Total Sugars, $\alpha$-amylase Activity, and Germination after Priming of Normal and Aged Rice Seeds

  • Osmoconditioning and hardening effects for the seed germination of normal and naturally aged rice seeds were studied through analyzing the total sugars and $\alpha$-amylase activity. The normal seeds which used to have high germination rate accelerated germination with the osmoconditioning at the suboptimal temperature of 17$^{\circ}C$. On the other hand, the aged seeds did not affect germination rate at $25^{\circ}C$, while increased germination rate and accelerated germination a little at 17$^{\circ}C$, Hardening of aged seeds increased germination rate by 10-15% compared with control seeds at both 17 and $25^{\circ}C$ and accelerated germination. Total sugar content and $\alpha$-amylase activity of normal seeds were higher than did aged seeds. The aged seeds with treatment of osmoconditioning and hardening increased total sugar content and $\alpha$--amylase activity, but hardening was more effective than osmoconditioning. The $\alpha$--amylase activity was positively correlated with the total sugar content and germination rate.

Effects of Microwaves on the Germination of Weed Seeds

  • Purpose: Weeds cause significant losses in agricultural production. In this study, we investigated the effect of microwaves on the germination rates of cress and arugula seeds to determine whether microwaves could be developed as an effective alternative to conventional chemical-based herbicides. Methods: Seeds were planted at equal depths (8-10 mm) in a soil-turf mixture, and seeds were exposed to microwaves for 126 s, 70 s, and 50 s in a device constructed specifically for this study. A microwave tunnel was built using a variable speed conveyor belt and 4 magnetrons with a combined output power of 2.8 kW. Seeds that were not exposed to microwaves were germinated with regulated irrigation, temperature, and humidity controls in parallel with the treated seeds, and the germination rates were compared among the treatment groups. Results: We found that the exposure of cress and arugula seeds to microwaves for 126 s inhibited germination close to 100%. Cress seeds treated with microwaves for 50 s showed 95% germination compared to 65% germination of the untreated cress seeds. Conclusions: We predict that the thermal effect of microwave energy affects the germination ratio and germination rate of seeds.

Changes of Germination Rate of Rice Seeds with Variable Status after 10-years of Storage Period

  • The seeds of rice with variable status were examined the germination rate after 10 years of long-term storage (-18℃) conservation. For seeds of wild relatives, 2 accessions were examined and germination rate of all accessions showed no changed after freezing conservation. For seeds of native germplasm, 1,259 accessions were examined and germination rate of 696 accessions was increased or showed no change after 10 years of long-term storage. Germination rate of 432 accessions was decreased with below 15% of initial germination rate. For 79 accessions of native germplasm, germination rate of seeds was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. For seeds of developed varieties, 873 accessions were examined and germination rate of 486 accessions of developed varieties was increased or showed no change after 10 years of long-term storage. Germination rate of 251 accessions was decreased with below 15% of initial germination rate. For 25 accessions of developed varieties, germination rate of seeds was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. For seeds of developed line, 2,131 accessions were examined and germination rate of 1,245 accessions of developed line was increased or showed no change after 10 years of long-term storage. Germination rate of 588 accessions was decreased with below 15% of initial germination rate. For 96 accessions, germination rate of seeds was decreased with above 15% of initial germination rate, which is needed to be rejuvenated.

Studies on the Improvement and Management of Hill Pasture (산지초지 개량과 관리에 관한 연구)

  • To improve germination and growth of forages of hill pasture seeds of several forages were osmotically primed with polyethylene glycol (PEG) under different PEG concentrations, treatment periods, and temperatures. Seeds were coated with several materials, and tested for germination. The best primed or coated seeds in germination tests were surface sow on hilly area. The results obtained are summarized as follows; 1. Osmotic priming with PEG accelerated germination of forages and reduced time taken for germination. 2. Germinability was best when the seeds of Tama ryegrass or orchardgrass were treated for 6-9 days at 30g PEG1100 ml water, red clover or white clover for 3, or 9 days in 20g PEG1100 ml water at $10^{\circ}C$. 3. Priming seeds of Tama ryegrass or orchardgrass in 30% PEG solution for 6 days, and seeds of red clover or white clover in 20% PEG solution for 6 days at $15^{\circ}C$ were most effective in germination, similar to priming at $10^{\circ}C$ . 4. Priming seeds of Tama ryegrass at 15C, and orchardgrass, red clover, or white clover at $10^{\circ}C$ were effective in germination than priming at other temperatures. 5. Osmotic priming with PEG accelerated germination of forages compared to coated seeds in Petri-dishes, while coated seeds germinated more slowly, but showed better emergence and superior growth to those of primed or intact seeds in the field. 6. Priming seeds increased yield slightly, and coating seeds significantly increased its yield on hilly area.

