송이균(松茸菌) (Tricholoma matsutake)의 배양환경(培養環境)에 대한 증식반응(增殖反應)에 관한 연구(硏究) (A Study on Multiplication Response of "Tricholoma matsutake" (Pine Mushroom) Conidio to Cultural Media Environment)

  • 송이(松茸)는 적송(赤松)의 세근(細根)에 활물기생(活物寄生)한다고 하나 그 본질(本質)이 아직 구명(究明)되지 못하고 있음으로 인공재배(人工栽培) 확립(確立)이 모색(模索)되고 있는 중이다. 본논문(本論文)에서는 일차적(一次的)으로 송이(松茸)에서 순수분리(純粹分離)한 균(菌)을 배양(培養)하여 생리적(生理的) 특성(特性)을 구명(究明)하고 인공원기(人工原基)를 만들어 인공적(人工的)인 송이재배(松茸栽培)의 본질(本質)을 구명(究明)하여 송이재배(松茸栽培) 방법(方法)을 돕고자 송이균(松茸菌)의 생리특성(生理特性)에 대(對)하여 조사한 결과(結果)는 다음과 같다. 1) 각종(各種) 배양기상(培養基上)에서의 송이균계(松茸菌系)의 발육(發育)은 봉밀(蜂蜜)+송이발생토양전즙(松茸發生土壤煎汁)+적송수근추출물(赤松鬚根抽出物)+건조효모(乾燥酵母)+$KH_2PO_4$+Inositol+엽산(葉酸)+Biotin의 배지(培地)가 가장 우수(優秀)하였다. 2) 송이포자(松茸胞子)의 발아(發芽) 및 균계배양(菌系培養)에서 최적온도(最適溫度)는 $24^{\circ}C$, 최적(最適) pH는 4.5였다. 3) 송이균계배양(松茸菌系培養)에 있어서 송이자실체(松茸字實體)의 조직(組織)에서 분리(分離)한 것과 포자(胞子)에서 분리(分離)한 것은 그 발율(發育)에 차이(差異)가 없었다, 4) Hamada 배지(培地)에 각종(各種) 미량중금속염류(微量重金屬鹽類) $ZnSO_4$, $MnSO_4$, $MgSO_4$, $CaCl_2$ 구연산철(枸櫞酸鐵) 등(等)의 첨가(添加)는 별효과(別效果)가 없었다. 5) 적송수근추출물(赤松鬚根抽出物) 첨가(添加)는 균계발육(菌系發育)에 확실(確實)한 효과(效果)가 있었다. 적송(赤松)에는 송이균계(松茸菌系)의 생장(生長)을 촉진(促進)하는 인자(因子)가 존재(存在)한다는 것을 알 수 있었다. 6) 송이균계(松茸菌系) 배양용(培養用) 인공배양기(人工培養基)의 C원(源)으로서 glucose보다 봉밀(蜂蜜)의 효과(效果)가 크다. 7) Fairy Ring이 가장 많이 존재(存在)하는 미생물(微生物)인 Mortierlla spp의 배양여액(培養濾液)을 송이균계(松茸菌系) 인공배양기(人工培養基)에 첨가(添加)한 것이 대조치(對照値)에 비(比)하여 균계(菌系)의 발육(發育)이 양호)良好)하였다. 8) 송이인공원기(松茸人工原基)의 배지(培地)에 적송수근추출물(赤松鬚根抽出物) lnsitol Biotin 엽산(葉酸)의 첨가(添加)는 효과(效果)가 크다. 균계(菌系)의 만연(蔓延)된 후(後) 온도(溫度)를 내려서 $19^{\circ}C$로 유지(維持)할 때 primordium의 형성(形成)을 확인(確認)하였다.

식물생장조절물질 말포민 B동족체의 화학구조 및 생리활성 (Chemical Structures and Physiological Activities of Plant Growth Substance, Malformin B's)

