Thermotoga neapolitana 유래 내열성 4-알파-글루칸전이효소의 효소적 특성 (Enzymatic Characterization of a Thermostable 4-α-Glucanotransferase from Thermotoga neapolitana)

  • Thermotoga neapolitana 유래 내열성 4-알파-글루칸전이효소(MgtA)가 산업적 응용성을 지닌 싸이클로아밀로스를 생산할 수 있는지를 검사하기 위하여 그 유전자를 클로닝하고 대장균에서 발현시켰다. MgtA는 HiTrap Q와 Sephacryl S-200 분배 크로마토그래피를 이용하여 순수한 형태로 정제되었으며, SDS-PAGE를 통하여 분자량이 약 52 kDa으로 아미노산서열로부터 계산된 분자량과 일치하였다. 효소활성의 최적 pH와 온도는 6.5와 $85^{\circ}C$였으며 알파1,4결합을 갖는 글루칸의 1,4결합을 효율적으로 가수분해함과 동시에 작은 크기의 올리고당을 말토덱스트린에 전이하는 전이활성을 가지고 있었다. 그러나 플루란, 글리코겐 및 1,4결합 이외의 다른 알파결합을 갖는 글루칸에는 활성을 나타내지 않았다. MgtA는 말토트리오스를 말토올리고당으로 전환할 수 있는 능력에서 그렇지 못한 Thermotoga maritima 의 효소와 구별할 수 있었으며, 반응 후 glucoamylase의 처리결과로부터 그 전이산물이 싸이클로아밀로스 대신에 긴 연쇄상의 말토올리고당을 생산하는 효소로 확인할 수 있었다.

Overproduction, Purification, and Characterization of Bacillus stearothermophilus Endo-xylanase A (XynA)

  • By using a T7 expression system, a large amount of Bacillus stearothermophilus endo-xylanase A (XynA) could be produced in Escherichia coli cells. The overproduced enzyme formed inclusion bodies, and so the protein could be more easily purified to homogeneity. The molecular weight of the purified enzyme was estimated to be 22 kDa by SDS-polyacrylamide gel electrophoresis and 43 kDa by Sephacryl S-200 gel filtration, suggesting that the native enzyme was a homodimer. The pI value was determined to be 8.4. The Michaelis constants for birchwood xylan and oat spelts xylan were calculated to be 3.83 mg/ml and 5.03 mg/ml, respectively, and the $V_{max}$ max/ values for both xylans were 2.86 $\mu mole$/min. The purified enzyme was most active at $55^{\circ}C$ and pH 8.0, and stable up to $60^{\circ}C$ and in the near neutral pH range. From the zymogram, Bacillus stearothermophilus was found to have at least three xylanases and the purified one was the smallest among them.

  • A ${\beta}$-ketothiolase was purified 180-fold from the cell extracts of Alcaligenes sp. SH-69 by a series of chromatography on DEAE-Dephadex A-50, Sephacryl S-200, and hydrozyapatitie columns, The optimum pH values of the partially purified enzyme were 7.5 for condensation reaction and 8.3 for thiolysis reaction were estimated to be 0.12mM and $18.7\;{\mu}M$, respectively. The $K_m$ valued for acetoacetyl-CoA and free CoASH in the thiolusis in the condensation reaction was 0.70mM. The condensation reaction of the ${\beta}$-ketothiolase was inhibited even by low concentrations of free CoASH($K_i=30.4{\mu}M$). Pretreatment of the enzyme with NADH and NADPH markedly inhibited the thiolysis reaction of the enzyme. The potent inhibition of the enzyme by sulfhydryl reagents suggests the involvement of cystein residue in the active site.

