Breeding of Tetraploid in Platycodon grandiflorum (Jacq.)A. DC. by Colchicine treatment

  • Present studies were carried out to produce tetraploid plants by colchicine treatment using seeds, seedlings and shoot tips of Platycodon grandiflorum in Campanulaceae. The most successful colchicine treatment for tetraploid production in P. grandiflorum was soaking treatment using 0.01 and 0.5% colchicine solution for 1 hour and 12 hours, respectively. Morphological characteristics of both diploid and tetraploid were similar, but tetraploid plants had more leaves. Compared to diploid, tetraploid had the larger stomata, but less number of stomata. Fresh weight of tetraploids was 20∼40% heavier than that of diploid.

  • When reactivity insertion such as refueling occurs in CANDU reactors, the power and the water levels are tilted in the upper outer zone of the LZCS (Liquid Zone Control System) and fluctuate unstably for a certain period of time (1-5 days). The instability described above is observed in most CANDU reactors in service around the world, but its root cause is unidentified and no solutions to this problem have been established. Therefore, this study attempted to prove experimentally and analytically that the root cause lies in the hold-up of light water on the top of the TSP (Tube Support Plate) due to the mismatch between net volumetric flow rate of light water and helium crossing the narrowed porous TSP installed within the LZCS compartment. Our method was to perform a hydrodynamic simulation of in/outflow of light water and helium. Two solutions for the aforementioned instability of LZCS are suggested. One is to regulate the compartment for both inflowing helium gas and outflowing light water; the other is to enlarge the flow paths of helium and light water within TSP. The former may be applicable to nuclear reactors in service and the latter to those planned for construction.

  • This study was intended to suggest HACCP-based standard recipe for bellflower roots, classified as no thermal cooking process, served in a university foodservice operation. The time-temperature and microbial contamination level in each cooking step were analyzed. The temperatures of bellflower root, peeled garlic and green onion at receiving were at 13.8$\pm$2.8, 12.6$\pm$2.9 and 13.7$\pm$$0.8^\circC$ respectively, which were above the temperature limit. The time consumed for pre-preparation was up to 90 min at room temperature having high microbial growth potential. The levels of total plate counts (TPC) of bellflower root and garlic were over the limit of $10^6$ CFU/g as were the numbers of coliforms in bellflower roots. There were no microbial reductions in pre-preparation and cooking, which resulted in over $10^5$- $10^6$ CFU/g of TPC at service step. Two CCPs identified were washing/sanitation at pre-preparation and service steps. The control measures were washing/sanitation and temperature control. It was verified that CCPs for no cooking process developed in preceding studies were applicable for the microbiological food safety of this menu item. The HACCP-based standard recipe was developed to produce a quantity for 100 servings by observing the critical limits established for CCPs. These results suggest that the selection of proper provider is imperative to control the microbial contamination of raw materials at purchasing step. Also, the sanitary education program should be developed for the employees to understand and comply the HACCP plan and standard recipe.

뿌리기업 자동화·스마트 공정을 위한 Plug-in 구조의 IOT 미들웨어 구축 방법 (Structure Method for IOT Middle Ware with Plug-in module for Automation & Smart processing of Ppuri Manufacturing Factory)

  • 우리나라 산업의 근간인 뿌리기업의 자동화 스마트 공정상 IOT 미들웨어는 사물인터넷 기기(센서 등)의 데이터 정보를 해석하고 관리하며 제어할 수 있는 중추적 역할이 요구된다. 특히 뿌리산업은 산업별 공정 흐름이 상이하고 기업별 데이터 처리요구도 다양해 이를 수용하기 위한 범용 IOT 미들웨어가 필요한 상황이다. 본 논문이 제시한 IOT 미들웨어 구조는 그에 대한 해결책으로 통신, 데이터 수집과 처리, 서비스 연계 등 미들웨어 기본 프로세스는 엔진부로 범용화 시키고 뿌리산업 분야별 사물인터넷 기기와 기업별 처리 요구사항은 개별적으로 대응할 수 있는 Plug-in 구조를 도입함으로서 유연하고 효과적인 뿌리산업 스마트 공정을 제안하였다. 또한 미들웨어 Plug-in과 연계 서비스 계층 간 네트워크 데이터에 대한 암호화를 통해 변조, 탈취 등에 대한 예방과 보안을 강화하는 방안을 제시하였다. 향후 MQTT, COAP, XAMP 등 다양한 네트워크 프로토콜에서 확장 가능할 수 있도록 뿌리산업에 특성화된 IOT 미들웨어 플랫폼으로 발전시킬 시스템을 제안하고자 한다.

  • 서비스 인적자원 운용의 효율성 제고와 부품 또는 시설 할당의 적정성 향상을 위해 서비스센터를 통해 접수되는 서비스 요청 건수를 보다 정확하게 예측하고자 하는 필요성이 제조업을 중심으로 증가하고 있다. 본 연구에서는 제품의 특성을 반영하여 제품수명주기 별로 제품들을 군집화하고 군집 별로 적절한 예측모형을 구축한 후 예측 값을 통합하는 개별예측방식을 LCD 모니터 제조사의 사례를 통해 제시한다. 또한 예측 결과를 총량방식 및 기존에 기업이 사용하고 있는 방식과 비교. 평가하여 우수성을 증명함으로써 제품이나 산업의 특성을 반영한 맞춤형 수요예측 기법 도입의 필요성을 부각하고, 그에 따른 이론적, 실무적 가이드라인을 제공한다.

