The Effect of Light, Temperature and GA Soaking Treatment on Seed Germination of Cool Season Turfgrasses (광, 온도 및 GA 침지처리가 한지형 잔디의 종자발아에 미치는 영향)

  • This study is aimed to understand the effects of temperature, light and GA soaking treatment on the seed germination of perennial ryegrass 'Dandy', creeping hentgrass 'Penlinks', tall fescue 'Arid' and Kentucky hluegrass 'Nasaw'. These experiments are carried in vitro, and their results obtained are summarized as the followings ; The optimum temperature for seed germination of perennial ryegrass 'Dandy' was found to he ranged at 20~30$^{\circ}C$, and the optimum temperature for germination of creeping bentgrass 'Penlinks', tall fescue 'Arid' and kentucky bluegrass 'Nasaw' seeds was found to be ranged at 20~25$^{\circ}C$, whereas the germination percentage under the below or above temperature of this range decreased. The germination of perennial ryegrass 'Dandy' and tall fescue 'Arid' seeds showed no effect on light, and while the germination percentage of creeping hentgrass 'Penlinks' increased more highly under the dark condition than under the light condition. The germination percentage decreased according to the higher temperature. The germination of Kentucky bluegrass 'Nasaw' seeds increased more highly under the dark condition than under the light condition at 20~25$^{\circ}C$, whereas the germination percentage under the dark condition at 15$^{\circ}C$ and 30$^{\circ}C$ decreased. In case of GA soaking treatment of seeds, all the cool season turfgrasses tested showed their own promoting effects on seed germination. The early germination of tall fescue 'Arid' was good without the soaking times. Creeping bentgrass 'Penlinks' and perennial ryegrass 'Dandy' had re-spectively the highest germination percentage at 10min and 1hr soaking treatment, and the longer the soaking time, the lesser the germination percentage. Kentucky bluegrass 'Nasaw' obtained the best results at 24has soaking treatment, and the longer the soaking time, the better the ger-mination percentage.

Present Status and Problems of Chemical Seed Treatment of Seedborne Diseases (종자소독의 현황과 문제점)

  • A wide variety of pathogens are known io be seedborne, carried either as infectious mycelium internally or as contaminants on the seed coat. When seed is infected with a pathogen, the seed nay be rendered nonviable or it may remain viable but produce weak seedling. In some cases, the Infected seedling nay not be severely weakened, but nay serve as a source of primary inoculum within a community of plants. A recent problem nay be the dissemination of seedborne pathogens occurring as a result of the massive movements of seed, as a part of the 'Green revolution' Disease of great danger to agriculture may be introduced with seed from other parts of world. Seed treatment with organic mercury compounds in liquid form had become popular since about 1955. Organic mercury compounds contributed considerably to the increase in production of many crops and vegetables. In 1975, however, the use of organic mercury compound was forbidden because of doubts regarding their residual mammalian toxicity in agricultural products. Benomyl-thiram mixture, thiophanate methyl-;hiram mixture and TCMB have now been registered as seed disinfectants for the use of rice blast, brown spot and Bakanae disease. Oxathiinsthiram mixture has been registered as seed disinfectant for barley and wheat loose smut and leaf stripe of barley. Agricultural techniques have made such rapid progress that the nursery methods changed from the use of paddy nursery to box nursery designed for machine-transplanting. The spread of rice transplanting machines has caused increase of seedborne diseases. Among seedborne diseases, Bakanae disease has remarkably increased and causes much damage recently. In order to counter this trend, seed disinfectants must also be diversified. First, effective non-selective disinfectants need to be developed, and second, appropriate control methods always need to be prepared in parallel with the development of new techniques for cultivation.

RAV1 Negatively Regulates Seed Development by Directly Repressing MINI3 and IKU2 in Arabidopsis

  • A plant-specific B3 domain and AP2 domain-containing transcription factor, RAV1 acts as a negative regulator of growth in many plant species and its transcription was down-regulated by BR and ABA. In this study, we found that RAV1-overexpressing transgenic plants showed abnormally developed ovules, resulting in reduced seed size, weight, and number in a silique. Interestingly, the endogenous expression of RAV1 fluctuated during seed development; it remained low during the early stage of seed development and sharply increased in the seed maturation stage. In plants, seed development is a complex process that requires coordinated growth of the embryo, endosperm, and maternal integuments. Among many genes that are associated with endosperm proliferation and embryo development, three genes consisting of SHB1, MINI3, and IKU2 form a small unit positively regulating this process, and their expression was regulated by BR and ABA. Using the floral stage-specific RNAs, we found that the expression of MINI3 and IKU2, the two downstream genes of the SHB1-MINI3-IKU2 cascade in the seed development pathway, were particularly reduced in the RAV1-overexpressing transgenic plants. We further determined that RAV1 directly binds to the promoter of MINI3 and IKU2, resulting in their repression. Direct treatment with brassinolide (BL) improved seed development of RAV1-overexpressing plants, but treatment with ABA severely worsened it. Overall, these results suggest that RAV1 is an additional negative player in the early stages of seed development, during which ABA and BR signaling are coordinated.

