STUDIES ON THE CYSTINE COMPONENT IN THE SERICULTURAL PROTEINS OF BOMBYX MORI L. (가잠사단백질의 각과정에서의 Cystine 성분에 대한 연구)

  • The purpose of this treatise is to prove the presence of cystine in silk fiber through wide sampling throughout all the sericultural processes of Bombyx mori.; also to show that disulfide cross linkages exist in the silk fiber. The conclusions reached were as follows: 1. Crystalline cystine was obtained from silk fibroin using Folin's Method. 2. Analytical data showing the cystine content of silk fiber and its related materials were obtained using Sullvan's Method as follows: Material Percent Cystine A. Mulberry leaf protein 0.175 B. Silkworm egg 0.33 C. Silkworm Body, matured, fat extracted, without silk gland 0.41 D. Silk gland, matured 1.23 E. Silkworm feces none F. Silkworm pupa, fat extracted 0.30 G. Silkworm moth, fat extracted 0.60 H. Raw Silk 0.22 I. Fibroin 0.175 J. Sericin 0.30 3. The presence of cystine in the silkworm was substantiated the existence of 0.175 % methionine in mulberry leaves and 0.12% methionine in the silk gland. 4. Part of the sulfhydryl compounds in the silk gland is believed to transfer to serine and methionine, with the former being secreted into the liquid silk finally as silk fiber and the latter used for nutritive purposes in the growing of silk gland tissue. 5. The cystine content is variable by mulberry species, silkworm species, sex, breeding process, and other culturing environments. 6. Hybrid silkworms require more nutritive amino acids for effective growth than the original parents, and secrete less of them as silk fiber. 7. From such an observation, the amino acid composition of silk fiber is believed to be fairly flexible. Cystine if included in the amorphous part of the fiber, especially in sericin. 8. The result from enriching the silkworm diet with pure cystine or wool cystine did not result in any advantage, therefore it is believed that the natural cystine and methionine contents in the mulberry leafaregoodenoughforsilkwormnutrition. 9. The disulfide cross linkage in silk fiber was verified by using the Harris Method. Contraction took place following the treatment of the fiber with various salts and acids. Comparisons were made with wool fiber. 10. During these experiments, the fibrious structure of silk fiber and the net-globular liquid form were photographed microscopically. It is believed that the globules of liquid silk are net-formed by the inter attraction of the OH ion of the globular peptide and the H ion of water as shown by the hair cracking behavior of the film. The net-globular protein precipitation from the mulberry protein solution showed that mulberry is a proper diet for the formation of fibrous protein in the silk fiber. 11. The significance of the presence of cystine in silk fiber as emphasized in this paper should result in modification of the general conception that cystine is absent from this fiber.   • In order to observe the contents of some essential amino acids in Korean foods, total and free tryptophan, and available lysine in fifty kinds of Korean foods were analyzed by the sfectrophotometry. The results obtained are summarized as follows : 1) The tryptophan contents per 100g of soybean, wet green laver and skim milk were over 250 mg, and the contents Per g nitrogen in ginger, dried persimmon and chestnut were over 150 mg. 2) The free tryptophan contents per 100 g of soybean, small red bean, dried persimmon, ginseng stem were over 100 mg, ana the contents per g nitrogen in dried persimmon was over 150 mg. 3) The available lysine contents Per 100g of soybean, wet green laver and skim milk, fish(auchovy), dried yeast, casein and silkworm pupa were over 1000 mg, and the contents per g nitrogen in potato, perilla(wild sesame), red pepper, sausage and skim milk were over 300mg. 4) The contents of tryptophan and available lysine in soybean, green laver and skim milk were higher than in other samples. 5) In general, the contents of tryptophan and available lysine were abundant in seaweeds.

