Antioxidants Supplementation on Acid Base Balance during Heat Stress in Goats

  • The effects of vitamin C and vitamin E with selenium on acid-base balance and some stress hormones were evaluated during heat stress in goats. Goats, 1.5 years of age, were divided into control, heat stress and antioxidant treatment groups 1, 2 and 3. Except for the control, all groups were exposed to a temperature of $40{\pm}2^{\circ}C$ with a relative humidity of 30% for 5 h/d for 21 days in a psychrometric chamber. Rectal temperature and respiratory rates were recorded daily post exposure. Blood samples were collected on every 3rd day for estimation of plasma vitamins C and E, total antioxidant activity and hormones, and separate blood samples were taken to estimate acid-base status. The rectal temperature and respiratory rates were increased (p<0.05) in the heat stress group only. Except for pH and $pO_2$, which were increased significantly (p<0.05) other parameters of acid-base balance such as $pCO_2$, $HCO_3^-$, $TCO_2$, BEb, BEcef, PCV and Hb were significantly decreased (p<0.05) in the heat stress group. An improvement in acid-base status was noted in the antioxidant supplemented groups. Prolactin and cortisol levels were significantly (p<0.05) higher and free T3 and T4 levels were significantly (p<0.05) lower in the heat stress group. Levels of prolactin and cortisol were decreased and free T3 and T4 were increased in antioxidant treatment groups. Different levels of antioxidant supplementation resulted in similar protection against heat stress.

Effect of Sodium Selenite and Zinc-L-selenomethionine on Performance and Selenium Concentrations in Eggs of Laying Hens

  • The objective of this study was to determine the effect of sodium selenite and zinc-L-selenomethionine on performance and egg Se concentration in laying hens. Two hundred and twenty-four CP Browns aged 71 weeks were divided according to a $2{\times}3$ factorial in a completely randomized design. One more group without additional Se supplementation was used as a negative control. Each treatment consisted of four replicates and each replicate contained eight laying hens. The dietary treatments were T1: basal diet; T2, T3 and T4: basal diets plus 0.3, 1.0 and 3.0 mg Se from sodium selenite/kg, respectively; T5, T6 and T7: basal diets plus 0.3, 1.0 and 3.0 mg Se from zinc-L-selenomethionine/kg, respectively. The findings revealed that feed conversion rate/kg eggs, egg production, egg weight, Haugh units and eggshell thickness were not affected by source and level of Se (p>0.05). Increasing level of dietary Se significantly increased (p<0.05 the Se content of eggs. Zinc-L-selenomethionine markedly increased p<0.05 egg Se concentration as compared with sodium selenite. The results indicated that Se source did not influence performance of laying hens. However, zinc-L-selenomethionine increased p<0.05 egg Se concentration more than sodium selenite.

Effects of Housing Systems and the Diet Supplements on the Slaughter Value and Concentration of Mineral Elements in the Loin Muscle of Lambs

  • The objective of this study was to investigate the effect of maintenance system as well as the effect of Se, Zn, and vitamin E supplementation of ram-lambs on the slaughter value and concentration of mineral elements in the loin muscle of lambs. The experiment was conducted on 72 Polish Merino ram-lambs divided into three groups: group C, indoor with no supplement, 19 lambs; S, indoor with supplement, 23 lambs; G, outdoor with no supplement, 30 lambs. From birth all the lambs were maintained indoor with their dams and then weaned at the age of 8 weeks. The rams from group C and S were placed in individual straw-bedded pens and fattened individually with concentrate mixture offered ad libitum until the age of 16 weeks. The lambs from group G were grazed every day from May to July (2 months). During the fattening period each lamb from the supplemented group S was administered per os 1 mL 0.1% $Na_2SeO_4$ (Se, 0.42 mg), 3 mL 10% $ZnSO_4$ (Zn, 68 mg), and 1 mL premix protect vitamin E (0.1 g ${\alpha}$-tocopherol, 5 mg lysine, 5 mg methionine) daily. A comparison of half carcasses across the groups has shown no difference between the control group and the one with supplements, while the weight of half carcasses in the grazing group was smaller in comparison with groups C and S (p<0.001). The meat content in the pelvic limb showed no differences across all groups under study. The pelvic limb of grazing lambs contained less fat compared to the control and supplemented groups (p<0.001). The concentrations of Se and Zn in the blood plasma of ram-lambs from the supplemented group were significantly higher than for the control and grazing lambs. Inorganic Se and Zn supplementation with vitamin E to the diet of lambs increased Se and Zn levels in loin muscle (p<0.001) to $0.46{\mu}g/g$ and $32.9{\mu}g/g$ in fresh tissue, respectively.