  • 말포민 B로부터 신규의 $B_3$, $B_4$$B_5$를 포함한 6종의 B동족체들이 단리 구조결정되었다. 본 연구에 의해 말포민 B동족체의 화학구조는, $B_{1a}$(1)가 cyclo-D-Cys-D-Cys-L-Val-D-Leu-L-allo-Ile, $B_{1b}$(2)가 cyclo-D-Cys-D-Cys-L-Val-D-Leu-L-Leu, $B_2$(3)가 cyclo-D-Cys-D-Cys-L-V al-D-Val-L-Leu, $B_3$(4)가 cyclo-D-Cys-D-Cys-L-Val-D-Ile-L-Leu, $B_4$(5)가 cyclo-D-Cys-D-Cys-L-Val-D-Ile-L-Ile, $B_5$(6)가 cyclo-D-Cys-D-Cys-L-Val-D-Val-L-Ile으로서 확정되었다. B's, $B_{1b}$ C 이들 중, 말포민 $B_{1b}$(2)의 화학구조는 말포민 $A_3$ 및 말포민 C와 동일한 것으로 판명되었다. 단리된 말포민 B동족체에 대해서 옥수수 근부굴곡활성과 녹두 상배축산장활성을 지표로 하여 생물검정을 실시한 결과, 모든 B동족체가 활성을 나타내었다. 옥수수 근부굴곡활성의 검정으로부터, 분자량 529의 동족체가 분자량 515의 동족체보다 높은 활성을 보여주었으며, 이는 HPLC상의 retention time에서 확인된 바와 같이, 전체 말포민분자의 극성 이 옥수수 근부굴곡활성에 대해서 영향을 마친다는 사실을 시사한다. 또한, 말포민의 구조를 유지시켜주는 -S-S-가교구조와 제4-제5 아미노산 잔기에 측쇄구조가 상이한 leucine과 isoleucine의 조합이 말포민 수용부위와의 결합에 있어서 최적의 형상을 취할 수 있다는 점에서 말포민류의 활성 발현에 중요한 구조적 요소들일 것으로 추정되어진다. 말포민 $B_{1a}$$0.25{\mu}m$에서 90%의 옥수수 근부굴곡활성을 나타내었다. 그러나, 녹두 상배축산장활성의 경우 분자량에 따른 활성의 차이는 인정되지 않았으며, 말포민 $B_5$의 처리시 $0.1{\mu}m$에서 대조구에 비해 1.54%의 신장촉진을 보여주었다.

미성숙 매복지치의 치낭, 치수, 치근유두 조직에서 다능성 줄기세포의 분리와 특성화에 대한 연구 (Isolation and characterization of human dental tissue-derived stem cells in the impacted wisdom teeth: comparison of dental follicle, dental pulp, and root apical papilla-derived cells)

  • Introduction: The first aim of this study was to isolate the dental tissue-derived stem cells from the dental follicle (DF), dental pulp (DP), and root apical papilla (RAP) of the extracted wisdom teeth. Second was to evaluate their characterization with the expressions of transcription factors and cell surface markers. Finally, their ability of the in vitro multi-lineage differentiations into osteogenic and adipogenic cells were compared, respectively. Materials and Methods: Dental tissues, including dental follicle, dental pulp, and root apical papilla, were separated in the extracted wisdom teeth. These three dental tissues were cultured in Dulbecco’s modified Eagle’s medium (DMEM) with supplements, respectively. After passage 3, the homogeneous shaped dental tissue-derived cells were analyzed the expression of transcription factors (Oct-4, Nanog and Sox-2) and cell surface markers (CD44, CD90 and CD105) with reverse transcription polymerase chain reaction (RT-PCR) and fluorescence-activated cell sorting (FACS) analysis. In order to evaluate in vitro multi-lineage differentiations, the culture media were changed to the osteogenic and adipogenic induction mediums when the dental tissue-derived cells reached to passage 3. The characteristics of these three dental tissue-derived cells were compared with immunohistochemistry. Results: During primary culture, heterogenous and colony formatted dental tissue-derived cells were observed in the culture plates. After passage 2 or 3, homogenous spindle-like cells were observed in all culture plates. Transcription factors and mesenchymal stem cell markers were positively observed in all three types of dental tissue-derived cells. However, the quantity of expressed transcription factors was most large in RAP-derived cells. In all three types of dental tissue-derived cells, osteogenic and adipogenic differentiations were observed after treatment of specific induction media. In vitro adipogenic differentiation was similar among these three types of cells. In vitro osteogenic differentiation was most strongly and frequently observed in the RAP-derived cells, whereas rarely osteogenic differentiation was observed in the DP-derived cells. Conclusion: These findings suggest that three types of human dental tissue-derived cells from extracted wisdom teeth were multipotent mesenchymal stem cells, have the properties of multi-lineage differentiations. Especially, stem cells from root apical papilla (SCAP) have much advantage in osteogenic differentiation, whereas dental follicle cells (DFCs) have a characteristic of easy adipogenic differentiation.