Impact of maternal crowd on the reproductive performance of an ecto-pupal parasitoid Nesolynx thymus (Hymenoptera: Eulophidae)

  • Nesolynx thymus Girault (Hymenoptera: Eulophidae) is one among 20 hymenopteran parasitoids reported to attack the uzi fly, Exorista bombycis, which in turn infests the silkworm, Bombyx mori accounting a cocoon yield loss of 10-20%. A host of factors viz., age and size of host, age and size of parasitoid, availability of host, temperature and quality of host are reported to influence the reproductive efficiency of the parasitoids. In the present study an attempt was made to record the influence of foraging parasitoid density on their reproductive performance. Three day-old uzi pupae were offered to varying numbers (1 - 10) of 2 day old Nesolynx thymus adults for parasitization. The progeny production per pupa (84.44 - 132.77) increased significantly with increase in the number of foraging parasitoid adults (1 - 10) however, progeny production per female decreased. The sex ratio (M : F) (1:25.49 - 1:1.53) decreased significantly with increase in foraging parasitoid adults. The morphometric parameters except female abdomen width of first generation parasitoid showed significant negative correlation with number of foraging adults. However, correlation between the parasitoid size in first generation and their reproductive performance was nonsignificant.

Status and prospect for development of insect foods (곤충식품 개발 현황 및 전망)

  • The reasons for selecting insects as future food are high growth and feed conversion rates, reproduce quickly, low environmental footprint, valuable source of nutrients, and source of a various undeveloped functional materials. Since 2014, Tenebrio molitor larva, Protaetia brevitarsis larva, Allomyrina dichotoma larva, and Gryllus bimaculatus were registered as new food in Korea because it has been scientifically proven that they are not harmful to eat and nutritious. Therefore they can be legally produced and sold as food. Accordingly, there are 7 species of edible insects including grasshopper, silkworm pupa, and Baekgangjam in Korea. To improve aversion to edible insects, using their powder, chop, and gravy hidden their morphology, we developed more than 100 different kinds of recipes for general food menu, held several times tasting events to be familiar with edible insect food, and published cookbooks. Moreover, we developed more than 50 kinds of recipes for patients. To eat the insect food more and more people, we have been analyzed various function of insects. If health food based on the results of functional analysis is developed, edible insect's value will be raised. If various insect foods are developed and consumed by people, edible insect market will grow up more than 100 billion won in 2020.

Supplement of High Protein-Enriched Diet Modulates the Diversity of Gut Microbiota in WT or PD-1H-Depleted Mice

  • Supplement of high-protein food plays an important role in improving the symptoms of malnutrition and the immune capacity of the body, but the association of high-protein diet and gut microbiota remained unaddressed. Here, we systematically analyzed the internal organs and gut microbiota in C57(WT) or PD-1H-depleted (KO) mice (T cells were activated) fed with pupae or feed for six weeks. We observed that the body weight gain in the mice fed with pupae increased less significantly than that of the feed group, while the villi and small intestine lengths in the pupa group were reduced compared with that of mice given feed. However, the average body weight of the KO mice increased compared with that of the WT mice fed with pupae or feed. Pupae increased the concentration of blood glucose in WT, but not in KO mice. Moreover, in the feed group, there was no difference in the weight of the internal organs between the WT and KO mice, but in the pupae-fed group, liver weight was decreased and spleen weight was increased compared with that of KO mice. The amounts/plural/amounts of Melainabacteria, Chloroflexi, and Armatimonadetes were specifically upregulated by pupae, and this upregulation was weakened or eliminated by PD-1H depletion. Some bacteria with high abundance in the feed-fed KO mice, such as Deferribacteres, Melainabacteria, Acidobacteria, Bacteroidetes, Spirochaetes and Verrucomicrobia, were decreased in pupae-fed KO mice, and Proteobacteria and Deinococcus were specifically enriched in pupae-fed KO mice. Bacteroidetes, Firmicutes and Akkermansia were associated with weight loss in the pupae-fed group while Lachnospiraceae and Anaerobiospirillum were related glucose metabolism and energy consumption. Based on high-throughput sequencing, we discovered that some gut bacteria specifically regulated the metabolism of a high-protein diet, and PD-1H deficiency improved life quality and sustained blood glucose. Moreover, PD-1H responses to high-protein diet through modulating the type and quantity of gut bacteria. These findings provide evidence about the association among gut microbiota, T cell activation (for PD-1H depletion) and high-protein diet metabolism, have important theoretical significance for nutrition and health research.

