Selenium의 첨가가 육계 및 산란계의 생산성 및 축적에 미치는 효과 (Effects of Dietary Selenium Sources on Performance and Selenium Retention in Broiler Chickens and Laying Hens)

  • 셀레늄의 첨가가 육계 및 산란계의 생산성과 축적에 미치는 영향을 구명하기 위하여 2차례의 실험을 수행하였다. 실험 1은 무기태 selenium 0.12, 0.24 ppm과 유기태 selenium 0.12, 0.24 및 0.6 ppm의 첨가가 육계의 사료 섭취량, 증체량, 사료 요구율 및 닭고기 내 selenium 축적에 미치는 영향을 조사하였다. 시험 전 기간의 증체량은 무기태 0.24 ppm 첨가구보다 유기태 selenium 0.24와 0.60 ppm 첨가구에서 높았다(P<0.05). 그러나 사료 섭취량과 사료 요구율은 대조구, 무기태 및 유기태 selenium의 첨가 수준에 의한 차이는 없었다. 가슴과 다리근육의 selenium 함량은 대조구에 비하여 유기태와 무기태 selenium 첨가구가 높았으며(P<0.05), 유기태 selenium의 첨가 수준에 따른 차이는 있으나(P<0.05), 무기태 selenium의 첨가 수준에 따른 차이는 없었다. 실험 2에서는 산란계 사료에 무기태 selenium 0.06, 0.12 ppm과 유기태 selenium 0.06, 0.12 및 0.30 ppm의 첨가하여 급여시에 산란율, 사료 섭취량, 사료 요구율, 난질 및 계란 내 selenium 함량에 미치는 영향을 조사하였다. 사료 요구율은 무기태 0.06 ppm 첨가구가 대조구보다 유의적으로 개선되었으나(P<0.05), 산란율, 난중, 1일 산란량, 사료 섭취량은 대조구, 무기태 및 유기태 selenium의 첨가 수준에 따른 차이는 없었다. Haugh unit는 모든 처리구에서 차이가 없었으며, 난황색은 유기태 selenium 0.30 ppm 첨가구가 대조구를 포함한 다른 처리구에 비하여 높았다(P<0.05). 난각 강도는 유기태 selenium 0.06 ppm 첨가구가 높았다.(P<0.05). 그러나 난각 두께는 대조구를 포함한 모든 처리구에서 차이는 없었다. 계란 중의 selenium 함량은 대조구에 비하여 selenium을 첨가한 모든 처리구에서 높았다(P<0.05). 이외에도 사료내 selenium 첨가 수준에 비례하여 계란에서 selenium 축적량도 증가하였으며(P<0.05), 특히 유기태 selenium 이 무기태 selenium보다 효과적이었다.

The Effect of Dietary Selenium Source and Vitamin E Levels on Performance of Male Broilers

  • Selenium and vitamin E are micronutrients essential for normal health and maintenance in poultry. They are necessary in preventing free radical damage to phospholipid membranes, enzymes and other important molecules. Two experiments were conducted in a semi-commercial environment to examine the effect of Se source and vitamin E level in diet on broiler performance and meat quality. Increasing vitamin E from 50 IU to 100 IU did not affect growth performance of broilers although the 24 h drip-loss was tended to be reduced (p=0.06). There was an interaction between vitamin E and the source of Se in glutathione peroxidase activity (GSH-Px) and Se concentration in excreta. Increasing vitamin E from 50 IU to 100 IU elevated GSH-Px and Se concentration in excreta by 42 IU/g Hb and 0.9 ppm for the organic Se group, respectively, but reduced GSH-Px and Se concentration in excreta by 16 IU/g Hb and 1.3 ppm for inorganic group, respectively. Vitamin E played no role in the feather coverage of the birds when scored on day 37. Organic Se is more effective in improving feather score and 24 h drip-loss, with a markedly higher deposition rate in breast muscle and a lower excretion rate in the excreta (p<0.05) compared to the inorganic Se source. Both vitamin E and the source of Se did not affect (p>0.05) the energy utilisation by birds.

The Effect of Pulse Electric Field on Accumulation of Selenium in Cells of Saccharomyces cerevisiae

  • Cultures of Saccharomyces cerevisiae were subjected to the effect of PEF (pulse electric field) and a source of selenium. The culture period after which yeast cells were subjected to PEF treatment was optimized, as was the duration of the exposure. Optimization of the nutrient medium composition in S. cerevisiae cultures resulted in an over 1.8-fold increase in selenium accumulation with relation to cultures on the initial substrate. Optimization of the pH value and of culture duration resulted in selenium accumulation increase by approximately 78%. A significant correlation was found between the accumulation of selenium in yeast cells and its concentration in the culture substrate. The highest accumulation of selenium in the biomass of yeast, approx. $240\;{\mu}g/g$ d.m., was obtained after 15-min exposure to PEF on a 20-h culture. An approx. 50% higher content of selenium in cells was recorded, as compared with the control culture without the application of PEF.

