살오징어(Todarodes pacificus)와 갑오징어(Sepia esculenta)에 대한 광대역 음향산란특성의 비교 (Comparison of Broadband Acoustic Scattering Characteristics Between Japanese Flying Squid Todarodes pacificus and Golden Cuttlefish Sepia esculenta)

  • Cephalopods, such as the Japanese flying squid Todarodes pacificus and golden cuttlefish Sepia esculenta, are an important food source for all toothed whales, particularly bottlenose dolphins in costal and offshore waters around the Korean Peninsula. A controlled laboratory experiment was conducted to investigate the broadband acoustic backscattering from live individuals of these two cephalopod species using linear chirp signals (100-200 kHz). The backscattered echo signal was measured at about 1° intervals from -45° (head down) to +45° (head up) in the dorsal plane. The species-specific, frequency-dependent scattering characteristics were investigated by comparing the relationship between the wavelength-normalized backscattering cross-section (σ/λ2) and the wavelength-normalized fish length L/λ for each species. The estimated σ/λ2 value for Japanese flying squid was 9.51 at an L/λ range of 12.79-30.27 (mean, 21.26). This was approximately 7% of the σ/λ2 value (136.1) for golden cuttlefish at an L/λ range of 9.07-25.49 (mean, 15.77).

오징어, 꼴뚜기류의 성장 및 섭이율과 이료의 전환효율 (On the Growth, Feeding Rates and the Efficiency of Food Conversion for Cuttlefishes and Squids)

  • Studies on the rate of growth, the rate of feeding and the efficiency of food conversion on the stage of new-born fries to the near adult size for three species of cuttlefishes , Sepia esculenta, Sepia subaculeata, Sepiella maindroni and two species of squids, Sepioteuthis lessomiana, Euprymna berryi were carried out in the process of artificial raising, and then argued about a feasibility of the propagation of cuttlefishes and squids. 1. The relation between the daily age (D) and the body weight(W) of Sepia exculent is expressed in a logarithmic equation, log W=3.0649 log D-4.2768. The daily rates of growth through 121 days of the raising period were 1.46 per cent for the man시 length and 1.67 percent for the body weight. The raipidest growth of Sepia esculenta is observed at the stage of 1 to 4 cm in the mantle length . At that time the daily rates of growth reach 3.3 to 5.5 percent for the mantle length and 10.4 to 12.0 percent for the body weight, respectively. The growth of Sepia esculenta varies a great deal to the bait. When fed on a dead bait the rates of growth decrease 17 per cent for the mantle length and 26 percent for the body weight compared with those fed on a live bait. 2. The relation between the daily age and the body weight of Sepia subaculeata is expressed in a logarithmic equation, log W=3.7447 log D-4.9003. The daily rates of growth through 110 days of the raising period were 1.63 percent for the mantle length and 1.83 percent for the body weight. The rapidest growth of Selia subaculeata is observed at the stage of 1.5 to 9.0 cm in the mantle length. At that time the daily rates of growth reach 3.1 to 7.4 percent for the mantle length and 6.8 to 16.7 percent for the body weight , respectively. 3. The relation between tehdaily age and the body weight of Sepiella maindroni is expressed in a ogarithmic equation , log W=2.9332 log D-3.8224 . The daily rates of growth through 133 days of the rearing period were 1.39 percent for the mantle length and 1.51 percent for the body weight . The rapidest growth of Sepiella maindroni is observed at the stage of 0.4 to 5.8 cm in the mantle length. At that time the daily rates for growth reach 4.6 to 7.3 percent for the mantle length and 8.5 to 15.4 percent for the body weight , respectively. 4. The daily rates of growth onthe stage of 0.5 to 6.0 comin the mantle length of Sepioteuthis lessoniana were 4.1 to 5.9 percent for the mantle length and 7.1 to 10.7 percent for the body weight . 5. During the rearing period of 31 days immediately after the hatching , the daily rateof feeding of Sepia esculenta marked 11.0 to 39.4 percent (28.2 percent in an average), and the efficiency of food conversion of this species reached 9.0 to 71.0percent (38.7percent in an average). Even at the more growing stage of 4.5to 6.2 cm in the mantle length, the daily rate of feeding of three species of cuttlefishes wee maintained 17.7 percent for Sepia esculenta, 30.8 percent for Sepia subaculeata and 34.7percent for Sepiella maindroni on an average. 6. The efficiency of food coversion of cuttlefishes and squids are larger than those of other fishes, and all the species are rapid in their growth. Four to five months are thought to be enough for their growing into a fair commercial size.

