Assessment of Seed Viability and Vigour in Neem (Azadirachta indica A. Juss.)

  • Rapid loss in viability of neem (Azadirachta indica A. Juss.) seed is a major problem. Present effort was undertaken for developing a set pattern for assessing of viability and vigour in seed of various mother tree age of neem (Age I-06 years, Age II-15 years, Age III-25 years and Age IV->30 years old). Various viability test viz. triphenyle tetrazolium chloride test, electrical conductivity, excised embryo test, and germination test have been performed on seeds obtained from mother tree age classes. Inconsistency was observed with the TTC and EC test in germination of seed in laboratory as well as nursery. While various vigour tests viz. cold test, chemical stress test (methanol stress test), and accelerated ageing test alongwith ageing index, germination test (G%, MGT and GV) and various seedling growth parameters like seedling length (cm), number of leaves, collar diameter (cm), total biomass (g) alongwith mathematical indices i.e. vigour index, sturdiness quotient, volume index, quality index, root shoot ratio in nursery as well have been taken for study and showed better consistency. On the basis present study results of various viability and vigour test indicated that mother tree age class II performed better in comparison to others and it can be recommended for seed collection. Further it is also recommended that viability of neem seed may be assessed using various laboratory tests like excise embryo test and germination test (G%, MGT and GV) and vigour test may be taken preferably by cold germination test, chemical (methanol) stress test, accelerated ageing test in laboratory and germination alongwith various seedling growth parameters seedling length (cm), number of leaves, collar diameter (cm), total biomass (g) alongwith mathematical indices like Vigour Index, Sturdiness quotient, Volume Index, Quality index, root shoot ratio in nursery as discussed in this study.

Effects Of the Kieserite Application on the Seedling Vigour and Yield of Grass/Clover Mixed Swards on Newly Reclaimed Hilly Soil (신개간 산지토양에서 Kieserite 시용이 혼파목초의 초기생육 및 수량에 미치는 효과)

  • This pot experiment was conducted te find out the effects of kieserite application ($T_1$; control, $T_2$; standard amount of $N-P_2$$O_{5}$ $-K_2$0, $T_3$; $T_2$+ kieserite) on the seedling vigour and yield of pass/clover mixed swards on newly reclaimed hilly soil. The results obtained are summarized as follows: At the $T_1$ treatment the seedling vigour and yield of mixed forages were very poor, whereas they were remarkedly enhanced by the treatments of $T_2$ and $T_3$. Comparing with the $T_2$ treatment the forage yields were increased 3.1% in orchardgrass, 27.5% in ladino clover, and 12.8% in mixed forages by the application of kieserite($T_3$). In the grass-clover mixed swards, the application of kieserite more remarkedly influenced the seedling vigour, botanical composition, and yield of clover than those of grass.

  • This pot experiment was conducted in order to observe the effects of double superphosphate and fused phosphate application(T$_1$: control. T$_2$: double superphosphate application, T$_3$; fused phosphate application) on the seedling vigour, botanical composition, and yields of grass/clover mixed swards on newly reclaimed hilly soil. The results obtained are summarized as fellows: At the grassland establishment. the seedling vigour and yields of grass-clover mixed swards were much better by the T$_2$than the T$_3$. These favorable effects were more enhanced in white clover than orchardgrass. Compared with the T$_2$, the T$_3$resulted in the gradual increase of botanical composition and relative yield of white clover according to the cutting order. With the consideration of these results, double superphosphate could be favorable for pasture establishment and fused phosphate could be favorable for pasture management fertilizers.

Variation of Seed Viability among Cone Harvest Times at Two Clonal Seed Orchards of Chamaecyparis obtusa

  • The timing of seed harvesting is an important decision in the management of seed orchards because it affects seed quality and yield. To investigate the effect of cone harvest time on seed quality and determine the optimal harvesting time, cones were regularly collected in seven times and germination tests were performed at each harvest time in two clonal seed orchards of Chamaecyparis obtusa. As cones developed, the percentage of seed germination increased before cone moisture content began to decrease significantly. The moisture contents of cones were highest at the first collection as 68.3% and 67.3% in Jeju and Gochang seed orchards respectively. At this time, germination speed was slowest, indicating poor seed vigour. The highest germination was found at the second stage in Jeju (36.5%) and at the seventh stage in Gochang (28.6%) seed orchard. The germination speed increased as cone moisture content decreased. Additionally, changes of seed vigour differed among the developmental stages in both seed orchards. Consequently, the optimal cone harvest time of C. obtusa seed orchards in Jeju was early September when high germination percentage was obtained. In Gochang seed orchards, late October was optimal cone harvest time when the germination speed was fast and the cone moisture content decreased.

Effects of Fungicides and Bioagents on Seed Mycoflora, Growth and Yield of Watermelon

  • Fungicides and antagonists were tested for their efficacy in the management of fungal pathogens of watermelon. The fungal species in different genera were isolated from the seeds of watermelon and their vulnerability was assessed against an array of chemicals and bioagents. Among the fungal pathogens, Fusarium species were effectively controlled by Bavistin. Topsin also showed the promising effects against all the fungal pathogens, and Dithane M-45 effectively controlled Didymella bryoniae. Seed treatment with antagonists like Trichoderma harzianum and T. viride improved the seed germination, seedling vigour and reduced the incidence of seed-borne fungal pathogens. Bavistin and Topsin among chemicals increased significantly the seed germination and vigour index. Trichoderma harzianum showed its efficacy against all Fusarium species and even stood effective than Captan and Blitox. However, Pseudomonas fIuorescens also showed promising effect against Didymella bryoniae over fungicides. Under field condition, Topsin and Dithane M-45 showed better yield than Bioagents.

