CRISPR as a strong gene editing tool

  • Clustered regularly-interspaced short palindromic repeats (CRISPR) is a new and effective genetic editing tool. CRISPR was initially found in bacteria to protect it from virus invasions. In the first step, specific DNA strands of virus are identified by guide RNA that is composed of crRNA and tracrRNA. Then RNAse III is required for producing crRNA from pre-crRNA. In The second step, a crRNA:tracrRNA:Cas9 complex guides RNase III to cleave target DNA. After cleavage of DNA by CRISPR-Cas9, DNA can be fixed by Non-Homologous End Joining (NHEJ) and Homology Directed Repair (HDR). Whereas NHEJ is simple and random, HDR is much more complex and accurate. Gene editing by CRISPR is able to be applied to various biological field such as agriculture and treating genetic diseases in human.

『황제내경(黃帝內經)』의 '관절(關節)'의 의미에 대한 고찰(考察) (A Study on the Meaning of 'GuanJie(關節)' in the Huangdineijing)

  • Objectives : This paper aims to examine the meaning of guanjie(關節) in the Huangdineijing. Methods : Meanings of guan(關) and jie(節) were examined within the Neijing, after which the meaning of guanjie was studied in reference to usages of guan and jie. Other terms used similarly to guanjie were also studied. Results & Conclusions : 1. The meaning of guanjie in the Huandineijing could be inferred to meanings such as 'vital point and node', 'node of entrance gate', 'entrance gate and node', 'bolt and node', 'node of bolt', etc. 2. Most usages of guanjie in the Huangdineijing referred to 'pivotal node', while those mentioned in relation to edema or joint pain from wind and dampness meant 'node of entrance gate.' 3. The four entrances, eight nodes, extremity node, eight hollows, eight nodes of the limbs, twelve nodes, which are used similarly to guanjie, manage the five zang, communicate intimately via qi, while shen qi or zhen qi enter and exit. 4. Guanjie is closely linked to the five zang, while it is the entrance gate or pivotal node through which shen qi roams and travels. Therefore when interpreting a verse that includes 'guanjie', one must be aware of the perspectives of the Huangdineijing on qi, keeping in mind the close relationship with the five zang, and that zhen qi enters and exits.

조후용신(調候用神)을 활용한 월별(月別) 장부간(臟腑間)의 호오관계 (Study on the Fancy Relationship of Viscera under the Diao-hou-yong-shen)

  • The human body and nature respond to each other. The change of nature influences to directly human body. One of the change is the weather change. and there is order in the change. The order is twelve months. To explain the weather change of the cold, hot. dry and wet. we can use Four seasons, The twenty-four seasons, or The twelve month. To explain the five elements of the chinese cosmogony, The four season is useful, and the change of weather, the twenty-four seasons is good. But, the viscera is explained by the ten celestial stems and the twelve horary sings. Thus the twelve horary sings is most useful to explain, because one year is composed by twelve months. The twelve season and fancy of the ten celestial stems is explained fully by Diao-hou-yong-shen. Diao-hou-yong-shen is expressed by the zodiac signs, and explained by the disposition of the zodiac sings. In addition to the oriental medical science is also explained and expressed by the zodiac signs. Therefore, I think that the change of viscera, according to weather, is based on the Diao-hou-yong-shen, and I get some results. In the acting of viscera, there are some relationship between organs. The relationship is not influenced directly by the weather of four seasons, unless there are some special details. If the energy of the month, which rules one specific organ, is weak, doctor should fill up the organ. If not, the organ tends to get sick. According to the full or weak of the five elements which is under the month, the useless or harmful elements should be taken off, and useful and needful elements should be serviced. The balance between cold, hot, dry and wet should be maintained. However, the fancy of the viscera is very different. So, doctor should be careful to maintain balance. The full and weak between Yin and Yang should be considered when doctor think about cold and hot in the month.

『삼국유사(三國遺事)』의 환웅(桓雄)과 염제신농(炎帝神農)과 기록학적 비교고찰 (A Philological Comparative Study on HwanWoong of Samgukyusa and YanDi-ShenNong)

  • "삼국유사(三國遺事)", 권일(卷一)의 "고조선(古朝鮮)"조(條)를 신화와 역사기록 두 부분으로 나누어, 먼저 단군신화(檀君神話) 속의 환웅(桓雄)과 중국의 염제신농(炎帝神農)의 신격(神格)을 비교 고찰하였는데, 그들은 매우 유사한 신격을 가지고 있었다, 즉 인류의 생활과 농업에 필수적인 태양신(太陽神), 농경을 가르친 농업신(農業神), 그리고 약초(藥草)의 성질을 밝혀내어 질병을 예방하고 치료했던 의약신(醫藥神) 신격과, 문물을 발명하고 문화를 창시했던 문화영웅(文化英雄)의 성격도 가지고 있었다. 신화시대에서 역사시대로 발전해 가는 단계상에서 신(神)들이 담담했던 역할은 창세(創世) ${\rightarrow}$ 교화(敎化) ${\rightarrow}$ 통치(統治) 과정으로 변천해 가는데, 환웅과 염제신농은 모두 교화(敎化)의 단계에 있었고, 교화의 내용은 모두 그들의 신격과 관련된 것들이었다. 그러므로 환웅신화와 염제신농신화는 고대 한국과 중국의 정신문화 발전과 물질문명 발달과정 중 같은 단계에서 발생한 신화인 것을 알 수 있다. 다음은 역사기록 부분을 통하여 "위서(魏書)"의 본질과 단군 환웅의 시대를 고찰하였다. "위서에 이르기를 지금으로부터 2천년 전에 단군왕검이 있었다"라는 기록은 "위서"의 저술 시점(時點)을 밝히는 것이고 '위(魏)'는 위(魏)는 (위(衛))만조선(滿朝鮮)을 가리키는 것이다. 위만조선으로부터 2천여 년을 거슬러 올라가면 본문기록의 "당요동시(唐堯同時)"설(說)과 매우 근접한다. 그러므로 "위서"는 중국의 사서(史書)가 아니라 위만조선의 역사서일 가능성이이 크다. 본문의 "1500년 동안 나라를 다스렸다"와 "기자를 조선에 봉하였다"는 단군 건국년대를 밝히는 중요한 기록이다. 기자동래(箕子東來)는 은주교체(殷周交替) 때(B.C 1122년)로, 단군이 이때까지 1500년 간 나라를 통치하였으므로 고조선 건국년대는 B.C 2622년이다. 이는 요(堯)임금보다 훨씬 이전인 왕제(黃帝)(B.C 2698-2358) 시대에 해당한다. 그러므로 단군의 부신(父神)인 환웅의 사대는 아무리 늦어도 B.C 3000년 전후일 것인 바, 이는 염제신농(B.C 3218-2699) 시대에 해당되므로 환웅과 염제신농이 교화활동을 한 것도 동시대였음을 알 수 있다. 이같이 "삼국유사 고조선"편은 우리민족의 상고시대(上古時代) 역사를 규명해주는 기록학적으로 의의가 깊은 매우 귀중한 자료이다.
