Placenta specific 8 (PLAC8, also known as ONZIN) is a multi-functional protein that is highly expressed in the intestine, lung, spleen, and innate immune cells, and is involved in various diseases, including cancers, obesity, and innate immune deficiency. Here, we generated a Plac8 knockout mouse using the CRISPR/Cas9 system. The Cas9 mRNA and two single guide RNAs targeting a region near the translation start codon at Plac8 exon 2 were microinjected into mouse zygotes. This successfully eliminated the conventional translation start site, as confirmed by Sanger sequencing and PCR genotyping analysis. Unlike the previous Plac8 deficient models displaying increased adipose tissue and body weights, our male Plac8 knockout mice showed rather lower body weight than sex-matched littermate controls, though the only difference between these two mouse models is genetic context. Differently from the previously constructed embryonic stem cell-derived Plac8 knockout mouse that contains a neomycin resistance cassette, this knockout mouse model is free from a negative selection marker or other external insertions, which will be useful in future studies aimed at elucidating the multi-functional and physiological roles of PLAC8 in various diseases, without interference from exogenous foreign DNA.

Downy mildew (DM), caused by several species in the Peronosclerospora and Scleropthora genera, is a major maize (Zea mays L.) disease in tropical or subtropical regions. DM is an obligate parasite species in the higher plants and spreads by oospores, wind, and mycelium in seed surface, soil, and living hosts. Owing to its geographical distribution and destructive yield reduction, DM is one of the most severe maize diseases among the maize pathogens. Positional cloning in combination with phenotyping is a general approach to identify disease resistant gene candidates in plants; however, it requires several time-consuming steps including population or fine mapping. Therefore, in the present study, we suggest a new combination strategy to improve the identification of disease resistant gene candidates. Downy mildew (DM) resistant maize was selected from five cultivars using the spreader row technique. Positional cloning and bioinformatics tools identified the DM resistant QTL marker (bnlg1702) and 47 protein coding genes annotations. Eventually, 5 DM resistant gene candidates, including bZIP34, Bak1, and Ppr, were identified by quantitative RT-PCR without fine mapping of the bnlg1702 locus. Specifically, we provided DM resistant gene candidates with our new strategy, including field selection by the spreader row technique without population preparation, the DM resistance region identification by positional cloning using bioinformatics tools, and expression level profiling by quantitative RT-PCR without fine mapping. As whole genome information is available for other crops, we propose applying our novel protocol to other crops or for other diseases with suitable adjustment.

The advance of technologies for human genome makes it possible that the analysis of association between genetic variants and diseases and the application of the results to predict risk or susceptibility to them. Many of those studies carried out in case-control study. For quantitative traits, statistical analysis methods are applied to find single nucleotide polymorphisms (SNP) relevant to the diseases and consider them one by one. In this study, we presented methods to select informative single nucleotide polymorphisms and predict risk for quantitative traits and compared their performance. We adopted two SNP selection methods: one considering single SNP only and the other of all possible pairs of SNPs.
The purpose of this study was to investigate eco-friendly measures to manage major diseases which cause heavy economic damages to ginseng. Morphological, physicochemical, and molecular biological species identification was carried out after isolating useful antagonistic bacteria from ginseng fields. In addition, optimal conditions for mass culture were established, and he efficacy of the bacteria in the prevention of the diseases was verified in the field. The results showed that about 150 bacteria were extracted from 150 ginseng fields in the whole county. Among them, B. pyrrocinia LA101 was finally selected, which had a strong antagonistic potency against Alternaria panax, Botrytis cinerea, Rhizoctonia solani, and Cylindrocarpon destructans on agar media. The B. pyrrocinia LA101 is a baculiform gram-negative bacterium identified as Burkholderia pyrrocinia according to results from an API(Analytical Profile Index) kit, 16S rRNA, and gyrase gene sequencing analysis. It was donated to the microbe bank of the Agricultural Genetic Resources Center at the National Academy of Agriculture Science under the Rural Development Administration on September 28, 2011 (Donation No. KACC91663P). A patent for the mass culture technology was granted in August 2012 (Patent No. 10-1175532).
After the outbreak of COVID-19, local infections were spread by foreign sailors of foreign ships entering the ports. However, the government had difficulties in designating temporary living facilities around port areas due to the opposition from local residents. Therefore, the purpose of this study was to plan a floating temporary living facility for foreign sailors in the port area to prevent the spread of COVID-19 by foreign sailors. In this study, location selection, a facility volumetric plan, a unit facility plan, and a unit facility block plan are presented as results of facility planning. Floating temporary living facilities are expected to be used to prevent the spread of first-class infectious diseases in port areas in the future.
The elevator installed in the building consists of an elevator call button and an input button for selection to the target floor. The elevator button is input only when the elevator user directly presses it. Such passenger input can be infected with an infectious disease due to contamination of the button. A non-contact button is required as a means for solving this problem, which detects the proximity of an object by applying a capacitive method. It implements a function of measuring the body's body temperature by attaching an infrared heat sensor, and provides a sterilization function of a button by attaching a UV-LED. A button was selected, a body temperature was measured through an infrared temperature measurement sensor, and UV-LED was turned on and sterilized when there was no user. The contactless elevator button is expected to be effective in preventing infectious diseases as it can prevent infection of viruses carrying infectious diseases and can detect body temperature to select positive patients of CIVID 19.