Germination of West African Ebony (Diospyros mespiliformis Hochst) Seeds: Effects of Dehydration and Different Pre-sowing Treatments

  • Diospyros mespiliformis is a highly valued and threatened tree species within the Sahelo-Sudanian zone of Africa, but its seed germination requirements under cultivation are not well researched. In a first experiment which aimed at determining germination response of seeds to dehydration, fresh seeds were dried at room temperature for 26 days during which their moisture content, their germinability, and their viability were monitored at two-day intervals. In the second experiment, 14 pre-germination treatments were tested for their effect on the germination of dried seeds. Results showed that fresh seeds had 52.7% moisture and achieved 97.7% germination. As seeds were dried, percentage germination gradually decreased with decreasing moisture content and reached 0% when moisture content had dropped to 18%. Meanwhile, seed viability remained at 100% over drying duration. Seeds that were not germinated after air dry also recorded 100% viability. The most effective treatment for inducing germination of dried seeds was scarification using 98% sulfuric acid for 30 min which resulted in 96.6% germination. This study reports for the first time in D. mespiliformis seeds a desiccation-induced dormancy which can be efficiently alleviated by acid scarification. This study provides useful information that will contribute to efficient management of D. mespiliformis seed resources for propagation.

Priming Effects on Germination of Aged Tobacco Seeds

  • Tobacco (Nicotiana tabacum L. cv KF109) seeds were artificially aged in a controlled environment of 45$^{\circ}C$ and 80% relative humidity condition for different duration up to 14 days before priming in polyethylene glycol 6000 solution of equivalent osmotic potential of -0.8 MPa for 8 days at $25^{\circ}C$. The seeds aged only and primed after aging were germinated at 15$^{\circ}C$ and $25^{\circ}C$ to observe the priming effects on the germination of aged seeds at different temperature. The germination percentage of the aged seeds was rapidly dropped starting from 8 days of aging and mean germination time (T$_{50}$) was greatly increased, particularly in germination at 15$^{\circ}C$. The germination capacity was greatly restored in the primed seeds after aging, particularly in the seeds of longer aging and germinating at 15$^{\circ}C$.>.

Effects of Provenances, Storage Temperature and Duration on Seed Germination of Bombax costatum Pellegr & Vuillet

  • Seed morphology, physiology and environmental conditions have influence on germination of any tropical seeds and their appropriate handling, processing and handling enhances seeds emergent. This study therefore investigated effects of storage durations under different temperatures on germination of Bombax costatum seeds from different provenances. Fresh 25 seeds of B. costatum in four replicates were collected from four different provenances (Aponmu, Oluwa, Ibadan and Oyo). They were surface sterilized, thoroughly washed in distilled water and stored at four different temperature regimes: 28℃, 21℃, 5℃ and -17℃. Samples were taken every 2 months for germination test for 18 months. Germination assessment was carried out daily and recorded. The experimental design was 4×4×10 factorial in 4 replicates. Data were subjected to percentages and analysis of variance (ANOVA). Results showed that there were significant differences among storage temperatures, storage durations and provenances on germination percentage. Freshly collected seeds from Aponmu and Oyo had the highest mean germination of 100% followed by seeds from Ibadan (89.3%). Seeds from Oyo provenance stored 5℃ had germination of 94.0%, 88.70% and 78.7% at 2nd, 4th and 6th month respectively. Seeds from Ibadan and Oyo stored at 28℃, 21℃ recorded 0.0% starting from 8 months of storage. Germination of B. costatum seeds from the four provenances decreased with increase in storage duration at different temperatures.

