Effects of Feeding Period of Organic Selenuim Supplementation on Meat Quality of Hanwoo Steers (유기 셀레늄 강화 버섯 폐배지 급여기간이 쇠고기육질에 미치는 영향)

  • Current study was conducted to investigate the effect of organic selenium supplementation originated from mushroom culture medium on meat quality of Hanwoo sleets. The result showed that organic selenium supplementation of 0.9 ppm (DM based ratio) for 2 (T1), 4 (T2) and 6 (T3) months had no effect on moisture content in longissimus muscle, with 63 to $66\%$ compared to non-supplemented control group Similarly, intramuscular fat content ranged from 11.7 to $16.4\%$ did not differ between the treatment groups (p>0.05). T3 group showed the highest protein content with $20.8\%$ while T2 group had the lowest content with $19.2\%$ The data indicated that organic selenium supplementation to the experimental concentration had indictable effect on proximate composition The treatments similarly had no influence on physical and biological characteristics of longissimus muscle, where cooking loss and shear force ranged from 20 to $21\%$ and from 3.6 to 4.4kg, respectively. On the other hand, muscle pH at 24 h postmortem showed 5.52, 5.57, 5.50, 5.50 for control, T1, T2 and T3, respectively, indicating that the longer feeding period resulted in the lower ultimate un A similar trend was observed from water-holding capacity (63.8, 64.4 and $64.2\%$ for T1, T2 and T3, respectively) which was significantly higher than con01 group of $59.5\%$ For sensory evaluation, juiciness did not differ between the treatment groups, but n and n (5.30 and 5.28, respectively) showed significantly tender mat Particularly, T2 group received significantly higher flavor score among the treatment groups including controls. Growth and substance transfer effect of Pleurotus cornucopiae by high concentration treatments of selenium(II) (고농도 셀레늄 처리가 노랑느타리버섯(Pleurotus cornucopiae) 생육과 물질전이에 미치는 영향(II))

  • The research for incubation period, mycelial density, day required for primordial formation after inoculation(below DPI), number of valid stipes, individual weight and accumulation amounts of organic selenium for P. cornucopiae by treating 600, 700, 800, 900, 1000(${\mu}g/50g$) of $Na_2SeO_3$ is following. Incubation periods of P. cornucopiae are 25~30 days per each treatment with $Na_2SeO_3$. Compared to the control which took 22 days of incubation period, it is increased 3 or 8 days for the treatment of $600{\sim}1000{\mu}g/50g$. Mycelial density of P. cornucopiae treated with $Na_2SeO_3$ between 600 and $1000{\mu}g/50g$ is very similar with control. DPI of P. cornucopiae treated with $Na_2SeO_3$ between 600 and $1000{\mu}g/50g$ was increased 3 or 8 days. Number of valid stipes of P. cornucopiae treated with $Na_2SeO_3$ between 600 and $1000{\mu}g/50g$ was between 10 and 16. It was decreased 2 or 8 as compared to 18 of control. Individual weight of P. cornucopiae treated with $Na_2SeO_3$ between 600 and $1000{\mu}g/50g$ was between 94 and 116g/850cc. It was decreased 5.7~23.5% as compared to 123g/850cc of the control. Accumulation amount of organic selenium for P. cornucopiae treated with $Na_2SeO_3$ between 600 and $1000{\mu}g/50g$ was $9.1{\sim}10.8{\mu}g/g/dry$. It was increased 182~216 times as the concentration increased when compared to $0.05{\mu}g/g/dry$ of the control. In conclusion, incubation period, mycelial density, DPI, number of valid stipes, individual weight and accumulation amounts of organic selenium for P. cornucopiae by treating $600{\sim}1000{\mu}g/g$ of $Na_2SeO_3$ was decreased. So that the optimal treatment was less $400{\mu}g/g$ than $600{\sim}1000{\mu}g/g$.

