Studies on the High Temperature Induced Stress on the Biochemical Profile and Fecundity of Daba and Laria Ecoraces of Tropical Tasar Silkworm Antheraea mylitta Drury (Lepidoptera: Saturniidae)

  • Tropical tasar silkworm Antheraea mylitta Drury experiences extreme temperature stress conditions during its life cycle particularly during diapauses and first crop. The present study witnessed the impact of high temperature on some biochemical profiles and egg production (fecundity) of semi-domesticated Daba and Shorea robusta (Sal) based wild ecorace Laria during seed cocoon (pupa) preservation. Cocoons of Daba and Laria were treated with high temperature at $40^{\circ}C$ for 10 days in a BOD incubator. The protein profile and carbohydrate content in the hemolymph and fat body and total haemocyte count (THC) in the hemolymph of pupa were investigated. Further, the fecundity and fertility of egg was assessed. Significant increase in the protein concentration was observed in the hemolymph with reduction in the fat body (p<0.05). The difference in protein concentration was highly significant between the semidomesticated Daba and wild ecorace Laria (p<0.05). High pupal mortality (20%) and reduced fecundity (10-15%) in Daba was noticed compared to wild Laria. Also an increased THC (>28000) was recorded in Laria. The study infers the potentials of wild ecoraces in sustaining the extreme temperature conditions and need of adopting suitable package of practices for the preservation of diapause seed cocoons during extreme summer conditions. There is possibility to introgression thermal stress resistant traits in the semi-domesticated races of tasar silkworm by resorting to conventional breeding plans with wild races and keeping the thermal stress induced response as markers.

  • Ecophysiologically diapause represents a syndrome of physiological and biochemical characteristics, all of which ensure survival during a long period of dormancy. Since, silkworm enters diapause as embryo at the early embryonic stage, the duration of egg life depends on the duration of embryonic diapause. The nature of diapause in silkworm, Bombyx mori, is primarily determined by genetic characters and endocrinologicnl mechanisms, mediated by environmental factors such as temperature and photoperiod. Hibernating potency value besides nucleic acid and carbohydrate metabolism, production and utilization of sorbitol are also equally responsible for induction, initiation, determination, maintenance and termination of diapause. Embryonic diapause in Bombyx moir, induced by active secretion of sub-oesophageal ganglion is attributed to hormonal system and metabolic adjustment, which serves to bring about a new physiological state. Metabolic conversion of trehalose to glycogen at induction, glycogen to sorbitol at initiation and sorbitol to glycogen at termination of diapause is correlated and in each metabolic shift a key enzyme becomes active in response to hormonal and environmental stimulation. An attempt has been made in this review article to discuss briefly the nature of embryonic diapause, influence of various factors on diapause nature, hormonal mechanism of diapause besides biochemical composition of egg, nucleic acid and carbohydrate metabolism, production and utilization of sorbitol in relation to induction, determination, maintenance, initiation and termination of diapause in the silkworm, Bombyx mori.

Quantitative analysis of rutin content using silkworm genetic resources

  • Rutin is an important bioflavonoid that is consumed in the daily diet. This study compared the functional components of rutin from various silkworm species using a gene database with those of rutin produced by silkworms selectively bred through cross-combinations. We made comparisons between the geographical origin and species of silkworm using a gene database and discovered that rutin activity was ranked in the following order by species, Chinese (C5)> miscellaneous varieties (Jamsaeng 1 Ho) >Japanese (Jam 115) > European (E58) >Korean (Sun 3 ho). However, rutin levels with respect to various genetic traits (blood color, silk color, and egg color) were consistent. In order to study rutin changes that occurred during the cross breeding of the silkworm gene, we bred cross-combinations utilizing Jam 115 and the 4051 silkworms. In conclusion, in order to provide information about the constituents of functional materials contained in silkworm powder, it is imperative that silkworm cross breeding occurs so that the database of functional materials extracted from silkworms will expand.

