30 nematode strains were analyzed by various RAPD bands with twenty primers. The 30 nematode strains were classified into six major subgroups on the basis of the genetic similarity coefficient of 0.853. The grouping by RAPD was agree with those of morphological taxa in discrimination of the higher group, however, was not completely agree in the subgroup. The family Steinernematidae belong to Rhabditida was clarified as closer to the Tylenchida, rather than the other Rhabditida of Heterorhabitidae, Rhabditidae, and Diplogatroidae in genetic distance valule. From the result of the morphological classification and RAPD of the genomic DNA showed that genetic relationship analysis furnish infurmation on phylogenetic classification and relationships of entomopathogenic nematodes. The application of genetic similarity will overcome the limitation of taxonomy and classification of morphologically simple nematode. Several primers were confirmed those utility of identification for individual nematode strains, the methods of molecular genetics secured the simplicity, rapidity and accuracy on the selection of entomopathogenic nematodes.
