Influence of Rainfall During the Ripening Stage on Pre-Harvest Sprouting, Seed Quality, and Longevity of Rice (Oryza sativa L.)

  • The influence of rainfall during the ripening stage on pre-harvest sprouting, seed viability, and seed quality was investigated in two Korean rice cultivars, Shindongjin and Hopum. When the rainfall was artificially treated in a greenhouse, HP started to pre-harvest sprouting at three days of rainfall treatment (DRT), but Shindongjin did not show pre-harvest sprouting at 40 DAH treatment and just 0.3~0.8% at 50 DAH, which was much lower than 15.3~25.8% of Hopum in the same treatment. After harvest, the seed germination of Hopum decreased about 10~25% compared to non-treated seeds, but that of Shindongjin decreased much little rate than that of Hopum. The seed longevity tested by accelerated aging decreased with prolonged rainfall period in both cultivars, but the varietal difference was clear; Shindongjin could withstand longer accelerated aging than Hopum. Shindongjin maintained its germination (>50%) ability after 15 days of accelerated aging regardless of the rainfall treatment period and time, but Hopum dropped below 50% germination ability after only 5 days of accelerated aging. In conclusion, rainfall during the ripening stage induced not only pre-harvest sprouting, but also reduced seed quality and longevity during storage, which varied between two cultivars.

Molecular Characterization of Granule-Bound Starch Synthase (GBSSI) gene of Waxy Locus Mutants in Japonica Rice (Oryza sativa L.)

  • Five mutants were investigated at the molecular level to determine the factors responsible for mutated endosperm types. They were classified as high (HA) or low amylose (LA) phenotypes based on the amylose content in endosperm. The five were previously produced from Ilpum and Shindongjin cultivar treated with N-methyl-N-nitrosourea and gamma-ray irradiation, respectively. Analysis of the genomic structure and expression of Granule-bounded Starch Synthase I (GBSSI) genes revealed that mutants generally showed a higher incidence of nucleotide transition than transversion, and the $A:T{\rightarrow}G:C$ transition was particularly prevalent. The rates of nucleotide substitution in HA mutants were generally higher than those in the LA mutants, leading to higher substitutions of amino acid in the HA mutants. Neither nucleotide substitutions interfering with intron splicing or causing early termination of protein translation were found, nor any large-sized deletions or additions were found in all the mutants. In principle, amylose content can be regulated by three factors: internal alterations of GBSSI protein, the strength of gene expression, and other unknown external factors. Our results indicate that the endosperm mutants from Shindongjin arose from internal alterations of GBSSI proteins, which may be the result of amino acid substitutions. On the other hand, the Ilpum mutants might be principally caused by the alteration of gene expression level. Analysis of another three glutinous cultivars revealed that the major factor leading to glutinous phenotypes is the 23-bp duplicative motif (5'-ACGGGTTCCAGGGCCTCAAGCCC-3') commonly found in exon 2, which results in the premature termination of protein translation leading to the production of a non-functional GBSSI enzyme.

Monitoring of Rice Quality for Jeonju Bibimbab

  • This study was carried out to investigate the current use and the demand for Jeonju-bibimbab rice and to suggest the suitable range of bibimbab rice. We conducted a survey on varieties and rice quality of Jeonju bibimbab restaurants from April to August 2016. In a survey of 12 bibimbab restaurants in Jeonju, the restaurants showed that 83% of the 10 restaurants were using shindongjin rice variety. Characteristics of rice suitable for bibimbab were showed low stickiness, high hardness, so it should keep the shape of rice when cooked and mixed with the ingredients. and people tend to prefer a large grain of rice. Also, the rice used in bibimbab restaurants were examined the average percentages of head rice ratio 89%, protein content 6% and moisture content 14.5%. These results suggest that the quality of rice, which is suitable for bibimbab, is similar to the range of common rice. Thousand grain weight over 24g is especially preferred. In conclusion, the quality and grain size of rice preferred for bibimbab rice was the level of Shindongjin.

Amylose, Tocopherol, Free Sugar and Fatty Acid Content in Selected Mutant Lines of Oryza sativa cv. Shindongjin