비타민 E와 불포화 지방과의 관계 - 들깨유(油)를 중심으로 한 동물의 비교 연구 - (Relationship between Vitamin E and Polyunsaturated Fat - A comparative animal study emphasizing perilla seed oil as a fat constituent -)

  • Perilla (frutescens) seed oil, which is widely used as a source of vegetable oil in Korea, contains a strikingly large amount (58.4% of total fatty acids) of polyunsaturated linolenic acid (18 : 3) which is one of the essential fatty acids. Our hypothesis was that vitamin E contained in this oil would not be enough to prevent peroxidation of this polyunsaturated oil. A comparative study was carried out using rats and chicks devided into seven groups with various diet combinations emphasizing fat sources for the period of four weeks. The level of fat in each diet was 15% and animals were fed ad libitum. Various diet combinations were as follows; perilla seed oil and sesame seed oil with and without vitamin E supplementation, tallow as a saturated fat source and perilla seed hull group (10% at the expense of carbohydrate). The fat constituents of control group were consisted of 50% vegetable oil and 50% animal fat. A few important findings are as follows: 1. Rats fed perilla seed oil lost their hair focally around the neck and suffered from a bad skin lesion at the same place. In chicks, yellow pigmentation both of feather and of skin was clearly observed only in groups fed perilla seed oil with or without vitamin E supplementation. The basis of biochemical mechanisms of this phenomena remains as an important research interest. 2. The mean value for hematocrit was significantly lower for the chicks fed perilla seed oil than for those fed control diet. This result seems to be attributable to the effect on the red cell membrane known as peroxidation-hemolysis of vitamin E deficiency. 3. The serum cholesterol level was higher for the rats fed perilla seed oil than for those fed control diet, whereas in chicks the group fed perilla seed oil showed lower value than the control group indicating that different animal species could vary in their responses to the same diet. 4. In pathological examinations, the sign of hepatic fibrosis was seen in the perilla seed hull group and it was noticeable that the level of hepatic RNA was significantly increased in the rat recovering from vitamin E deficiency. It is hoped that more detailed studies on perilla seed oil and hulls will soon be carried out in many aspects especially i) at various levels of fat in the diet, ii) in relation to dietary selenium level and iii) to find an optimum level of dietary essential fatty acids in terms of P/S ratio using various animal species. In the mean time, the public should be informed to preserve this particular oil with care to minimize fatty acid oxidation and should be discouraged from overconsuming this oil. In-feed organic and inorganic manganese supplementation on broiler performance and physiological responses

  • Objective: A trial was conducted to investigate the effects of supplemental levels of Mn provided by organic and inorganic trace mineral supplements on growth, tissue mineralization, mineral balance, and antioxidant status of growing broiler chicks. Methods: A total of 500 male chicks (8-d-old) were used in 10-day feeding trial, with 10 treatments and 10 replicates of 5 chicks per treatment. A 2×5 factorial design was used where supplemental Mn levels (0, 25, 50, 75, and 100 mg Mn/kg diet) were provided as MnSO4·H2O or MnPro. When Mn was supplied as MnPro, supplements of zinc, copper, iron, and selenium were supplied as organic minerals, whereas in MnSO4·H2O supplemented diets, inorganic salts were used as sources of other trace minerals. Performance data were fitted to a linearbroken line regression model to estimate the optimal supplemental Mn levels. Results: Manganese supplementation improved body weight, average daily gain (ADG) and feed conversion ratio (FCR) compared with chicks fed diets not supplemented with Mn. Manganese in liver, breast muscle, and tibia were greatest at 50, 75, and 100 mg supplemental Mn/kg diet, respectively. Higher activities of glutathione peroxidase and superoxide dismutase (total-SOD) were found in both liver and breast muscle of chicks fed diets supplemented with inorganic minerals. In chicks fed MnSO4·H2O, ADG, FCR, Mn balance, and concentration in liver were optimized at 59.8, 74.3, 20.6, and 43.1 mg supplemental Mn/kg diet, respectively. In MnPro fed chicks, ADG, FCR, Mn balance, and concentration in liver and breast were optimized at 20.6, 38.0, 16.6, 33.5, and 62.3 mg supplemental Mn/kg, respectively. Conclusion: Lower levels of organic Mn were required by growing chicks for performance optimization compared to inorganic Mn. Based on the FCR, the ideal supplemental levels of organic and inorganic Mn in chick feeds were 38.0 and 74.3 mg Mn/kg diet, respectively.