A Study on Heavy Metal and Selenium Levels in Dried Seafoods (건조수산물의 중금속 및 셀레늄 함량)

  • The heavy metal concentrations in dried seafoods commonly consumed in Gyeonggi-do were analyzed. Concentrations of lead (Pb), cadmium (Cd), mercury (Hg) and selenium (Se) were measured in 95 samples with an inductively-coupled plasma-mass spectrometer (ICP-MS) and an Hg analyzer. The average concentration [mean±SD(minimum-maximum) mg/kg] of heavy metals were as follows: Pb 0.062±0.071(0.002-0.428), Cd 0.083±0.100(0.004-0.540), Hg 0.012±0.012(N.D-0.054) and Se 0.839±0.371(0.362-2.124). All the levels were below the recommended standards of the MFDS (Ministry of Food and Drug Safety). In the comparison of heavy metal content by anchovy size, it was shown that mercury was significantly higher in large anchovy (P<0.05). Selenium levels were found to differ significantly in small anchovy and large anchovy (P<0.05). Pb, Cd and Hg were significantly higher in Tiger prawn among shrimp (P<0.05). The heavy metal and selenium levels of anchovy and shad were highest in the intestine. However, the heavy metal and selenium levels of shrimp were highest in the head. In addition, weekly (monthly) intake of mercury and cadmium from dried seafoods was found to be 0.712% and 2.978% of PTW(M)I (Provisional Tolerable Weekly(Monthly) Intake) respectively. Therefore, it was found that dried seafoods were safe for consumption.

  • The wide distribution of redox-sensitive elements (RSE) as arsenic (As), cadmium (Cd), selenium (Se), and strontium (Sr) in the top soil of Siwaqa area are related to the weathering action of alkaline surface and groundwater on the parent rocks. The bioavailability, distribution, sorption, and ecotoxicity of As, Cd, Se, and Sr, of the wild plants and top soils in the study area were investigated. A total number of 23 surface soil samples and 23 plant samples were collected and analyzed for the most toxic elements. The uptake of elements by plants was dependent on the plant species and the concentration of elements in the soil. For example, Sr was the highest concentration in soil samples and plants, while Se was the lowest concentration in soil samples and pants. For the plants, the results showed that Bellevalia sp. had the highest elements uptake, while Allium rothii had the lowest elements uptake. The results of this work provide a valuable knowledge for understanding the bioavailability of some toxic elements in the soil and plants of Central Jordan. The results are expected to be of great help for the Jordanian Uranium Mining Company during their environmental risk assessments.

In Vitro Production of Bovine Embryos by Modification of Simple Defined Culture Medium (단순한정배양액의 성분조정에 의한 소 수정란의 체외생산)

  • In this study, we investigated the effects of three kinds of culture medium (Charles and Rosenkrans; CRlaa, Tyrode's; TALP, synthetic oviduct fluid: SOF), insulin transferrin + selenium complex (ITS), macromolecules(polyvinyl alcohol: PVA, fetalb-ovine serum: FBS) and NaCl on the development of early bovine embryos. In experiment 1, there were no differences in embryo development among three kinds of embryo culture medium (CR $l_{aa}$ , TALP, SOF). In experiment 2, BSA, FBS and PVA were added each in TALP as macromolecule sources. The developmental rates of embryos in BSA or FBS added TALP were significantly higher than in PVA added one (p〈0.01), but there was no difference between BSA and FBS added groups. In experiment 3, bovine embryos were cultured in TALP with the following supplements: BSA alone(1, 3 or 8 mg/ml, each) or BSA(1, 3 or 8 mg/ml, each)+ITS (10$\mu\textrm{g}$/m1 insulin, 5 $\mu\textrm{g}$/ml transferrin, 5 ng/ml selenium). In higher concentration of BSA and ITS supplemented groups, the developmental rates over compacted morula were higher than others, but there was a significant effect of ITS only in 1 mg/ml of BSA added group (p〈0.05). In experiment 4, the effect of reduced concentration of NaCl was evaluated. The developmental rate over compacted morula in the medium containing 90 mM of NaCl was higher than in 114 mM group (p〈0.05). In conclusion, BSA could be used as a macromolecule source in bovine embryo culture, and ITS, as a serum substitute, could be used for improving of embryonic development. Also, reduction of NaCl concentration from 114 mM to 90 mM may improve the development of bovine embryos.bryos.

