지초뿌리 유래의 기능성 물질의 탐색

  • 지초(Lithospermum efthrorhizon)뿌리는 예로부터 한방약제로 이용되어져 왔으며, 지초 뿌리에 함유된 naphtoquinon계 색소물질인 shikonin은 항암, 항균, 항바이러스, 항염증 등의 효과가 있다고 보고되었다. 그러나 지초뿌리에 함유된 기능성 물질에 관한 연구는 미비한 실정이다. 이에 본 연구에서는 지초뿌리에서 기능성 물질의 탐색 및 기능성 해명연구를 위하여 지초뿌리를 ethanol로 추출한 후 이 추출물을 n-hexane, EtOAc, MeOH로 순차적으로 추출하여 MeOH추출물을 얻었다. 이 MeOH추출물을 silica gel adsorption column chromatography, ODS column chromatography, Sephadex LH-20 column chromatography로 순차 정제한 후 Shodex Asahipak column을 이용한 GPC-HPLC에 의해 활성 물질을 분리하였다. 분리된 물질들은 1H-NMR, 13C-NMR, COSY, HSQC, HMBC, FAB-MS 등의 기기분석을 통해 당 관련 화합물인 것으로 판명되었다.

DMNQ S64 exerts antitumor activity on A549 cells via COX-2 inhibition

  • We synthesized naphthazarin derivatives from shikonin, a major compound from Lithospermum erythrorhion Sieb et ZUCC. Of derivatives, DMNQ S64, 2-or 6-(l-hydroxyiminoalkyl) effectively showed antitumor activity on A549, human lung cancer cells (IC$\sub$50/= 30 ${\mu}$M). It significantly inhibited prostaglandin E$_2$(IC$\sub$50/= 10 ${\mu}$M). We also confirmed it selectively downregulated the expression of cyclooxygenase 2(COX-2), while it didn't affect COX-1. The induction of apoptosis by DMNQ S64 is underway.

오디 및 자초추출물의 피부 광노화 보호효과 (Protective Effect of Mulberry and Lithospermum erythrorhizon Extracts on Anti-aging against Photodamage)

  • 본 연구에서 적색의 색소 성분을 함유한 오디 및 자초 추출물의 피부 광노화 보호효과를 조사하였다. 오디 추출물의 총 안토시아닌 함량을 분석한 결과 4.92 mg/g이었고 HPLC 분석을 통한 자초 추출물의 shikonin 함량은 9.58 mg/g으로 나타났다. 오디 및 자초추출물의 전자공여능은 각각 84.32%와 57.46%로 분석되어 오디가 우수하였으며, superoxide radical 소거능은 오디 추출물이 76.34%로 자초추출물 61.55%보다 항산화 활성이 높았다. 오디 및 자초추출물의 ORAC 값은 각각 $545.37{\mu}moles$ TE/g과 $427.18{\mu}moles$ TE/g으로 분석되어 오디 추출물의 항산화 활성이 높음을 알 수 있었다. HS68 세포에서의 UVB 조사군과 비교 시 MMP-1 생산 억제율은 오디 및 자초 추출물 $200{\mu}g/mL$ 첨가 시 각각 68.6%, 32.7%로 분석되었다. 무모쥐를 이용한 UVB 광노화 피부 동물 모델에서 오디 및 자초 추출물의 피부조직의 변화를 조사한 결과, 시료 처리군이 무처리군에 비해 홍반, 주름 두께 및 깊이, 표피 비후, 콜라겐 각질층, 표피층이 감소되었으며 수분함량은 증가하였다. 총 주름 깊이의 경우, 오디 추출물 7% 처리군과 자초 추출물 5% 처리군에서 대조군과 비교 시 30% 가량 억제되어 in vitro에서 뿐만 아니라 in vivo에서도 오디 및 자초 추출물이 UVB에 의한 피부 광노화 보호효과가 있음을 확인하였다.

  • During the past two decades, China has seen her great progress in plant biotechnology. Since the Chinese market of herb medicine is huge, while the plant resources are shrinking, particular emphasis has been placed in plant tissue and cell cultures of medicinal plants, this includes fast propagation, protoplast isolation and regeneration, cell suspension cultures and large scale fermentation. To optimize culture conditions for producing secondary compounds in vitro, various media, additives and elicitors have been tested. Successful examples of large scale culture for the secondary metabolite biosynthesis are quite limited : Lithospermum ery throrhizon and Arnebia euchroma for shikonin derivatives, Panax ginseng, P. notoginseng, P. quinquefolium for saponins, and a few other medicinal plants. Recent development of genetic transformation systems of plant cells offered a new approach to in vitro production of secondary compounds. Hairy root induction and cultures, by using Ri-plasmid, have been reported from a number of medicinal plant species, such as Artemisia annua that produces little artemisinin in normal cultured cells, and from Glycyrrhiza uralensis. In the coming five years, Chinese scientists will continue their work on large scale cell cultures of a few of selected plant species, including Taxus spp. and A. annua, for the production of secondary metabolites with medicinal interests, one or two groups of scientists will be engaged in molecular cloning of the key enzymes in plant secondary metabolism.

