The objective of this study was to examine the effect of root pruning intensity in combination with different initial seedling size on the growth of transplanting seedlings of Quercus acutissima. One-year-old seedlings were divided into three groups depending on their height, i.e. small (< 15 cm), medium (25-35 cm) and large size (35 cm <). Root of seedlings was pruned by three intensity such as, leaving 5 cm (severe), 10 cm (medium) and 15 cm (light) of taproot from the root-collar. After one year, we investigated survival rate, height and root-collar diameter (RCD) increment and final shoot dry weight. Also we measured characteristics of newly developed lateral roots such as number, total length, dry weight and diameter. Severe root pruning showed the lowest survival rate in all seedling size. Height increment, RCD increment and shoot dry weight were decreased with increasing intensity of root pruning. Seedlings of medium and light root pruning showed similar above-ground growth and dry weight of lateral roots. More large seedlings showed good survival rate, height increment and final shoot dry mass in all root pruning intensity. Therefore, one-year-old seedlings of Q. acutissima should be pruned taproot by 10 cm and transplanted to obtain excellent performance and increase the efficiency of transplanting work. Based on the findings of this study, it is important that applying to different root pruning intensity depending on initial seedling size for producing 2-year-old seedlings with excellent growth and high quality.

This experiment was carried out in order to estimate the recovery days of root and stubble to the days after cutting, and contribution of specific stubble weight on the regrowth was examined using the relationships between the dry weight of shoot and yield components, and regrowth parameters by the days after cutting. The varieties examined were Maprima, Manhattan, Tove, Peramo, Caliente, Tempo and P-2 grown under individual plant basis. The results are may be summarized as follows: 1. Dry weight of root and stubble were recovered up to 13.5 and 11 days after cutting, respectively. 2. Dry weight of shoot(regrowth parts+stubble) was affected significantly by the varieties, stages of regrowth and variety x stage of regrowth. 3. The variety with tiller weight type showed higher average productivity of shoot than those of the variety with tiller number type. 4. Absolute growth rate(AGR) of shoot was correlated significantly with regrowth parts, stubble, root and weight of a tiller at the early stage of regrowth(up to 12 days after cutting), and correlated with regrowth parts, stubble, weight of tiller and stubble area at the late stage of regrowth(up to 20 days after cutting). 5. Contribution of specific stubble weight to absolute growth rate of shoot was different between the stages of regrowth. Thus, regrowth parts per specific stubble weight(RP1SSbW) and weight of tiller per specific stubble weight(WT1SSbW) contributed to absolute growth rate of shoot at the early stage of regrowth, and efficiency of specific stubble weight(ESSbW), regrowth parts per specific stubble weight (RPISSbW) and weight of a tiller per specific stubble weight(WT1SSbW) contributed to absolute growth rate of shoot at the late stage of regrowth. 6. Regrowth utilization rate(RUR) was one of the useful regrowth parameter to indicate the regrowth potential of grasses.
We micropropagated pear (Pyrus species) using shoot tips and nodal explants from three pear genotypes. The ability to establish shoot tip cultures, proliferate shoots, induce rooting, and acclimatize the resulting plantlets are all elements of in vitro micropropagation. Shoots were induced from shoot tips on Murashige and Skoog medium (MS) with five different plant growth regulator combinations. The highest shoot formation rates were achieved for the three genotypes using MS supplemented with 1.0 mg/L N6-benzyladenine (BA) and 0.1 mg/L gibberellic acid (GA3). The maximum shoot number and shoot length for the three cultivars were recorded with 2.0 mg/L BA and 0.2 mg/L indole-3-butyric acid (IBA) in multiplication medium using nodal explants produced from microshoots. Nodal explants with one or two axillary buds cultured for three weeks initiated roots on medium supplemented with various concentrations of 1-naphthaleneacetic acid (NAA) or/and IBA in half-strength MS medium for adventitious rooting. The highest rooting response was with the combination of 0.2 mg/L NAA and 0.2 mg/L IBA. A combination of NAA and IBA resulted in a significant increase in the rooting ratio over NAA or IBA alone. In this medium, the root formation rate according to ranged from 68.9% for the BaeYun No. 3 genotype to 51.8% for the Hwanggeum genotype. We also investigated the influence of the concentration the polyamine phloroglucinol in rooting medium. For all three genotypes, the highest rooting ratio, longest root length, and greatest root number were observed in the treatments with 75-150 mg/L phloroglucinol. Most rooted plants were acclimatized successfully.

