ICR 마우스를 이용한 삼소음 및 발효삼소음의 급성독성 연구 (Acute toxicity on Samsoeum and fermented Samsoeum in ICR mice)

  • Objectives : This study aims to investigate the acute oral toxicity and safety of Samsoeum (Shensuyin) extracts and fermented Samsoeum extracts. Methods : For that objective, we used ICR mice. ICR mice were administerd orally with dosage of 1250mg/kg, 2500mg/kg and 5000mg/kg of Samsoeum extracts and fermented Samsoeum extracts. Results : We daily examined number of deaths, clinical signs, body weights and gross findings for 2 weeks. 1. The results of acute oral toxicity using ICR mice showed that LD50 of value over 5000 mg/kg. 2. Samsoeum extracts and fermented Samsoeum extracts not affect on bodyweight gross findings of ICR mice. 3. The results of Serum chemistry analysis and Complete Blood Count(CBC) through the autopsy were showed normal range values. Conclusions : Samsoeum extracts and fermented Samsoeum extracts did not show any toxic effects in ICR mice. And oral LD50 value was over 5000mg/kg in ICR mice and it is very safe for ICR mice.

One-Step Simultaneous Immunochromatographic Strip Test for Multianalysis of Ochratoxin A and Zearalenone

  • Individual immunochromatographic assays (ICG) for ochratoxin A (OTA) and zearalenone (ZEA) were optimized and used in the development of a one-step simultaneous immunochromatographic assay (OS-ICG) for the rapid multianalysis of two mycotoxins in corn samples. The nitrocellulose membrane of the OS-ICG was treated with OTA-bovine serum albumin (BSA), ZEA-ovalbumin (OVA), and anti-mouse IgG in the OTA test, ZEA test, and control zones, respectively. Monoclonal antibody-gold conjugates (OTA3 MAb-gold and ZEA2C5 MAb-gold) were sprayed onto the conjugate pad. The visual detection limits were 2.5 and 5 ng/ml for OTA and ZEA, respectively, and the results were obtained within 15 min after starting the analysis. An efficient, simple, and rapid extraction method using 30% MeOH/PBS was established and validated by analyzing the corn samples spiked with OTA/ZEA mixtures (0/0, 5/10, 10/20, and $20/30\;{\mu}g/kg$). The cut-off values of the OS-ICG for the spiked corn were 5 and $10\;{\mu}g/kg$ for OTA and ZEA, respectively. Natural corn samples were analyzed by OS-ICG, direct competitive enzyme-linked immunosorbent assay (DC-ELISA), and HPLC. Results of the OS-ICG were in good agreement with those obtained by DC-ELISA and HPLC. The developed OS-ICG offers a rapid, easy-to-use, and portable analytical system and can be used as a convenient qualitative tool for the on-site simultaneous determination of OTA and ZEA in cereals, food, and agricultural products in one analytical cycle.

흑연로 원자흡수분광법에 의한 혈중 카드뮴 분석시 매트릭스 개선제(matrix modifier)의 역할 (The role of matrix modifier and for the determination of cadmium in blood by graphite furnace atomic absorption spectrometry)

  • 본 연구에서는 혈액 중 저농도의 카드뮴 분석을 위하여 시료를 1% Triton X-100으로 5배 희석시킨 다음에 흑연로 원자흡수분광법을 이용하여 분석하였다. 매트릭스 개선제로서 2% $(NH_4)_2HPO_4$와 L'vov platform이 삽입된 pyrocoated 흑연로를 사용하여 비교적 높은 회화온도 ($600^{\circ}C$)에서 카드뮴의 손실을 최대한 억제시키면서, 혈액내의 방해 성분으로 인한 영향을 감소시켰다. 분석의 정확성을 평가하기 위하여 NIST 표준물질(혈청 중 Cd) 및 외부 분석 기관(일본)에서 분석한 결과를 비교 검토하였다. 이 실험에서 얻은 상대 표준편차는 1.0ng/ml 농도에서 10% 정도이며 검출한계값은 0.1ng/ml였다.

