A Jopock (Jpk), a trans-acting factor associating with the position-specific regulatory element of murine Hoxa-7, has shown to have a toxicity to both prokaryotic and eukaryotic cells when overexpressed. Since Jpk protein harbors a transmembrane domain and a putative endoplasmic reticulum (ER)-retention signal at the N-terminus, a subcellular localization of the protein was analyzed after fusing it into the green fluorescent protein (GFP): Both N-term (Jpk-EGFP) and C-term tagged-Jpk (EGFP-Jpk) showed to be localized in the ER when analyzed under the fluorescence microscopy after staining the cells with ER- and MitoTracker. Since ER stress triggers the ER-stress mediated apoptosis to eliminate the damaged cells, we analyzed the expression pattern of Jpk under ER-stress condition. When MCF7 cells were treated with the ER-stress inducer such as DTT and EGTA, the expression of Jpk was upregulated at the transcriptional level like that of Grp78, a molecular chaperone well known to be overexpressed under ER-stress condition. In the presence of high concentration of ER-sterss inducer (10 mM), about 70 (DTT) to $95\%$ (EGTA) of cells died stronly expressing ($10\~12$ fold) Jpk. Whereas at the low concentration ($0.001\~1.0\;mM$) of the inducer, the expression of Jpk was increased about 2.5 (EGTA) to 5 fold (DTT), which is rather similar to those of ER chaperone protein Grp78. These results altogether indicate that the ER-stress upregulated the expression of Jpk and the excess stress induces the ER-stress induced apoptosis and the concomitant expression of Jpk.
The physiological signals measured by Oriental Medicine instruments have been analyzed quantitatively in the view of the rule of promoting and counteracting relation of five evolutive phases theory. We tried to reduce the physiological signals measured by EAV(Elec-tro-Acupuncture according to Voll) and IR thermography to the representation of five evelutive phases. The EAV index and local skin temperature on acupuncture points of each phases measured and normalized so that the total value of five phases became unity. We assumed that the normalized EAV index and local skin temperature mean the deficiency or excess of Qi for each phases. The state of Qi distribution for each phases were approximately agree with the diagnostic pattern of O. M. doctor. Taking account of the Qi distribution state of·or the five evolutive phases, we performed a proper needle insertion on acupuncture points to induce the distinct change of Qi for each phases. We compared the measured results with the predictions of Qi variation by the rule of pro- moting and counteracting relation over the five evolutive phases. For all cases, the variation of Qi in the own phase on which a needle insertion was performed were exactly same to the theoretical prediction and partial agreement was shown for the other four phases. The same analysis was carried to the results of skin temperature measurements at accupoints. We found that the local skin temperature at accupoints of each phases shelved a finite change by the needle insertion and the behavior- of its change were strongly correlated to the rule of promoting and counteracting relation of five evolutive phases.
Although the physiological roles of calretinin have not been established, it may simply work as a calcium buffer or may actively work in calcium-mediated signal transduction. Calretinin plays a little role in the transport and physiological buffering of calcium in the adult photoreceptor cells, bipolar cells and horizontal cells of the human retina. We identified the calretinin-immunoreactive neurons in the inner nuclear cell layer and ganglion cell layer and the distribution pattern of the labeled neurons in the retina of a bat, Rhinolophus ferrumequinum, in this study. We observed the existence of calretinin-immunoreactive AII amacrine cell in the inner nuclear layer and ganglion cells in the ganglion cell layer of bat retina through this study. This observation must be significant along with our previous studies as we need to study for more understanding about the unsolved issue of a bat vision and the unique behavioral aspects of bat flight maneuverability.

Henoch-Shonlein purpura(HSP) is a systemic small-vessel vasculitis that primarily affects the skin, gastrointestinal tract, joints, and kidneys. The nervous system may be involved, less commonly than other organs. When the central nervous system(CNS) was involved, headache, changes in mental status, seizures, and focal neurologic deficits have been reported. Hypertension, uremic encephalopathy, metabolic abnomalities, electrolyte abnormalities, or cerebral vasculitis were suggested as possible causes of the neurologic manifestation. Diagnosis of vasculitic involvement of CNS is difficult. Magnetic resonance imaging of the brain is the modality of choice for the evaluation of the CNS disease. Steroid or plasmapheresis are used in treatment of cerebral vasculitis. We experienced a case of 9-year-old boy who had presented with Henoch-Schonlein purpura nephritis complicating encephalopathy accompanied by hypertension and cerebral vasculitis. Brain MRI showed multiple small nodular-linear pattern enhancing lesions in whole cerebral hemispheres and focal increased T2 signal in the right basal ganglia. We used intravenous immunoglobulin in treatment of cerebral vasculitis. We report this case with a brief review of related literature.