한국산 해산어류의 Anisakid유충 감염률 및 형태학적 분류 (Morphological Classification and Infection Rate of Anisakid Larvae in Marine Fishes)

  • Anisakiasis occur after the ingestion of raw marine fish and squid containg anisakid larvae. In recent years about 40 cases of anisakiasis have been recorded in Korea. Considering, however, the Korean style of eating raw fish, many more cases would exist and prevention is necessary. We describe the infection rate of anisakid larvae in 13 species of marine fish and squid which were caught in the Korean sea. And each extracted larva is classified according to morphological characters. The results are following ; Scomber japonicus, Pseudosciaena manchurica, Trichiurus haumela showed high infection rate of anisakid larvae. Although Sepia esculenta showed low infection rate, most of anisakid larvae found in Sepia edulis are embedded in muscles. So relative high frequent rate of anisakiasis may developed by Sepia esculenta. Five type (Anisakis Type I, Terranova Type B, Raphidascaris sp., Contracaecum Type A, Contracaecum Type D) of anisakid larvae are classified according to their morphological characters.

황해산 두족류의 가용성 단백질에 대한 연구(II) (Soluble Proteins Analysis of Class Cephalopoda in the Yellow Sea(II))

  • 황해산 두족류 6종(참갑오징어, Sepia esculenta ; 쇠갑오징어, Sepiella japonica : 한치꼴뚜기, Loligo chinensis : 살오징어, Todarodes pacificus : 참꼴뚜기, Loligo beka : 낙지, Octopus minor)의 안구단백질을 추출하여 항원-항체 교차반응을 실시하였다. 쇠갑오징어, 살오징어와 낙지의 안구단백질로 항혈청(antiserum)을 제작하였으며 갑오징어목, 살오징어목 및 낙지목에 속하는 두족류 6종의 안구단백질을 항원(antigen)으로 사용하였다. 갑오징어목 속하는 쇠갑오징어 및 참갑오징어에서 항원-항체 반응 양상이 매우 뚜렷하였으며 종간 비교에 기준이 될 수 있고 또한 유의성있는 침강선을 볼 수 있었다. 또한 gel filtration chromatography 방법으로 살오징어의 안구단백질을 분획하여 crystallin을 분리하였으며, 주 분획을 SDS-PAGE 전기영동 방법으로 분자량을 추정해 본 결과 약 20-35 KDa 였다.

갑오징어의 갑에 대한 모델 예측과 측정 반사강도의 비교 (Comparison of the Model-predicted and Measured Target Strength of Cuttlebones from Golden Cuttlefish Sepia esculenta)

  • The purpose of this study was to compare the model-predicted and experimentally measured target strength (TS) of golden cuttlefish Sepia esculenta cuttlebones. Ultrasonic signals used to estimate frequency-dependent TS and the speed of sound in cuttlebones were measured by pulse-echo and through-transmission techniques, using a chirp sonar system and an ultrasonic pulser/receiver system under controlled laboratory conditions. The model appeared to slightly underestimate the predicted TS values in the frequency range of 100-160 kHz. However, there was good agreement between the predicted and measured TS values in the frequency range of 160-200 kHz. The significant similarity between the model-predicted and experimentally measured TS values supports the use of the Kirchhoff-ray mode (KRM) model for acoustic scattering analysis of cuttlebones. Accordingly, we concluded that the KRM model can be used as a tool to evaluate the frequency-dependent variability of TS due to changes in golden cuttlefish swimming depth.