인삼(人蔘)의 Ether추출물(抽出物)의 성분(成分)에 관한 연구(硏究)(I) -유리지방산(遊離脂肪酸)에 관한 연구(硏究)- (Studies on the Components in the Ethereal Extract of Panax ginseng(I) -Studies on the Free Fatty Acids-)

  • 인삼(人蔘)의 ether 추출물(抽出物)로부터 유리(遊離) 지방산(脂肪酸)을 분리(分離)하고 이것을 diazomethane으로 처리하여 methyl ester를 만든 다음 초산수은 및 column chromatography를 이용(利用)하여 포화(飽和) 지방산(脂肪酸)과 불포화(不飽和) 지방산(脂肪酸)으로 분리(分離)하였다. 분리(分離)된 지방산(脂肪酸)을 G. L. C.에 의하여 분석(分析)하여 다음과 같은 결론(結論)을 얻었다. 1) 한국산(韓國産) 인삼(人蔘) 6년생근중(年生根中)에는 유리(遊離) 지방산(脂肪酸)이 0.28% 함유(含有)되어 있다. 2) 인삼중(人蔘中)에서 24 종류(種類)의 유리(遊離) 지방산(脂肪酸)이 발견(發見)되었으며 그 중(中) 22 종류(種類)는 G. L. C.에 의(依)하여 확인(確認)되었으나, 나머지 2 종류(種類)는 G. L. C. data 만 가지고는 확인(確認)할 수 없었다. 미확인(未確認)의 두 지방산(脂肪酸)은 그 양(量)이 다량(多量)이었으며 천연(天然)에 흔히 존재(存在)하지 않는 unusual fatty acids 라고 사료(思料)된다. 3) $L_{EE}$ 등(等)은 인삼중(人蔘中)에 n18:3이 존재(存在)한다고 보고(報告)한 바 있으나 본실험(本實驗)에서는 n18:3의 존재(存在)를 확인(確認)할 수 없었으며 그 대신(代身) peak XVI이 n18:2와 n18:3의 사이에서 나타남을 보여준다. peak XVI은 미확인(未確認) 지방산(脂肪酸)이다.

Diversity of Root-Associated Paenibacillus spp. in Winter Crops from the Southern Part of Korea

  • The genus Paenibacillus is a new group of bacilli separated from the genus Bacillus, and most of species have been isolated from soil. In the present study, we collected 450 spore-forming bacilli from the roots of winter crops, such as barley, wheat, onion, green onion, and Chinese cabbage, which were cultivated in the southern part of Korea. Among these 450 isolates, 104 Paenibacillus-like isolates were selected, based on their colony shape, odor, color, and endospore morphology, and 41 isolates were then finally identified as Paenibacillus spp. by 16S rDNA sequencing. Among the 41 Paenibacillus isolates, 23 were classified as P. polymyxa, a type species of the genus Paenibacillus, based on comparison of the 16S rDNA sequences with those of 32 type strains of the genus Paenibacillus from the GenBank database. Thirty-five isolates among the 41 Paenibacillus isolates exhibited antagonistic activity towards plant fungal and bacterial pathogens, whereas 24 isolates had a significant growth-enhancing effect on cucumber seedlings, when applied to the seeds. An assessment of the root-colonization capacity under gnotobiotic conditions revealed that all 41 isolates were able to colonize cucumber roots without any significant difference. Twenty-one of the Paenibacillus isolates were shown to contain the nifH gene, which is an indicator of $N_{2}$ fixation. However, the other 20 isolates, including the reference strain E681, did not incorporate the nifH gene. To investigate the diversity of the isolates, a BOX-PCR was performed, and the resulting electrophoresis patterns allowed the 41 Paenibacillus isolates to be divided into three groups (Groups A, B, and C). One group included Paenibacillus strains isolated mainly from barley or wheat, whereas the other two groups contained strains isolated from diverse plant samples. Accordingly, the present results showed that the Paenibacillus isolates collected from the rhizosphere of winter crops were diverse in their biological and genetic characteristics, and they are good candidates for further application studies.

두릅나무 25개 선발 클론의 형태적 특성과 유연관계 (Morphological Characteristics and Classification of 25 Selected Clones of Aralia elata Seem)

  • 본 연구에서는 기능성 약용식물로 부가가치가 높은 두릅나무 선발클론을 대상으로 잎, 가시 그리고 동아의 형태적 특정을 조사하고 다변량 분석 방법을 통하여 선발클론 간 유연관계를 분석하였다. 주성분 분석 결과, 제4 주성분까지의 누적 기여율은 76%로 나타났으며, 유집분석 결과 거리 수준 2.0을 기준으로 I그룹은 강원 연곡 등 15클론, II그룹은 경기 여주 등 5클론, III그룹은 봉화와 울릉 클론, IV그룹은 용문과 보성 클론 그리고 V그룹은 신구 클론으로 총 5개 그룹으로 구분되었다.