  • Protein phosphatase 2C (PP2C) is one of the four major serine/threonine phosphatases which is dependent on $Mg^{2+}$ for its activity. PP2C was purified from rat liver cytosol and its characteristics were investigated. The substrate employed for routine assay was $[^{32}P]casein$ phosphorylated by PKA. The purification process involved DEAE chromatography, ammonium sulfate fractionation, phenyl sepharose chromatography, sephacryl 5-200 gel filtration, and histone agarose chromatography. The SDS-PAGE of PP2C showed one major single protein band at a position corresponding to a molecular mass of 43 kd and the purification fold was 637. The enzyme showed a pH optimum of 8 and $K_M$ value was $1.9\;{\mu}M$. However, when the substrate was changed to $[^{32}P]histone$, the pH optimum was shifted to 7 and $K_M$ value was $2.3\;{\mu}M.\;Mg^{2+}$ was essential to the enzyme activity and okadaic acid did not exert any inhibitory effect on the enzyme. To examine residue in the active site of PP2C effects of some protein-modifying reagents were tested.

  • Acetylxylan esterase II was produced by Escherichia coli HB101 harboring the recombinant plasmid pKMG7 which contained the estII gene of Bacillus stearothermophilus. Optimal medium for the production of the acetylxylan esterase by E. coli HB101/pKMG7 was determined to contain 0.5% galactose, 1% yeast extract and 1% NaCl. The enzyme produced was purified to homogeneity using a combination of 20-50% ammonium sulfate precipitation, DEAE-Sepharose CL-6B chromatography and Sephacryl S-200 gel filtration. The temperature and pH optimum of the esterase were 45$\circ$C and pH 6, respectively. The essential amino acids for the esterase activity were found to be methionine, serine, and cysteine. Molecular weight of the esterase was determined to be 28 kDa by SDS-polyacrylamide gel electrophoresis, and 120 kDa by gel filtration. This suggests that the functional enzyme is a homomeric tetramer. The esterase had an isoelectric point of pH 3.4. The N-terminal amino acid sequence of the enzyme was Ala-Leu-Phe-Glu-Ser-Arg-Phe-Phe-Ser-Glu-Val-Leu-Gly-Leu.

Partial characterization of a 17 kDa protein of Clonorchis sinensis

  • A 17 kDa protein from Clonorchis sinensis adults was purified by a procedure including Sephacryl S-200 HR gel filtration and Q-Sepharose anion exchange chromatography. The protein was proved to be a cysteine protease as it showed hydrolytic activity toward Cbz-Phe-Arg-AMC in the presence of dithiothreitol and was inhibited by specific inhibitors such as iodoacetic acid or trans epoxy-succinly-L-leucyl-amido(4 guanidino) butane. The polyclonal antibody raised against the protein reacted to 17 kDa proteins of trematodes such as Paragonimuf westermani, Fasciola hepatica, Opisthorchis viverrini, Gymnophalloides seoi, and Metagonimus yokogawai. The antibody recognized the 17 kDa and 16 kDa cysteine proteases purified from C. sinensis, P. westemani, and G. seoi as well. These results suggest that the 17 kDa protein may be a cysteine protease commonly present in trematodes.

Purification and Properties of Laccase of the White-rot Basidiomycete Coriolus hirsutus

  • Laccase produced by Coriolus hirsutus was purified to electrophoretic homogeneity by acetone precipitation, Sephacryl S-2000 HR chromatography, DEAE Sepharose CL-6B chromatography, and Mono Q HR 5/5 chromatography. The purification of laccase was 46.6-fold with an overall yield of 23.7%. Laccase from this fungus was a monomeric glycoprotein with 16% carbohydrate content, and has an isoelectric point of 4.2, and molecular mass of 78 kDa, respectively. The N-terminal amino acid sequence of the enzyme showed significant homology to hoste of laccases from Coriolus versicolor, Pycnoporus cinnabarius, and an unidentified basidiomycete, PM1. The highest rate of 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonate) (ABTS) oxidation by laccase was reached at 45$^{\circ}C$, and te pH optima of the enzyme varied depending on the substrate in the range of 2.0 to 4.5. The enzyme was stable at 60$^{\circ}C$ for 5 h and lost 80% activity at 80$^{\circ}C$ in 30 min. The enzyme oxidized a variety of usual laccase substrates including lignin-related phenol, and had the highest affinity toward ABTS. Under standard assay conditions, the apparent Km value of the enzyme toward ABTS was 8.1 ${\mu}$M. The enzyme was completely inhibited by L-cysteine and sodium azide, but not by potassium cyanide, SDS, ad thiourea.