Bolting Response of Various Lettuce Cultivars Affected by Seed Treatments (종자처리에 의한 상추 품종간 추대반응 차이)

  • A series of experiments were performed to evaluate effects of lettuce seed treatment with low temperature and dry heat treatment (DHT) on bolting response in the case of spring cultivation. During spring production in greenhouse, bolting response of the plants produced from the treated seeds was faster in the order of low temperature treatment, control and DHT. Plant height was increased 5-10 cm higher by low temperature treatment, but plant growth of some cultivars was inhibited by DHT. Difference on lettuce plant growth between low temperature treatment and DHT was gradually diminished with the lapse of time after transplanting. Fresh weight of plant was not significantly different among all treatments. 'Red Gyeolku' showed very early bolting response and plant height was significantly promoted by low temperature treatment. However, 'Cheongchima' showed low bolting percentage after various seed treatments, so it is considered late-bolting cultivar.

Establishment of Seed Treatment for Healthy Production of Peanut Sprout (청정 땅콩나물 생산을 위한 종자처리기술 확립)

  • The present study was conducted to develop seed treatment for the production of healthy and clean peanut sprout. Dry heat treatment of peanut seeds reduced the incidence of the rot. The seed treatment condition at $52^{\circ}C$ for 10 h. was the most efficient without inhibiting seed viability significantly. Seeds were dark cultured at $27^{\circ}C$ for up to 9 days. The treatment of Indole-B and gibberellic acid influenced germination, T50, fresh, dry weight, hypocotyl length, hypocotyl length diameter, root length, number of lateral root and epicotyl of peanut sprout. There were no differences in the germinability of peanut seeds between gibberellic acid treatment methods but higher fresh weight was observed in the GA3 solution spray after 2 hour water soaking. The general growth and lateral root development of peanut sprouts were suppressed by Indole-B which is used for inhibiting root formation and promoting hypocotyls. The treatment of gibberellic acid promoted hypocotyl elongation, but it did not influence on the growth of hypocotyls and root system.

Studies on the Acceleration of Germination in Carrot Seed (II) Effects of X-ray and Ultraviolet light in the Germination of Carrot Seed (당근종자 발아촉진에 관한 연구 (제2보) 당그종자 발아에 미치는 X-ray, Ultraviolet Light의 영향)

  • The treatment of X-ray was manipulated with the large focus of X-ray which was produced the capacity of 90 kilovoltage and 30 milliamphere in the distance of 50 centimeter. The germination of each gruop irradiated with X-ray for 5, 6, and 7 seconds was more delayed in the growth of carrot seed than those of the control group. However the germination of each group irradiated with X-ray for 2, 3, and 4 seconds was rather accelerated than those of the control group. The irradiation of ultraviolet light was carried out in the distance of 20 centimeter by Toshiba germidicial lamp which appeared the wavelength of 2537A$^{\circ}$. The germination of each group irradiated with ultraviolet light for 9, 15, 20, and 40 minutes was more delayed than those of the control group, but the germination of carrot seed was accelerated by adequate dosage of ultraviolet light. Consuquently it was suggested for the author that the inhibited substance contained in carrot seed would be certain fatty substance, becasue the germination of carrot seed was more accelerated by the treatment of acetone than those of X-ray and ultraviolet light.

