남해안 일부해역 해수의 세균학적 위생안전성 평가 (Evaluation of the Microbiological Safety for the Seawater in a Part of the South Coast, Korea)

  • 이 연구는 패류가 다량으로 양식되고 있는 남해안 일부해역 해수의 세균학적 위생안전성을 평가하기 위하여 조사해역 내에 영향을 줄 수 있다고 판단되는 해수 14곳을 선정하여 2014년 3월부터 10월까지 총 6차례 조사를 실시하였으며 그 결과는 다음과 같다. 해수는 수온이 평균 $21.2^{\circ}C$, 염분농도는 32.43‰, pH는 7.92, 용존산소는 7.83 mg/L으로 조사되어 우리나라 수질환경기준으로 볼 때 1등급으로 조사되었다. 전도도는 44,600 us/cm, 탁도는 1.93 NTU로 조사되었다. 남해안 일부 조사해역 해수의 위생학적 안전성을 조사한 결과 대장균군은 평균 < 1.8~790 MPN/100 mL, 90번째 값이 168.1 MPN/100 mL, 기하학적 평균이 12.8 MPN/100 mL으로 나타났고, 분변계대장균은 < 1.8~790 MPN/100 mL, 90번째 값이 50.5 MPN/100 mL, 기하학적 평균값이 6.5 MPN/100 mL로 조사되어 정착성 수산물 생산해역 위생관리기준 중 청정해역의 기하학적 평균 기준치인 14 MPN/100 mL를 넘지 않아 청정해역으로 나타났다. 그러나 5월과 7월에 43 MPN/100 mL를 초과하는 시료가 35.7%와 28.6%로 KSSP 기준치인 10%를 초과하여 계절에 따라서 다소 차이가 있는 것으로 나타났다. 조사해역은 폐쇄성 내 만으로 적은 오염원에도 크게 영향을 받을 수 있으므로 계절에 따라서 철저한 해역의 관리가 요구되었다.

남해 창선해역의 해수 및 진주담치의 위생학적 안전성 평가 (Microbiological Quality of the Shellfish-growing Waters and Mussels in Changseon, Namhae, Korea)

  • A sanitary survey was conducted to evaluate the water quality and mussel (Mytilus edulis) conditions of two administrative shellfish growing waters: those designated as shellfish growing water for export, and adjacent waters on the east coast of Changseon Island, Namhae, Korea. In all, 1,656 seawater and 166 mussel samples were collected at 46 stations for seawater and five stations for the shellfish from January 2007 to December 2009. Both seawater and mussels were examined for total coliforms and fecal coliforms. The standard plate count and most probable number of Escherichia coli were also determined for the shellfish samples. The range of the geometric means and the estimated 90th percentiles of fecal coliform for seawater samples at each station were <1.8-4.1 MPN/100 mL and <1.8-22.3 MPN/100 mL, respectively. The sanitary conditions for both shellfish growing areas, the designated shellfish growing area and adjacent area in Changseon, met the 'Approved area' criteria of the United States National Shellfish Sanitation Program. The range of E. coli detected in mussels in these areas was <20-500 MPN/100 g; and only 3 of 166 samples exceeded 230 MPN/100 g, the European standard for the consumption of raw bivalves. Therefore, the mussel farms in the Changseon area are classified as 'Class A' according to criteria set in European Community regulations. However, the levels of E. coli in mussels in parts of the designated shellfish growing area and adjacent area exceeded the bacterial limits after rainfall exceeding 45 mm. Further studies are needed to fully define the conditions leading to a temporary closure to harvest after a rainfall event.

