Characterization of a novel Cotesia vestalis polydnavirus (CvBV) gene containing a ser-rich motif expressed in Plutella xylostella larvae

  • Cotesia vestalis is an endoparasitoid of Plutella xylostella larvae and injects a polydnavirus (CvBV) into its host during oviposition. In this report we characterize the gene, CvBV3307, and its products. CvBV3307 is located on segment S33 of the CvBV genome, is 517 bp, and encodes a putative protein of 122 amino acids, including a serine-rich region. The expression pattern of CvBV3307 in parasitized larvae and the subcellular localization of CvBV3307 only in granulocytes indicated that it might be involved in early protection of parasitoid eggs from host cellular encapsulation and in manipulating the hormone titer and developmental rhythm of host larvae. Western blot analysis showed that the size of the immunoreactive protein (about 55 kDa) in parasitized hosts at 48 hours post parasitization (h p.p.) is much larger than the predicted molecular weight of 13.6 kDa, which suggests that CvBV3307 undergoes extensive post-translational modification in hosts.

치마버섯 Mating Locus(Y-region)의 비교분석에 관한 연구 (Studies on the Comparative Analysis of Mating Locus (Y-region) of Schizophyllum commune)

  • 북미산 S. commune UVMl-34의 A $\alpha$ 3 mating locus를 지니는 cosmid clone pSC13의 mating activity에 필요한 sequence 함유여부를 확인한 후 sequencing을 행하고 이에 대한 비교분석을 실시하였다. 그 결과 blast program을 사용한 전체 nucleotide 염기서열의 homology비교분석에서 남미산 1-71에 패하여 약 96%의 높은 homology를 나타내었으며, 1-71 A $\alpha$ 3 mating locus에서 모두 7개로 추정되는 exon의 염기서열의 비교실험에서도 거의 96% 이상에 이르는 homology를 나타내었다. 부분적으로는 AR (acidic rich region)에서 약 97%, HD (homeodomain)에서는 약 99%, BR (basic rich region)에서 약 97%, 그리고 Ser (serine rich regon)에서 약 95%의 높은 homology를 나타내어 지역간에 유전자상의 큰 변화는 나타나지 않는 것으로 확인되었다. 그리고 translated polypeptide sequence 를 이용하여 자생지가 다른 남미산 S. commune A $\alpha$ 3 mating locus 내의 Y-region은 비롯하여 다른 $A\alpha$ alleles내의 Y-region과 비교 분석한 결과 남미산에 대해 전체적으로 약 97%의 높은 homology를 나타내고 있지만 그 외 $A\alpha$ mating alllele gene의 Y-region과는 41~49%의 낮은 homology로 mating activity에 관여하고 있는 것으로 나타났다. 특히 mating에 있어 transcription regulator로 알려진 homeodomain에서는 약 98%의 homology가 나타남으로서 Z-region의 74%에 비하여 대륙간에 상당히 높은 유전자 보존상이 확인되었다. 또한 AR에서 97%, BR에서 100% 그리고 Ser에서도 98%의 상당히 높은 homology를 지니는 사실을 확인하였다. 이러한 결과로 보아 남미와 북미에 자생하는 같은 mating allele type간에는 상당히 높은 비율의 염기서열 보존이 이루어지고 있음을 알 수 있었으며, 비록 다른 $A\alpha$ alleles간의 비교 이지만 다른 mating alleles간의 약 50% homology와 비교할 때 보다 상당히 높은 결과로 보인다. 특히 homeodomain motif의 비교에서 Y1을 비롯한 다른 mating allele gene과도 85% 이상의 높은 homology를 나타내었으며 그 외 AR, BR, Ser 부위에서는 10~50%에 이르는 낮은 비율로 나타났다.

Description of Nearly Completed Mitochondrial Genome Sequences of the Garden Chafer Polyphylla laticollis manchurica, Endangered in Korea (Insecta: Coleoptera)

  • In this study, we present the nearly complete mitogenome sequences of the garden chafer, Polyphylla laticollis manchurica, which is listed as an endangered species in Korea. The P. l. manchurica mitogenome, which includes unfinished whole A+T-rich region and a partial srRNA was 14,473-bp long, possessing typical sets of genes (13 PCGs, 22 tRNA genes, and 2 rRNA genes). Gene arrangement of the P. l. manchurica mitogenome was identical to the common one found in the majority of insects. The 5 bp-long motif sequence (TAGTA) that has been suggested to be the possible binding site for the transcription termination peptide for the major-strand was also found in the P. l. manchurica mitogenome between $tRNA^{Ser}$(UCN) and ND1. The start codon for COI gene and ATPase8 was designated as a typical TTG. All tRNAs of the P. l. manchurica showed a stable canonical clover-leaf structure of other mt tRNAs, except for $tRNA^{Ser}$(AGN), DHU arm of which could not form stable stemloop structure. As has been previously determined, the high A/T content was unanimously observed in P. l. manchurica in terms of A/T bias in the third codon position (73.5%) compared with the first (66.4%) and second codon position (66.2%). The PCGs encoded in major-strands are slightly T-skewed, whereas those of the minor-strand are A-skewed, indicating strand asymmetry in nucleotide composition in the Coleoptera including P. l. manchurica.

Complete Mitochondrial Genome Sequence of the Yellow-Spotted Long-Horned Beetle Psacothea hilaris (Coleoptera: Cerambycidae) and Phylogenetic Analysis among Coleopteran Insects

  • We have determined the complete mitochondrial genome of the yellow-spotted long horned beetle, Psacothea hilaris (Coleoptera: Cerambycidae), an endangered insect species in Korea. The 15,856-bp long P. hilaris mitogenome harbors gene content typical of the animal mitogenome and a gene arrangement identical to the most common type found in insect mitogenomes. As with all other sequenced coleopteran species, the 5-bp long TAGTA motif was also detected in the intergenic space sequence located between $tRNA^{Ser}$(UCN) and ND1 of P. hilaris. The 1,190-bp long non-coding A+T-rich region harbors an unusual series of seven identical repeat sequences of 57-bp in length and several stretches of sequences with the potential to form stem-and-loop structures. Furthermore, it contains one $tRNA^{Arg}$-like sequence and one $tRNA^{Lys}$-like sequence. Phylogenetic analysis among available coleopteran mitogenomes using the concatenated amino acid sequences of PCGs appear to support the sister group relationship of the suborder Polyphaga to all remaining suborders, including Adephaga, Myxophaga, and Archostemata. Among the two available infraorders in Polyphaga, a monophyletic Cucujiformia was confirmed, with the placement of Cleroidea as the basal lineage for Cucujiformia. On the other hand, the infraorder Elateriformia was not identified as monophyletic, thereby indicating that Scirtoidea and Buprestoidea are the basal lineages for Cucujiformia and the remaining Elateriformia.