Effect of Photoperiod, Temperature and True-leaf Stage in Bolting Rate of Chicory (Cichorium intybus L. var. sativus)

  • Root chicory (Cichorium intybus L. var. sativus) is potential alternative medicinal and sugar crop which accumulates a high amount of linear polyfructan, inulin in its roots. A problem in root production is that over-wintered stock plants often flower. Once the plant becomes reproductive, stem elongation and root growth slows and floral buds arise from every node, rendering the plants useless for propagation. The objectives of this research was to examine the effectiveness of manipulating environmental factors containing photoperiod, temperature and number of leaf states. The experiment was performed in growth chamber to create two photoperiods (8 h, and 16 h) with three temperature regimes (5$^{\circ}C$/3$^{\circ}C$, 1$0^{\circ}C$/8$^{\circ}C$ and 15$^{\circ}C$/13$^{\circ}C$ day/night temperature) for a total of six treatments on three type of true-leaf stage of plant. Data of bolting rate, shoot and root length, shoot and fresh weight was invetigated in each treatments. This is the first report on changes in bolting rate and shoots and roots production during a whole growing season and differences in the effect of cold and photoperiod treatment depending on the true-leaf stage of plant.

Nitrate enhances the secondary growth of storage roots in Panax ginseng

  • Background: Nitrogen (N) is an essential macronutrient for plant growth and development. To support agricultural production and enhance crop yield, two major N sources, nitrate and ammonium, are applied as fertilizers to the soil. Although many studies have been conducted on N uptake and signal transduction, the molecular genetic mechanisms of N-mediated physiological roles, such as the secondary growth of storage roots, remain largely unknown. Methods: One-year-old P. ginseng seedlings treated with KNO3 were analyzed for the secondary growth of storage roots. The histological paraffin sections were subjected to bright and polarized light microscopic analysis. Genome-wide RNA-seq and network analysis were carried out to dissect the molecular mechanism of nitrate-mediated promotion of ginseng storage root thickening. Results: Here, we report the positive effects of nitrate on storage root secondary growth in Panax ginseng. Exogenous nitrate supply to ginseng seedlings significantly increased the root secondary growth. Histological analysis indicated that the enhancement of root secondary growth could be attributed to the increase in cambium stem cell activity and the subsequent differentiation of cambium-derived storage parenchymal cells. RNA-seq and gene set enrichment analysis (GSEA) revealed that the formation of a transcriptional network comprising auxin, brassinosteroid (BR)-, ethylene-, and jasmonic acid (JA)-related genes mainly contributed to the secondary growth of ginseng storage roots. In addition, increased proliferation of cambium stem cells by a N-rich source inhibited the accumulation of starch granules in storage parenchymal cells. Conclusion: Thus, through the integration of bioinformatic and histological tissue analyses, we demonstrate that nitrate assimilation and signaling pathways are integrated into key biological processes that promote the secondary growth of P. ginseng storage roots.

  • This paper examines the service ideal preached by Bruce Barton, a bestselling American author in the 1920s. Based on archival sources and other historical materials, this paper presents Barton's endorsement of service ideal less as an adaptation to the looming consumerist society than an endeavor to revive the old producer ethos that was more coercive than therapeutic. Revisiting Barton's messages to Americans, this paper traces the intellectual root of Barton's service ideal to the Social Gospel and illustrates the ways his producer ethos evolved into a commonplace ideology of Christian meritocracy that endorsed individualism and small government.

Bacterial Mixture from Greenhouse Soil as a Biocontrol Agent Against Root-Knot Nematode, Meloidogyne incognita, on Oriental Melon

  • The biological control efficacy of a greenhouse soil bacterial mixture of Lactobacillus farraginis, Bacillus cereus, and Bacillus thuringiensis strains with antinematode activity was evaluated against the root-knot nematode Meloidogyne incognita. Two control groups planted in soil drenched with sterile distilled water or treated with the broad-spectrum carbamate pesticide carbofuran were used for comparison. The results suggest that the bacterial mixture is effective as a biocontrol agent against the root-knot nematode.

  • 실내에서의 위치기반 서비스 및 상황인식 서비스를 위해서는 실내에서의 정밀한 위치인식 기술이 필수적이다. 신호원과 여러 개의 신호 수신기와의 거리 차이를 이용하여 신호원의 상대위치를 추정하는 TDOA(Time Difference of Arrival) 기술이 사용되고 있는데, 거리 차이 측정의 오차에 따른 위치 추정 오차가 발생하므로 이를 최소화하기 위한 연구가 많이 진행되어 왔다. 본 논문에서는 신호원과의 거리 차이값을 이용하여 계산된 여러 근중에서 실제의 근을 선별하고 이를 평균내어 신호원의 위치를 추정하는 알고리즘이 제안되었으며, 기존의 방법에 비해 오차가 개선되었다.