This study was conducted to examine effects of plant growth regulators and light resources on the formation of multiple shoot and plant regeneration of Hovenia dulcis var. koreana Nakai. Stem and shoot tip were cultured on MS medium or WPM supplemented with various plant growth regulators. At the single treatment, the highest shoot formation was obtained when stem explants were cultured on WPM supplemented with kinetin $1.0mg{\cdot}L^{-1}$. MS medium containing NAA 0.1 and TDZ $0.1mg{\cdot}L^{-1}$ gave the best results for shoot induction rate and shoot growth in combination treatments. Of the BAP and kinetin tested, BAP $0.5mg{\cdot}L^{-1}$ on WPM was found to be more effective for shoot growth from shoot tip. Under white fluorescent light treatment, shoot growth was much higher than blue, red LED treatments. Root induction from in vitro growth of plantlet was the best on WPM supplemented with $1.0mg{\cdot}L^{-1}$ IBA. The results suggest that selection of plant growth regulators and light resources could be important factor to achieve an efficient in vitro growth.
This study was undertaken to know the possibility of in vitro propagation of Stevia through axillary bud culture and the results indicated that: (1) Addition of NAA (0.01-0.05 mg/l) alone on Murashige-Skoog basal medium promoted shoot differentiation and growth rate. And also additional of kinetin of 0.5-1.0 mg/1 alone showed the same trend as that of NAA: (2) Addition of both NAA (0.01-0.05 mg/l) and kinetin (0.5-1.0mg/l) to MS medium promoted better shoot formation. An efficient system for high frequency plant regeneration was established through investigating various factors such as plant growth regulator combinations, explant types and ages, and addition of $AgNO_3$ influenced on shoot regeneration in Brassica juncea L. cv. BARI sarisha-10. Murashige and Skoog (MS) medium supplemented with 0.1 mg/L NAA (1-naphthaleneacetic acid) and 1 mg/L BA (6-benzyladenine) showed the maximum shoot regeneration frequency (56.67%) among the different combinations of NAA and BA. Explant type, explant age, and addition of $AgNO_3$ also significantly affected shoot regeneration. Of the four type of explants (cotyledon, hypocotyl, root, and leaf explants)- cotyledon explants produced the highest shoot regeneration frequency and hypocotyls explants produced the highest number of shoots per explant, whereas root explants did not produce any shoot. The cotyledonary explants from Four-day-old seedlings showed the maximum shoot regeneration frequency and number of shoots per explant. Shoot regeneration frequency increased significantly by adding $AgNO_3$ to the medium. Two mg/L $AgNO_3$ appeared to be the best for shoot regeneration with the highest shoot regeneration frequency (86.67%) and number of shoots per explant (7.5 shoots). Considerable variation in shoot regeneration from cotyledonay explants was observed within the B. juncea L. genotypes. The shoot regeneration frequency ranged from 47.78% for cv. Shambol to 91.11% for cv. Rai-5. In terms of the number of shoots produced per explant, B. juncea L. cv. Daulot showed the maximum efficiency. MS medium supplemented with 0.1 mg/L NAA showed the highest frequency of rooting. The regenerated plantlets were transferred to pot soil and grown to maturity in the greenhouse. All plants were fertile and morphologically identical with the source plants.

To select the section with shoot formation ability, the calli and shoot formation from three sections (first leaf including cotyledonary node, hypocotyl and cotyledon explants) of 5-days-seedlings of soybean were induced on MS medium supplemented with 1.0 mg/L BAP, 3% sucrose, and 0.3% gelrite for one month. The first leaf section exhibited the highest shoot formation rate (51%), followed the hypocotyl section (10%) and the cotyledon section (0%). The shoot formation rates and shoot number of the four excised sections (whole first leaf, a half of the first leaf, a third of the first leaf and only node) of the first leaf were also investigated on the same medium. A half of the first leaf explant and the third of the first leaf explant had higher shoot formation rates (76-80%) and numbers (3-4 / explants) than those in other two explants. Effects of six cytokinins (kinetin, zeatin, BAP, 2iP, PBA, and TDZ) on shoot formation were determined, using the half of the first leaf explants. Zeatin (1.0 mg/L) exhibited the highest in shoot formation rate (94%) and numbers (8 / explant). In addition, the combined effects of three cytokinins (zeatin, BAP, and TDZ; 0.5, 1.0, 2.0 mg/L, respectively) and an auxin (IAA; 0.0, 0.5, 1.0, 2.0 mg/L) were determined. The combination (1:1, v/v) of zeatin (1.0 mg/L) and IAA (1.0 mg/L) exhibited the highest in shoot formation rate (96%) and numbers (16 / explant), twice more than zeatin (1.0 mg/L) alone. The shoot cuttings were transferred and cultivated on the rooting media supplemented with only auxin, IBA at various concentrations. The highest root formation (8 / shoot) was achieved on the medium supplemented with 1.5 mg/L. After 4 weeks of cultivation, the plantlets with an extensive root system were transplanted in pots with a soil mixture of vermiculite and fine sand. Transferred to field, about 75% of the plantlets survived.