Effects of functional nutrients on chicken intestinal epithelial cells induced with oxidative stress

  • The objective of this study was to investigate the protective effects of functional nutrients including various functional amino acids, vitamins, and minerals on chicken intestinal epithelial cells (cIECs) treated with oxidative stress. The cIECs were isolated from specific pathogen free eggs. Cells were exposed to 0 mM supplement (control), 20 mM threonine (Thr), 0.4 mM tryptophan (Trp), 1 mM glycine (Gly), 10 μM vitamin C (VC), 40 μM vitamin E (VE), 5 μM vitamin A (VA), 34 μM chromium (Cr), 0.42 μM selenium (Se), and 50 μM zinc (Zn) for 24 h with 6 replicates for each treatment. After 24 h, cells were further incubated with fresh culture medium (positive control, PC) or 1 mM H2O2 with different supplements (negative control, NC and each treatment). Oxidative stress was measured by cell proliferation, whereas tight junction barrier function was analyzed by fluorescein isothiocyanate (FITC)-dextran permeability and transepithelial electrical resistance (TEER). Results indicated that cell viability and TEER values were less (p < 0.05) in NC treatments with oxidative stress than in PC treatments. In addition, FITC-dextran values were greater (p < 0.05) in NC treatments with oxidative stress than in PC treatments. The supplementations of Thr, Trp, Gly, VC, and VE in cells treated with H2O2 showed greater (p < 0.05) cell viability than the supplementation of VA, Cr, Se, and Zn. The supplementations of Trp, Gly, VC, and Se in cells treated with H2O2 showed the least (p < 0.05) cellular permeability. In addition, the supplementation of Thr, VE, VA, Cr, and Zn in cells treated with H2O2 decreased (p < 0.05) cellular permeability. At 48 h, the supplementations of Thr, Trp, and Gly in cells treated with H2O2 showed the greatest (p < 0.05) TEER values among all treatments, and the supplementations of VC and VE in cells treated with H2O2 showed greater (p < 0.05) TEER values than the supplementations of VA, Cr, Se, and Zn in cells treated with H2O2. In conclusion, Thr, Trp, Gly, and VC supplements were effective in improving cell viability and intestinal barrier function of cIECs exposed to oxidative stress.

한약재내의 미량원소의 의의와 치료효과에 대한 고찰 (Therapeutical Effect with Trace Elements in Herbal Medicine)

  • Trace element are involved in enzymatic activities, immunological reactions. physiological mechanisms. Deficiency in some trace elements, such as iron and iodine. is still an important health problem, The role of trace elements deficiency is suspected in various clinical situations and is now confirmed by well designed supplementation studies. However, the importance of trace elements as chinese herbal constituents is not sufficiently appreciated by the oriental medical profession, although in recent years a significant increase of new finding on their essential character in chinese herbal medicine occurred. It is well known that herbal medicine contains a variety of trace elements which would show therapeutic effects with active components in herbal medicine . In china, recent work showed some positive correlation between trace element and traditional chinese medicine (TCM) in terms of therapeutic effects even if their role in therapeutic effects is still obscure. In korea, not much attention has been on the therapeutic importance of trace element contained in herbal medicine Here, the therapeutic effects of trace element in TCM were reviewed and summarized. 1. Iron, copper, zinc and manganese are mainly contained in TCM. In addtion, chromium, magnesium, molybdenum, nickel, alminium, cobalt, arsenic and selenium has been studied for their therapeutic effects 2. Zinc, is decreased in patients who have deficiency of kindney(腎虛) and chronic disease. Fe is decreased in patients who have deficiency of blood(血虛). However copper is increased in patients who have chronic disease and hepatic disease.3 Iron concentration is high in herbs used for tonifying and nourishing yin or blood(補陰補血藥) Zinc concentration is high also in herb used for tonifying kidiney and vital essence(補腎補精藥). In addition. copper concentration Is high in herb used for replenishing qi(補氣藥) 4 In herbal drugs, the therapeutic substances in TCM are not only organic but also inorganic. It seems that trace elements would be one of components in herb for its therapeutic effects. This indicates that therapeutic effects of TCM should be extended not only to herb itself, bur also to trace elements contained in herb.