  • To assess the potential as biofortified rice varieties, new endosperm and grain mutant lines were selected from $M_4$ generation seeds of the rice cultivar Shindongjin, which were either $\gamma$-irradiated or treated with N-methyl-N-nitrosourea(MNU) and lipid, sugar, and tocopherol content were analyzed. Amylose content in non-waxy mutants ranged from 8.8% in SM-4, a dull-type mutant, to 29.5% in SM-51, compared to 18.9% in the parental variety, Shindongjin. SM-23, a floury-type mutant, contained 0.09 ${\mu}g/g$ $\alpha$-tocopherol(40.9% of total tocopherol), was three times higher than in the parental variety. SM-32, a giant embryo-type mutant, had a 2.2-fold higher total tocopherol content, 2.1-fold higher $\alpha$-tocopherol, and 5.5-fold higher $\delta$-tocopherol content(47.3% of total tocopherol) than the parental variety(0.13 ${\mu}g/g$). Total free sugar content was elevated in all selected mutants and 1.2-8.6 times higher than in the parental variety(11.38 ${\mu}g/g$). These increased sugar levels were due to increase in sucrose concentration. SM-23(floury-type mutant) and SM-51(high amylose-type mutant) had 4.6 and 7.0 times more sugar, respectively, than the parental variety(11.38 ${\mu}g/g$). With relatively high concentrations, most mutants showed elevated fatty acid content in the SM 32(giant embryo-type) and SM-51(high amylose-type) mutants, at 124.56 and 89.59 mg/g, respectively. All selected mutants displayed valuable characteristics for the development of new varieties in rice-breeding programs.

Calibration of cultivar parameters for cv. Shindongjin for a rice growth model using the observation data in a low quality (저품질 관측자료를 사용한 벼 생육 모델의 신동진 품종모수 추정)

  • Crop models depend on a large number of input parameters including the cultivar parameters that represent the genetic characteristics of a given cultivar. The cultivar parameters have been estimated using high quality data for crop growth, which require considerable costs and efforts. The objective of this study was to examine the feasibility of using low quality data for the parameter estimation. In the present study, the cultivar parameters for cv. Shindongjin were estimated using the data obtained from the report of new cultivars development and research from 2005 to 2016. The root mean square errors (RMSE) of the heading dates were less than 3 days when the parameters associated with phenology were estimated. In contrast, the coefficient of determination for yield tended to be less than 0.1. The large errors incurred by the fact that no growth data collected over a season was used for parameter estimation. This suggests that detailed observation data needs to be prepared for parameter calibration, which would be aided by remote sensing approaches. The occurrence of natural disasters during a growing season has to be considered because crop models cannot take into account the effects of those events. Still, our results provide a reasonable range for the parameters, which could be used to set the boundary of a given parameter for cultivars similar to cv. Shindongjin in further studies.

Evaluation of Various Characteristics of High Quality Rice Varieties That Could Potentially be Grown on Reclaimed Land in Jellabuk Province, Korea (전북지역 간척지에서 최고품질 벼 품종의 작물학적·이화학적 특성 비교 및 선발)

  • The main objective of this study was to identify the best quality rice for cultivation on reclaimed land in Jeollabukdo-Province based on yield, head rice ratio, protein content, amylose content, and Toyo taste value evaluations. Seven varieties of rice were grown for two years on the reclaimed land. The Sukwang variety had the earliest heading date. The tiller number for the Younghojinmi variety was 19 on average, and its variation in tiller number was the lowest, which showed that the safety for cultivation of the variety was high on the reclaimed land with respect to that of the other varieties. The culm and panicle lengths of the Sukwang and Hyunpum varieties were the highest at 81.8 and 21.3 cm, respectively. Shindongjin produced the highest thousand grain weight at 27.7 g, whereas Haepum produced the lowest at 22.3 g. The number of grains per panicle for 7 all varieties was statistically equal for both years. The yield order for the varieties was Shindongjin (590) > Hopum (575) > Younghojinmi (552) > Sukwang (551) > Hopum (543) > Mipum (534) > Haepum (498 kg/10a), which showed that there was no significant difference in average yield between the varieties when both years are taken into consideration. However, the annual difference in Haepum yield between 2015 and 2016 was the highest at 108 kg/10a, indicating low cultivation safety. The head rice ratio of Younghojinmi was higher than for the other cultivars in both 2015 and 2016. However, there was no significant difference between the 2 years when the averages for all varieties were analyzed. There was was a considerable difference in protein and amylose content between the varieties in the two years, although this was not apparent in the results when the average over 2 years for each variety was calculated. There was no significant difference between the two years with respect to the average head rice ratio, and the protein and amylose contents of the seven varieties. The Toyo taste values for the Sukwang and Mipum varieties were higher than for the other varieties. Based on the principle characteristics, such as Toyo value, the Sukwang variety is a suitable substitute for Shindongjin, which has been previously used to produce high-quality rice on the reclaimed land.