Effect of Feeding a Mixed Microbial Culture Fortified with Trace Minerals on the Performance and Carcass Characteristics of Late-fattening Hanwoo Steers: A Field Study

  • This study was conducted to determine the effects of feeding a trace minerals-fortified microbial culture (TMC) on the performance and carcass characteristics of late-fattening Hanwoo steers. A mixture of microbes (0.6% [v/w] of Enterobacter sp., Bacillus sp., Lactobacillus sp., and Saccharomyces sp.) was cultured with 99% feedstuff for ensiling and 0.4% trace minerals (zinc, selenium, copper, and cobalt). Sixteen late-fattening steers (mean age, 21.8 months) were allocated to two diets: a control diet (concentrate mix and rice straw) and a treated diet (control diet+3.3% TMC). At a mean age of 31.1 months, all the steers were slaughtered. The addition of TMC to the diet did not affect the average daily weight gain of the late fattening steers, compared with that of control steers. Moreover, consuming the TMC-supplemented diet did not affect cold carcass weight, yield traits such as back fat thickness, longissimus muscle area, yield index or yield grade, or quality traits such as meat color, fat color, texture, maturity, marbling score, or quality grade. However, consumption of a TMC-supplemented diet increased the concentrations of zinc, selenium, and sulfur (p<0.05) in the longissimus muscle. With respect to amino acids, animals consuming TMC showed increased (p<0.05) concentrations of lysine, leucine, and valine among essential amino acids and a decreased (p<0.05) concentration of proline among non-essential amino acids. In conclusion, the consumption of a TMC-supplemented diet during the late-fattening period elevated the concentrations of certain trace minerals and essential amino acids in the longissimus muscle, without any deleterious effects on performance and other carcass characteristics of Hanwoo steers.

Effect of Organic Selenium (Sel-Plex) on Thermometabolism, Blood Chemical Composition and Weight Gain in Holstein Suckling Calves

  • The objective of this study was to determine the effects of milk supplemented with Sel-Plex on thyroid hormones, rectal temperature, plasma glucose and cholesterol concentrations, and body weight in suckling calves. Ten Holstein suckling male calves of approximately 1 month of age were selected and randomly allocated to one of two groups and fed either unsupplemented milk (control) or milk supplemented with 0.3 mg/kg DM Se from Sel-Plex (treated). The animals received treated milk and free choice starter for two months. Dry matter intake for each individual animal was recorded daily throughout the study. The calves were weighed and blood samples were taken weekly. Plasma concentrations of triiodothyronine ($T_{3}$), thyroxin ($T_{4}$) and $T_{3}$-Uptake were determined using radioimmunoassay kits. Plasma cholesterol and glucose concentrations were determined by enzymatic-colorimetric methods. Rectal temperature was recorded at blood sampling time. During the experiment, ambient temperature and relative humidity were recorded at the time of blood sampling and every four hours. Treatment significantly (p<0.01) affected the concentration of $T_{3}$, free $T_{3}$ index ($FT_{3}I$) and ratio of $T_{3}$:$T_{4}$ in plasma. The plasma concentrations of $T_{3}$ in treated calves were 33% higher than in the control. The plasma concentrations of $T_{4}$ (p<0.05), glucose (p<0.01) and cholesterol (p<0.01) were decreased in the treated group. Calves fed Sel-Plex had higher rectal temperature (p<0.01). Plasma free $T_{4}$ index (FT4I) did not differ significantly between the two groups. An increase in body weight was observed (p<0.09) in the treated group. The results indicated that milk fortified with Sel-Plex could increase tissue conversion of $T_{4}$ to $T_{3}$ and therefore improve thermometabolism in suckling calves.

