Pathological Properties of Cryptococcus pseudolongus on the Mycelia and Fruit Body of Lentinula edodes

  • Recently, Cryptococcus pseudolongus has been reported as a new pathogen of shiitake (Lentinula edodes). However, its pathological properties are not much known. To further understand its impact on the mushroom, we investigated the pathogen's interactions with the mycelium of shiitake, histopathological properties, host range, and sensitivity to diverse antifungal agents. The strain C. pseudolongus DUCC 4014 inhibited the mycelial growth of L. edodes strain (cultivar Sanjo 701ho) and caused browning in the mycelia confronted with the yeast on PDA. Spray inoculation of the yeast caused an abnormal browning symptom on the cap and/or gills of three shiitake cultivars grown on sawdust media in vinyl bags. Scanning electron microscopic images of the abnormally browned parts of shiitake fruit body illustrated that mushroom tissues were loosed and dispersed in the middle and edge of the cap and the arrangement of basidiospores borne on basidia in the gills was disturbed compared to those of normal shiitake fruit body. Spray inoculation also led to developing abnormal browning on the harvested fruit body, indicating C. pseudolongus could be a problem during mushroom storage. But the yeast was not able to induce abnormal browning on mushrooms of Pleurotus ferulae, Pleurotus fostreatus, and Agaricus bisporus. But it induced browning only on button mushroom (A. bisporus) when they were inoculated after wounding. Tests with 16 kinds of fungicides revealed that the cell growth of C. pseudolongus could be inhibited by benzalkonium chloride at MIC 7 ㎍/ml and benomyl at MIC 3 ㎍/ml.

Determination of Glucan Contents in the Fruiting Bodies and Mycelia of Lentinula edodes Cultivars

  • Shiitake mushrooms (Lentinula edodes) containing ${\beta}$-glucans may be beneficial for human health; they have been used in the treatment of cancer, hypertension, and high cholesterol levels. The objective of this study was to determine the ${\beta}$-glucan content in different sections of the fruiting bodies and mycelia of ten shiitake mushroom cultivars. The measured ${\beta}$-glucan content ranged from $20.06{\pm}1.76%$ to $44.21{\pm}0.13%$ in the pileus sections, and from $29.74{\pm}1.40%$ to $56.47{\pm}4.72%$ in the stipe sections. The results of this study indicate that the variance in ${\beta}$-glucan content dependent on the shiitake cultivar, and that the ${\beta}$-glucan content is higher in the stipe than in the pileus.

Breeding and Screening of Lentinula edodes Strains Resistant to Trichoderma spp.

  • Trichoderma spp. cause large crop losses of the cultivated shiitake mushroom, Lentinula edodes. We bred several shiitake strains that are resistant to Trichoderma spp. using di-mon mating to establish a useful method for controlling the greenmold disease. We examined the competitive ability of L. edodes against Trichoderma spp. using a dual culture system to select resistant strains. By screening Trichoderma-resistant strains, we found that among 11 parental strains, 4 strains, including KFRI 36, were confirmed resistant strains. They showed especially strong resistance to T. harzianum, which formed deadlock after mycelial contact and then invaded into the territory of T. harzianum. KFRI 171 also showed resistance to T. atroviride strains. Among 13 strains, which were made by hybridization of shiitake strains, 5 were confirmed to be resistant to Trichoderma, including KFRI 58-1. Their resistance was not correlated to the resistant activity of their parents’ strains. Two strains lose resistance and two strains acquire resistance compared to their parents’ strains. In SEM observation, the mycelium of L. edodes at the interaction zone of Lentinula-Trichoderma was rugged and swollen by T. harzianum.

Identification of Trichoderma, a Competitor of Shiitake Mushroom (Lentinula edodes), and Competition between Lentinula edodes and Trichoderma species in Korea

  • During investigating of shiitake mushroom competitors, 289 isolates of Trichoderma spp. were collected from shiitake mushroom farms in different districts and the Forest Mushroom Research Center of Korea, among which 29 representative strains were selected. Based on the DNA sequences of the rpb2 and tef1 genes and the ITS rDNA, and their morphological characteristics, they were identified as T. atroviride, T. citrinoviride, T. harzianum, T. longibrachiatum, and two undescribed species, Trichoderma spp. 1 and 2, which are considered to be the candidate of new species. Competition tests between Lentinula edodes (Sanjo302) and the Trichoderma species indicated that the six species of Trichoderma were significantly different from each other in terms of their ability to invade the mycelial blocks of shiitake. In both of dual cultures on potato dextrose agar and sawdust media, Trichoderma spp. 1 and 2 strongly invaded the mycelial blocks of shiitake. Our results suggest that the two Trichoderma species may cause potentially serious economic losses in shiitake cultivation of Korea.

