Isolation of Ginsenoside Rh1 and Compound K from Fermented Ginseng and Efficacy Assessment on Systemic Anaphylactic Shock

  • Ginsenosides are responsible for the pharmacological and biological activities of ginseng. In this study, ginsenoside Rh1 and compound K were isolated and purified from fermented ginseng substrate and their anti-allergic effects were assessed in compound 48/80-induced anaphylactic shock model. The fermented ginseng substrate was extracted by methanol and ginsenoside Rh1 and compound K were efficiently purified by preparative high performance liquid chromatography (prep HPLC). Their quality and quantity were analyzed by liquid chromatography-mass spectrometer (LC-MS) and HPLC. Ginsenoside Rh1 showed better anti-allergic effects than compound K in compound 48/80-induced anaphylactic shock model. This study suggested that fermented ginseng extracts with enriched Rh1 may be utilized as a potential biomaterial of functional food for the alleviation of allergic symptoms.

  • In order to reduce the shock which may occur during shipping of a drumtype container, rubber mounts were designed using a commercial FEM package and manufactured, and then the performance was checked by static and dynamic test. According to the design specifications, the container system was tested by dropping. The experimental results are compared with the theoretical ones.

  • To assess the seismic performance of Plate-Shell Integrated Concrete Liquid-Storage Structure (PSICLSS), a scaled test model was constructed. This model incorporated a hybrid isolation system, which combined shape memory alloy (SMA), lead-cored rubber isolation bearing (LRB) and sliding isolation bearing (SB). By conducting shaking table test, the dynamic responses of both non-isolated and hybrid-isolated PSICLSS were analyzed. The results show that the hybrid isolation system can effectively reduce the acceleration and displacement responses of the structure. However, it also results in an increase in local hydrodynamic pressure and liquid sloshing height. Under extreme earthquake action, the displacement of isolation layer is small. When vertical ground motion is taken into account, the shock absorption rate of horizontal acceleration decreases. The peak hydrodynamic pressure increases significantly, and the peak hydrodynamic pressure position also changes. The maximum displacement of isolation layer increases, the residual displacement decreases.

  • Rocket, held using the CCA for the mission, a plurality of recording devices, and navigation equipment. If shock is transmitted directly to the CCA through the body, it can be caused malfunction of payloads, resulting in failure of the mission of the projectile. In order to ensure the stability against shock, in this paper, Calculating a target resonacne frequency of the CCA, and verified through modal test and analysis. Maximum acceleration position of CCA is checked by SRS analysis. In addition, effectiveness of shock isolation system through shock analysis.

  • Launch vehicles and satellites experience severe vibration and pyroshock during the flight phase. These severe dynamic loading could result in the malfunction of electric devices which equipped in the launch vehicle and satellite. In this paper, mesh washer isolators are applied to attenuate these severe shock and vibration and isolation performances are enhanced by applying pseudoelasitic SMA wire material. Through random vibration and ground pyroshock tests, outstanding isolation performances are studied. Especially, comparison of isolation performances due to the change of pre-compressive displacement of mesh washer itself are suggested and applicablity to the adaptive vibration control are confirmed.

  • In this study, the isolation system for multi-mechanism HIF (high impulsive force) device has been investigated. For this purpose, parameter optimization process has been performed based on the simplified isolation system model under constraints of moving displacement and transmitted force. The design parameters for multi-mechanism HIF device have been derived with respect to HIF system I and HIF system II, respectively. In order to implement the dynamic absorbing system, the dual stage hydro-pneumatic damper and magnetorheological damper with semi-active control scheme are considered. Finally, the performance of the designed prototype isolation system has been evaluated by experimental works under actual operating conditions.

Performance Evaluation of Advanced Municipal Wastewater Tretment by Phased Isolation Intrachannel Clarifier Ditch (침전지내장형 상분리 산화구공정에 의한 하수 고도처리특성 평가)

  • Phased isolation intrachannel clarifier ditch process developed in this study is an enhanced biological nutrient removal process employing two ditches with intrachannel clarifiers. Bench-scale phased isolation ditch process was used to evaluate the system performance on municipal wastewater and detailed assessment of internal behavior in a ditch and each reactions. When the system was operated at the HRTs of 6~12hours, SRTs of 9~31 days, and cycle times of 4hours, the system showed removals of BOD, TN, and TP as high as 88~97%, 73~78%, and 65~90%, respectively. The internal behavior were well matched on each reactions such as nitrification, denitrification, and phosphorus release and uptake. As the SRT became longer, TN removal increased gradually, whereas TP removal decreased contrarily. However, the system was capable of producing an effluent TP concentration 1mg/L or less even at longer SRTs except the case of solids discharge by malfunction of intra-clarifier occurred by its geometrical limit. The system performance slightly decreased by hydraulic shock loading(increasing of influent flowrate and decreasing of system HRT). However, the higher system performance could be achieved again after four cycles. Thus, the system reliability could be successfully achieved short-term hydraulic shock loading that occurred in medium- and small-sized wastewater treatment plants suffering fluctuation of influent quality and flowrate during wet season.