  • An 8-week feeding experiment was conducted to evaluate three types of squid Sepia esculenta liver powder (SLP) as a dietary protein source replacing fish meal (FM) in juvenile olive flounder Paralichthys olivaceus. To replace FM, six experimental diets were formulated with three types (A, B, C) of SLP at the 5 and 10%: SLP-A5, SLP-A10, SLP-B5, SLP-B10, SLP-C5, and SLP-C10. One control diet contained 100% FM as the main protein source and another was a commercial diet (Com). Fish with an average body weight of $22.8{\pm}0.4$ g ($mean{\pm}SD$) were allocated randomly to aquaria in groups of 20 fish and fed the experimental diets in triplicate to satiation. The weight gain (WG) and specific growth rate (SGR) of fish fed the SLP-C10 diet were lower than those of fish fed the FM and SLP-B5 diets. No significant difference was observed in the WG and SGR among fish fed the diets other than SLP-C10. The feed efficiency (FE) and protein efficiency ratio (PER) of fish fed each SLP diet did not differ from those fed the control diet. However, fish fed SLP-C5 and SLP-C10 had a lower FE and PER than the fish fed commercial, SLP-A5 and SLP-B5 diets. Each SLP diet except for SLP-C10 could replace up to 10% of FM for juvenile olive flounder. The results of this experiment provide information that will assist in formulating an inexpensive practical diet containing SLP for juvenile olive flounder.

Target Strength Measurements of Live Golden Cuttlefish Sepia esculenta at 70 and 120 kHz

  • Cuttlefish Sepia esculenta are commercially important in Korea. Assessments of their biomass currently depend on fishery-landings data, which may be biased. Towards fishery-independent acoustic surveys of cuttlefish, target strength (TS) measurements at 70 and 120 kHz were made of 23 live cuttlefish, in early May 2010. The fish were caught by traps in the inshore waters around Geojedo, Korea. The TS were measured using split-beam echosounders (Simrad ES60 and EY500, respectively). The cuttlefish mantle lengths (L) ranged from 15.6 to 23.5 cm (mean L=17.8 cm) and their masses (W) ranged from 335 to 1020 g (mean W=556.1 g). Their mean TS values at 70 and 120 kHz were -33.01 dB (std=1.39 dB) and -31.76 dB (std=2.15 dB), respectively. The mean TS at 70 kHz was 0.17 dB higher than the TS-length relationship resulting from a least-squares fit to the data ($TS=24.67{\log}_{10}L(cm)-64.03$, $r^2$ = 0.52, N=23). The mean TS at 120 kHz was 0.45 dB higher than the fitted TS-length relationship ($TS=40.59{\log}_{10}L(cm)-82.96$, $r^2$ = 0.58, N=23). The differences between the mean TS values and an equation regressed from all of the TS measurements at both frequencies ($TS=24.92{\log}_{10}L(cm)-4.92{\log}_{10}{\lambda}(m)-22.82$, $r^2$ = 0.86, N=46) was 0.22 dB at 70 kHz and 0.31 dB at 120 kHz, respectively.

Genetic Differences and Geographic Variation in Cuttle Fish (Sepia esculenta Hoyle)