To determine the appropriate concentrations of nutrients and growth regulators for shoot proliferation and root initiation, several rose hybrid tea cultivars were cultured. Cultured shoot tips and lateral buds from different cultivars proliferated multiple shoots on Murashige and Skoog (MS) medium supplemented with 0 to 4 mg/L BA and 0 to 0.05 mg/L NAA. The ability of the explants to proliferate shoots and initiate roots was affected by genotype, the nodal position of explant, the strength of MS basal medium and growth regulators used. The buds nearest the apex exhibited the slowest rate of development. Most cultivars had the highest shoot proliferation when cultured on MS medium with 2 mg/L BA and 0.01 mg/L NAA, but the degree varied by cultivars. Root development was enhanced by lowering the concentration of MS salts.
This study was conducted to establish practical micropropagation of jujube cultivars ('Geumsumg', 'Bokjo') by axillary bud culture. The results are summerized as follows : 1. Addition of activated charcoal to half-strength Murashige and Skoog(MS) medium supplemented with 0.5mg/l benzylaminopurine(BAP) enhanced shoot and root growth. At 500mg/l activated charcoal level 'Geumsung' showed best result, and shoot length and the number of multiple shoot were 6.4cm and 10.0, respectively. At 1,000mg/l activated charcoal level 'Bokjo showed best result, and shoot length and the number of multiple shoot were 7.5cm and 12.4, respectively. 2. As indole-3-butyric acid(IBA) concentration increased, rooting and callus growth of microshoot were enhanced. The optimum IBA concentration for shoot elongation and multiplication was 1.0mg/l. 3. Growth responses of shoot-tip and axillary bud segments between two jujube cultivars were different. 'Geumsung' showed that axillary bud explants were about twice better than shoot-tip explants for shoot multiplication, but 'Bokjo' showed that shoot-tip explants mere better than axillary bud explants for shoot elongation and multiplication. 4. In acclimatization processes of plantlets produced in vitro, the survival of plantlets with only root primordia in soil medium was better than that of plantlets with several routs resulting in 97.8%. 5. In cutting of in vitro-derived microshoot, paclobutrazol was more effective than IBA, naphth-aleneacetic acid(NAA) and $Rooton^{(R)}$ in rooting and root growth.
This study was carried out to find the effects of the mixtures of earthworm cast, peatmoss, and vermiculite as a vegetable plant growth medium on the growth pepper seedlings. The mixed ratios of earthworm cast-peatmoss-vermiculite were 40-20-40, 40-30-30, 40-40-40, 50-20-30, 50-30-20, 60-10-30, 60-20-20 and 60-30-10%. The results of the study are as follows: 1, There was a significant difference of plant length, leaf area, shoot dry weight, root dry weight and biological yield per plant for growth stages and mixed ratios(p<0.05). But there was no significant interactions for both of them. 2. The nursery soil with earthworm cast was generally higher than the control treatment in shoot dry weight, root dry weight adn biological yield per plant. The shoot dry weight and biological yield per plant were high in the treatment including 60% of the earthworm cast to the 3rd week and in the one including 50% from the 4th week. But in root dry weight, the treatment including 40% of it was higher than treatment of the others. 3. The shoot dry weight per plant in treatments consisting both of 40% of earthworm, 40% of peatmoss and 20% of vermiculite and of 60%, 10% and 30% was more significant than that in the control treatment(p<0.05). 4. The average relative growth rates of shoot dry weight, root dry weight and biological yield for all treatments were higher than the ratio of control treatment except the ration of the treatment consisting of 60% of earthworm, 20% of peatmoss and 20% of vermiculite.