In 2000, $F_3$ and $F_4$ lines or populations developed from the respective $F_2$ or $F_3$ selections in the previous year were evaluated for resistance to both diseases. A significant progress in resistance to both diseases was achieved by selection.

Chronic diseases such as hypertension require a differentiated approach according to age and life cycle. Chronic diseases such as hypertension require differentiated management according to the life cycle. It is also known that the cause of hypertension is a combination of various factors. This study uses machine learning prediction techniques to analyze various factors affecting hypertension by life cycle. To this end, a total of 35 variables were used through preprocessing and variable selection processes for the National Health and Nutrition Survey data of the Korea Centers for Disease Control and Prevention. As a result of the study, among the tree-based machine learning models, XGBoost was found to have high predictive performance in both middle and old age. Looking at the risk factors for hypertension by life cycle, individual characteristic factors, genetic factors, and nutritional intake factors were found to be risk factors for hypertension in the middle age, and nutritional intake factors, dietary factors, and lifestyle factors were derived as risk factors for hypertension. The results of this study are expected to be used as basic data useful for hypertension management by life cycle.

Background: Tsutsugamushi, leptospirosis and hemorrhagic fever with renal syndrome (HFRS) are the prevalent diseases among the acute febrile illnesses in Korea. Pulmonary involvement in the patients with these diseases remains poorly recognized in endemic regions, and this is despite reports of recent outbreaks and epidemic episodes. Pulmonary involvement and a higher CRP level as clinical manifestations show a more severe form of infection. The aim of this study is to analyze the correlation of pulmonary involvement and the CRP level in patients with acute febrile illnesses. Methods: We retrospectively reviewed the clinical records of 105 patients who were diagnosed with tsutsugamushi, HFRS and leptospirosis from January 2002 to May 2008 in Chuncheon Sacred Heart Hospital. The radiographic images were retrospectively analyzed by two radiologists. We analyzed the pulmonary complications of the patients with these febrile diseases and we checked the CRP level at admission. Results: The study included 105 patients who were diagnosed with febrile diseases. Of these patients, 32 patients had hantaan, 10 patients had leptospirosis and 63 patients had tsutsugamushi disease. 42 (40%) patients had pulmonary complications, 20 patients had pulmonary edema, 20 patients had pleural effusion and 2 patients had interstitial pneumonitis. The patients with pulmonary involvement showed a more severe form of infection and a higher CRP level than that of those patients without pulmonary involvement (p=0.0073). Conclusion: Pulmonary involvement in patients with acute febrile diseases might be correlated with a higher CRP level. Identification of this factor on admission might provide useful selection criteria for the patients who need early intensive care.
Objective: Porcine respiratory disease is one of the most important health problems causing significant economic losses. To understand the genetic basis for susceptibility to swine enzootic pneumonia (EP) in pigs, we detected 102,809 single nucleotide polymorphisms in a total of 249 individuals based on genome-wide sequencing data. Methods: Genome comparison of susceptibility to swine EP in three pig breeds (Jinhua, Erhualian, and Meishan) with two western lines that are considered more resistant (Duroc and Landrace) using cross-population extended haplotype homozygosity and F-statistic (FST) statistical approaches identified 691 positively selected genes. Based on quantitative trait loci, gene ontology terms and literature search, we selected 14 candidate genes that have convincible biological functions associated with swine EP or human asthma. Results: Most of these genes were tested by several methods including transcription analysis and candidate genes association study. Among these genes: cytochrome P450 1A1 and catenin beta 1 (CTNNB1) are involved in fertility; transforming growth factor beta receptor 3 plays a role in meat quality traits; Wnt family member 2, CTNNB1 and transcription factor 7 take part in adipogenesis and fat deposition simultaneously; plasminogen activator, urokinase receptor (completely linked to AXL receptor tyrosine kinase, r2 = 1) plays an essential role in the successful ovulation of matured oocytes in pigs; colipase like 2 (strongly linked to SAM pointed domain containing ETS transcription factor, r2 = 0.848) is involved in male fertility. Conclusion: These adverse genes susceptible to swine EP may be selected while selecting for economic traits (especially reproduction traits) due to pleiotropic and hitchhiking effect of linked genes. Our study provided a completely new point of view to understand the genetic basis for susceptibility or resistance to swine EP in pigs thereby, provides insight for designing sustainable breed selection programs. Finally, the candidate genes are crucial due to their potential roles in respiratory diseases in a large number of species, including human.
