  • The objectives of this research were to determine the effects of brown rice (BR) on food intake, digestion, energy value, and fecal characteristics. Three replacement levels which BR replaced 0, 15 and 30% of wheat flour were tested. Six female Miniature Schnauzer (8~9 month age, initial mean body weight 5.0 ${\pm}$ 0.3 kg) were assigned to treatments in replicated $3{\times}3$ Latin square design. Total tract digestibilities of DM, OM, acid hydrolyzed fat and gross energy except CP increased linearly (P<0.01), and observed digestible energy and metabolizable energy values also increased linearly (P=0.001 and P=0.006, respectively) with increasing BR replacement level. Wet and dry fecal output decreased linearly (P<0.001, P=0.004) with increasing BR inclusion to the diets and BR 30% treatment reduced wet fecal output up to 21% of that of control. Quadratic (P<0.01) effects was observed in fecal score for dog fed BR and fecal ammonia concentration tended to increase linearly (P=0.07) in response to increasing BR replacement level. It seems that the increase in fecal ammonia concentration may be partially related to the decrease in shortchain fatty acid concentration (P=0.001). This study clearly demonstrates that BR improves nutrients digestibility and fecal characteristics of dog.

  • Deer antler velvet is widely used in traditional medicine for its anti-aging, antioxidant, and immunity-enhancing effects. However, few studies have reported on the discovery of probiotic strains for deer antler fermentation to increase functional ingredient absorption. This study evaluated the ability of probiotic lactic acid bacteria to enhance the concentrations of bioactive molecules (e.g., sialic acid and gamma-aminobutyric acid [GABA]) in extracts of deer antler velvet. Seventeen strains of Lactobacillus spp. that were isolated from kimchi and infant feces, including L. sakei, L. rhamnosus, L. brevis, and L. plantarum, and those that improved the life span of Caenorhabditis elegans were selected for evaluation. Of the 17 strains, 2 (L. rhamnosus LFR20-004 and L. sakei LFR20-007) were selected based on data showing that these strains increased both the sialic acid and GABA contents of deer antler extract after fermentation for 2 d and significantly improved the life span of C. elegans. Co-fermentation with both strains further increased the concentrations of sialic acid, GABA, and metabolites such as short-chain fatty acids and amino acids. We evaluated the biological effects of the fermented antler velvet (FAV) on the antibacterial immune response in C. elegans by assessing worm survival after pathogen infection. The survival of the C. elegans conditioned with FAV for 24h was significantly higher compared with that of the control worm group fed only normal feed (non-pathogenic E. coli OP50) exposed to E. coli O157:H7, Salmonella typhi, and Listeria monocytogenes. To evaluate the protective effects of FAV on immune response, cyclophosphamide (Cy), an immune-suppressing agent was treated to in vitro and in vivo. We found that FAV significantly restored viability of mice splenocytes and immune promoting-related cytokines (interleukin [IL]-6, IL-10, inducible nitric oxide synthase [iNOS], interferon [IFN]-γ, and tumor necrosis factor [TNF]-α) were activated compared to non-fermented deer antlers. This finding indicated the protective effect of FAV against Cy-induced cell death and immunosuppressed mice. Taken together, our study suggests that immune-promoting antler velvet can be produced through fermentation using L. rhamnosus LFR20-004 and L. sakei LFR20-007.

  • Objective: The current study investigated the effects of mulberry 1-deoxynijirimycin (DNJ) on the digestion ability, intestinal morphology, and intestinal barrier of rabbits. Methods: A total of 36 New Zealand White rabbits (male) about 45 days old (mean body weight of 1.05±0.04 kg) were reared and commercial diets were employed, and afterwards divided into three groups (n = 12) with different levels of DNJ extract additive in feed: T0 (0 g/kg), T1 (0.35 g/kg), T2 (0.7 g/kg) for 28 d. Results: The results demonstrated that T2 decreased the average daily gain (p<0.05). T1 and T2 decreased villus height and inflammatory factor levels as compared with T0 (p<0.05). DNJ significantly decreased the content of valeric acid (p<0.05). The content of acetic acid, propionic acid, iso butyric acid, iso valeric acid in T1 were higher than those in T0 and T2 (p<0.05). The content of butyric acid in T2 was lower than it in T0 and T1 (p<0.05). The content of caproic acid was firstly improved then reduced as the DNJ concentration improved (p<0.05). T2 significantly increased the abundance of dgA-11_gut_group and Christensenellaceae_R-7_group while decreased Bacteroide and Ralstonia as compared with T0 (p<0.05). Compared with T0, T1, and T2 significantly improved the gene expression of JAM2, JAM3, mucin4, mucin6 (p<0.05), T1 significantly decreased the expression of occluding while T2 significantly increased (p<0.05), T2 significantly increased the expression of claudin1 and claudin2 (p<0.05). Conclusion: DNJ at high level changed microbiome compositions, inhibited inflammation, and improved intestinal barrier while it decreased the growth performance and shorted villus height in rabbit jejunum by regulating short chain fatty acid compositions in rabbits.