Effect of supplementation and withdrawal of selenium-enriched kale sprouts on productivity and egg selenium concentration of laying hens

  • Objective: The aim of this trial was to investigate the effect of supplementation and withdrawal of selenium-enriched kale sprouts (SeKS) on productivity and egg Se concentration of laying hens. Selenium from commercial Se-enriched yeast (SeY) was used as a comparative Se source. Methods: One-hundred and eighty 61-week-old laying hens were randomly divided into 5 treatment groups with 4 replicates (9 hens each) in a 2×2+1 Augmented Factorial Experiment in a completely randomized design. The experimental diets were basal diet, basal diet supplemented with 0.2 and 0.4 mg Se/kg from SeKS and SeY, respectively. The 8-week feeding trial was divided into 2 periods, namely the Se supplemental period (week 1 to 4) and the Se withdrawal period (week 5 to 8). Results: Productive performance, egg quality and egg Se concentration of laying hens were not affected by sources of Se (SeKS and SeY) during both, the Se supplemental and withdrawal periods. Egg production and egg Se concentration increased (p<0.05) with increasing levels of Se supplementation. The egg Se concentration increased and reached a peak 1 week after Se supplementation. However, concentration of Se in eggs of hens fed Se from both sources decreased rapidly from the second week of the Se withdrawal period to reach the same egg Se concentration of hens fed the basal diet by the fourth week of the Se withdrawal period. Conclusion: The efficacy of Se from SeKS on productivity and egg Se concentration in laying hens was comparable to commercial SeY. Thus, SeKS can provide an alternate organic Se source for production of Se-enriched eggs.

  • Selenium is an antioxidant trace element linked to cardiovascular disease and cancer. Although diet is a major source, relatively little else is known about independent determinants of selenium levels in free-living humans. In this study, we aimed to investigate the independent demographic. lifestyle, and dietary determinants of selenium levels in 1,997 men and 1,905 women in two large prospective U.S. cohorts. Toenail selenium levels were quantified using neutron activation analysis. Diet, geographic residence, demographic, and environmental factors were assessed by validated self-administered questionnaires. Multivariate generalized linear models were conducted to assess the independent relations of these factors with toenail selenium levels, correcting for measurement error in the diet. In multi variable-adjusted analyses, independent predictors of higher selenium were male gender (6.3% higher levels); living in West and Northern-Midwest U.S. regions (8.9% and 7.4% higher than Southern-Midwest regions, respectively); consumption of beef and bread products (between 0.7 - 2.5% higher per daily serving); and selenium supplement use (6.9% higher than non-users); whereas cigarette smoking (5-10% lower than never smokers), older age (0.6% lower per 5 years), and consumption of eggs, white rice, dairy products, coffee, and alcohol (between 0.1 to 2.0% lower per daily serving) were associated with lower selenium. Multiple dietary and non-dietary factors independently predicted selenium levels, suggesting that both consumption and non-dietary processes (e.g.. related to oxidant status) may affect levels. Significant geographic variation in selenium levels exists in the US.

  • A novel technology has been developed for the synthesis of thioglycolic acid (TGA)-capped CdSe quantum dots (QDs) in aqueous medium. The reaction was carried out in air atmosphere with one-pot by using $SeO_2$ to replace Se or $Na_2Se$. The technological parameters including refluxing time, pH values and molar ratios of selenium to cadmium had significant influence on the luminescence properties of CdSe QDs. Furthermore, the obtained QDs were characterized by fluorescent spectroscopy, X-ray powder diffraction (XRD) and transmission electron microscopy (TEM), respectively. The results demonstrated that the CdSe QDs were of zinc-blended crystal structure in a sphere-like shape.

Effects of Dietary Selenium Supplementation on Growth Performance, Selenium Retention in Tissues and Nutrient Digestibility in Growing-finishing Pigs

  • This experiment was conducted to investigate the effects of selenium (Se) sources and levels on growth performance, nutrient digestibility and Se retention in growing-finishing pigs. A total of 56 crossbred pigs ([$Landrace{\times}Yorkshire$]${\times}$Large White) with average $28.5{\pm}0.2kg$ BW were allotted to 7 treatments on the basis of sex and weight in two replicates and four pigs per pen. A $2{\times}3$ factorial arrangement of treatments was used in a randomized complete block (RCB) design. Two sources of Se (selenite Se or Se-enriched yeast) were added at 0.1, 0.3 and 0.5 mg/kg to each treatment diet. A basal diet without Se supplementation was the seventh treatment group. Three pigs per treatment were randomly selected and samples of loin, liver, pancreas and a kidney were collected, frozen and later analyzed for Se. The digestibility trial was conducted to evaluate the apparent absorption and retention of Se and availability of other nutrients. Growth performance was not affected by dietary sources and levels of Se. No growth retardation was observed in the 0.5 mg/kg dietary Se treatment group regardless of Se sources. The Se concentration of serum in Se supplemented groups was increased compared with the control group (p<0.01). During the growing and finishing phase, Se in serum was clearly increased when organic Se was provided (p<0.01). Interaction of Se source ${\times}$ Se level was observed in Se concentration of loin, liver and pancreas of the pigs at the end of experiment. Selenium retention in the liver, kidney, pancreas and loin of pigs was increased as dietary Se level increased and was higher when pigs were fed organic Se resulting in an interaction response (p<0.01). Nutrient digestibilities were not affected by dietary Se sources or levels. No dietary Se source ${\times}$ Se level interaction was observed in nutrient digestibility. The results from this experiment indicated that dietary Se sources and levels affected the distribution of Se in the body of growing-finishing pigs. Organic source of Se, such as Se-enriched yeast resulted in higher serum and tissue Se concentration compared to inorganic form, while no beneficial effects on nutrient digestibility were observed from dietary Se supplementation in growing-finishing pigs.