Effect of Some Herbal Extracts on Entomopathogenic Nematodes, Silkworm and Ground Beetles (몇 가지 한약재 추출물이 곤충병원성선충과 누에 및 먼지벌레에 미치는 영향)

  • Effect of four nematicidal herbal extracts (Daphne genkwa, Eugenia caryophyllata, Quisqualis indica and Zingiber officinale) and 3 acricidal herbal extracts (Pharbitis nil, Xanthium strumarium, and Desmodium caudatum) on entomopathobenic nematodes [Steinernema carpocapsae Pocheon strain (ScP) and Heterorhabditis sp. Gyeongsan strain (HG)], silkworm (Bombyx mori), and ground beetles (Synuchus sp.) were investigated in the laboratory and field. D. genkwa was highly toxic to SCP and HG (100% mortality) at the concentration of 5,000 ppm in X-plate. All the infective juveniles of HG were dead after 3 days by E. caryophyllata and Q. indica. The mortality of ScP and HG was below 10% by D. genkwa, D. caudatum, E. caryophyllata, Q. indica and Z. officinale at the concentration of 1,000 ppm two days after treatment while mortality of HG was 62.8% by D. genkwa at the concentration of 1,000 ppm in X-plate. However, 1,000 ppm had not effect on nematode survival and pathogenicity of ScP in sand column. On the contrary, E. caryophyllata had effect on pathogenicity of HG. Mean number of dead Galleria mellonella larva of HG was 0.5 in E. caryophyllata treatment. Q. indica did not effect silkworm reared on mulberry leaves at the treatment of 1,000 ppm in 10 days after treatment. However, there were 20.0 and 100% mortalities in the treatment of D. genkwa 3 and 10 days after treatment, respectively. The weight of silkworm was low in D. genkwa and did not pupate. The weight of pupa and cocoon were not different in E. caryophyllata, P. nil, Q. indica, X. strumarium and Z. officinale. D. genkwa, E. caryophyllata, P. nil, Q. indica and Z. officinale had no effect on ground beetles, Synuchus sp. in forest soil.

Different Ratio between Male and Female in F$_1$, Hybrids and Parents of Silkworm; Bombyx mori. L. (가잠의 원종 및 교잡종의 자웅별 생존율의 차)

  • In the F$_1$ hybrids and parents of silkworms, the silkworms of 47 blocks in summer and 68 blocks in fall in 1965 were reared in order to know the effect of mortality on the difference of sex ratio. The results analyzed are as follows; 1. In Moran of Japanese strain-parent and Morean x Daedong of F$_1$ hybrid, there is of no effect of an increase and decrease of mortality on the sex ratio. So male is not always stronger than female. 2. In Daedong of Chinese strain-parent, the more the mortality in the blocks is, the higher the survival ratio of male pupa becomes. (Y=0.233x-41.67). 3. As the results mentioned above, the present investigation method of cocoon quality of Chinese strains with the same number of cocoons of both sexes will have to be considered again, according to the cocoon crops, as far as concerned with the reasonable sampling from the population.

Antitumor Activity of Paecilomyces japonica is Mediated by Apoptotic Cell Death

  • The aqueous extract from the cultural mycelium of Paecilomyces japonica showed cytotoxicity against several tumor cells including Jurkat, U937, HL-60, HepG2, BW5147.G.1.4, and NIH3T3. When the aqueous extract was fractionated by sequential organic solvent extractions using n-hexane and ethyl acetate, the ethyl acetate fraction appeared to contain the most cytotoxic activity, and the $IC_{50}$ values for various tumor cells were in the range from 1.5 to $10.0{\;}\mu\textrm{g}/ml$. To elucidate the cellular mechanism underlying the induced cytotoxicity, the apoptotic DNA fragmentation along with the cell cycle proression was examined in Jurkat T cells following the ethyl acetate fraction treatment. In the presence of $2.5{\;}\mu\textrm{g}/ml$ of the ethyl acetate fraction, apoptotic DNA fragmentation of the cells was detected within 1 h and increased upto 24 h in a time-dependent manner. Under the same conditions, a sub-G1 peak was detectable by flow cytometry. These results indicate that the cytotoxic effect of P. japonica on tumor cells is attributable to the induced apoptosis.

