  • The present study was performed to explore the antioxidant effect of selenium on damaged brain induced by organic mercury. Male ICR mice were given consecutively 7 injections for 7 days of : (I) sodium selenite 1 mg/kg s.c. alone, (2) methylmercuric chloride 10mg/kg s.c. alone, (3) methylmercuric chloride simultaneously in combination with sodium selenite, and (4) saline alone as control respectively. Based on the above protocol, we monitored various oxyradical scavenging system as well as the finding of electron microscopy. The results from brain tissues were as follows` 1. Selenium inhibited the rate of generation of superoxide radical.

  • Investigations of the prevalence of SalmoneIla organisms in chicken of Suwon area and the effect of selenite broth on the growth of Sal. pullorum were made. The results obtained were summarized as foIlowings. 1. Salmonella organisms was not isolated from fecal samples of 357 chicken from 6 poultry farms of Suwon area. 2. In selenite broth, the growth of Sal. pullorum was completely inhibited may due to the toxic effect of the medium when the inoculum contained less than $5{\times}10^3$ organisms. However, enrichment was attained overcoming the inhibitory action of the medium when $5{\times}10^3$ or more organisms were inoculated.

  • Autometallography method with intraperitoneal injedlon of sodium selenite was employed to investigate the localization of somata of zinc enriched (ZEN) neurons in the medulla oblongata The distribution patterns of the labeled neurons were variable from rostral to caudal regions. The labeled cells by the method were found in Cl adrenaline cells, gigantocellular reticular nucleus, inferior olive, paragigantocellular nucleus, prepositus hypoglossal nucleus, raphe obscurus nucleus, and reticular nucleus regions.

  • The protective of influences of sodium selenitc ($Na_2SeO_3$) against the methemoglobinemia with sodium nitrite were investigated on hemoglobin, methemoglobin, glutathione peroxidase and NADH-methemoglobin reductase activity in rabbits which were given 0,1,3 and 9ppm sodium selenite of drinking water for a week. Dietary selenium did not alter total hemoglobin in the blood of rabbits. Selenium was found to decrease nitrite-induced methemoglobin in a dose-dependent manner. The glutathione peroxidase activity was also increased by selenium in all the experimental groups. However, the NADH-methemoglobin reductase activity by selenite did not show significant differences as concerns the methemoglobinemia. These results showed that selenium could inhibit nitrite-induced methemoglobinemia. Its influence of inhibition is suggested that the effect of the reduction of methemoglobin was greatly stimulated by glutathione peroxidase activity.

  • The objective of this study was to determine the effect of Se-enriched bean sprout, Se-enriched yeast and sodium selenite on productivity, egg quality and egg Se concentrations in laying hens. Using a Completely Randomized Design, 144 Rohman laying hens at 71 weeks of age were divided into four groups. Each group consisted of four replicates and each replicate contained nine hens. The dietary treatments were T1: control diet, T2: control diet plus 0.3 mg Se/kg from sodium selenite, T3: control diet plus 0.3 mg Se/kg from Se-enriched yeast, T4: control diet plus 0.3 mg Se/kg from Se-enriched bean sprout. The results showed that there was no significant difference (p>0.05) in feed intake, egg production and egg quality among treatments. Selenium supplementation from Seenriched yeast and Se-enriched bean sprout markedly increased (p<0.05) egg Se concentration as compared to the control and sodium selenite groups. The results indicated that Se-enriched bean sprout could be used as an alternative Se source in diets of laying hens.

  • Removal of dissolved selenium by D. michiganensis, a iron-reducing bacterium, and effects of dissolved metal elements such as iron, sulfate, and copper were investigated. Selenide that was reduced from selenite (2 mM) by D. michiganensis was gradually removed from the aqueous medium. As the reduced selenide was combined with aqueous iron, it was precipitated as a nanoparticulate iron-selenide. Sulfate and copper negatively affected the microbial selenite reduction, and the copper was especially toxic to the bacterium, inhibiting a microbial removal of dissolved selenite. These results show that it should be carefully biotreated for a selenium-contaminated site considering in situ sulfate or copper distribution and concentration. Consequently, the formation of iron-selenide by bacteria will be an important measure for preventing a long-distance migration of selenium in the subsurface environments.

  • Much interest has been recently focused on the production of large quantities of hydrogen, due to its potential importance in our economy and needs in the petroleum and chemical industries. Formate dehydrogenase H (FDH-H) from Escherichia coli containing selenocysteine that oxidizes formate to carbon dioxide with the release of hydrogen is a component of the anaerobic formate hydrogen-lyase complex of E. coli. To make full use of FDH-H, we need effective expression condition. In this approach, we investigated the effect of pH on FDH-H stability and observed the effect of selenite and formate concentration on the activity of FDH-H. Additionally, coexpression of selenocysteine insertion genes were tried to improve the expression of FDH-H. The highest level of FDH-H expression was achieved by coexpression of selenocysteine insertion genes (pSUABC) as well as by the addition of $10\;{\mu}M$ selenite and 10 mM formate. At this optimized condition, a 2.6 fold elevation of expression of FDH-H was achieved.