  • All types of dynamic excitation, periodical, pulse or transient in vertical, horizontal or all three directions can be effectively reduced by vibration isolation systems. Typical elements for vibration isolation control are spring units consisting of a group of helical compression springs. In all cases of shock, transient or random excitation energy absorbing dampers have to be added to the spring units in order to reduce system response in the frequency range near the natural frequency of the isolation system. The same isolation system of spring units and viscos-dampers has been used since 1979 for passive protection of buildings and structures has been proved to by very advantageous for vibration and structure borne noise control. Not only because of high vertical flexibility of the spring units, compared for example with typical rubber or neoprene mounts out also because of the horizontal of flexibility, which can be adapted by modifying the spring dimensions to nearly every requirement. It is just normal to use the same basic elements for passive isolation as for active isolation.

Ethnography on Isolation Unit for Hematopoietic Stem Cell Transplantation: Focusing on Patients (조혈모세포이식 병동에 관한 문화기술지: 환자를 중심으로)

  • Purpose: The purpose of the study was to understand how patients experience everyday life in an isolation unit for hematopoietic stem cell transplantation (HSCT). Method: The data were collected from 25 patients with HSCT at the isolation unit from January to March in 2008 in one general hospital in Korea. The data were collected by participant observations and ethnographic interviews and were analyzed using ethnographic method. Results: Four themes regarding environmental area emerged: 'barrier pulling up the drawbridge', 'very strange world', 'small and restricted space tied by IV and other treatment lines', and 'loud noise in a silent space.' Three themes regarding patients emerged: 'facing fear and anxiety', 'continuation of loneliness and lethargy', and 'compromising with a very long, dull, and boring time'. These themes describe how patients with HSCT suffer from continuous physical and psychosocial problems in a confined space, while endeavoring to control these problems and to search for hope for a new life. Conclusion: The results of the study provide an in-depth understanding of the experience and culture of patients in an isolation unit for HSCT. They would be used in developing practical programs to decrease patient's culture shock including fear and anxiety at isolation unit for HSCT.

Different Responses in Brain Regions upon Heat Shock in Adult Zebrafish (Danio rerio)

  • HSP70 has widely been induced in in vivo hyperthermia conditions in various organisms to study gene regulation and recently neuroprotectve roles of the induced gene expression under varying conditions. We investigated different responses among various tissues in zebrafish under heat shock to evaluate whether spatial and temporal expression pattern of zebrafish (z) hsp70 in transcriptional and translational level under heat shock stress in different brain regions. Heat shock groups were given for 1 h at $37^{\circ}C$ after recovery by transferring the treated animals back to $28^{\circ}C$ for 1, 2 and 24 h for recovery, respectively. Control (CTRL) group was kept at $28^{\circ}C$. At the end of treatments, five animals were collected and used for isolation of total RNAs and peptides from the corresponding tissues. Expression of zhsp70 mRNA showed different patterns in recovery periods in the tissues including the brain, eye, intestines, muscles, heart and testis by RT-PCR. Unlike the RT-PCR analysis, Northern blot analysis demonstrated nearly 30-fold increase in zhsp70 at 1 h heat shock, suggesting that RT-PCR may not be appropriate in unmasking regulation of the time-dependent zhsp70 expression. In the experiment involving different brain regions, the cerebellum showed gradual activation at 1 h to R1h and decreases in R2h and R24h, while the medulla oblongata and optic tectum showed gradual increase at R1h and decrease at R24h, indicating that different brain tissues respond specifically to heat shock in inducing zhsp70 and recovering from the heat shock status. Western blot analysis also demonstrated that the intracellular levels of zHSP70 in three different brain regions including the cerebellum, medulla oblongata and optic tectum are differently induced and recovered to normal state. These results clearly demonstrate that different regions of the body and the brain tissues are responding differently to heat shock in the aspects of its level of expression and speed of recovery.