  • The Aspergilli can be isolated from many of damaged domestic fowl's lung which were collected at fowl slaughter house from July to August 1973, during two months at Dapsimri, Dongdaemoon-ku, Seoul city. The survey of incubation condition for carbon source and nitrogen source were summerized as follows: 1. One of the Aspergillius fumigatus group were isolated from damaged domestic fowl's lung. 2. In the preparation of isolation media, adding the 4 gs of silkworm pupa and Meju to Czapek's agar medium is more preferable than Austick's media (2% malt extract agar) 3. In survey of incubation condition, the growth rate and sporulation shown that higher differences according to varieties and amounts of C-source. And in the case of 20% C-source concentration shown that low growth rate than lower concentration of C-source. 4. In N-source, the growth rate indicated that some difference according to varieties of N-source, difference of growth rate demonstrated almost not shown at various amounts differences of mycelium and sporulation was only detected reversely.

Comparison of Physicochemical Properties and Analysis of sEquence Structure Relationships of Commercial Dongchongxiacao of Three Species in Korean Market

  • To compare the quality of manufactured goods distributed in the domestic markets, 6 isolates of Dongchongxiacao products, namely, 4 Paecilomyces tenuipes specimens (J2P, 901A, 901B, and 901C), 1 Cordyceps militaris specimen (901D), and 1 Cordyceps sinensis specimen (CI-1), were analyzed for their physicochemical properties and sequence-structure relationships. P. tenuipes (J2P), a kind of Dongchongxiacao, was successfully inoculated on silkworms by percutaneous infection of Rural Development AdminstraionNam et al., 1999); fruiting bodies were then formed on the complete surface of the pupa. Since P. tenuipes (J2P) from silkworm larva was also proved to have remarkable pharmacological activities, it has been produced in bulk and has been successfully sold to buyers in the Korean market. Additionally, imitation products such as 901A, 901B, 901C, 901D, and CI-1 were sold simultaneously, resulting in deterioration of product quality. This research focuses on establishing quality standards to discriminate between the original and imitation products circulating in the market. The products obtained for the experiments included J2P, 901A, 901B, 901C, 901D, and CI-1; proximate analysis was performed for these products. The hosts and methods of conidia inoculation for proliferation of mycelia differed among the products. P. tenuipes (J2P) was proliferated in live silkworm larvae, and dead silkworm pupae were used to produce 901A, 901B, and 901C. On the other hand, 901D was produced on hulled rice medium. Quality analysis of C. sinensis revealed that CI-1, which was imported from China, smelled bad and proved to be a counterfeit with the fruiting body glued to the insect by twigs. The results of the proximate analysis of 901A, 901B, 901C, and 901D were similar to those of J2P with respect to the moisture content. Otherwise, J2P contained higher crude protein than 901A, 901B, 901C, and 901D, but contained very low fat. C. militaris (901D) and C. sinensis (CI-1) had low crude protein content-12.79% and 9.78% respectively-as compared to that of J2P, which was 62.38%. In contrast to the crude ash content of 6.4% in J2P, the crude ash content of CI-1 was 18.51% and this specimen was found to contain many impurities. phylogenetic analysis of P. tenuipes revealed that the sequence similarity of J2P, 901B, and 901C was in the range of 92.3~92.7%. Additionally, differences in the sequences were found at the positions 65 bp, 436 bp, 441 bp, 463 bp, etc.