  • The gDNA isolated from Korean cuttle fish (Sepia esculenta Hoyle) from Sockcho (SOCKCHO), Seocheon (SEOCHEON), Incheon (INCHEON) and Vietnamese cuttle fish (VIETNAM), were amplified by PCR. Here, the seven selected primers (BION-A07, BION-A09, BION-A11, BION-A20, BION-B04, BION-B06, and BION-B14) were used to generate the unique shared loci to each population and shared loci by the four cuttle fish populations. In this study, the primer BION-A11 detected 112 shared loci by the four populations, major and/or minor fragments of sizes 300 bp, 400 bp, 700 bp and 1,000 bp, respectively, which were identical in all samples. The dendrogram obtained by the seven primers indicates five genetic clusters: cluster 1 (SOCKCHO 01-SOCKCHO 07), cluster 2 (SEOCHEON 08-SEOCHEON 10), cluster 3 (SEOCHEON 11-SEOCHEON 14), cluster 4 (INCHEON 15-INCHEON 21), and cluster 5 (VIETNAM 22-VIETNAM 28). The shortest genetic distance that displayed significant molecular differences was between individuals 25 and 26 from the Vietnamese cuttle fish (0.025), while the longest genetic distance among the twenty-eight cuttle fishes that displayed significant molecular differences was between individuals SOCKCHO no. 02 and SEOCHEON no. 12 (0.640). Individual of Seocheon and Incheon cuttle populations was somewhat closely related to that of Vietnamese cuttle fish population. Even though it could not be affirmed by a single case, such a result seems to be closely connected that the Korean peninsula is subject to climate changes by global warming. In conclusion, our PCR analyses revealed a significant genetic distance among the four cuttle fish populations.

Fish with an average body weight of $6.50{\pm}0.03g$ ($mean{\pm}SD$) were allocated randomly in triplicate groups of 50 to aquaria and fed the experimental diets until satiation. The weight gain (WG) and specific growth rate (SGR) of fish fed the FM, SLP-$A_5$, SLP-$A_{10}$, and SLP-$B_5$ diets were higher than those of fish fed the Com, SLP-$B_{10}$, SLP-$C_5$ and SLP-$C_{10}$ diets. No significant differences were observed in WG and SGR among the diet groups, with the exception of the SLP-$C_{10}$ diet group. The feed efficiency (FE) and protein efficiency ratio (PER) of fish fed the SLP-$A_5$, SLP-$A_{10}$, and SLP-$B_5$ diets did not differ from those fed the control diet. However, fish fed SLP-$B_{10}$ and SLP-$C_{10}$ had lower FE and PER than the fish fed the FM, commercial, SLP-$A_5$, SLP-$A_{10}$, and SLP-$B_5$ diets. Both the SLP-A and SLP-B diets, but not the SLP-C diets, replaced up to 10% and 5% of FM for juvenile black rockfish, respectively. The results of this experiment provide information that will assist in formulating an inexpensive and practical diet containing SLP for juvenile black rockfish.

  • 황해서식 두족류 (class Cephalopoda)의 가용성 단백질에 대한 연구를 위해, 인천 및 목포 연근해에서 채집된 두족류 3목 5종의 (오징어목 : 참갑오징어 (Sepia esculenta) 및 쇠갑오징어 (Sepiella japonica), 살오징어목 : 한치꼴뚜기 (Loligo chinensis) 및 참꼴뚜기 (Loligo beka), 문어목 : 낙지 (Octopus minor)의 안구단백질, 근육단백질 및 간조직을 추출하여, 각종 전기영동 (Davis-PAGE 및 SDS-PAGE, Exponential gradient SDS-PAGE, 등전점 전기영동, 2차원 전기영동) 방법에 의한 단백질 분리 양상을 통해 두족류 종사이의 유전적 근연관계를 분석하였다. 시료의 안구 및 근육 단백질에 대한 exponential gradient SDS-PAGE 전기영동 결과 대략 분자량 35-50 KDa 사이에서 단백질 분리 양상의 차이를 볼 수 있었으며, 등전점 전기영동 방법(IEF)에 의해서는 pI값 7.5-8.5 사이에서 종간 특이성을 갖는 단백질 분리 양상을 볼 수 있었다. 특히 유의성이 있다고 판단된 시료의 안구 단백질을 2차원 전기영동 방법에 의해 분리 해 본 결과 대부분 분자량 30-50 KDa 사이에 분포하고 있어 exponential gradient SDS-PAGE 전기 영동에 의한 결과와 일치함을 알 수 있었다.