진도홍주색소의 사용기준에 관한 연구 (Studies on the standard method of Jindo Hongju pigments)

  • Jindo Hongju is a traditional liquor in Jindo island of Korea. The characteristics of Hongju are its unique flavour by fermetation and red color of gromwell(Lithospermum erythrorhizon) root. However, the evaluation of red pigment is different from one manufactures to other manufactures and from place to place, also the standard method is not established. An attempt has made to compare the quality of gromwell root from different places and to standardize the extracting method. The results obtained from this study are summerized as follow, The chemical properties and composition of gromwell root from Jindo and other areas were compared. There were no difference among the samples in moisture content, content of naphtoquinone derivatives and absoption spectra. These results indicate that the pigments from Jindo and other region products seems to be the same quality. For efficient extraction of gromwell pigment, more than 40% ethanol as solvent and at least 10 hours extraction time was required. According to the visual test for Hongju pigment, the most preferable color was that it shows absorbance of 1.0 (contents of shikonin was 3.90 mg/45% EtOH 20 ml). From this visual test it can be proposed that the may be applied absorbance at 1.0 for the quality control of pigment.

견직물의 자초 염색 시 합성탄닌의 매염 효과 (Effect of Tannin Mordanting on Gromwell-dyed Silk Fabric)

  • The purpose of this study was to check color change depending on dyeing conditions when silk fabrics was dyed with gromwell and to investigate mordant effect when synthetic tannin was used as a mordant. First, when silk fabrics were dyed with gromwell, the change of color, brightness and chroma were examined by measuring K/S value, a, b value, L value and C value depending on dyeing condition. Second, color, brightness and chroma differences which appeared after mordanting with synthetic tannin were investigated and muti-functional mordanting effect was confirmed with dyeing fastness, antibiosis. As a result of color analysis of gromwell, it was proved that the main pigment of gromwell was shikonin. Color was red purple at pH 3 when silk fabrics were dyed with gromwell. But It became greenish and bluish as temperature rose and time passed by. And it grew reddish and yellowish as concentration level reached higher. The color became purple at pH 5 and purple blue at pH 7. Both at pH 5 and pH 7, it became greenish and yellowish, as temperature went up and time ran by. And it became reddish with concentration increasing. Brightness and chroma decreased with temperature and concentration increasing as well as time passing by. After mordanting with synthetic tannin, the color became less reddish at pH 3 and reddish-yellowish at pH 5 and pH 7. The brightness increased and the chroma decreased. At all pH, the color, the brightness and the chroma became similarly by synthetic tannin mordanting. Dyeing fastness of synthetic tannin mordanted fabric was higher than that of non-mordanted fabric. Especially, wet fastness at pH 3 improved from 1 grade before synthetic tannin mordanting to $3{\sim}4$ grade after tannin mordanting. Antibiosis was improved by gromwell dyeing and synthetic tannin mordanting.

천연염색 직물의 환경조건에 따른 변.퇴색 및 물성변화에 관한 연구 (Effect of Storage Conditions on the Color and the Mechanical Properties of Fabrics dyed with Natural Dyes)

  • Cotton and silk fabrics dyed with brazilein(Caesalpinia Sappan), berberine (Phellodendron Amurense), and shikonin(Lithospermum Erythrorhizon) were stored in the air, in the water, and under the ground for about one year. The change of the color, the flexibility, and the breaking strength were measured at the intervals of few weeks. The results are as follows; 1. The color of the fabrics changed most extensively in the fabrics that were under the ground, then in the order of the ones that were in the water, and in the air. The color of the fabrics changed to the achromatic color over the time. 2. The flexibility change among the dyes was similar. All the fabrics became stiff under the ground and in the water over the storage time. 3. In general, cotton and silk fabrics dyed with berberine showed better strength retention than the fabrics dyed with other substances. 4. The strength retention of cotton fabrics was high in the order of the fabrics which were in the air, in the water, and under the ground. Only minuscule change occurred in the strength of the cotton fabrics. The strength of the fabrics that were in the water and under the ground decreased remarkably after 30 weeks and 20 weeks respectively. 5. The strength retention of silk fabrics was high in the order of the fabrics which were in the air, under the ground, and in the water. In the air, the strength on silk fabrics decreased rapidly after 30 weeks. In the water, the strength of silk fabrics decreased more rapidly than that of the cotton fabrics. Under the ground, the strength retention of silk was higher than that of cotton.