  • Hepatocyte damage caused by medications or herbal products is one of the important problem when these compounds are chronically administrated. Thus, improving hepatocyte survival during treatment offers a wide range of opportunities. Valproic acid (VPA), a branched short-chain fatty acid derived from naturally occurring valeric acid, is commonly used to treat epilepsy and seizures. Although VPA exerts numerous effects in cancer, HIV therapy, and neurodegenerative disease, its effects on the liver and its mechanism of action have not been fully elucidated. Here, we demonstrated that VPA caused moderate liver cell toxicity and apoptosis. Interestingly, VPA treatment increased transcription levels of PPAR alpha (PPAR-α) and fibroblast growth factor 21 (FGF21) in murine (Hepa1c1c7) hepatoma cells in a time and concentration dependent manner. VPA-induced FGF21 expression was significantly weaker under PPAR-α silencing condition than in cells transfected with non-targeting control siRNA. Subsequent experiments showed that cell viability was significantly lowered when the FGF21 signaling pathway was blocked by FGF receptor antagonist. Finally, we further determined that AMPK phosphorylation was not responsible for VPA-induced FGF21 expression and PPAR-a increments. These results indicate that increases of FGF21 expression alleviate VPA-induced hepatic toxicity, thereby making FGF21 a potential biomarker for predicting liver damage during VPA treatments.

  • This study was performed to compare the level of odorous compounds in feces and urine of finishing pigs. Feces and urine from 16 finishing pigs were separately collected for 28-d. Concentrations of volatile organic compound (VOC; phenols and indoles) and volatile fatty acid (VFA; SCFA and BCFA) were measured in feces and urine. Amount of phenols and p-cresol was higher (P<0.05) in urine than in feces. Urinal levels of phenols and p-cresol were 257.8 ppm and 250.9 ppm, and those of fecal phenols and p-cresol were 0.50 ppm and 0.05 ppm, respectively. There was no difference in concentration of indoles from feces (1.0 ppm) and urine (1.8 ppm). Short chain fatty acid (SCFA) level in urine was higher (P<0.05) than in feces showing 4,547 ppm and 863 ppm, respectively. Proportion of acetic acid to total SCFA was higher in urine (94%) than in feces (66%). However, level of branched fatty acid (BCFA) was greater in feces (118 ppm) compare to that of urine (87 ppm). Odorous compounds analyzed in the current study, phenols and SCFA, were contained more in urine than in feces. Greater amount of VFA is typically found in feces than in urine since it is generated in the large intestine. However, urine contained more VFA than feces in the current study. Therefore, it will be necessary to exploit odor reducing techniques especially for pig urine as grassland fertilizer.

  • This study was conducted to investigate the effect of storage time of manure on the concentration of odorous compounds. Levels of odorous compounds were measured from manure incubated in $20^{\circ}C$ for 6 wk in pilot chamber whose structure is similar to slurry pit. Levels of short chain fatty acids were decreased (p<0.05) by 4,159, 1,925, 844, and 483 ppm as storage time increased as 0, 2, 4, and 6wk, respectively. Transfatty acid level was not changed for 2wk but decreased (p<0.05) afterwards (levels were 250, 248, 151, and 61 ppm at 0, 2, 4, 6wk, respectively). Levels of phenol compounds were decreased (p<0.05) by 68, 48, 26, and 9 as storage time increased as 0, 2, 4, 6wk, respectively. Phenol concentration was increased whereas p-cresol level was decreased as storage time increased showing ratios of phenol and p-cresol were 6:94, 34:66, 51:49, and 67:33 at 0, 2, 4, and 6wk, respectively. Concentration of indole compounds was not different for 2wk but increased (p<0.05) after 4wk. The ratios of indole and skatole were 71:29, 42:58, 28:72, and 36:64 at 0, 2, 4, and 6wk, respectively. Skatole concentration was increased as storage time increased. Therefore, our current results indicate that levels of volatile fatty acid and phenol compounds were deceased but indole compounds were increased as manure storage time was increased.

