Comparative study on bone regeneration between silk mat incorporated 4-hexylresorcinol and collagen membrane

  • The purpose of this study was (1) to demonstrate the anti-microbial properties of 4-hexylresorcinol (4HR) loaded silk mat and (2) comparison of bone formation between 4HR incorporated silk mat and collagen membrane. Anti-microbial properties of 4HR incorporated silk mat was done after sterilization of silk mat (autoclaving and ethylene oxide gas). For the evaluation of bone formation, bilateral bony defects (size: 8 mm) were prepared in the parietal bone of the rabbits (n=10). 4HR incorporated silk mat (size: $10{\times}10mm$) was applied on the right defect. For the comparative purpose, the same size of commercial collagen membrane was applied on the left defect. The anti-microbial properties of 4HR incorporated silk mat were maintained after sterilization process. When compared bone mineral density and bone volume, there was no statistically significant difference between groups at 4 weeks and 8 weeks after operation (p>0.05). In conclusion, 4HR incorporated silk mat could be autoclaved without concern of anti-microbial properties loss. In addition, 4HR incorporated silk mat showed similar bone regeneration to collagen membrane. Therefore, 4HR incorporated silk mat might be considered for the application of open membrane technique.

  • When 4-hexylresorcinol (4HR) incorporated silk mat is used for clinical application, sterilization process is essential. In this study, 4HR incorporated silk mat was sterilized with ethylene oxide (EO) gas or autoclave, and then antibacterial property was evaluated. For the examination of micro-structure after the sterilization, scanning electron microscopic (SEM) exam and Fourier Transform Infrared (FT-IR) spectroscopy were also done. There was no gross difference after sterilization in SEM exam and FT-IR spectra. Antibacterial property of 4HR was also maintained after the sterilization. In conclusion, EO gas or autoclave did not damage the antibacterial property of 4HR in the silk mat.

Soluble fraction from silk mat induced bone morphogenic protein in RAW264.7 cells

  • The objective of this study was to evaluate the changes in gene expression after incubation of cells with soluble fraction from different silk mat layers. A silk cocoon from Bombyx mori was separated into 4 layers of equal thickness. The layers were numbered from 1 to 4 (from the inner to outer layer). Each silk mat was placed into normal saline and collected soluble fraction. They were administered to RAW264.7 cells, and changes in the expression of genes were evaluated by cDNA microarray analysis. Layer 1 and 4 groups showed significantly higher expression of BMP-2 at 8 h after administration of soluble fraction (P < 0.05). Runx2 expression was significantly higher in Layer 4 group at 8h (P < 0.05). The silk mat from the innermost and outermost portion of the silkworm cocoon showed a significant change in the expression of genes that are associated with osteoinduction such as BMP-2 and runx2.

  • Background: Silk mats have been approved for clinical trials by the Korean Food and Drug Administration as membranes for guided tissue regeneration (GTR). In this study, silk mat application was compared to high-density polytetrafluoroethylene (dPTFE) membrane application or no membrane group. Methods: To compare the silk mat group to the dPTFE group or the no membrane group, a retrospective sample collection was conducted. Bony defects were measured at the time of extraction (T0) and then at 3 months (T1) and 6 months after extraction (T2) on a digital panoramic view. Bone gain (BG) was calculated by subtracting from the bony defect at T0 to the bony defect at each follow-up. Results: The BG at T2 was 2.44 ± 2.49 mm, 4.18 ± 1.80 mm, and 4.24 ± 2.05 mm in the no membrane group, silk mat group, and dPTFE group, respectively. Both membrane groups had significantly higher BG than BG in the no membrane group at T2 (P < 0.05). Conclusions: Both membrane groups showed higher BG than the no membrane group.

  • Background: The objective of this study was to evaluate the changes in gene expression after incubation of cells with proteins released from different silk mat layers. Methods: A silk cocoon from Bombyx mori was separated into four layers of equal thickness. The layers were numbered from 1 to 4 (from the inner to the outer layer). The proteins were released by sonication of a silk mat layer in normal saline. The concentration of proteins was determined by spectrophotometry. They were incubated with RAW264.7 cells, and changes in the expression of genes were evaluated by cDNA microarray analysis and quantitative reverse transcriptase-polymerase chain reaction (qRT-PCR). Results: Layer 1 and 4 groups had higher protein concentrations compared to those in layer 2 and 3 groups. The genes associated with inflammation and angiogenesis showed significantly higher expression in layer 1 and 4 groups. The results of qRT-PCR were in agreement with those of the cDNA microarray analysis. Conclusions: The silk mat from the middle portion of the silkworm cocoon yielded a lower protein release and caused an insignificant change in the expression of genes that are associated with inflammation and angiogenesis.