할로아세틸시코닌 유도체의 합성 및 항암성 평가 (Haloacetylshikonin Derivatives : Synthesis and Evaluation of Antitumor Activity)

  • The secondary hydroxy group at side chain of shikonin structure was selectively acylated with various haloacetic acids in presence of dicyclohexylcarbodiimide and 4-dimethylamin opyridine to produce haloacetylshikonin derivatives. The cytotoxicity of monohaloacetylshikonin derivatives against L1210 cells increased in the following order; monochloroacetylshikonin ($ED_{50}$, 0.142${\mu}$g/ml) > monobromoacetylshikonin ($ED_{50}$. 0.158${\mu}$g/ml) > monoiodoacetylshikonin ($ED_{50}$, 0.173${\mu}$g/ml). Introduction of larger halogen atoms decreased the cytotoxic activity, presumably due to steric hinderance. The cytotoxicity of chloroacetylshikonin derivatives was dependent on the number of chlorine atom, thus increasing in the following order; trichloroacetylshikonin (0.032${\mu}$g/ml) > dichloroacetylshikonin (0.059${\mu}$g/ml) > monochloroacetylshikonin ($ED_{50}$, 0.142${\mu}$g/ml). Thus, the electron withdrawing effect seems to be important for the cytotoxicity of chloroacetylshikonin derivatives. Consistent with the above, dichloroacetylshikonin (T/C. 182%) and trifluoroacetylshikonin (195%) showed higher T/C values than monochloroacetyl-(T/C, 122%), monobromoacetyl-(T/C, 154%) and monoiodoacetylshikonin (T/C, 117%) derivatives. Haloacetylshikonin derivatives showing lower cytoxic activities against L1210 cells exhibited lower T/C values. It seems that there is a relationship between the cytoxicity of haloacetylshikonin and their antitumor activity.

자초 뿌리를 이용한 직물의 염색에 관한 연구 (The Fabrics Dyeing using Gromwell Roots)

  • For the efficiency in extracting gromwell colorant, methanol, ethanol and aceton were used as solvents. Also, to compare the results when dyed in various conditions and on various fabrics, the ratio of water-dye mixture, pH, temperature, and varieties of fabrics -wool, silk, cotton, nylon, ramie- were selected. L, a, b, ΔE, munsell and K/S value of each sample was measured and compared for the practical use. Water-dye mixture of 5:5 ratio showed the optimum dyeability. Not to mention wool and silk, but also cotton showed a satisfying dying result at the acidity of pH4. As the temperature increased, the dyeability of all three fabrics improved greatly. Especially, wool showed the highest improvement in terms of dyeability as the temperature increased. K/S values for wool and nylon showed superior results to the others in the gromwell colorant. The resulting color turned out somewhat different depending on the solvents used and the sort of fabric that was tested on. When the gromwell colorant extracted with methanol or ethanol was applied, the color of fabrics came out as purple or purpleblue. When the colorant extracted with acetone was used, the color of fabrics came out to be redpurple. The colorfastness to light showed low glades regardless to whichever solvents were used. The grades of colorfastness to laundering were recorded low numerical values, and its record became even lower when the gromwell colorant extracted with aceton was applied. The staining grade of the colorfastness to laundering showed a good grade in the range of 4 to 5. All the dyed fabrics showed a excellent drycleaning fastness.

Acute and 28-Day Subacute Toxicity Studies of Hexane Extracts of the Roots of Lithospermum erythrorhizon in Sprague-Dawley Rats

  • Lithospermum erythrorhizon has long been used as a traditional oriental medicine. In this study, the acute and 28-day subacute oral dose toxicity studies of hexane extracts of the roots of L. erythrorhizon (LEH) were performed in Sprague-Dawley rats. In the acute toxicity study, LEH was administered once orally to 5 male and 5 female rats at dose levels of 500, 1,000, and 2,000 mg/kg. Mortality, clinical signs, and body weight changes were monitored for 14 days. Salivation, soft stool, soiled perineal region, compound-colored stool, chromaturia and a decrease in body weight were observed in the extract-treated groups, and no deaths occurred during the study. Therefore, the approximate lethal dose (ALD) of LEH in male and female rats was higher than 2,000 mg/kg. In the subacute toxicity study, LEH was administered orally to male and female rats for 28 days at dose levels of 25, 100, and 400 mg/kg/day. There was no LEH-related toxic effect in the body weight, food consumption, ophthalmology, hematology, clinical chemistry and organ weights. Compound-colored (black) stool, chromaturia and increased protein, ketone bodies, bilirubin and occult blood in urine were observed in the male and female rats treated with the test substance. In addition, the necropsy revealed dark red discoloration of the kidneys, and the histopathological examination showed presence of red brown pigment or increased hyaline droplets in the renal tubules of the renal cortex. However, there were no test substance-related toxic effects in the hematology and clinical chemistry, and no morphological changes were observed in the histopathological examination of the kidneys. Therefore, it was determined that there was no significant toxicity because the changes observed were caused by the intrinsic color of the test substance. These results suggest that the no-observed-adverse-effect Level (NOAEL) of LEH is greater than 400 mg/kg/day in both sexes.