  • Lipid-lowering effects of the inulin have been demonstrated in animal, yet attempts to reproduce similar effects in humans have generated conflicting results. In this study, the lipid-lowering potential of inulin and especially its effect on bile acid and neutral sterol excretion were investigated in Korean postmenopausal women. Nineteen postmenopausal women were randomly divided into two groups in a double-blind parallel design and consumed one of two supplements for 12 weeks; placebo of 8g maltodextrins/sucrose mixture (placebo group) or 8g inulin (inulin group). There were no significant changes in body weight during the supplementation period in either inulin or placebo group. Dietary consumption of animal fat in both group tended to decrease after 12 weeks of experiment. Intake of cholesterol was lower in placebo group, whereas the decrease of cholesterol intake in inulin group did not reach statistical significance after 12 weeks. The levels of serum total cholesterol (TC) and LDL-cholesterol (LDL-C) were significantly decreased in both placebo (p<0.05) and inulin group (p<0.01) after supplementation for 12 weeks compared with the baseline. The levels of serum triglyceride (TG) and HDL-cholesterol (HDL-C) were not significantly affected by inulin supplements, but atherogenic index (AI) and LDL-C/HDL-C ratio (LHR) as a predictor for coronary heart disease were improved (p<0.01) significantly after inulin supplementation. Therefore, inulin supplement may decrease the risk of cardiovascular disease via improving blood cholesterol level. Fecal weight and pH were not changed after 12 weeks of supplementation. There were no statistically significant changes for the fecal short-chain fatty acids (SCFAs). In inulin group, fecal deoxycholic acid (DCA) was significantly lowered compared with the baseline (p<0.05) whereas other bile acids were not changed. During the 12 weeks of intervention, no differences were found in fecal excretion of neutral sterol in the two groups. In summary, dietary inulin decreases serum TC, LDL-C, AI, LHR and lowers excretion of fecal DCA in the Korean postmenopausal women. These results support the use of inulin for reducing risk factors for hyperlipidemic postmenopausal women. However, the exact mechanism (s) responsible for the blood lipid lowering action of inulin including altered fecal bile acid remain to be elucidated.

  • This study showed that thermophilic anaerobic acid fermentation of food wastes had an enhanced hydrolysis capability and improvement of acidification efficiency. Influence of pH on the anaerobic hydrolysis and acidogenesis was investigated to determine the proper alkalinity in the thermophilic fermentation of food wastes. The results of putting NaOH as alkali to evaluate hydrolysis and acid fermentation efficiency In acid fermentation process of food wastes showed that the food wastes pretreated with 0.05 g NaOH/g TS had the maximum 12,600 mg/L of VFAs concentration during HRT 3 days in $55^{\circ}C$ thermophilic condition and the maximum 9,700 mg/L of VFAs concentration during HRT 5 days in $35^{\circ}C$ mesophilic condition. The accomplishment of high VFAs concentration resulted from that the main component of food wastes such as cellulose, lignin and etc. is performed active chemical decomposition by alkali in thermophilic condition. The major components of VFAs produced from the thermophilic acid fermentation process of food wastes were the short chain fatty acids such as acetic acid, butyric acid, and propionic acid.

  • In the present work, the in vitro prebiotic activity of xylooligosaccharides (XOS) derived from corn cobs combined with Lactobacillus plantarum, a probiotic microorganism, was determined. These probiotics exhibited different growth characteristics depending on strain specificity. L. plantarum S2 cells were denser and their growth rates were higher when cultured on XOS. Acetate was found to be the major short-chain fatty acid produced as the end-product of fermentation, and its amount varied from 1.50 to 1.78 mg/ml. The antimicrobial activity of XOS combined with L. plantarum S2 was determined against gastrointestinal pathogens. The results showed that XOS proved to be an effective substrate, enhancing antimicrobial activity for L. plantarum S2. In vivo evaluation of the influence of XOS and L. plantarum S2, used both alone and together, on the intestinal microbiota in a mouse model showed that XOS combined with L. plantarum S2 could increase the viable lactobacilli and bifidobacteria in mice feces and decrease the viable Enterococcus, Enterobacter, and Clostridia spp. Furthermore, in the in vitro antioxidant assay, XOS combined with L. plantarum S2 possessed significant 2,2-diphenyl-1-picrylhydrazyl, 2,2'-azino-bis, and superoxide anion radical-scavenging activities, and the combinations showed better antioxidant activity than either XOS or L. plantarum S2 alone.

  • The objective of the present study was to compare the chemical and sensory properties of regular cream cheese (control) and cholesterol-reduced cream cheese manufactured using crosslinked ${\beta}$-cyclodextrin (${\beta}$-CD) or powdered ${\beta}$-CD. Crosslinked ${\beta}$-CD was made using adipic acid. The composition of cream cheese treated by the crosslinked ${\beta}$-CD was similar to the regular cream cheese. Approximately 91% of cholesterol-reduction was observed in the cheeses that were treated using ${\beta}$-CD, which was not significantly different between powdered vs. crosslinked ${\beta}$-CD treatments. Total amount of short-chain free fatty acids was significantly lower in both ${\beta}$-CD-treated cheeses than in the control cheese throughout the storage. The cheeses made by ${\beta}$-CD-treated cream produced much lower amounts of individual free amino acids than the control in all periods. Most rheological characteristics, except cohesiveness, decreased dramatically in the control compared with the cholesterol-reduced cream cheeses. In sensory attributes, both wateryness and spreadability in ${\beta}$-CD-treated cheeses were significantly higher than in the control during 8 wk storage. Sensory scores for sourness increased significantly in the control from 4 to 8 wk storage, however, those in the cream cheese made by crosslinked-${\beta}$-CD treated cream increased slowly during 8 wk storage, which was shown in the control during a 4 wk period. Therefore, the present study showed the possibility of cholesterol-reduced cream cheese manufacture.