시판 냉동굴(Crassostrea gigas)의 위생 안전성 평가 (Safety Evaluation of Frozen Oyster Crassostrea gigas as a Raw Material for Seafood Products)

  • This study was conducted to investigate the suitability of frozen oysters as a raw material for the preparation of seafood products by measuring the concentrations of harmful microorganisms and chemicals in thawed flesh. The microbial concentrations in thawed oysters were 2.3-5.0 log CFU/g for viable cell counts, not detected (ND)-1.0 log CFU/g for coliform bacteria, and ND for Escherichia coli and pathogenic bacteria such as Staphylococcus aureus, Salmonella spp., Listeria monocytogenes, Vibrio parahaemolyticus, Enterohemorrhagic Escherichia coli (EHEC), and Clostridium perfringens. In frozen oysters, the heavy metal concentration for viable cell counts was ND-0.030 mg/kg, for lead was ND-0.393 mg/kg, and for cadmium was 0.021-0.597 mg/kg. Benzo(a)pyrene, shellfish poison (paralytic shellfish and diarrhetic shellfish poisons), and radioactivity were not detected in the thawed oysters. These results suggest that frozen oysters can be safely used as a raw material for the preparation of seafood products.

Applicability Evaluation of Male-Specific Coliphage-Based Detection Methods for Microbial Contamination Tracking

  • Outbreaks of food poisoning due to the consumption of norovirus-contaminated shellfish continue to occur. Male-specific (F+) coliphage has been suggested as an indicator of viral species due to the association with animal and human wastes. Here, we compared two methods, the double agar overlay and the quantitative real-time PCR (RT-PCR)-based method, for evaluating the applicability of F+ coliphage-based detection technique in microbial contamination tracking of shellfish samples. The RT-PCR-based method showed 1.6-39 times higher coliphage PFU values from spiked shellfish samples, in relation to the double agar overlay method. These differences indicated that the RT-PCR-based technique can detect both intact viruses and non-particle-protected viral DNA/RNA, suggesting that the RT-PCR based method could be a more efficient tool for tracking microbial contamination in shellfish. However, the virome information on F+ coliphage-contaminated oyster samples revealed that the high specificity of the RT-PCR- based method has a limitation in microbial contamination tracking due to the genomic diversity of F+ coliphages. Further research on the development of appropriate primer sets for microbial contamination tracking is therefore necessary. This study provides preliminary insight that should be examined in the search for suitable microbial contamination tracking methods to control the sanitation of shellfish and related seawater.

양식산 굴(Crassostrea gigas)의 생굴 및 가공소재용으로서 화학적 및 생물학적 위생 특성 (Chemical and Biological Properties on Sanitary of Cultured Oyster Crassostrea gigas Intended for Raw Consumption or Use in Seafood Products)

  • Oysters Crassostrea gigas are a globally popular shellfish for human consumption. As filter-feeding bivalve mollusks, oysters may harbor many microorganisms and chemicals that could pose potential human health risks. The objective of this study was to investigate the suitability of cultured oysters for raw consumption or use in seafood products by measuring concentrations of harmful microorganisms and chemicals in their flesh. Microbial concentrations in cultured oysters were found to be: $1.0{\times}10^2-6.0{\times}10^4CFU/g$ (viable cell counts), not detected $(ND)-5.4{\times}10^3CFU/g$ (coliform bacteria), $ND-1.3{\times}10^2CFU/g$ (E. coli), and $ND-4.6{\times}10^3CFU/g$ (Vibrio parahaemolyticus). Other pathogenic bacteria, including Enterohemorrhagic E. coli (EHEC), Listeria monocytogenes, Staphylococcus aureus, and Salmonella spp., were not detected in any samples. Heavy metal concentrations of cultured oysters were ND-0.239 mg/kg (total mercury), ND-1.091 mg/kg (lead), ND-0.968 mg/kg (cadmium). The concentrations of benzo(a)pyrene ranged from $0.280-0.880{\mu}g/kg$. Paralytic shellfish poison ranged from ND-0.58 mg/kg, while diarrhetic shellfish poison was not detected. No radioactivity was detected. These results suggest that oysters intended for raw consumption or use in seafood products should be subjected to chemical and biological controls.