Short-term effects of dietary selenium on lactating sows to improve litter performance, milk composition and tissue selenium retention in piglets

  • Objective: This study was conducted to evaluate the short-term effects of dietary selenium supplementation on lactating sows on the physiological response, litter performance, milk composition, and tissue selenium retention in piglets when selenium was provided by different sources and at different levels in a lactation diet. Methods: A total of 48 multiparous sows (Yorkshire×Landrace) with average body weight, backfat thickness, and parity were assigned to one of the four treatments with 12 sows per treatment using a 2×2 factorial arrangement in a completely randomized design. Inorganic or organic Se sources were added to the diet at 0.30 ppm and 0.50 ppm Se. Treatments were as follows: i) IS30, basal diet + inorganic Se 0.30 ppm; ii) IS50, basal diet + inorganic Se 0.50 ppm; iii) OS30, basal diet + organic Se 0.30 ppm; and iv) OS50: basal diet + organic Se 0.50 ppm. Results: At Day 21 of lactation, a high tendency of litter weight (p = 0.08) and litter weight gain (p = 0.09) were observed when sows were fed an organic Se source. The milk Se concentration in the organic Se treatment was higher than that in the inorganic Se treatment at Day 21 of lactation (p<0.05). The serum Se concentrations of sows and piglets at Day 21 of lactation were significantly higher when lactating sows were fed organic Se instead of inorganic Se (p<0.01). During the suckling period, the kidney and muscle Se concentrations of piglets at Day 21 of lactation were significantly higher when the sow dietary Se source was organic (p<0.05). Liver Se concentrations were affected by Se source and level (p<0.05). This also resulted in an interaction response at 21 days of lactation (p<0.05). Conclusion: The supplementation of dietary organic Se in a lactating diet could improve sow feed consumption, piglet performance, milk Se level, and the Se status of sows and piglets.

Nano-Selenium Microcapsule의 항산화에 관한 연구 (Study on the Antioxidant Effects of Nano-Selenium Microcapsule)

  • 항산화 기능이 있는 것으로 알려진 셀레늄의 약물전달 효율을 높이기 위하여 마이크로 캡슐 피막제로 키토산(0.1~0.9%)을 사용하여 chitosan nano-selenium을 제조하여 chitosan nan-oselenium의 모양과 형태는 주사전자현미경(SEM)을 이용하여 관찰하였고, 입자의 크기는 ELS-8000으로 측정하였으며, chitosan nano-selenium의 항산화 효능과 세포독성을 조사하였다. Chitosan nano-selenium의 모양과 형태를 SEM(JSM-5400, Japan)으로 관찰한 결과 구형을 이루고 있어 마이크로 캡슐을 형성한 것을 알 수가 있었고, chitosan nano-selenium의 입자의 크기를 ELS-8000으로 측정한 결과, 입자 크기는 대략 50~500 nm로 분포되었으며, chitosan nano-selenium은 chitosan 농도가 높을수록 입자의 크기도 커지는 것을 알 수가 있었다. 입자 크기는 0.1% chitosan nano-selenium은 $55{\pm}5$ nm가 많이 분포가 되어 있고, 0.3%는 $60{\pm}5$ nm가 많이 분포되어 있고, 0.5%는 $70{\pm}5$ nm, 0.7%는 $100{\pm}5$ nm, 0.9%는 $450{\pm}5$ nm가 많이 분포되어 있는 것을 확인하였다. 0.1% chitosan nano selenium 전자공여능은 50 ${\mu}g/m{\ell}$ 농도에서 0.1%, 0.3%, 0.5%, 0.7% 및 0.9% chitosan nano selenium은 90.28.0%, 90.36%, 89.34%, 89.33% 및 89.49%로 나타나 항산화효능의 높은 값을 보였으나, chitosan 함량에 따른 항산화 효능의 차이는 볼 수 없었다. Chitosan nano-elenium은 0.1%, 0.3%, 0.9% chitosan nano-selenium에서의 독성이 강하게 나타나지 않았으며, 0.7% chitosan nano-selenium에서는 50 ${\mu}g/m{\ell}$ 농도에서 61.60% 생존율을 보여 약간의 독성이 있었지만 200 ${\mu}g/m{\ell}$에서 85.44%로 생존율이 증가하였고, 0.9% chitosan nano-selenium에서 생존율이 90.08~98.71%로 가장 높은 값을 보였다.