A cDNA clone encoding phytocystatin was isolated from Brassica rapa seedlings, through rapid amplification of cDNA ends (RACE). This gene (name as Brcpi1; GenBank accession no.: EF079953) had a total length of 881 bp with an open reading frame of 609 bp, and encoded predicted polypeptide of 203 amino acid (aa) residues including a putative N-terminal signal peptide. Other relevant regions found its sequence included the G and PW conserved aa motifs, and the consensus LARFAV sequence for phytocystatins and the reactive site QVVAG. The BrCPI1 protein shared 95, 94, 81, 80 and 78% identity with other CPI proterins isolated from Brassica oleracea (BoCPI-1), Arabidopsis thaliana (AtCY SB), Glycine max (GmCPI), Oryza sativa (OsCYS-2) and Zea may (ZmCPI) at amino acid level, respectively. Southern blot analysis showed that Brcpi1 was a low copy gene. Expression pattern analysis revealed that Brcpi1 was a tissue-specific expressing gene during reproductive growth and strongly expressed at mature seedling stages. Furthermore, overexpression of Brcpi1 in transgenic Arabidopsis was enhanced tolerance to salt and cold stresses. Meanwhile the juvenile seedling of Brcpi1 transgenic plants was not affected by various concentrations ABA in MS medium. Taken together, the results showed that Brcpi1 functioned as a cysteine protease inhibitor and it exhibited a protective agent against diverse types of abiotic stress, which induced this gene in a tissue- and stress-specific manner.

It is very essential to get the accurate classification of defects in primary pressure vessel and piping welds for the safety of nuclear power plant. Ultrasonic testing has been widely applied to inspect primary pressure vessel and piping welds of nuclear power plants during PSI / ISI. Classification of flaws in weldments from their ultrasonic scattering signals is very important in quantitative nondestructive evaluation. This problem is ideally suited to a modern ultrasonic Pattern recognition technique. Here, a brief discussion on systematic approach to this methodology is presented including ultrasonic feature extraction, feature selection and classification. A stronger emphasis is placed on Fuzzy-UTSCS (UT signal classification system) as efficient classifiers for many practical classification problems. As an example Fuzzy-UTSCS is applied to classify flaws in ferrite pressure vessel weldments into two types such as linear and volumetric. It is shown that Fuzzy-UTSCS is able to exhibit higher performance than other classifiers in the defect classification.
Glioblastoma is the most frequent and most aggressive brain tumor in adults. Solute carrier family 8 member 2 (SLC8A2) is only expressed in normal brain, but not present in other human normal tissues or in gliomas. Therefore, we hypothesized that SLC8A2 might be a glioma tumor suppressor gene and detected the role of SLC8A2 in glioblastoma and explored the underlying molecular mechanism. The glioblastoma U87MG cells stably transfected with the lentivirus plasmid containg SLC8A2 (U87MG-SLC8A2) and negative control (U87MG-NC) were constructed. In the present study, we found that the tumorigenicity of U87MG in nude mice was totally inhibited by SLC8A2. Overexpression of SLC8A2 had no effect on cell proliferation or cell cycle, but impaired the invasion and migration of U87MG cells, most likely through inactivating the extracellular signal-related kinases (ERK)1/2 signaling pathway, inhibiting the nuclear translocation and DNA binding activity of nuclear factor kappa B ($NF-{\kappa}B$), reducing the level of matrix metalloproteinases (MMPs) and urokinase-type plasminogen activator (uPA)-its receptor (uPAR) system (ERK1/2-$NF-{\kappa}B$-MMPs/uPA-uPAR), and altering the protein levels of epithelial to mesenchymal transitions (EMT)-associated proteins E-cardherin, vimentin and Snail. In addition, SLC8A2 inhibited the angiogenesis of U87MG cells, probably through combined inhibition of endothelium-dependent and endothelium-nondependent angiogenesis (vascular mimicry pattern). Totally, SLC8A2 serves as a tumor suppressor gene and inhibits invasion, angiogenesis and growth of glioblastoma.