  • 1-Deoxynojirimycin(1-DNJ or DNJ), a component in silkworm powder, prevents glucose from being absorbed into the bloodstream from the small intestine by inhibiting ${\alpha}$-glucosidase activity. This study compared the functional components of 1-DNJ from various silkworm species using a gene database with those of 1-DNJ produced by silkworms bred through cross-combinations. We utilized comparisons of geographical origins and species of silkworms using a gene database and discovered that 1-DNJ activity was ranked in the following order by species, Japanese (SK-1) > Chinese (C48) > European (Rock191). 1-DNJ constituted varying percentages of silkworm organs in the following order, blood > epithelial tissue > body fat > silk glands. With regard to sex, 1-DNJ, levels were higher in males than females. However, 1-DNJ levels with respect to various genetic traits (blood color, silk color, and egg color) were consistent. In order to study 1-DNJ changes that occurred during cross breeding of the silkworm gene, we bred cross-combinations utilizing SK-1 and C48 silkworms. In conclusion, in order to provide information about the constituents of functional materials contained in silkworm powder, it is imperative that silkworm cross breeding occurs so that the database of functional materials extracted from silkworms will expand.

Impact of Mating Duration on the Fecundity of the Newly Evolved Races

  • Of all factors that govern productivity in sericulture, the role of silkworm egg is of prime importance. The production of silkworm seed involves a long chain of interdependent and highly specialized conditions. Female moths of Bombyx mori L. commence egg deposition immediately when the moths were decoupled. More than 60% of eggs are deposited within 4 hrs after decoupling i.e. 62.33% in CSR-3, 61.67% in CSR-6, 62.50% in CSR-16 and 63.28% in CSR-17. Almost 90% of eggs were laid in the first 12 hrs (93.13% in CSR-3, 90.81% in CSR-6, 89.71% in CSR-16 and 93.17% eggs in CSR-17) after decoupling. Almost 8 to 10% of more eggs were laid when oviposition period was prolonged upto 48 hrs instead of 24 hrs. The present investigation showed that a significant increase of 45 (8.34%), 50 (9.98%), 41 (8.26%) and 45 (8.30%) (P < 0.01) numbers of more eggs were found between 24 to 48 hrs (T6 to T9) in CSR-3, CSR-6, CSR-16 and CSR-17 races respectively. The findings of present investigation is very important for sericulture industry while producing layings to keep for different hibernation schedule especially for autumn/late autumn crop, female may be allowed for prolonged oviposition to get 8 -10% more eggs and farmer may fetch 8 -10 kg. more cocoon yield/l00 dfls with more income.

한성계통의 전견중, 견층중, 견층비율에 대한 성차 (Sexual Differences of Cocoon weight, Cocoon Shell Weight, and Cocoon Shell Percentage in the Sex- limited Silkworm Strains, Bombyx mori L.,)

  • 한성반문 및 한성란색 계통잠에 있어서 전좌염색체로 인한 생리적 결함이 전견중, 견층중 및 견층비율에 미치는 영향을 이들에 관한 성차를 지표로 하여 검토한 결과, 한성 및 비한성 계통견의 전견중의 성차 평균치는 각각 127%, 129%, 견층중의 성차는 각각 107%, 110%, 견층비율의 성차는 공히 85%였다. 이상의 결과에서 한성반문 및 한성 난색계통잠은 전견중, 견층중, 견층비율 등의 실용형질에 관한 한 전좌된 염색체편에 기인하는 생리적 결함은 극히 미소한 것으로 생각된다.

Breeding of Two New Silkworm Varieties with Peculiar Laval Mark, "Eolrukmal" and "Hukpyobeom"

  • The Eolrukmal and Hukpyobeom varieties were bred for special purpose such as experience and education. The Eolrukmal variety was selected and succeeded from the $F_1$ of Huka$\times$Galwon in 2004 summer. It has stripes and brown half-moon body marks. The Hukpyobeom variety was selected and succeeded from the $F_1$ of Remone$\times$Hukho in 2004 summer. It has black body color and bright yellow segments. They are healthy yellow cocoon spining varieties showing over 93% of high practical hatching ratio and 93.1% of emergence ratio. The Eolrukmal moth laid 450 egg/batch and the Hukpyobeom moth laid 440 egg/batch.

