Studies on the Solubility Phenomenon and Activities of Silk Cocoon Sericin through the Filature Water Conditions. (제사용수의 수질이 견층세리신의 용해에 미치는 거동구명에 관한 연구)

  • This study was carried out ill an attempt to investigate the properties and activities of sericin obtained from silk cocoon shells in silk reeling water through various instrumental analyses. In addition, the effects of the characteristics in sericin solubility on the reeling process and silk qualities were also studied on the basis of tile above conditions. The results obtained are as follows: I. The sericin properties and activities through various analytical instruments. 1. The water solubility of each amino acid such as serine, glycine. glutamic and aspartic acids against the pH modified water by using automatic amino acid analyzer, showed the lowest solubility at PH 5, but it increased according to drifting toward the alkalinity. 2. When the obtained sericin particles by water pH variation were observed with the electronic microscope, it was found for the sericin Particles to expand in the alkaline regions. 3. The IR spectrum results showed the differences among the pH modified sericin solutions at the range of 2,100cm-1 and 1.890cm-1 of wave number. 4. The existence of sericin with in silk fabrics made differences in the X-ray interference intencity, that is, the non-degummed fabrics had the interference peak at 2$\theta$=14$^{\circ}$, 17$^{\circ}$, 24$^{\circ}$, and the degummed ones had it at 2$\theta$=17$^{\circ}$, 18$^{\circ}$, 20$^{\circ}$, 23$^{\circ}$, 26$^{\circ}$. II. The results of sericin solubility for silk reeling process. 1. The sericin solubility and swelling had a tendency to increase up with high M-alkalinity and pH value of water. But in case of acidity. water hardness and concentration of ethylene glycol were high, the sericin solubility and swelling were decreased. 2. With the filature experiments, the best conditions of filature orator are summarized as fellows ; a. pH; 6.9${\pm}$0.2 d. acidity; below10ppm b. total hardness; 55:5ppm e. Fe ion; none c. M-alkalinity; 40${\pm}$10ppm

Structure and properties of silk sericin obtained from different silkworm varieties

  • Recently, many researchers have studied silk sericin because of its high water retention, good wound healing, good cyto-compatibility, and blood-glucose- and cholesterol-lowering effects. Although sericin film can be used in wound dressing and cosmetic packs, its poor mechanical properties have prevented its use in industrial fields. In the present study, sericin was obtained from different silkworm varieties, and the effect of silkworm variety on the structure and properties of sericin was examined. Except for a small difference in serine content, no significant difference in sericin was noted among the silkworm varieties. In addition, silkworm variety almost had no effect on solution viscosity, implying that it does not influence the molecular weight of sericin. Mechanical properties of sericin film were strongly affected by silkworm variety. Wonwon 126 showed the best mechanical properties, while N74 and Geumgwangju displayed the worst properties.

  • Silk Sericin(SS) is a natural protein extracted from cocoon of bombix mori and shows moisturizing effect to the skin due to a number of hydroxyl groups in the structure. But its application to cosmetics is limited due to its poor solubility in water. In order to solve this drawback and expand its application to cosmetics, polyethyleneglycol(PEG) was conjugated with sericin by reacting activated polyethyleneglycol(ActPEG). Reaction site of sericin is tyrosine residue, which was determined by using $^1$H-NMR. Random coil structure of sericin was transformed to beta-sheet structure by conjugating polyethyleneglycol. It was confirmed that melting point of sericin-PEG conjugate was lowered compared to that of each sericin and PEG due to the interaction between sericin and PEG in crystalline structure. Self-assembled sericin-PEG nanoparticle was obtained by dialyzing with alcohol solution of sericin-PEG conjugate against water. The particle is spherical and has 200-400nm of size. The moisturizing ability of sericin-PEG nanoparticle was much higher than that of sericin itself. Incorporation of vitamin A into sericin-PEG nanoparticle was carried out by diafiltration method. The content of incorporated Vitamin A in sericin-PEG nanoparticle was 8.9 wt%. Releasing behaviour of vitamin A incorporated into nanoparticle was tested in phosphate buffer, pH 7.4 at 37$^{\circ}C$. and half-life of Vitamin A release was 43hrs. Sericin-PEG nanoparticle exhibited higher moisturing effect than sericin itself and distilled water, respectively. No toxicity and irritation were observed in animal tests. It can be expected that the self-assembled sericin-PEG nanoparticle can be developed for cosmetics.

Half-degumming Behaviors of Raw Silk Yarns Degummed with Soap and Alkalis (비누와 알칼리에 의한 실크 생사의 반숙정련 거동)

  • To investigate the half-degumming of raw silk, the degumming was carried out with soap or sodium carbonate or sodium bicarbonate and some mixed agents at $80^{\circ}C$ for 15 and 30 minutes. With soap of 20~25%o.w.f. degumming losses were about 7~12%. Degumming losses with sodium carbonate of 2~5%o.w.f. were about 6~15%. And about 6~12% wright losses were observed in the degumming with sodium bicarbonate of 20~30%o.w.f. In the degumming with the mixed degumming agents of soap and sodium carbonate, degumming losses were 9~15% with 5%o.w.f. soap and 2~5%o.w.f. sodium carbonate. Sodium carbonate was more effective on the removal of sericin from raw silk than soap. During drying generally half degummed silk yarns stuck together to result in harsh and hard lumps by the adhesion and solidification of the residue sericin of partially degummed silk. Sodium hydrosulfite in degumming agent effectively protected the adhesion of half degummed silk yarns.