  • Alkalophilic Cephalosporium sp. RYM-202 produced multiple xylanases extracellularly. One of these xylanases was purified to electrophoretical homogeneity by chromatography with DEAE-Sephadex A-50, Sephacryl S-200 HR and Superose 12 HR. The purified xylanase differed from most other microbial xylanases in that it had low-molecular weight and acidic isoelectric point. The molecular weight of the xylanase in that it had low-molecular weight and acidic isoelectric point. The molecular weight of the xylanase was 23 kDa by SDS-polyacrylamide electrophoresis and 24 kDa by gel permeation chromatography, and the isoelectric point was 4.3. The xylanase had the highest activity permentation chromatography, and the isoelectric point was 4.3. The xylanase had the highest activity permeation chromatography, and the isoelectric point was 4.3. The xylanase had the highest activity at pH 8.0 and 50 .deg.C. It was stable over a wide range of pH and retained more than 80% of its original activity after 24 h of incubation even at pH 12. The Km values of this enzyme on birchwood xylan and oat spelts xylan were 2.33 and 3.45 mg/ml, respectively. The complete inhibition of the enzyme of n-bromosuccinimide suggests the involvement of tryptophan in the active site. The sylanase lacked activity towards crystalline cellulose and carboxymethyl cellulose.

Purification and Characterization of Mouse Liver Rhodanese

  • Rhodanese from mouse liver was purified to near homogeneity by ammonium sulfate precipitation, CM-Sephadex ion exchange, hydroxyapatite and Sephacryl S-200-HR gel filtration chromatographies with a purification of 776 folds. The molecular weight was determined by Sephadex G-150 gel filtration and found to be 34.8 KDa. SOS-PAGE showed molecular weight 34 KDa and two identical subunits splitting by aging for 3 weeks at $-70^{\circ}C$ the molecular weight of which was 17 KDa. The optimal pH of enzyme activity was 9.4 and the pI value of the enzyme was 6.6. Rhodanese showed the optimal reaction temperature of $25^{\circ}C$ and near linear increasing pattern until 10 min. incubation. $K_m$ values of rhodanese for KCN and $Na_{2}S_{2}O_{3}$ as substrates were 12.5 mM and 8.3 mM, respectively. Rhodanese activity was inhibited by more than 70% at a concentration of 100 ${\mu}M$ of $Ni^{2+}$, $Zn^{2+}$, $Cd^{2+}$, $Hg^{2+}$ and $Cu^{2+}$. Other metal ions, such as $Mn^{2+}$, $Mg^{2-}$, $Ca^{2+}$, and $Fe^{2+}$ showed no effect on rhodanese activity.

Serratia marcescens ATCC 25419가 생산하는 Acetolactate Synthase Isozyme의 특성 (The Properties of Acetolactate Synthase Isozyme Produced by Serratia marcescens ATCC 254 19)

  • Serratia marcescens ATCC 25419를 질소원이 풍부한 BHI 배지에서 황산 암모늄 분별 침전을 시킨 후 DAEA-Sephacel chromatography, Phenyl-Sepharose hydrophobic chromatography, Sephacryl S-400 gel filtration, native gel elution을 거쳐 ALS isozyme Rf 0.83을 분리하였다. 분리한 ALS isozyme Rf 0.83의 native 형태는 gel filtration을 이용하여 분자량을 측정한 결과, 531,400이었고, SDS-PAGE를 수행한 결과 55,000의 large subunit와 38,900의 small subunit로 구성된 multimer임을 알 수 있었다.
