SELDI-TOF MS Combined with Magnetic Beads for Detecting Serum Protein Biomarkers and Establishment of a Boosting Decision Tree Model for Diagnosis of Pancreatic Cancer

  • Aim: New technologies for the early detection of pancreatic cancer (PC) are urgently needed. The aim of the present study was to screen for the potential protein biomarkers in serum using proteomic fingerprint technology. Methods: Magnetic beads combined with surface-enhanced laser desorption/ionization (SELDI) TOF MS were used to profile and compare the protein spectra of serum samples from 85 patients with pancreatic cancer, 50 patients with acute-on-chronic pancreatitis and 98 healthy blood donors. Proteomic patterns associated with pancreatic cancer were identified with Biomarker Patterns Software. Results: A total of 37 differential m/z peaks were identified that were related to PC (P < 0.01). A tree model of biomarkers was constructed with the software based on the three biomarkers (7762 Da, 8560 Da, 11654 Da), this showing excellent separation between pancreatic cancer and non-cancer., with a sensitivity of 93.3% and a specificity of 95.6%. Blind test data showed a sensitivity of 88% and a specificity of 91.4%. Conclusions: The results suggested that serum biomarkers for pancreatic cancer can be detected using SELDI-TOF-MS combined with magnetic beads. Application of combined biomarkers may provide a powerful and reliable diagnostic method for pancreatic cancer with a high sensitivity and specificity.

Biochemical Indices of Vitamin E, Ascorbic Acid and Iron Status : Relation to Diet, Supplement Use and Other Lifestyle Variables in Urban and Rural (Amish) Populations

  • The relation of food and supplemental intake of iron, vitamin E and ascorbic acid and other lifestyle variables to packed cell volume (PCV) and serum vitamin levels was studied in urban and rural (71% Amish) communities. Subjects were interviewed (24-h dietary recalls) on three occasions over 18-months, and blood samples were taken (maximum observations = 442). Mean PCV was lower in rural males (43.3) than in urban males (45.4) despite higher man food iron intake (18.7 and 14.4 mg/day, respectively). Mean meal iron availability was higher at lunch and lower at breakfast and dinner for rural than for urban subjects. Smoking was the number one variable in males and females explaining variance in PCV. Supplemental vitamin E and ascorbate intakes explained the most variance in serum vitamin E and ascorbate levels, respectively. Serum vitamin E was also associated with supplemental ascorbate intake (r=0.29). Serum ascorbate was also associated with food ascorbate intake (r=0.28) and body weight (r=-0.24).

Bioavailability of Digoxin Tablets in Healthy Volunteers

  • The bioavailability of digoxin generic tablets manufactures in Korea (formulations A & B) wwere compared to a standard (formulation C; Lanoxin brand digoxin, Burroughs Wellcome, USA) in 12 healthy Korean male volunteers (mean age 31.4 years) in a single dose, randomized, complete block crossover study. Using a latin square design, each of the subjects was randomized to the order number and allocated to each of the three treatments of 0.5mg oral digoxin. Digoxin conc4ntrations in serum and urine samples collected for 48 hours after dosing were measured by fluoprescence polarization immunoassy and radioimmunoassy, respectively. Treatments were compared by using nonlinear least squares regession analysis to evaluate the following pharmacokinetic parameters : maximum serum concentation $(C_{max})$; time of maximum serum concentation $(T_{max})$; area under the serum concentration-time curve $AUC_{0-12}$, $C_{max}$\;and\;(AUC_{0-12})$; and cummulative urinary excretion for 0-48 hours $(CLE_{0-48}.\;Mean\;AUC_{0-12},\;C_{max},\;and\;CUE_{0-48}$ values for formulations B and C were significantly different from formulation A (P<0.001), but not significantly diffeerent form each other. Basede on $AUL_{0-12}\;and\;CUE_{0-48}$ respectively, the relative availability of formulation B was 87.5% and 89.6% and the relative availability of formultation A was 43% and 35% when compared to formulation C(the standard).