Effect of osmotic potential on germination of tomato seed

  • Seeds of Tomato (Lycopersicon esculentum Mill.) have demanded high quality because of their high cost of seed. The optimization of the seed priming techniques that have positive effect on fast and uniform germination becomes important at the commercial level. Several factors such as solution composition, osmotic potential, and treatment duration affect seed priming response. In this study, osmotic potentials of priming solution and germination characteristics of primed seed were investigated to clarify the effects different inorganic salt types and the duration. Tomato seeds were primed in osmotic solutions that were osmotic potential ranged -1.54 to -0.45 MPa in an aerated solution of PEG 8000 (17%, 22%, 27%), and inorganic salt solution of $KNO_3$, $Na_2SO_4$, and $K_2SO_4$ (100, 200, 300mM). The seeds were treated at $20^{\circ}C$ for 2, 4, and 6 days. After each treatment, the seeds were dried to moisture content ranged 5-8% at $25^{\circ}C$. Four replications of 25 seeds per each treatments were placed in 10-cm petri dishes containing two filter papers and 3 ml of $dH_2O$ and incubated at $20^{\circ}C/30^{\circ}C$ and $15^{\circ}C$ and seedlings evaluated for abnormality after 14 days of incubation. Seed water potential (${\psi}$) was correlated with water potential of priming solution ($r^2=0.86$). Seeds primed in 100mM $KNO_3$ resulted the highest germination rate (GR, $63.9 %{\cdot}day^{-1}$) and lowest mean germination time (MGT, 2.0 days) comparing to untreated control ($23.9%{\cdot}day^{-1}$ of germination rate and 4.1 days of MGT) at $20/30^{\circ}C$, even though 96% of germination percentage were not different. Seeds primed in 100mM $KNO_3$ (${\psi}=-0.45MPa$) for 4 days showed ${\psi}=-0.38MPa$. Priming in $Na_2SO_4$, $K_2SO_4$, and PEG solution for 6 days improved MGT and GR, but not significantly than 4 days of treatment. Additionally, stepwise osmotic solution treatment with 100mM and 300mM concentration for 6 day did not showed differences with single treatment. In relation to osmotic potentials, identical osmotic potential in different inorganic salt solution showed different effect on germination characteristics.

Effect of Replacing Beef Fat with Poppy Seed Oil on Quality of Turkish Sucuk

  • Sucuk is the most popular dry-fermented meat product. Sucuk has a relatively high fat. Poppy seed oil as animal fat replacer was used in Turkish sucuk and effects of its use on sucuk quality were investigated. There was a significant (p<0.5) treatment × ripening time interaction for moisture, pH (p<0.05) and 2-thiobarbituric acid reactive substances (TBARS) values (p<0.01). Increasing poppy seed oil level decreased (p<0.05) TBARS values. Addition of poppy seed oil to the sucuks had a significant effect (p<0.01) on hardness, cohesiveness, gumminess, chewiness and springiness values. Cholesterol content of sucuks decreased (p<0.05) with poppy seed oil addition. Using pre-emulsified poppy seed oil as partial fat replacer in Turkish sucuk decreased cholesterol and saturated fatty acid content, but increased polyunsaturated fatty acids. Poppy seed oil as partial animal fat replacer in Turkish sucuk may have significant health benefits.

Comparison of Antioxidant Activity of Vegetable Oil by Using Adsorbents (식물성 압착오일의 흡착제에 따른 항산화 활성 비교)

  • This study was designed to analyze the chemical composition and antioxidant activity of various vegetable oils (pumpkin seed oil, camellia seed oil, red pepper seed oil and peanut oil) using adsorbents (active carbon, acid clay, kaolin). Their chemical composition was analyzed by GCMS. Their antioxidant activity was evaluated by measuring their DPPH and ABTS radical scavenging activity. After the treatment with the adsorbents, the contents of most of the fatty acids and active ingredients contained in the four kinds of vegetable oils were reduced. After the treatment with the three adsorbents, the linoleic acid and erythrodiol contents of the pumpkin seed oil were reduced. In the case of the camellia seed oil, the fatty acids content was decreased, but there was no loss of vitamin E after the acid clay treatment. The content of the compound capsaicin, which forms part of the spicy component of red pepper seed oil, was reduced by 53.33% after the acid clay treatment. The peanut oil showed the lowest loss of sitosterol compound in the group treated with active carbon. The antioxidant activity was observed to be in the order of pumpkin seed oil (kaolin>acid clay>active carbon), camellia seed oil (acid clay>kaolin>active carbon), red pepper seed oil (kaolin>acid clay>active carbon) and peanut oil (active carbon>acid clay>kaolin).

Effect of Grapefruit Seed Extract and Ascorbic Acid on the Spoilage Microorganisms and Keeping Quality of Soybean Sprouts (Grapefruit Seed Extract와 Ascorbic Acid의 혼합 처리가 콩나물 변패 미생물과 저장 품질에 미치는 영향)

  • The antimicrobial effect of mixed solutions of grapefruit seed extract(250ppm) and ascorbic acid(1%) on the spoilage microorganisms such as Staphylococcus epidermis, Escherichia coli, Pseudomonas syringae, Candida albicans and Corynebacterium xerosis isolated from the spoiled soybean sprouts were investigated. Cell wall and membrane were partially destroyed and the contents of the destroyed cell were exuded after treatment. Packages with 30 m cast polypropylene(CPP), 16 m polyolefin(RD 106) and 10 m high density polyethylene(HDPE) were applied for soybean sprouts dipped in mixed solutions respectively. Oxygen and carbon dioxide concentration inside packages were dependent on the kind of films during storage at 5oC. The antimicrobial activity of mixed solutions was maintained for 5 days at CPP package. Package with HDPE showed a severe browning than the others after 5 days. Ascorbic acid content of mixed solution treatment was higher than that of control for each package.