  • 종래의 말포민류에 대한 화학구조 연구에 있어서의 난제들이 최신의 각종 기기분석 및 새로운 정제 방법의 적용에 의해 해결되어졌으며, 이 결과 말포민 A로부터 신규의 $A_4$를 포함한 4종의 A동족체들이 단리 구조결정되었다. 본 연구에 의해 말포민 A동족체의 화학 구조는, $A_1$(1)이 cyclo-D-Cys-D-Cys-L-Val-D-Leu-L-Ile, $A_2$(2)가 cyclo-D-Cys-D-Cys-L-Val-D-Leu-L-Val, $A_3$(3)가 cyclo-D-Cys-D-Cys-L-Val-D-Leu-L-Leu, $A_4$(4)가 cyclo-D-Cys-D-Cys-L-Val-D-Ile-L-Val으로서 확정 되었다. 이들 중, 말포민 $A_3$(3)는 말포민 C와 동일한 화합물로 확인되었다. 단리된 말포민 A동족체에 대해서 옥수수 근 부굴곡활성과 녹두 상배축신장활성을 지표로 하여 생물검정을 실시하였다. 옥수수 근부굴곡 활성의 검정으로부터, 모든 A동족체가 활성을 나타내었으며, 특히 분자량 529의 동족체는 분자량 515의 동족체보다 높은 활성을 보여주었다. 그러나, 녹두 상배축신장활성의 검정에서는 분자량에 따른 차이가 인정되지 않았으며, 이들 중 말포민 $A_4$는 활성형이 아닌 것으로 확인되었다. 양검정법에서 모두 최고 수준의 활성을 나타낸 말포민 $A_1$은, $0.25{\mu}m$ 에서 83%의 옥수수 근부굴곡활성을 나타냈으며, 녹두 상배축신장활성의 경우, $0.1{\mu}m$에서 대조구에 비해 165%의 신장촉진을 보였다.

편백나무(Chamaecyparis obtusa) 추출물의 항산화, 미백효과에 관한 연구 (Studies on the Antioxidant and Whitening Effects of Chamaecyparis obtusa Extract)

  • 본 연구는 화장품 소재로서 편백나무(Chamaecyparis obtusa)의 부위별 이용 가능성을 알아보기 위하여 잎·가지(Leaf), 수피(Bark), 목재(Wood), 뿌리(Root)로 분리하여 99.9 % 에탄올에 추출한 시료로 항산화, 미백효능을 확인하였다. 이를 위해 다음과 같은 연구를 진행하였다. 항산화 평가는 11-diphenyl-2-picrylhydrazyl (DPPH) 라디칼 소거능, 2 2′-azino-bis (3-ethylbenzothiazoline-sulfonic acid) diammonium salt (ABTS+) 라디칼 소거능으로 확인하였다. 미백 효과를 평가하기 위해서 mushroom tyrosinase 저해 활성 평가, MTT assay를 통한 편백나무 추출물의 세포독성평가를 진행하였으며 cellular tyrosinase 저해율 측정과 melanin 생합성량을 확인하였다. 항산화 활성 결과 편백나무 부위 중 수피(Bark) 추출물이 가장 효능이 우수하였으며 B16F10 melanoma cell내 편백나무 수피(Bark)는 tyrosinase 활성 억제 및 멜라닌 생합성 억제 효과를 나타내었다. 따라서 편백나무 수피 에탄올 추출물의 안전하고 우수한 미백 기능성 화장품의 천연물질 소재로서의 개발 가능성을 확인하였다.