  • Chalcogenides (Te,Se) and pnictogens(Bi,Sb) materials have been widely investigated as thermoelectric materials. Especially, Bi2Te3 (Bismuth telluride) compound thermoelectric materials in thin film and nanowires are known to have the highest thermoelectric figure of merit ZT at room temperature. Currently, the thermoelectric material research is mostly driven in two directions: (1) enhancing the Seebeck coefficient, electrical conductivity using quantum confinement effects and (2) decreasing thermal conductivity using phonon scattering effect. Herein we demonstrated influence of annealing temperature on structural and thermoelectrical properties of Bismuth-telluride-selenide ternary compound thin film. Te-rich Bismuth-telluride-selenide ternary compound thin film prepared co-deposited by thermal evaporation techniques. After annealing treatment, co-deposited thin film was transformed amorphous phase to Bi2Te3-Bi2Te2Se1 polycrystalline thin film. In the experiment, to investigate the structural and thermoelectric characteristics of Bi2Te3-i2Te2Se1 films, we measured Rutherford Backscattering spectrometry (RBS), X-ray diffraction (XRD), Raman spectroscopy, Scanning eletron microscopy (SEM), Transmission electron microscopy (TEM), Seebeck coefficient measurement and Hall measurement. After annealing treatment, electrical conductivity and Seebeck coefficient was increased by defect states dominated by selenium vacant sites. These charged selenium vacancies behave as electron donors, resulting in carrier concentration was increased. Moreover, Thermal conductivity was significantly decreased because phonon scattering was enhanced through the grain boundary in Bi2Te3-Bi2Te2Se1 polycrystalline compound. As a result, The enhancement of thermoelectric figure-of-merit could be obtained by optimal annealing treatment.

  • SANGNYANGHYOLTANG(SYT) is one of the most important prescription that has been used in oriental medicine for diabetes mellitus. The sudy was done in order to elucidate the anti-diabetic effect of SYT. After pretreatment of SYT(1,000mg/kg) for 6 weeks, the effect of of SYT was prevented on serum liver function test and hepatic lipid peroxide content in rats i.v. injected with streptozotocin(STZ, 50mg/kg, tail vein) 5 weeks after pretreatment of SYT. The hepatic microsomal cytochrome P-450 and aniline hydroxylase were significantly decreased, and aminopyrine N-demethylase activity was significantly increased in SYT-STZ group as compared with control group. Changes in aldehyde oxidase, xanthine oxidase, superoxide dismutase, catalase, epoxide hydrolase, UDP-glucuronyltransferase and sulfotransferase activities were not significantly different in any of the group. The cytosolic glutathione S-transferase activity was significantly decreased in SYT-STZ group as compared with control group. The selenium-independent glutathione peroxidase was significantly increased in SYT-STZ group as compared with control group, but there was no significant difference in selenium-dependent glutathione peroxidase in any of the groups. The hepatic glutathione concentration was significantly increased in SYT-STZ group as compared with control group, and ${\gamma}-glutamylcystein$ synthetase and glutathione reductase activities were not significantly different in any of the groups. The hepatic lipid peroxide content, serum aminotransferase and sorbitol dehydrogenase activities were slightly decreased in significantly in SYT-STZ groups.

  • A total of 100 seafoods commonly consumed in Gyeonggi-do were investigated to determine the concentration of lead (Pb), total mercury (Hg), methyl mercury (MeHg), cadmium (Cd) and selenium (Se). Concentration of heavy metals and selenium was measured by using mercury analyzer, ICP-MS and GC-ECD. The average content (mg/kg) of heavy metals in the seafood samples was as follows; Pb 0.0915 (0.0021-0.4490), Cd 0.0084 (ND-0.1773), and Hg 0.0412 (0.0013-0.3032). All the levels were below the recommended standards of the MFDS in Pb (0.5 mg/kg), Cd (0.2 mg/kg), Hg (0.5 mg/kg). The methylmercury was detected in the hairtail (0.0677 mg/kg) and cod (0.2941 mg/kg). After the average content of heavy metals in seafood was determined, the exposure assessment for heavy metals was conducted. Relative hazardous levels compared to PTWI were lower than the official standards of the JECFA for Pb (0.97%), Hg (3.42%) Cd (0.45%). In conclusion, the levels presented in this study are presumed to be safe for consumption.