THI 52 Inhibits Inducible Nitric Oxide Synthase Gene Expression in RAW 264.7 Cells and Rat Lung Tissue by Lipopolysaccharide

  • Previously we reported that THI 52 inhibits tumor necrosis factor $(TNF)-{\alpha}$ mRNA expression in mouse peritoneal macrophages exposed to LPS plus $IFN-{\gamma}.$ In the present study, the effects of THI 52 on vascular reactivity ex vivo, and iNOS protein expression (rat lung) were investigated in LPS-treated rats. Treatment of THI 52 concentration-dependently reduced not only serum nitrite production but also the expression of iNOS protein in rat lung tissues. Thoracic aorta taken from LPS injected rat for 8 h ex vivo resulted in suppression of vasoconstrictor effects to phenylephrine (PE), which was restored by THI 52 (20 mg/kg) 30 min prior to LPS. When measured iNOS activity, treatment of THI 52 concentration-dependently reduced the enzyme activity in RAW 264.7 cells activated with LPS plus $IFN-{\gamma}.$ Likewise, iNOS activity was significantly reduced in lung tissues taken those rats that were injected THI 52 prior to LPS injection compared with LPS injection alone. These results strongly suggest that THI 52 can suppress iNOS gene expression induced by LPS, and restore the vascular contractility to PE. Thus, THI 52, a new synthetic isoquinoline alkaloid, may be beneficial in inflammatory disorders where production of NO is excessed by iNOS expression.

개에서 발생한 구리 축적성 간경화에 D-penicillamine, SAMe, DBB로 병용 치료하여 장기간 생존한 1례 (Long-term Management of Copper-associated Hepatic Cirrhosis with D-penicillamine, SAMe, and DBB in a Dog)

  • 4살령의 아메리칸 코가 스파니엘이 식욕부진, 몸떨림 그리고 복부 팽만증세로 내원하였다. 혈청학적 검사, 방사선 검사, 초음파 검사 그리고 간생검을 통한 조직병리학적 검사를 통해 경화로 진행된 만성 간염으로 진단할 수 있었으며, 특수 염색법을 통해 대부분의 간세포내에 구리가 축적되어 있는 것을 확인할 수 있었으며, 구리 농도는 1460 ppm으로 매우 높음을 알 수 있었다. 이러한 검사 결과를 바탕으로 구리 축적으로 인한 간염이 간경화까지 진행된 말기 병변으로 진단하고 D-penicillamine, s-adenosymethionine, biphenyl-dimethyl-dicarboxylate과 대증 치료로 본 환자를 치료 하였으며, 본 환자는 진단 후 35개월을 생존하는 것에 성공하였다.

척추측만증이 있는 개에서 안구후방마취 후 발생된 안구심장반사 1례 (Oculocardiac Reflex Caused by Retrobulbar Block in a Dog with Scoliosis)

  • Enucleation of a 9-year-old, spayed female Pekingese's right eye was scheduled because of recurrent eyeball rupture caused by chronic corneal ulcer and descemetocele. Scoliosis of the thoracic vertebra was observed on thoracic radiography. Complete blood count, electrolytes, serum chemistry profiles were within normal ranges except of alkaline phosphatase, which was markedly improved, compared with that of five months ago. Severe respiratory sinus arrhythmia was observed before induction of anesthesia, it disappeared after induction. Retrobulbar block was performed with 0.5% bupivacaine, 2% lidocaine, 0.1% epinephrine combination (4 : 1 : 0.2 ratio of volume) before start of surgery. After retrobulbar block, heart rate decreased from 110 to 76 beats/min and sinus arrhythmia recurred. It was considered as oculocardiac reflex caused by increase of intraorbital pressure from retrobulbar block, atropine (0.025 mg/kg, IV) was administered and intermittent positive pressure ventilation was started. Three minutes after atropine administration, abnormal waveform of the electrocardiograph was appeared, it suspected as ventricular or supraventricular tachycardia, so lidocaine (2 mg/kg, IV) was administered. Then, heart rate was maintained around 130 beats/min, and the surgery was finished without the other problems. Although, retrobulbar block is performed to provide analgesia and to prevent oculocardiac reflex in ophthalmic surgery, occasionally it could induce oculocardiac reflex by infiltrated volume.

개에서 난관 선암종, 질 섬유종 및 자궁축농증 병발 1예 (Concurrence of Uterine Tube Adenocarcinoma, Vaginal Fibroma, and Pyometra in a Dog)

  • 12년령 암컷 혼혈견이 외음부에 돌출된 질 종양으로 내원하였다. 이 환자는 정상적으로 변과 오줌을 배출하였고 임신 경험은 없었다. 혈청생화학적인 소견상 질소혈증, 경미한 고나트륨 혈증과 alkaline phosphatase 값의 상승이 있었다. 혈액학적 검사 소견은 정상이었다. 초음파 검사에서 좌 우측 자궁각의 내강에 존재하는 액체로 인하여 무에코성에서 저에코성으로 나타났고 심하게 종대되어 있었다. 또한, 우측 난관의 종양 내에 국소적으로 저에코성 영역이 나타났다. 컴퓨터단층촬영에서는 우측 난소낭, 액체로 인해 확장된 자궁, 확장된 질과 질의 종양이 관찰되었다. 본 환자에 대해 난소자궁적출술과 회음부 절개술을 통한 종양절제술을 실시하여 난관과 질에 존재하는 종양을 제거하였다. 본 환자는 난관 선암종, 질 섬유종, 자궁축농증이 병발한 첫 번째 사례로 여겨진다.