Propagation of Bell Pepper (Capsicum annuum var. grossum) through tissue culture

  • Leaf discs and apical meristems were cultured in Murashige and Skoog (MS) medium supplemented with cytokinin and auxin at different concentrations. Callus production was observed in all tested media after six days of incubation. Callus produced in the presence of high concentration of NAA (2.0mg/1) was fragile in texture and yellow in colour. Highest callus formation was observed from leaf discs in the medium supplemented with 1.0mg/1 NAA and 0.5 mg/l BAP in dark at $25{\pm}1{\circ}C$. Percentage of callus formation was 95% and mean callus fresh weight was 654.88 43.53 mg. Shoots were induced from the callus after 4 weeks in 1/2MS medium supplemented with BAP and kinetin both at 0.5mg/1. When elongated shoots were separated and transferred into multiplication medium (MS+0.5mg/1 BAP+0.5mg/1 kinetin) multiplication rate was 6.4 after 6 weeks. Higher concentrations of BAP caused callus production at the base. Direct shoot induction was observed from apical meristems in MS medium in the presence of 0.175 mg/1 IAA + 2.25mg/1 BAP and 0.175 mg/1 IAA + 3.0 mg/1 BAP in 16 hour day at $25{\pm}1{\circ}C$. Explants (apical meristems) elongated to form a single shoot forming a callus at the base. Adventitious buds were sprouted out from the base. Percentage explants which producing shoots was 28.57 and 65.5 respectively. Multiple shoot induction was also observed in the same media. Highest multiple shoot production was observed in the presence of 0.175 mg/l IAA and 3.0mg/l BAP, Mean number of shoots per explant was 5.36 and the mean shoot length was $16.66{\pm}4.15$mm. Shoots (20 30m length) were tested for root induction. Excised shoots were transferred into rooting media, which contains different concentrations of NAA and IAA. Best rooting performance was observed in 1/2MS medium supplemented with 0.1mg/1 NAA after 10 days of incubation in 16 hr photoperiod at $25{\pm}1{\circ}C$. Mean number of roots per shoot was 6 and the mean root length was 252mm. Rooted plantlets were transferred into sterile coir dust:sand (1:4) mixture and maintained in a humid chamber for two weeks, They were gradually exposed to the natural environment. After three weeks they were transferred to pots containing coir dust:sand (1:2) mixture for further development where the 90% survival was observed.

백서에서 Depulpin®과 Formocresol에 대한 치수와 치근단 조직의 반응 (PULPAL AND PERIAPICAL REACT10N TO FORMOCRESOL AND DEPULPIN® IN THE RAT TEETH)

  • One fifth dilution of formocresol is usually used for pulpotomy of the primary teeth and emergency pulpotomy of the permanent teeth. However the use of formaldehyde has been subjected to criticism because it may be absorbed into the blood stream and become distributed systemically, it nay also alter the pulp tissue rendering it immunologically active, and have carcinogenic potential. Recently Depulpin$^{\circledR}$(VoCo., Germany) gains popularity as a devitalizing agent during root canal therapy in spite of high concentration of 49 % paraformaldehyde because it facilitate devitalization of pulp and make root canal therapy easier But there have been not enough publications about the reaction of pulp and periapical tissue caused by Depulpin. This study was performed to evaluate the histological changes in pulp and periapical tissue of rats after pulpotomy using formocresol and Depulpin and to elucidate the toxic effects of these agents. Thirty six Sprague-Dawley rats were anesthetized by intraperitoneal injection of ketamine Maxillary first molar teeth were used for pulpotomy with formocresol and Depulpin. Rats were sacrificed after 2 days, 4 days, 1 week, 2 weeks, 3 weeks and 4 weeks respectively. Specimens were histologically observed by light microscope changes in pulp and periapical tissue. The obtained results were as follows. 1. Formocresol group A zone of fixed tissue. in which odontoblasts could clearly be defined, was present directly underneath the pulpotomy dressing in almost all teeth of this group. This was followed by an area of necrotic tissue which resembled dried out fibrous tissue with no cellular detail except some pyknotic nuclei. In the specimens of after 2 days, 4 days, 1 week, 2 weeks in which vital tissue was present, it was separated from the fibrous area by a zone of inflammation. In the specimens of after 3 weeks and after 4 weeks, inflammatory infiltrate was in the periodontal ligament adjacent to the apical foramina of the teeth. 2. Depulpin$^{\circledR}$ group The area of necrotic tissue which had no cells and fibers, was present adjacent to the dressing. This was followed by dried out fibrous tissue with no cellular details except some pyknotic nuclei, A short stump of vital pulp with odontoblasts was present at the end of the canal after 2 days. Inflammatory infiltrate was in the periodontal ligament after 4 days and after 1week. Severe root resolution and necrosis of periapical tissue opposite the root resorption site were defined after 2 weeks and after 3 weeks. Periapical lesion which consist of necrotic tissue surrounded by a fibrous connective wall, was found after 4 weeks. The results indicated that Depulpin can cause more adverse reaction to the dental pulp and periapical tissue than formocresol, and further studies are needed for its clinical use with safety.