3D 프린팅 기술을 이용한 누에 알 미세주입 기술 개선 (Improvement of Silkworm Egg Microinjection Using 3D Printing Technology)

  • 오래전부터 산업곤충으로서 이용되어온 누에는 최근 바이오 신소재 생산을 위한 생체공장으로써 주목받고 있다. 소재생산을 위해서는 주로 형질전환 기술을 이용하게 되며, 이는 배아가 있는 알 속으로 목적 유전자를 삽입하는 마이크로인젝션(microinjection) 방식으로 이루어진다. 마이크로인젝션을 위해서는 알을 고정하는 과정이 필수적이며 시간 소모 및 피로도가 높은 작업이다. 따라서 본 연구에서는 시간 소모 및 피로도 개선을 통하여 형질전환 효율을 높이고자 3D 프린팅을 이용하여 알의 정렬 및 고정에 도움을 줄 수 있는 알 배열판(egg liner) 및 접착제 줄눈판(glue drawer)을 3DCADian 프로그램을 이용하여 모델링하고, Fusion 360를 이용하여 3차원 도면을 제작 후 프린팅하여 제작하였다. 제작된 두 도구를 이용하여 슬라이드 글라스에 알을 고정하고, 소요된 시간을 분석한 결과 도구를 사용하지 않았을 때에 비하여 2가지 도구를 이용했을 때 작업시간이 약18.6% 감소하였으며, 연구자의 작업 편의성을 향상시키고 마이크로인젝션을 위한 현미경 및 로봇 팔(manipulator) 조작을 유리하게 하였다. 따라서 알의 배열 수 또는 조작 편의성을 개선할 수 있는 추가적인 연구 및 개량이 이루어진다면, 알 배열판 및 접착제 줄눈판이 누에 형질전환 효율성 개선 및 다른 산업 곤충의 형질전환 연구에도 이용될 수 있을 것으로 판단된다.

Review on Silkworm (Bombyx mori) Sex Control in China

  • Normally, silkworms, Bombyx mori, generate offspring by sexual activity. As we known, the hybrids of the first generation of the silkworm have higher cocoon production than pure lines. During the sericulture production, many processes are related with sex control. For example, sex sorting in the egg grainages, rearing of only male silkworm to save the mulberry leaf consumption and increase silk output and quality. Therefore it is very interested in understanding the sex control of the silkworm in theory and practice. Chinese sericultural scientists have been being engaged in the researches in the fields of artificifial parthogenesis, dispermic androgenesis, sex-limited varieties, sex linkage balanced lethal strain and high temperature sensitive male stocks for several decades and gained substantial achievement. Some of the achievements have been used in the commercial production. In this review, the authors introduced that the methods for control of the silkworm sex, and regulate the silkworm sex ratio according to different producing aim in the world and especially in China.

Biomolecular Variations in Poly and Bivoltine Strains and Their Hybrids of Bombyx mori during Embryonic Development

  • Biometabolic assessment was made in early and late embryonic stage (just before hatching) of one poly, one bi and their hybrids (DP, YPe, DP ${\times} Ype, and YPe ${\times} DP respectively ) of Bombyx mori to observe the racial differences. Protein and nucleic acid (RNA and DNA) concentrations were recorded to be significantly higher in bivoltine breed (YPe) and also in the hybrid than the polyvoltine (DP) strain in both the stages of embryonic development. The single egg weight of polyvoltine race was lower as compared to that of bivoltine and the hybrid studied. Age specific changes in all the biomolecules were evident where protein and RNA concentrations were elevated sharply in prehatched larvae while in case of DNA it was observed to be just reverse. The differences in protein, RNA and DNA composition between breeds and hybrids reflect the racial variations in biometabolic demands responsible for differential growth and development of the breeds and hybrids.