진해만 서부 용남·광도해역의 세균학적 수질에 미치는 육상 오염원의 영향 (Impact of Pollution Sources on the Bacteriological Water Quality in the Yongnam-Gwangdo Shellfish Growing Area of Western Jinhae Bay, Korea)

  • To evaluate the bacteriological water quality in Yongnam-Gwangdo, located in western Jinhae Bay, seawater samples were analyzed using sanitary indicator bacteria at 57 sampling stations. According to survey results from January 2007 to December 2009, the range of the geometric mean and the estimated 90th percentile for coliforms and fecal coliforms in the samples were <1.8-16.5 and 1.8-246.8 MPN/100 mL and <1.8-7.1 and 1.8-74.8 MPN/100 mL, respectively. The samples, including those taken from stations located in Wonmunman, Gwangdo, and Dangdong, showed high levels of microbial contamination caused by the climate and weather patterns in the marine environment. The bacteriological water quality in the area met Korean criteria for a designated shellfish growing area for export and National Shellfish Sanitation Program criteria for an approved shellfish growing area, except at station #49. A total of 24 direct pollution sources were discharged into the shellfish growing area. The radius of impact was calculated for each pollution source to assess the effect on the shellfish growing area. The calculated radius of impact for most of the pollution sources was below 300 m. However, the radius of impact for the combined pollution sources in Kyeonnaeryang was 93-1973 m. There were significant differences between the calculated closed sea area and actual monitoring results. The closed sea area values calculated from the fecal coliform load in drainage water tended to be higher than the actual monitoring results. Tidal currents and environmental factors such as salinity, water temperature, sunlight, and microbiological factors affect the survival of fecal indicator bacteria in seawater.

강우에 따른 적양항과 장포항 육상오염원이 창선해역에 미치는 세균학적 영향평가 (The Effects of Inland Pollution Sources around the Port of Jeokyang and Jangpo after Rainfall Events on Bacteriological Water Quality in the Changseon Area, Korea)

  • The influences of inland pollution sources nearJeokyang and Jangpoports following rainfall events on the bacteriological water quality and safety of commercial shellfish were investigated in the Changseon area, Korea. Stream flow rates exhibited 1.9- to 5.7-fold increases after rainfall events and then fell to 68~81% of that level after 24 h. The calculated impact area of inland pollution sources was 0.47 $km^2$ in the Jeokyangport area and 0.27 $km^2$ in the Jangpoport area at 24 h following 11 mm of rainfall. When the flow rate of inland pollution increased, the level of male-specific bacteriophage (MSB) decreased, and no MSB could be detected in seawater samples, whereas 30 PFU/100 g was detected in shellfish samples. Fecal coliform levels in seawater and shellfish samples did not exceed their respective tolerance levels (4.5 MPN/100 mL) and 130 MPN/100 g, respectively and thus complied with the standard for approved shellfish growing area. The sanitary conditions of areas adjacent to Jeokyang and Jangpoports under conditions of rainfall below 29 mm met the criteria for approved growing area under the United States National Shellfish Sanitation Program and the EC Regulations.

경남 한산·거제만해역에서 생산된 패류의 위생학적 안전성 평가 (Evaluation of Sanitary Safety for Shellfish in Hansan·Geojeman, Korea)