  • Serum chemical values were determined in blood samples obtained from 179 normal, healthy Jindo dogs of both sexes that ranged in age from 3 months to 10 years. Female dogs had higher total protein and albumin values than their male counterparts. Age-related differences were detected for serum proteins, inorganic phosphate, glucose, and blood urea nitrogen levels. The GGT level Was within the normal range in dogs.

Effect of Family Type on the Nutrient Intake and Nutritional Status in Elderly Women (가족형태에 따른 여자 노인의 영양소 섭취 및 영양 상태에 대한 연구)

  • This study was performed to investigate the effect of family type on the nutrient intake and nutritional status in elderly women. Blood samples and anthropometric data of elderly women aged from 60 to 78 years were collected in 1996, who visited a health examination center in Seoul to screen their health status. They were divided into two groups by their family type, one was extended family(extended family elderly, n=37) and the other was independently living(independently living, n=29). Nutrients intake was measured by interview using a simplified questionaire. Albumin, cholesterol, triglyceride(TG), hemoglobin (Hb), hematocrit, Ca, Mg, and alkaline phosphatase activity of serum were analyzed. Intakes of energy, protein, carbohydrate, Fe, vitamin A, niacin, thiamin, riboflavin, and vitamin C were not different between the two groups. However, fat and Ca intakes were higher(p<0.05) in extended family elderly than indepen dently living elderly. There are no significant difference in hematocrit value and serum concentrations of Hb, TG, and Ca. Serum Mg and cholesterol levels tended to be higher in extended family elderly. However, serum albumin level was lower(p<0.05) in extended family elderly. These results show that the indepen dently living elderly women had intakes of daily energy, fat and Ca compared to the extended family elderly. Therefore, the nutritional education for the independently living elderly will be focused on the health risk resulted from less intakes of Ca and total energy.

  • A reverse-phase high performance liquid chromatography method with electrospray ionization and detection by mass spectrometry is described for the simultaneous determination of doxifluridine and its active metabolite 5-flu-orouracil (5-FU) in monkey serum. The method has greater sensitivity and simpler process than previous published methods with good accuracy and precision. A proper liquid/liquid extraction was used to extract simultaneously doxifluridine and 5-FU which has considerable difference in the polarity. Extracts were analyzed using LC/MS/MS providing a short analysis time within 5 min. The lower limit of quantification was validated at 10.0 ng/ml of serum for both doxifluridine and 5-FU. Accuracy and precision of quality control (QC) samples for both analytes met FDA Guidance criteria of ±15% for average QC accuracy with coefficients of variation less than 15%. The method will be applicable for preclinical studies and bioequivalence studies.

The Zinc Nutriture and Effect of Zinc Supplementation on the Elderly Living in Ulsan (울산지역 노인의 아연 영양상태 및 아연 보충의 효과)

  • This study was intended to examine the zinc status and effect of zinc supplementation on the zinc nutritional status of the elderly living in the Ulsan area. The zinc intake of 207 subjects(male 97, female 110) was measured by a 24-hour dietary recall and food frequency method. Biochemical analysis were conducted from blood and urine samples to evaluate the changes of zinc nutriture with zinc supplementation. The average dietary zinc intake of subjects was $7.7\pm{2.8mg}$ for male and $7.5\pm{2.6mg}$ for female, which were 51.3% and 62.3% of Korean RDA respectively. The first source of zinc was cereal and grain(36%), and the second was eggs and milk group(27%). After 8 weeks of zinc supplementation, the serum zinc content was significantly increased(p<0.01), although the serum copper content was not significantly decrease. Serum HDL- cholesterol level was not significantly decreased with zinc supplementation. Serum alkaline phosphatase(ALP) activity and urinary zinc excretion were significantly increased(p<0.05). The urinary Zn/Cr was not significantly increased. It is suggested from the results that the daily zinc supplementation can be effective to improve zinc nutriture.