Upgrading the Quality of Silk Worm Oak(Quercus acutissima Carr.) Fiberboard through Paraformaldehyde Treatment by the Dry Forming Process (파라포름알데히드 처리(處理)에 의한 상수리나무 건식섬유판(乾式纖維板)의 재질개량(材質改良))

  • This study was carried out to determine the possibility of making fiberboard through the dry forming process, utilizing the tannin-like material of the Asplund pulp from the silk worm oak as a binder. The fiberboard was made through paraformaldehyde treatment, spray of NaOH solution and adjustment of mat moisture content, by hot-pressing at $220^{\circ}C$, and 50-6-50kg/$cm^2$ pressure with 6-min. (0.6-1.8-3.6) three-stage pressing cycle. The results are summarized as follows: 1. The modulus of rupture value of fiberboard treated with NaOH solution was greater than that of untreated. The value was increased in proportion to the paraformaldehyde content from I to 5%, but there was no increasing between 5 and 7% paraformaldehyde content. The value became higher along with the increase of mat moisture content from 15% to 25%. 2. The water adsorption of fiberboard treated with NaOH solution was lower than that of untreated. The value was de"creased in proportion to the paraformaldehyde content from 1 to 5%, but there was no decreasing between 5 and 7% paraformaldehyde content.. 3. A good quality fiberboard (modulus of rupture value of about 250kg/$cm^2$) was made through 5% paraformaidehyde treatment and 3% treatment of 10% NaOH solution, when the mat moisture content was 25%. was 25%.

Comparison of the biodegradability in the membranes for the guided bone regeneration: preliminary study

  • In this study, 4 different types of GBR membrane were undergone for bio-degradability test; Silk mat from silkworm cocoon (TDI), silk mat from flatwise-spun (FS), collagen membrane (OssGuide), and dPTFE membrane (Cytoplast). Each material was segmented in 10.00 mm length and 0.3 mm thickness. The samples were put into the normal saline at $37^{\circ}C$ for 2 weeks. After that, yield strength and tensile strain were measured and compared them with those of before treatment. The morphology of the membranes was observed by SEM. Tensile strain of FS was significantly increased at 2 weeks' normal saline treatment (P=0.018). When compared to OssGuide, TDI and FS showed significantly higher tensile strain at 2 weeks' normal saline treatment (P<0.05). In the SEM images, there were no significant changes in Cytoplast, TDI, and FS after 2 weeks' treatment. However, OssGuide showed damaged surface after 2 weeks' treatment. In conclusion, both TDI and FS did not have any evidence of biodegradability at 2 weeks' observation in normal saline treatment. However, OssGuide showed more than 20 % decrease in yield strength and tensile strain.

Comparison of unprocessed silk cocoon and silk cocoon middle layer membranes for guided bone regeneration

  • Background: Silk cocoon is composed of multiple layers. The natural silk cocoon containing all layers was cut as a rectangular shape as defined as total group. The inner and outermost layers were removed from the total group and the remained mat was defined as the middle group. The objectives of this study was to compare the total group with the middle group as a barrier membrane for the guided bone regeneration. Methods: The effects of these materials on the cellular proliferation and alkaline phosphatase (ALP) expression of MG63 cells were explored. For comparing bone regeneration ability, bilateral bone defects were created in calvarial areas in ten adult New Zealand white rabbits. The defects were covered with silk membranes of the middle group, with silk membrane of the total group used as the control on the contralateral side. The defects were allowed to heal for 4 and 8 weeks. Micro-computerized tomography (${\mu}CT$) and histological examination were performed. Results: The middle group exhibited a higher MTT value 48 and 72 h after treatment compared to the total group. ALP expression was also higher in the middle group. The results of ${\mu}CT$ and histologic examination showed that new bone formation was significantly higher in the middle group compared to the total group 8 weeks postoperatively (P < 0.05). Conclusions: In conclusion, the middle layer of the silk cocoon supports guided bone regeneration better than unprocessed silk cocoon.

A Study on Survey of The Actual Use of Bedding - Comparisons Between Taegu and Other Areas - (침구류(寢具類)의 사용(使用) 실태(實態)에 관(關)한 조사(調査) 연구(硏究) - 대구지역(大邱地域)과 전국(全國) 8대(大) 지역(地域)의 비교(比較) -)

  • Sleeping is important for human being in that it has a great effect on their daily activities. Considering that an average adult sleeps seven to eight hours a third of our life is spent in bed. The present study intends to make consumers aware of the important of the bedding science and help the manufactures develop efficient, up-to-date beds. For these two purposes, consumers' recognitions and opinions on the features of the bedding were surveyed. In the study, a questionnaire consisting of various items on the features of the bedding was designed and distributed to consumers in 8 regions & Taegu in Korea to survey the patterns in which they buy, use, and maintain their bedding. The result of the study are as follows; Air conditioner is widely used popular because of hot weather in Taegu. People in Taegu mostly purchase their bedding from market, in every 5~6 years. The important point to be considered for bedding purchases were humidity absorption, air permeability, light weight in summer comforters, thermal insulation, flexibility, color figure design in winter comforters, humidity absorption, flexibility, color figure design in mats. As summer bedding, a sheet of single-layer quilt and rush mat were most popular which indicates climate of hot and humid weather in Taegu. Bedding uses in winter were in order a sheet of cotton, silk, wool quilt, and silk quilt showed high level in contrast to other areas. Dissatisfactions with summer quilts were humidity absorption, air permeability, heaviness and in winter quilts they were dissatisfied with thermal insulation, heaviness, flattening. In details about bedding managements, people in Taegu most frequently disinfected their bedding by sunlight in one month, and refinished every 5 years.
