Ultrastructural Study on the Effects of $^{60}Co$ $\gamma-irradiation$ on the seminiferous tubules in the Pheasant(Phasianus colchicus) ($^{60}Co$ 감마선 조사가 꿩의 정세관에 미치는 영향에 관한 전자현미경적 연구)

  • This study was undertaken to investigate ultrastructural changes according to the radiosensitivity in the spermatogenic cells and Sertoli cell of the seminiferous tubules in Korean native pheasants. During spermatogenetic period, testes were collected from male adult Korean native pheasant and they were used as experimental and control birds. The experimental group was divided into a single-dose whole body irradiation group (400, 600, 800 and 1,000 rads) and a split-dose whole body irradiation groups(400/2, 600/2, 800/2 and 1,000/2 rads). The experimental birds were sacrificed at 24 and 72 hrs after irradiation and the control pheasants were sacrificed at the same time. Ultrastructural changes of Sertoli cells and spermatogonia were investigated by ultrathin section with electron microscope. The results obtained are summarized as follows; 1. The apoptosis was observed after 72 hrs group of the single-dose irradiation of 400 rads. 2. The cytoplasmic organelles of spermatogonia were severely damaged more than that of sertoli cell in 72 hours group of split-dose irradiation of 800 rads. 3. The cytoplasmic organelles of Sertoli cell were severely damaged except the nuclear membrane of Sertoli cells in 72 hrs group of split-dose irradiation of 1,000 rads.

Diagnosis of immunohistochemical marker expressed by a canine Sertoli cell tumor case (개 세르토리세포종 케이스에서 면역조직화학적 마커를 통한 진단)

  • Sertoli cell tumor (SCT) of the testicle arises from the supporting cells within the seminiferous tubules. SCT is common in dogs, especially in cryptorchid testicles, but also has been reported in the stallion, ram, cat, and bull. Sertoli cell tumor sample was collected from 7-years male german shepherd. In this study, SCT arose from one testicle. Sample size is approximately 1.7 cm in diameter and it has a round form. In the microscopic, cells within the tumor variably resemble Sertoli cells (SCs) that normally populate the seminiferous tubules and interstitial area. There is abundant stroma of dense, mature fibrous connective tissue in SCT. In the immunohistochemical staining, cytokeratin AE1/AE3 was not expressed in the control and SCT. S-100 protein was expressed by SCs, germ cells and fibrous connective tissue of SCT. Melan A was expressed by leydig cells (LCs) of SCT. A study by using S-100 and melan A in canine SCT was almost never carried out. S-100 and melans A is considered to suggest for diagnosis and pathogenesis of canine SCTs. Inhibin-alpha and Vimentin were well known as the marekers of SCTs. Also, they were expressed by Sertoli cells and LSs of SCT in this study.

Gallic acid caused cultured mice TM4 Sertoli cells apoptosis and necrosis

  • Objective: The study was designed to determine the cytotoxic effect of gallic acid (GA), obtained by the hydrolysis of tannins, on mice TM4 Sertoli cells apoptosis. Methods: In the present study, non-tumorigenic mice TM4 Sertoli cells were treated with different concentrations of GA for 24 h. After treatment, cell viability was evaluated using WST-1, mitochondrial dysfunction, cells apoptosis and necrosis was detected using JC-1, Hoechst 33342 and propidium iodide staining. The expression levels of Cyclin B1, proliferating cell nuclear antigen (PCNA), Bcl-2-associated X protein (BAX), and Caspase-3 were also detected by quantitative real-time polymerase chain reaction and Western-blotting. Results: The results showed that 20 to $400{\mu}M$ GA inhibited viability of TM4 Sertoli cells in a dose-dependent manner. Treatment with $400{\mu}M$ GA significantly inhibited PCNA and Cyclin B1 expression, however up-regulated BAX and Caspase-3 expression, caused mitochondrial membrane depolarization, activated Caspase-3, and induced DNA damage, thus, markedly increased the numbers of dead cells. Conclusion: Our findings showed that GA could disrupt mitochondrial function and caused TM4 cells to undergo apoptosis and necrosis.