Ergothioneine Contents of Shiitake (Lentinula edodes) Fruiting Bodies on Sawdust Media with Different Nitrogen Sources

  • Ergothioneine is a natural compound with strong antioxidant properties. In this study, the effects of different nitrogen sources including ammonium nitrate, ammonium sulfate, sodium nitrate, and histidine in sawdust media were investigated to enhance ergothioneine contents in Shiitake (Lentinula edodes) fruit bodies. The addition of 0.2% ammonium sulfate in the growth media showed the highest enhancement of ergothioneine content in shiitake fruit bodies which was 1.7-fold higher than the control. On the other hand, histidine, a building block of ergothioneine decreased the concentration of ergothioneine significantly. Our results demonstrate that the cultivation of shiitake in sawdust media with suitable nutrients was effective to enhance its ergothioneine contents.

Identification of Single Nucleotide Polymorphism Markers in the Laccase Gene of Shiitake Mushrooms (Lentinula edodes)

  • We identified single nucleotide polymorphism (SNP) markers in the laccase gene to establish a line-diagnostic system for shiitake mushrooms. A total of 89 fungal isolates representing four lines, including Korean registered, Korean wild type, Chinese, and Japanese lines, were analyzed. The results suggest that SNP markers in the laccase gene can be useful for line typing in shiitake mushrooms.

  • Shiitake mushroom (Lentinula edodes) is usually cultivated on the oak log. Log cultivation of this mushroom is getting difficult to get oak logs and has a weak point of its long cultivation period. Recently sawdust cultivation is getting increase. It is important to make mycelia browning on the substrate surface. This browned surface in sawdust cultivation plays an important role like as artificial bark of the oak log, which protects the other pests and suppresses water evaporation in the substrate. The period for mycelia browning is so long that the sawdust cultivation of Shiitake mushroom can not spread well into the mushroom farms. The development of methods for the rapid mycelia browning is quite required. In this article we would like to discuss about the enzymatic activities related mycelia browning and search the methods of cultivation period reduction.

국내 표고(Lentinula edodes) 주산지인 장흥지역에 적합한 국산 원목품종 선발 (Selection of domestic shiitake strains suitable for the cultivation in Jangheung region)

  • 본 연구에서는 원목재배용 표고(Lentinula edodes) 국산 품종에 대한 장흥지역에서 적합한 표고품종 선발을 위해 국내산 총 5개(천백고, 수향고, 백화향, 풍년고, 산조303호) 품종과 일본산 2개(모리290, 모리유지로) 품종을 선택하여 적합품종 선발을 위해 재배시험을 실시하였다. 생산량 조사결과 산조303호가 40.9 kg으로 모리290 39.2 kg, 모리유지로 37.7 kg 보다 우수한 생산량을 보였다. 표고의 품질은 백화향이 갓이 반구형으로 형태가 우수하였으며, 화고형으로 발생하는 특징을 가진 것으로 확인되었다. 기존 재배중인 일본산 품종과 발생특성이 유사하며 생산량이 우수한 산조303호와 고품질의 표고를 생산하는데 유리한 백화향이 장흥지역에서 적합한 국산 표고품종으로 선발하였다.

Negative Effects of Water Extracts from Pinus densiflora Sawdust on Mycelium Growth of the Shiitake Mushroom Lentinula edodes

  • Extracts from sawdust of Pinus densiflora were showed antifungal activity against Lentinula edodes. It was extracted by hot water and then successively extracted by n-hexane, ethyl acetate, and methanol. The yields of the n-hexane-soluble, ethyl acetatesoluble, methanol-soluble and methanol-insoluble fractions of water extracts were 8.2%, 10.6%, 32.0%, and 49.2%, respectively. The ethyl acetate-soluble fraction showed the greatest antifungal activity against L. edodes: 41.5% inhibition at 1,000 ppm. However, there were not significant differences of antifungal activities between n-hexane-soluble fraction and methanol-soluble fraction at a concentration of 1,000 ppm. The hot water extracts showed 23.5% of antifungal activity against L. edodes at a concentration of 1000 ppm. The four antifungal compounds were separated from ethyl acetate fraction by thin layer chromatography.

Influence of Storage Temperature on Levels of Bioactive Compounds in Shiitake Mushrooms (Lentinula edodes)

  • Shiitake mushroom (Lentinula edodes) hold high nutritional and medicinal value as they contain an abundance of health-promoting compounds. However, the effect of long-term postharvest storage on the variation in the levels of health-promoting compounds has not been extensively studied. In this study, we investigated the changes in the levels of phenolic compounds, antioxidants, eritadenine, and ergothioneine in shiitake mushrooms stored at three different temperatures (1, 3, and 5 ℃) for 4 weeks. Compared to mushrooms stored at lower temperatures, those stored at 5 ℃ exhibited a higher level of total phenolics in their pileus after 2 weeks of storage; however, storage at 5 ℃ also increased the deterioration of the fruiting body of these mushrooms. In mushrooms stored at all temperatures, the eritadenine content in the pilei tended to increase up to 2 weeks of storage. In contrast, the ergothioneine content in the pileus decreased during storage, with a significantly lower level detected in mushrooms stored at 5 ℃ for 4 weeks. Together, these results suggest that the mechanisms underlying the accumulation of phenolics and eritadenine may be related to mushroom deterioration during storage. Our findings indicate that the levels of health-promoting compounds in shiitake mushrooms are influenced by storage temperature, suggesting the potential to control adjustments of specific bioactive compounds by regulating storage conditions.