Protective effect of resveratrol on arsenic trioxide-induced nephrotoxicity in rats

  • BACKGROUND/OBJECTIVES: Arsenic, which causes human carcinogenicity, is ubiquitous in the environment. This study was designed to evaluate modulation of arsenic induced cancer by resveratrol, a phytoalexin found in vegetal dietary sources that has antioxidant and chemopreventive properties, in arsenic trioxide ($As_2O_3$)-induced Male Wistar rats. MATERIALS/METHODS: Adult rats received 3 mg/kg $As_2O_3$ (intravenous injection, iv.) on alternate days for 4 days. Resveratrol (8 mg/kg) was administered (iv.) 1 h before $As_2O_3$ treatment. The plasma and homogenization enzymes associated with oxidative stress of rat kidneys were measured, the kidneys were examined histologically and trace element contents were assessed. RESULTS: Rats treated with $As_2O_3$ had significantly higher oxidative stress and kidney arsenic accumulation; however, pretreatment with resveratrol reversed these changes. In addition, prior to treatment with resveratrol resulted in lower blood urea nitrogen, creatinine and insignificant renal tubular epithelial cell necrosis. Furthermore, the presence of resveratrol preserved the selenium content ($0.805{\pm}0.059{\mu}g/g$) of kidneys in rats treated with $As_2O_3$. However, resveratrol had no effect on zinc level in the kidney relative to $As_2O_3$-treated groups. CONCLUSIONS: Our data show that supplementation with resveratrol alleviated nephrotoxicity by improving antioxidant capacity and arsenic efflux. These findings suggest that resveratrol has the potential to protect against kidney damage in populations exposed to arsenic.

로즈힙의 영양학적 기능 및 생리활성에 대한 문헌적 고찰 (The nutritional functions and physiological activities of rose hip (Rosa canina fruits): A systematic review)

  • 본 총설은 로즈힙의 영양학적 기능성에 대한 선행 연구결과들을 조사하여 로즈힙의 다양한 효능들을 체계적으로 정리함으로써 로즈힙을 효과적으로 사용하기 위한 기초 자료를 제공하기 위해 작성되었다. 본문에서 언급한 것처럼 로즈힙에는 유용한 비타민 및 무기질 성분들이 풍부하고 β-카로틴, β-시토스테롤, 카테킨, 라이코펜, 폴리페놀, 플라보노이드, 프로안토시아닌, 퀘르세틴, 탁시폴린, 엘라그산 및 살리실산 등의 기능성 성분들이 다량 함유되어 있어 항산화 효과가 뛰어나며, 골관절염 개선, 체지방을 감소, 피부의 노화를 방지, 심혈관 질환률 감소, 항균·항당뇨 및 항암 효과가 뛰어나 기능성 식품으로서 활용 가능성이 매우 높다. 따라서 다양한 질환에 노출되고 있는 현대인들에게 화학적 합성물질이나 약물과 비교하여 천연 기능성 소재로써 비교적 안전하게 사용될 수 있는 로즈힙의 장점을 활용하기 위하여 로즈힙에 대한 연구가 지속될 필요가 있으며, 로즈힙은 천연 기능성 소재로서 다양한 질환을 예방 및 개선하는데 효과적으로 사용될 수 있다고 사료된다.

Selenium과 비타민 E의 급여가 비육돈의 육질 특성 및 저장성에 미치는 영향 (Effects of Selenium and Vitamin E Supplementation on Meat Quality and Shelf-Life in Finishing Pigs)

  • 본 연구는 비육돈 사료내 Se과 비타민 E를 첨가하였을 때 육질 특성 및 저장성을 알아보기 위해 실시하였다. 3원 교잡종($Landrace{\times}Yorkshire{\times}Duroc$) 비육돈 80두를 공시하였으며, 시험개시시의 체중은 74.74 kg이었다. 시험설계는 Se의 첨가수준과 첨가형태에 따라 1) CON(basal diet), 2) ISE2 (basal diet + 0.2 ppm inorganic Se + 100 ppm vitamin E), 3) ISE4(basal diet + 0.4 ppm inorganic Se + 100 ppm vitamin E), 4) OSE2(basal diet + 0.2 ppm organic Se + 100 ppm vitamin E) 및 5) OSE4(basal diet +0.4 ppm organic Se + 100 ppm vitamin E)로 5처리를 하여 처리당 4반복, 반복당 4마리 씩 완전임의 배치하였다. 가열 감량은 ISE4 처리구가 CON 처리구와 비교하여 유의적으로 낮게 나타났다(p<0.05). 조지방 함량은 OSE2 처리구가 다른 처리구들과 비교하여 유의적으로 높게 나타났다(p<0.05). 적색도를 나타내는 $a^*$-값은 저장 10일째 OSE2 처리구가 다른 처리구들과 비교하여 유의적으로 높게 나타났다(p<0.05). TBARS에서도 저장 10일째에 OSE4 처리구가 CON과 ISE2 처리구와 비교하여 유의적으로 낮게 나타났다(p<0.05). 결론적으로, 비육돈 사료내 Se와 비타민 E를 첨가하였을 때 가열감량, 육색 및 지방산패도에 미치는 영향은 미미한 것으로 사료된다.