It is often crucial to obtain a map of flood inundated area with more accurate and rapid manner. This study attempts to evaluate the potential of satellite synthetic aperture radar (SAR) data for mapping of flood inundated area in Imjin river basin. Multitemporal RADARSAT SAR data of three different dates were obtained at the time of flooding on August 4 and before and after the flooding. Once the data sets were geometrically corrected and preprocessed, the temporal characteristics of relative radar backscattering were analyzed. By comparing the radar backscattering of several surface features, it was clear that the flooded rice paddy showed the distinctive temporal pattern of radar response. Flooded rice paddy showed significantly lower radar signal while the normally growing rice paddy show high radar returns, which also could be easily interpreted from the color composite imagery. In addition to delineating the flooded rice fields, the multitemporal radar imagery also allow us to distinguish the afterward condition of once-flooded rice field.

Double patterning technology (DPT) has been suggested as a promising candidates of the next generation lithography technology in FLASH and DRAM manufacturing in sub-40nm technology node. DPT enables to overcome the physical limitation of optical lithography, and it is expected to be continued as long as e-beam lithography takes place in manufacturing. Several different processes for DPT are currently available in practice, and they are litho-litho-etch (LLE), litho-etch-litho-etch (LELE), litho-freeze-litho-etch (LFLE), and self-aligned double patterning (SADP) [1]. The self-aligned approach is regarded as more suitable for mass production, but it requires precise control of sidewall space etch profile for the exact definition of hard mask layer. In this paper, we propose etch end point detection (EPD) in spacer etching to precisely control sidewall profile in SADP. Conventional etch EPD notify the end point after or on-set of a layer being etched is removed, but the EPD in spacer etch should land-off exactly after surface removal while the spacer is still remained. Precise control of real-time in-situ EPD may help to control the size of spacer to realize desired pattern geometry. To demonstrate the capability of spacer-etch EPD, we fabricated metal line structure on silicon dioxide layer and spacer deposition layer with silicon nitride. While blanket etch of the spacer layer takes place in inductively coupled plasma-reactive ion etching (ICP-RIE), in-situ monitoring of plasma chemistry is performed using optical emission spectroscopy (OES), and the acquired data is stored in a local computer. Through offline analysis of the acquired OES data with respect to etch gas and by-product chemistry, a representative EPD time traces signal is derived. We found that the SE-EPD is useful for precise control of spacer etching in DPT, and we are continuously developing real-time SE-EPD methodology employing cumulative sum (CUSUM) control chart [2].
To investigate the mechanism of salt tolerance of gram-positive moderately halophilic bacteria, two-dimensional gel electrophoresis (2-D PAGE) was employed to achieve high resolution maps of proteins of Halobacillus dabanensis $D-8^T$. Approximately 700 spots of proteins were identified from these 2-D PAGE maps. The majority of these proteins had molecular weights between 17.5 and 66 kDa, and most of them were distributed between the isoelectric points (pI) 4.0 and 5.9. Some protein spots were distributed in the more acidic region of the 2-D gel (pI <4.0). This pattern indicated that a number of proteins in the strain $D-8^T$ are acidic. To understand the adaptation mechanisms of moderately halophilic bacteria in response to sudden environmental changes, differential protein profiles of this strain were investigated by 2-D PAGE and $Imagemaster^{TM}$ 2D Platinum software after the cells were subjected to salt shock of 1 to 25% salinity for 5 and 50 min. Analysis showed 59 proteins with an altered level of expression as the result of the exposure to salt shock. Eighteen proteins had increased expression, S proteins were induced, and the expression of 33 proteins was down-regulated. Eight of the up-regulated proteins were identified using MALDI-TOF/MS and MASCOT, and were similar to proteins involved in signal transduction, proteins participating in energy metabolism pathways and proteins involved in stress.