Lectine-binding patterns of spermatogenic cells in the Jindo dog (진도견 정자형성계 세포들의 Lectin-binding patterns)

  • The lectin-binding patterns in the testis of the sexually matured Jindo dog were investigated to study the distribution of glycoconjugates in the seminiferous tubule under light and transmission electron microscopy. Positive reactions to Wheat germ agglutinin(WGA) and Dolichos biflorus agglutinin (DBA) were observed in the Sertoli cell and in the residual body of spermatid with a stronger reaction in the Sertoli cell to the lectins than in the residual body. Strong reactions to Soybean agglutinin(SBA) and Peanut agglutinin(PNA) were observed in the acrosome vesicles of the Golgi- and cap-phase spermatid, while a moderate reaction was observed in the acrosome-phase, maturation-phase spermatid and the residual body. The acrosome area of the spermatid reacted intensively to Griffonia simplicifolia agglutinin( GS-I) when the cell was in the acrosome-phase and maturation-phase, and the same reaction to the GS-I was observed in the residual body. However, the seminiferous tubule did not react to Ulex europeus agglutinin I(UEA-I). The gold-labelling of the Sertoli cells with DBA resulted in positive reactions of the Sertoli cell column and processes when observed under the electron microscopy, while the Golgi-, cap- and acrosome-phase spermatids reacted positively to SBA in the peripheral low-dense area of the acrosome vesicle of spermatid. Based on these results, we concluded that differences in the lectin-binding pattern of the seminiferous tubules were recognized in the Jindo dog compared to other animals.

Malignant Sertoli cell tumor in Shih Tzu dog

  • Malignant Sertoli cell tumor was diagnosed in a 5-year-old male Shih Tzu dog. Clinical features of the dog were anorexia, urinary incontinence, constipation, anemia, alopecia, and epistaxis. The dog also had unilateral cryptorchid testis in the abdomen. Several abdominal and thoracic masses were identified on radiography. Grossly, the cryptorchid testicular mass was markedly enlarged to 8 cm in diameter. On cut surface, firm and well demarcated milk-white neoplastic areas were irregularly separated by white fibrous bands. Histologically, the testicular mass was diagnosed as tubular pattern Sertoli cell tumor. In addition, abdominal and mediastinal lymph nodes metastasis were found. Immunohistochemically, the tumor cells were strongly positive for vimentin and neuron specific enolase, but negative for S-100 and cytokeratin.

Testosterone/Estradiol Ratio in a Dog with Sertoli Cell Tumor-Seminoma Combination

  • A 8-year-old male Golden Retriever dog with bilateral cryptorchidism presented for evaluation of symmetric alopecia and gynecomastia. Abdominal radiography and computed tomography revealed bilateral enlargement of the testicles in the abdominal cavity. The concentrations of estradiol and testosterone in the blood were evaluated, and the results revealed a low testosterone/estradiol ratio despite a normal concentration of estradiol. For correction, cryptorchid testicles were surgically removed. Grossly, the bilateral retroperitoneal cryptorchid testicles were enlarged, firm, and spherical. On histopathologic examination, the right abdominal cryptorchid testicle was diagnosed as a Sertoli cell tumor (SCT)-Seminoma (SEM) combination. The contralateral testicle in the abdomen was diagnosed as a Sertoli cell tumor. The clinical signs of feminization were improved after surgery. To our knowledge, this is the first report of a case of simultaneous SCT on one side and SCT-SEM combination on the other side with sex hormone profiles in a dog.

Concurrent Occurrence of Seminoma and Sertoli Cell Tumor in a Dog with Ctyptorchidism and Monorchidism (개의 잠복고환된 단 고환에서 정상피종과 세르토리 세포종의 동시 발생)

  • A 9-year-old male poodle dog exhibited a white to yellow 3.0$\times$2.0$\times$3.9 cm mass in the left testicle with cryptorchidism and monorchidism. Grossly, necrosis and hemorrhage were observed on the cut surface of the mass. Histopathologically the mass consisted of seminoma and sertoli cell tumor (SCT). Seminoma was characterized with diffuse solitary pattern in which tumor cells had polygonal nuclei and clear cell border. SCT was composed of compact neoplastic cells with small nuclei and indefinite cytoplasm. The tumor was occupied mainly with SCT. SCT and seminoma were observed occasionally in the same lesion. From these results, this case was diagnosed as concurrent seminoma and SCT in the left cryptorchidism and monorchidism. To our knowledge, this is the first report of concurrent occurrence of seminoma and SCT in a dog with cryptorchidism and monorchidism in Korea.