마우스에 대한 Surfactin C의 급성경구독성시험 (Acute Oral Toxicity of Surfactin C in Mice)

  • This study was carried out to investigate the acute toxicity of surfactin C in mice. Surfactin C was administered orally at does of 0, 381, 610, 977, 1562 and 2500 mg/kg. Number of deaths, clinical signs, body weights, feed and water consumptions, and biochemical examinations were investigated for 14 days after single oral administration of surfactin C. $LD_{50}$ value was over 2500mg/kg in mice. In addition, no differences were found between control and treated groups in clinical signs, body weight gains, hematology, serum chemistry, feed and water consumptions. The results indicate that surfactin C did not show any toxic effects at 2500 mg/kg in mice.

Streptozotocin 유발 당뇨 흰쥐에서 Tabebuia avellandae의 항당뇨 효과 (Hypoglycemic Effect of Tabebuia avellandae on Streptozotocin-Induced Diabetic Rats)

  • Hypoglycemic effect of Tabebuia avellandae was investigated in the streptozotocin(STZ)-induced diabetic rats. Diabetes was induced in male Sprague-Dawley rats by injections of STZ (45 mg/kg, i.v.). Rats weighing 200-250 g were divided into 6 groups: normal, STZ-control, hexane fr., CHCl$_3$fr., BuOH fr. and $H_2O$ fr. group. Normal and STZ-control rats received 3% Tween 80 only. Four groups of diabetic rats were administered orally at doses of 100, 400, 300 and 400 mg/kg/day of hexane, CHC1$_3$, BuOH and $H_2O$ fr. respectively. Fractions were administered orally to the rats for 7 days after STZ injection. All rats were anesthetized with ether, blood samples were taken by cardiac puncture for clinical chemistry and the rats were killed by exsanguination. Liver, kidney, heart and spleen were removed, weighed and analyzed. We measured glucose, protein, cholesterol and triglyceride levels in the plasma and glycogen, cholesterol and triglyceride levels in liver. The extent of blood glucose decrement in rats administered $H_2O$ fraction was greater than that in the STZ-control rats. The serum cholesterol and triglyceride levels were significantly lowered by administration of $H_2O$ fraction compared with those of STZ-control group. Treatment of rats with Tabebuia avellandae fractions caused decreases in STZ-induced elevation of cholesterol and triglyceride. Liver triglyceride level was significantly lowered hexane and BuOH fraction group compared with STZ-control group. These results suggest that $H_2O$ fraction of Tabebuia avellandae has the hypoglycemic action against STZ-induced diabetic rats.

칡소 송아지의 생후 2개월간 혈액화학치 양상에 관한 연구 (Blood chemistry profiles in indigenous Korean striped calves from birth up to the age of 2 months)

  • The present study was undertaken to clarify the changes in blood chemical values of indigenous Korean striped calves according to age. Blood samples were collected from 15 calves (8 females and 7 males) reared at the Gyeongsangbukdo Livestock Research Institute and the levels of various chemicals in serum were analyzed. The mean total protein, albumin and globulin concentration in calves increased from 2 weeks after birth, and the levels in females were higher than those in males at 8 weeks after birth. The mean glucose concentration increased significantly at 2 weeks after birth. The mean blood urea nitrogen concentration increased from 2 weeks after birth, whereas the mean creatinine concentration decreased significantly from 2 weeks after birth. The mean uric acid concentration increased gradually from 2 weeks after birth. The mean glutamic oxaloacetic transaminase and glutamic pyruvic transaminase concentration increased significantly from 2 weeks after birth. The mean total bilirubin concentration tended to decrease from 2 weeks after birth. The mean triglyceride, total cholesterol and high density lipoprotein cholesterol concentration increased significantly from 2 weeks after birth. In conclusion, the data obtained from this study may be valuable as a standard for interpreting results of biochemical analyses in indigenous Korean striped calves.