  • Recently, sericin has attracted increasing attention in biomedical and cosmetic research because of its useful properties including acceleration of wound healing, improvement of cell attachment, and inhibition of ultraviolet-B induced apoptosis. However, sericin films have poor mechanical properties, which restricts the application to those fields. In this study, cellulose nanofibril (CNF)/sericin composite films were fabricated by solvent casting, and the effects of ultrasonication time and CNF content on the solution turbidity, molecular conformation, and film mechanical properties of sericin film were examined. As the ultrasonication time increased, the turbidity of the CNF/sericin suspension decreased. Conversely, as the CNF content increased, the turbidity increased. However, ${\beta}$-sheet crystallization and mechanical properties remained almost unchanged by varying the ultrasonication time and CNF content, indicating that CNF is not effective to improve the mechanical properties of sericin films.

  • In this study we have evaluated the separation characteristics and concentration of sericin using tubular type ultrafiltration membrane in silk degumming solution that extracted from electrolytic reduction water process. Ultrafiltration membranes have used in sericin separation performance and the separation characteristics of membrane satisfied typical Hagen-Poiseuille equation. It had the increase of flux according to the increase of feed pressure and temperature in occasion of pure water flux. And also the flux and solute rejection had about $25{\sim}60{\ell}/m^2{\cdot}h$ and more than 95% in sericin feed solution with concentration 1.00~1.89% at feed pressure force of $3{\sim}8kgf/cm^2$ respectively. In addition, the separation performance of tubular type ultrafiltration membrane for silk degumming solution was very steady-state with long experiment time.

The Effect of Fixing Agents and Softner on Sericin Fixation of Trimethylolmelamine (트리메틸올멜라민의 세리신 정착에 있어 정착제와 유연제의 영향)

  • The fixing behaviors of raw silk yarns treated with melamine and formaldehyde at a molar ratio of 1:3 for trimethylolmelamine were investigated. Sericin was fixed during the fixing process, but a part of sericin I was removed simultaneously by hot water. The weight losses by fixing and the degumming losses by degumming greatly decreased with increasing concentrations of melamine and formaldehyde. The silk yarns fixed with 0.011 M melamine and 0.033M formaldehyde were significantly degummed due to the insufficient fixation of sericin and the alkaline hydrolysis of sericin by sodium carbonate during the degumming process. On the other hand, the silk yarns fixed with 0.055M melamine and 0.165M formaldehyde were degummed slightly (the degumming losses of 3-8%) due to the strong fixation of sericin, which might result from the many cross-linkages between the sericin I molecules, which were formed by trimethylolmelamine. Those fixed with the fixing solution containing 15% owf softener showed the lowest weight and degumming losses because under the condition of 15% owf softener, the cation of the softener can effectively form ionic bonds with the negatively charged side chain of aspartic acid in sericin. In addition, van der Waals' forces may be also formed between the hydrophobic tail of the softener and the hydrophobic region of sericin, which may help inhibit the removal of sericin I.

Silk fibroin (SF), another main component of cocoon filament, has been investigated by many researchers due to its good physicochenucal properties.   • First of all, the properties imparted to PET fabrics are resistance to and recovery from creasing or wrinkling when wet or dry; high resistance to stretch in the filament yarns but not in the staple; high abrasion resistance; good texture and appearance; resistance to heat ageing; good chemical resistance and good resistance, behind glass, to sunlight. But, the low moisture regain of PET fabric conduces to static troubles in textile processing. Furthermore, garments made from PET may, during wear, develop electric charges which attract to the fabric particles of soil(dirt, swarf, dust) flying in the air, so that the cuffs of shirts, for example, become soiled quickly and are not easily laundered clean. The sericin constitutes 25$\sim$30% of silk protein and surrounds the fibroin fiber with sticky layer that supports the formation of a cocoon. The useful biochemical properties of sericin protein are oxidation resistant, antibacterial, UV resistant, hydrophilic property, and good affinity with hydrophobic material. These properties can be used as an improving reagent or a coating agent for natural and synthetic fibers, fabrics, and other intermediate products. The sericin is also applied to cross-link, and can be blended with other materials. In this study, we modified the surface of PET fabric by mixture of sericin finishing agent; sericin, polyuretane binder and 1,2,3,4-butanetetracarboxylic acid (BTCA) cross-link agent. Also, we investigated the finshing effect; moisture regain, stiffness, handle, drape and electrostatic. The moisture regain of PET fabric treated with sericin finishing agent was higher than that of untreated PET fabric. As a result of evaluating influence about handle of PET fabrics treated with sericin finishing agent, it was confirmed that the sericin finishing agent could be use as a linen like finishing agent.

Enzymatic Hydrolysis of Silk Sericin and Its Anti-oxidative Effect (효소에 의한 실크 세리신의 가수분해와 항산화 효과)

  • In this study, we hydrolyzed hot-water extracted sericin with single or two enzymes and investigated anti-oxidative effect on DPPH free radical and inhibitory effect on tyrosinase activity of the sericin hydrolysates. Alcalase, flavourzyme, and protamex were effective in hydrolyzing sericin. Sericin was degraded into the range of 20 ${\sim}$ 30 kDa. The sericin hydrolysate was shown to have stronger antioxidant properties than the original sericin. In the case of flavourzyme and protamex combination, the scavenging effect of sericin hydrolysate on DPPH radical was increased up to about 85 %. However, the inhibitory effect on tyrosinase activity of enzymatic hydrolysates was lower than that of the original sericin. After fractionation of sericin hydrolysates, we found that F2 and P3 fraction has higher inhibitory effect on tyrosinase activity compared to other fractions.