Effect of Seed Leachates of 'Vernal' Alfalfa on Inhibition of Alfalfa Germination and Root Growth

  • Most parts of alfalfa plant have been reported to contain autotoxic substances that inhibit seed germination and early seedling growth, however, the chemical(s) is not still studied much. Effect of seed leachates of 'Vernal' alfalfa (Medicago sativa L.) was evaluated for inhibition of alfalfa germination and root growth through bioassay. Alfalfa seeds were extracted in 1 L deionized water for 1 h after soaking and the leachates caused to reduce root length of alfalfa significantly as the soaking time increased. Crude seeds at 4 g L$L^{-1}$ exudated autotoxins that reduce significantly root length by 34 % compared to the control, when the seeds soaked in deionized water for 24 h. However, the extracts did not affect final germination as well as speed of germination. Extracts from ground seeds significantly reduced speed of germination (GT 50) and root length. The results indicate that release of autotoxic substances from seeds during seed imbibition was increased with increase of soaking time and seed amount, and that autotoxicity was more occurred in ground seeds than in crude seeds.

Effect of Seed Treatment on Improved Germinability of Agastache rugosa O. Kuntze (배초향 종자처리가 발아성 향상에 미치는 영향)

  • This study was carried out to establish optimal conditions for breaking dormancy of Agastache rugosa O. Kuntze seeds. A series of experiments according to seed maturity and treatment with plant growth regulators were performed to improve germination percentage and synchronize germination of the seeds. In addition, it was conducted to test whether the useful effect of seed treatment before sowing leads to healthy seedling and early vigorous growth. The average seed size was 1.85 mm (length) × 0.82 mm(width). The seed size was much smaller than other vegetable seeds. Seeds colorappeared dark brown, the shape of the seeds was oval and the weight of 1,000seeds was 352.8 mg. The optimum germination temperature was 22℃. Light exposure during germination did not affect germination promotion, suggesting that A. rugosa seeds are a kind of dark germinating seeds. Seed dormancy lasted for 40 days after harvesting, and GA3 treatment of dormant seeds could break dormancy. There were significant differences in germination percentage and rate according to the maturity of seeds. The germination percentage of mature seeds was 10 - 18% higher than that of immature seeds, and germination rate was 2 days faster. GA3 treatment during growth regulator treatment improved germinability, but BAP or ethephone treatment did not. The optimal growth regulator concentration of for germination was the combination treatment of 100 mM GA3 + 100 mM BAP.

The Effect of Seed Coat Scarification with Sodium Hypochiorite on Germination of Zoysiagrass Seed (Zoysiagrass 종자 발아에 미치는 Sodiym Hypochlorite의 종피처리 효과)

  • Research was conducted to obtain t he optimum treatment Of sodium hypochlorite(NaOC I) at various temperatures in t he seed scarification for stimulating germination of zoysiagrass (Zocysia Japonica Steud ) seed. Morphological changes of seed coat were also examined by scanning electron micros cop(SEM). l. Differences in temperature of scarification with 2 .4% NaOCI showed little influence on promoting germination of seeds but seeds treated with 1% solution at l5˚C germinated less than that of higher temperatures. The promotion effect of 4% solution on germination was diminished when seeds were treated for 8 hours of more. The most favorable seed scarification unaffected hy temperature for enhancing germination was 4-6hours treatment at 4% solution in fresh seeds. 2. $GA_3$, treatment did not enhance germination of water-pretreated control seeds hut germination of seeds pretreated with NaOCI l was increased additional 10% or more hy$ GA_3$, Water pretreated control seeds treated with 50 mM hydrogen peroxide(H'O )germinated about 44%. In NaOCI treated seeds. $H_2$$O_2$ treatment increased germination additional l 0% or more. 3. NaOCI l and KOH treatment softened the seed coat and formed the pores hy removing the scab-like thikenings attached to the seed surface. These results suggest that the modes of action of NaOCl in the promotion of seed germination reside in it increase of the permeability of the seed coat, and in the provision of additional oxygen to the seed.