Effect of Seed Coating with Polymers on Seed Vigour and Seedling Stand in Direct Seeded Rice

  • These experiments were conducted to investigate water uptake, electrical conductivity, germination percentage, seedling growth, and seedling establishment rate in direct seeding cultivation of rice. The rice seeds of six japonica type varieties were coated with 12 kinds of polymers in a standard concentration of 0.2% using seed coating machine. The water absorption of the polymer-coated seeds under saturation conditions was not different among varieties, and was the highest in kulcel, maltrin, and waterlock on the polymer-coated seeds. The electrical conductivity with waterlock (55.0 $\mu scm^{-1}g^{-1}$) was higher than the control plot (45.6 $\mu scm^{-1}g^{-1}$) and other treatments. The germination of the polymer-coated seeds was 95.9% at control plot, 92.7% at low temperature and 35.7% at high temperature. The total dry weight of seed decreased in the order of low temperature, control plot, and high temperature, and was effective in pvp (polyvinyl pyrrolidone), opadry, and sacrust. The seedling establishment rate in direct seeding cultivation ranged from 74.9 to 81.0% in flooded paddy surface, and ranged from 64.7 to 76.6% in dry paddy. In both cases, it decreased in the order of early, medium and medium-late varieties, but was enhanced in daran 8600, sepirect, and sacrust. According to this study the recommended polymers for direct seeding cultivation of rice are daran 8600, sepirect, and sacrust.

A Simple Method of Seedling Screening for Drought Tolerance in Soybean

  • Water deficit is a serious constraint to soybean [Glycine max L. (Merr.)] production in rainfed regions of Asia, Africa, and America. This study was conducted to develop a simple and effective screening method for drought tolerance in soybean. Fifteen soybean cultivars, eight identified to be drought-tolerant and seven drought-sensitive in previous studies, were used for the evaluation of drought tolerance under the new screening conditions. The seedling screening method was consisted of a treatment in a PEG solution and drought treatment in parafilm-layered pots. 5-day-old seedlings were treated in a 18% PEG solution for 4 days and their wilting and hypocotyl browning were recorded. Three seedlings grown in a parafilm-layered pot containing peat moss were drought-stressed by withholding water from the third day after seedling emergence, and root and seedling growth were examined. Degree of drought tolerance were rated based on seedling vigor in the PEG solution and drought-stressed parafilm-layered pots, and also on the penetration ability of roots through parafilm layer. Most of seedlings of the drought-tolerant cultivars showed higher vigour and root penetration than those of the drought-sensitive cultivars under the new screening conditions. Our results indicate that the new method can be used as a simple and effective screening procedure for drought tolerance in soybean breeding programs.

Identification of quantitative trait loci for root development during seedling stage in rice

  • Vigorous root growth at the seedling stage in dry direct-seeded conditions is considered as a critical trait because it is involved in seedling emergence, early vegetative vigour, nutrient uptake as well as drought tolerance. In this study, we performed QTL mapping using the recombinant inbred lines obtained from the cross between Tongil-type Dasan and temperate japonica TR22183 (DT-RILs) to identify QTL underlying early root development. TR22183, which was previously reported to have high nitrogen utility and cold tolerance, showed vigorous root growth at the seedling stage in semi-drought conditions. Root length, fresh weight and dry weight of TR22183 were significantly higher than in Dasan. By QTL analysis with genotyping-by-sequencing method, we identified two QTLs for root fresh weight (RFW) in chromosome 7 and root dry weight (RDW) in chromosome 8, explaining phenotypic variances of 13.5% and 10.6%, respectively. These QTLs would be used to develop rice varieties adapted to direct-seeded cultivating system.

Changes in Quality and Vigour of Cucumber and Paprika Transplants as Affected by Storage Temperature under Dark Conditions

  • Cucumber and paprika transplants were stored at 9, 12, 15, and $18^{\circ}C$ under dark conditions for 15 days and then grown in a greenhouse for 14 days after transplanting. To determine the effects of low storage temperature and long-term continuous darkness on the quality and vigour of transplants, we investigated the quality of transplants during storage and the growth of stored transplants after transplanting. In cucumber transplants, decreasing storage temperature reduced stem elongation and decrease in SPAD value. The quality of cucumber transplants stored at $9^{\circ}C$ was well preserved during storage, but they did not survive after transplanting due to chilling damage. Growth and development after transplanting were significantly greater when cucumber transplants were stored at $12^{\circ}C$. In paprika transplants, the quality of transplants did not significantly differ before and after storage. After transplanting, there was no significant difference in the survival rate and growth, but the number of flower buds was greater in the paprika transplants stored at lower temperatures (9 and $12^{\circ}C$). These results indicate that the responses of transplants to the conditions of low temperature and darkness differed between cucumber and paprika, and storage temperature in darkness must be controlled carefully considering species-specific responses to reduce quality deterioration during storage and improve the recovery of transplants after transplanting.