A study on the development of high functional food protein ingredient from rice bran (고기능성 쌀단백질 소재 개발 연구)

  • Rice bran proteins from different cultivars(Youngan, Sindongjin, Suwon 511) were extracted with Xylanase using orthogonal analysis method and their functional properties were investigated. The optimum extraction conditions, based on protein content in the extract found to be at 1 wt% xylanase, pH 7 and 50:1, solvent to rice bran ratio(v/w %). Nitrogen solubility indices(NSI) of rice bran protein concentrates were shown a minimum value at pH 4 ranged 2~23%, varied with different cultivars and a maximum (NSI${\geq}$90% for all cultivars) at pH 10. As for water adsorption and fat adsorption capacity, rice bran protein concentrates were shown to be better than Na-caseinate and isolated soy protein, respectively. Emulsifying activities were observed high in order of Na-caseinate>Youngan rice bran protein>Shindongjin rice bran protein>Suwon 511 rice bran protein>isolated soy protein. In general, the surface tension of rice bran protein solution($10^{-3}$ wt%, 5 mM bis-tris, pH 7) was increased with increasing concentrations and found a minimum value near pI. On heating, it was decreased slightly with increasing temperatures up to $70^{\circ}C$ and then increased above $80^{\circ}C$. Addition of sodium chloride was made the surface tension decrease. In conclusion, with Xylanase, rice bran protein concentrate can be successfully extracted from the rice bran of different cultivars and the Youngan rice bran protein was thought to have best functionality among rice cultivars tested. It might be used as a milk protein substitute.

Evaluation of Rice Nitrogen Utilization Efficiency under High Temperature and High Carbon Dioxide Conditions

  • According to the 5th Climate Change Report, global average temperature in 2081~2100 will increase 1.8℃ based on RCP 4.5 and 3.7℃ based on RCP 8.5 from the current climate value (IPCC Working Group I AR5). As temperature is expected to increase due to global warming and the intensity and frequency of rainfall are expected to increase, damage to crops is expected, and countermeasures must be taken. This study intends to evaluate rice growth in terms of nitrogen utilization efficiency according to future climate change conditions. In this experiment, Oryza sativa cv. Shindongjin were planted at the SPAR facility of the NICS in Wanju-gun, Jeollabuk-do on June 10, and were planted and grown according to the standard cultivation method. Cultivation conditions are high temperature, high CO2 (current temperature+4.7℃·CO2 800ppm), high temperature (current temperature+4.7℃·CO2 400ppm), current climate (current tempreture·CO2 400 ppm). Nitrogen was varied as 0, 9, 18 kg/10a. The N content and C/N ratio of all rice leaves, stems, and seeds increased at high temperature, and the N content and C/N ratio decreased under high temperature and high CO2 conditions com pared to high temperature. Compared to the current climate, NUE increases by about 8% under high temperature and high CO2 conditions and by about 2% under high temperature conditions. This seems to be because the increase in temperature and CO2 induced the increase in biomass. ANUE related to yield decreased by about 70% compared to the current climate under high temperature conditions, and decreased by about 45% at high temperature and high CO2, showing a tendency to decrease compared to high temperature. This appears to be due to reduced fertility and poor ripening due to high temperature stress. However, as the nitrogen increased, the number of ears and the number of grains increased, slightly offsetting the production reduction factor.

Pre-Harvest Sprouting Variation of Rice Seeds Located on Each Panicle Position According to Grain Filling Days (벼 등숙일수에 따른 이삭 착생 부위별 종자의 수발아 변이)

  • This research was implemented to investigate pre-harvest sprouting characteristics in two Korean rice cultivars, Shindongjin (SDJ) and Hopum (HP). The panicles of both varieties were sampled after 15 days after heading (DAH) to 60 DAH at intervals of 5 days. As soon as sampled, the panicles were imbibed in water for 14 days at $25^{\circ}C$ to determine the vigor and germination ability of seeds according to location on panicle. To investigate the cause of non-germination of seeds in panicles, tetrazolium test and dormancy breaking were performed. The preharvest sprouting of HP started at 20 DAH while that of SDJ began at 30 DAH. The germination of seeds located in high rank branches within a panicle and the terminal seeds within a branch were earlier and faster and the germination patterns were same in both cultivars. The times at more than 50% of germination in a panicle were 35 DAH (57.0%) in HP and 45 DAH (56.8%) in SDJ. Preharvest sprouting was more than 80% at 50 DAH (82.6% of HP, 92.3% of SDJ) and more than 99% at 60 DAH (99.5%, 99.4%, HP and SDJ). These results indicated that the rate of PHS in a panicle increased with accumulation of the days after heading. The cause of non-germinated seeds at 15-25 DAH in panicle was immature embryo. After 30 DAH, however, when the non-germinated seeds were hulled, they started to germinate due to dormancy breaking, in which the germination percentage was higher in SDJ than HP. In conclusion, the pre-harvest sprouting varied according to days after heading, the seed position on panicle, and the dormancy intensity of varieties.