Sertoli Cell Tumor Accompanied by Pancytopenia in a Dog

  • 11 year old male Yorkshire terrier was referred to Haemaru Referral Animal Hospital with signs of hematuria, petechia, and gynecomastia. Blood works revealed severe leukopenia, moderate anemia and severe thrombocytopenia. On ultrasonography and radiography, mixed echo texture mass was found in abdomen. The abdominal mass was surgically removed, and submitted for histopathology. Histopathologic features of the tissues were consistent with malignant Sertoli cell tumor. Bone marrow aspirates were hypocellular. Serum estrogen concentration was 72.80 pg/ml (normal range for females <15 pg/ml) after surgery. Clinical signs of feminization and hemorrhagic diathesis were attributed to hyperestrinism caused by the tumor. The dog was put on fluid therapy, antibiotics and palliative drugs and survived 2 more weeks after surgery without clinical improvement.

Testicular Cell Indices and Peripheral Blood Testosterone Concentrations in Relation to Age and Semen Quality in Crossbred (Holstein Friesian×Tharparkar) Bulls

  • Present study analyzed the changes in peripheral blood testosterone concentrations and testicular cytogram in relation to age and semen quality in crossbred males. Three different age groups of crossbred males viz. bull calves (6 months, n = 5), young bulls (15 months, n = 5) and adult bulls (4 to 6 years, n = 8) were utilized for the study. Testicular fine needle aspiration cytology technique was used to quantify testicular cytology and their indices. Peripheral blood testosterone concentrations were measured using enzyme-linked immunosorbent assay method. Semen samples collected from adult bulls were microscopically evaluated for quality parameters. Mean peripheral blood testosterone concentrations in bull calves, young bulls and adult bulls were $2.28{\pm}0.09ng/mL$, $1.42{\pm}0.22ng/mL$ and $5.66{\pm}1.08ng/mL$ respectively, and that in adult bulls were significantly different (p<0.01) from young bulls and bull calves. There was no significant difference between the proportion of different testicular cells in bull calves and young bulls. Between young and adult bulls, significant differences (p<0.01) were observed in the proportion of spermatocytes, spermatozoa, and sperm: Sertoli cell ratio. The proportions of Sertoli cells showed a significant difference (p<0.01) between the three age groups. The number of primary spermatocytes had a positive correlation with peripheral blood testosterone concentrations in bull calves (r = 0.719, p<0.01). Number of Sertoli cells per 100 germ cells was negatively correlated with blood testosterone concentration in young bulls (r = -0.713, p<0.01). Among different semen parameters in adult bulls, ejaculate volume (r = 0.790, p<0.05) had positive relationship, and sperm motility had significant negative correlation (r = -0.711, p<0.05) with testosterone concentrations. The number of Sertoli cells and Sertoli cell index had a positive correlation with various semen quality parameters (p<0.001). Results of the present study conclude that number of Sertoli cells and Sertoli cell index are good indicators of semen quality, but peripheral blood testosterone concentrations may not have a direct relationship with various seminal attributes in crossbred bulls.

Cytological analysis of pregnancy-associated plasma protein-A expression in porcine neonatal testis (미성숙 돼지 정소에서 pregnancy-associated plasma protein-A의 발현의 세포학적 분석)

  • The identification of biomarkers of a living tissues is essentially required to understand specific functions of the cells. In previous study, we reported IGFBP 3 as one of the putative biomarkers, by showing specific expression at porcine spermatogonial stem cells (SSCs) of early stage of porcine testis. In this study, we analyzed the expression of seven members of IGFBP family (IGFBPs) in SSCs and histological expression pattern of pregnancy-associated plasma protein-A (PAPP-A), which plays a role on the growth promoting enzyme by cleavage of IGFBPs in testis of 5 days old pig. RT-PCR analysis showed that IGFBP 1, 2, 3, 4, and 6 were expressed at high level specifically in porcine SSCs compared with whole testis. We performed immunohisotochemical staining of testis sections with PAPP-A and protein gene product 9.5 (PGP9.5) which are the known biomarkers for SSCs. We were not able to find co-expression of PAPP-A and PGP9.5; PAPP-A was expressed only in Sertoli cells and PGP9.5 expression was confirmed in spermatogonium. Additionally, we were able to confirm the GATA4 expression in Sertoli and Leydig cells as a regulator of Sertoli cell function was not detected PGP9.5 expressing cells, indicating indirect evidence of that cytolocalization of PAPP-A expression is limited in Sertoli cells. These results suggested that the PAPP-A expressed in Sertoli cells may play role on regulation of development and differentiation of testicular cells through the IGF axis in neonatal porcine testis.