A Study on the Zinc Concentration in Serum and Hair of Korean Female Collegians (한국 여대생의 혈청과 두발중 아연 함량에 관한 연구)

  • Due to the fact that blood gathering is not readily accepted by people, it has been difficult to make a full-scale study on zinc through blood analysis. For that reason, in this paper, a correlation of zinc contents in serum and hair was studied to find out whether only hair, which is relatively easy to collect, can be used for a measurement of zinc content. The subjects were 47 healthy female collegians selected at random whose average age was 22, mean height 159.1cm and average weight 49.7kg. The mean zinc value in serum of subjects was ${107.1}\pm{29.0}{\mu}g{/100ml}$, while the mean value in hair was ${150.2}\pm{42.7}ppm$. Correlation analysis shows that there was a significant correlation in zinc contents of serum and hair ( p<0.01). The results shows that measuring zinc level by using only hair samples would be satisfactory. But the use of hair sample for a measurement of zinc level has one demerit. That is, the method is more likely to be subjected to contamination because it makes more complicated procedures in sample preparation than the method of using blood sample.

Development of monoclonal antibody capture ELISA for the detection of antibodies against transmissible gastroenteritis virus

  • Transmissible gastroenteritis (TGE) is a disease confined to pigs of all ages, and can be a significant cause of economic loss in breeding herds, primarily because of the very high piglet mortality. The causative agent is a coronavirus, an enveloped positive strand RNA virus and closely related but non-enteropathogenic porcine respiratory coronavirus (PRCV). Although the TGEV has declined with its innocent relative, PRCV, further genome changes could not be excluded. Therefore, the herd-level immunity against this virus is important for the prevention of disease and should be carefully monitored. The aim of this study is to develop monoclonal antibody capture enzyme-linked immunosorbent assay (MAC-ELISA) which can rapidly and accurately determine a large numbers of serum samples for surveillance purpose, and to compare the ELISA with a TGEV-specific serum neutralization test. The MAC-ELISA was sufficiently achieved, and the comparison with the virus-specific serum neutralization assays for 713 sera from pig farms showed a high correlation ($r^2=0.812$, P<0.001). The specificity and sensitivity of MAC-ELISA for the serum neutralization test 91.9% and 91.6%, respectively, which means that the antibody detected by the MAC-ELISA could be said to be protective antibodies. In conclusion, the developed MAC-ELISA would be very helpful in evaluating protective antibodies against TGEV.

Investigation of serum biomarkers for neuropathic pain in neuromyelitis optica spectrum disorder: a preliminary study

  • Background: We aimed to investigate candidates for serological biomarkers of neuropathic pain in individuals with neuromyelitis optica spectrum disorder (NMOSD). Methods: We analyzed 38 sera samples from 38 participants with NMOSD in National Cancer Center. Neuropathic pain was evaluated using the painDETECT questionnaire. Pain with neuropathic components (painDETECT score ≥ 13) was observed in 22 participants, among whom 17 had definite neuropathic pain (painDETECT score ≥ 19). The remaining 16 participants had non-neuropathic pain (painDETECT score < 13). Serum glial fibrillary acidic protein (GFAP) levels were assessed using a single-molecule array assay. Several cytokines, including tumor necrosis factor-alpha (TNF-α), interleukin (IL)-6, IL-10, and IL-17A, were measured by a multiplex bead-based immunoassay. Results: In comparison of NMOSD participants with neuropathic pain components (or definite neuropathic pain) and those with non-neuropathic pain, the absolute values of serum GFAP, TNF-α, IL-6, and IL-10 levels were higher in participants with neuropathic pain components (or definite neuropathic pain), but these findings did not reach statistical significance. Conclusions: Further larger-scale investigations to find reliable serological biomarkers for neuropathic pain in NMOSD are warranted.