  • 본 연구는 2013년부터 2017년까지 경남 한산 거제만 해역에서 생산되는 패류에 대한 위생지표세균, 마비성 패류독소, 설사성 패류독소를 분석하여 세균학적 독물학적 안전성을 평가하였다. 굴 시료 404점에 대한 분변계대장균은 < 18~330 MPN/100 g의 범위를 나타내었으나, 대장균 230 E. coli MPN/100 g을 초과한 시료는 없었다. 굴 수확시기 동안 분석한 대장균의 기하학적 평균치는 24.3 MPN/100 g으로 매우 낮게 나타났다. 한산 거제만해역에서 생산되는 굴의 세균학적 안전성을 평가한 결과, 식품위생법의 생식용 굴에 대한 위생기준을 만족하였고, EU의 패류생산해역 A등급 기준에 부합하였다. 한산 거제만해역의 독물학적 평가를 위해 굴 532개 시료와 패류독소 지표종인 지중해담치 268개 시료에 대한 분석을 실시하였다. 마비성패류독소는 2013년 4월에 지중해담치 3개 시료에서 1.20~2.29 mg/kg 범위로 기준을 초과하여 검출되었으나, 굴시료에서는 전혀 검출되지 않았다. 총 120개 시료에서 설사성패류독소를 분석하였으나, 정량한계 이하로 매우 낮게 나타났다. 독물학적 안전성 평가결과, 굴 시료에서는 마비성 및 설사성패류독소가 전혀 검출되지 않았으나, 지표종인 지중해담치에서는 마비성패류독소 기준치를 초과한 것으로 나타나 지속적인 모니터링과 안전관리가 요구되었다.

자란만.사량도 패류생산해역의 위생학적 안전성 평가 (Evaluation of the Bacteriological Safety for the Shellfish Growing Area in Jaranman.Satyangdo Area, Korea)

  • Bacteriological examination of a 12,700 ha area within Jaranman Saryangdo area, located in Jaran bay and Saryangdo island, was conducted with 35 fixed (designated areas and adjacent areas) seawater sampling stations between January 2006 and December 2008. According to results, the geometric mean range and the estimated 90th percentile range of fecal coliform counts in sea water samples collected in the designated area were <1.8-2.4 and <1.8-8.6 MPN/100mL, respectively. The estimated 90th percentile range of fecal coliform counts in sea water samples collected from 6 sampling stations in the adjacent areas were 6.1-34.6 MPN/100mL. Based on these results, bacteriological water quality of the designated areas in the Jaranman. Saryangdo area meet the NSSP (National Shellfish Sanitation Program) guidelines for approved area. The bacteriological sea water quality in Jaranman.Saryangdo area has been shown to be favorable at all investigated sampling stations except for July and August which coincided with heavy rainfall. In fact, the bacteriological water quality was not affected if rainfall was less than 30 mm rainfall. However, the degree of bacteriological contamination increased rapidly and the water quality exceeded approved NSSP guidelines set for rainfall above 60 mm.

가막만해역 패류의 세균학적·독물학적 안전성 평가 (Evaluation of the Bacteriological and Toxicological Safety for the Shellfish Growing Area in the Kamakman Area, Korea)

  • 본 연구는 2012년부터 2016년까지 전남 가막만해역에서 생산되는 패류에 대하여 위생지표세균, 마비성 및 설사성패류독소를 분석하여 세균학적, 독물학적 안전성을 평가하고자 하였다. 총 194점의 굴 시료에서 분변계대장균과 대장균을 분석한 결과 모두 230 MPN/100 g을 초과하지 않았다. 굴 수확시기 동안 분석한 분변계대장균의 기하학적 평균치는 19.6 MPN/100 g으로 비수확시기(26.5 MPN/100 g) 보다 안정적인 것으로 나타났다. 가막만해역의 독물학적 평가를 위해 굴 77개 시료와 패류독소 지표종인 지중해담치 350개 시료에 대한 분석을 실시하였다. 마비성패류독소는 4월말부터 6월초 사이에 지중해담치 13개 시료에서 $40{\sim}46{\mu}g$/100 g 범위로 매우 낮게 검출되었으며, 굴 시료에서는 전혀 검출되지 않았다. 설사성패류독소는 총 180개 시료 중 2개 시료에서 검출되었으나, 기준치에 미치지 못하는 것으로 확인되었다. 따라서 가막만해역에서 생산되는 패류의 세균학적 안전성을 평가한 결과, 식품위생법의 패류 위생기준에 적합하였고, EU의 패류생산해역 A등급 기준에 부합하는 것으로 확인되었다. 또한 마비성 및 설사성패류독소는 기준치보다 매우 낮은 수준으로 검출되어 독물학적 안전성이 확보된 패류생산해역으로 평가되었다.
