The Germination Characteristics of Seed by Storage and Germination Temperature in Astragalus membranaceus (저장 및 발아 온도조건에 따른 황기 종자의 발아 특성)

  • This study was investigated to evaluate germination rate of Astragalus membranaceus B. in Korea as affected by storage temperature, germination temperature and storage period of seed. The highest germination rate was obtained from condition of $25^{\circ}C$ in germination temperature. Seeds were stored at $-20^{\circ}C$ and $5^{\circ}C$ for 8 weeks has showed higher germination rate than one at room temperatures. The germination rates showed significantly difference by harvested year of 2010, 2011 and 2012. The seed of A. membranaceus in harvested year of 2011 and 2012 had germinated well. On the other hand, seeds in harvested year of 2010 were not nearly germinated. Consequently, the longer storage period after seed harvest lower germination rate and seed vigor as well.

  • The differences in seed germination rates of Korean Native Plants were observed depending on the harvest years, the seed companies, the storage periods, the drying methods and the plant morphological types. The seed germination rates were changed significantly by the harvest years and the seed companies in all 4 species tested. The seed germination rates were decreased after 1 year storage at $4^{\circ}C$ in all 8 species tested and three of them did not germinate at all. A rapid drying method at $35^{\circ}C$ with a convection oven was not suitable for Chrysanthemum spp., but there were no significant changes of germination rates in Aster spp. The germination rates were decreased more at seeding in soil than at lab. test with Dicotyledon but no significant changes with Monocotyledon. Especially, the germination rates of fine seed on 5mm soil covering treatment were significantly decreased.

  • The change in fatty acid compositions of the glycolipid and phospholipid of sesame seed was analyzed during the storage for 4, 9, and 15 months at four different storage conditions, that is, storage under light with linen bag(LA), storage under light with polyethylene bag(LS), storage in dark with linen bag(DA), and storage in dark with polyethylene bag(DS). In the fatty acid composition of glycolipid, palmitic and stearic acid decreased during the storage but the linoleic acid increased significantly, particularly, between 4 to 9 months of storage. The content of palmitic and stearic acid in phospholipid decreased until 9 months, then increased until 15 months of storage, but reverse tendency was, observed in the content of oleic and linoleic acid. The ratio of unsaturated to saturated fatty acids in glycolipid increased during the storage of 15 months in all the storage conditions. The ratio of unsaturated to saturated fatty acids of phospholipid in DA and LA was slightly higher than that in DS and LS conditions during the storage for 15 months.

  • In order to find the effect of the storage time on the characteristics and sterol composition of the sesame seed, the characteristics, unsaponifiable matters, fractionation pattern of sterol and sterol compositions of 4-desmethylsterol on the sesame seed oil were analysed at the harvest time and after 56 months storage. The sample seed was kept in cotton bag within the natural dark store room. The oil content was decreased from 50.3g per 100g sesame seed at harvest time to 45.5g at 56 months stored. The acid value was increased sharply from 1.8 to 13.4 during the storage of sesame seed and iodine value was increased small, but saponification value and refractive index were hardly changed. The amount of unsaponifiables from 24.3mg to 20.9mg per gram on basis during storage time, and the 4,4-dimethylsterol and 4-monomethylsterol from unsaponifiables were decreased, otherwise, 4-desmethylsterol was increased. In the changes of sterol composition in the 4-desmethylsterol fraction from unsaponifiables, the compesterol and stigmasterol content were not nearly changed, otherwise, the sitosterol content was decreased from 51.0% to 48.2%, but the content of ${\Delta}^5-avenasterol$ was increased from 15.2 % to 18.4% during the storage of sesame seed within 56 months.

Identification and Pathogenicity of Microorganisms Associated with Seed-Rhizome Rot of Gingers in Underground Storage Caves (토굴저장 생강의 부패에 관여하는 미생물의 동정 및 병원성)

  • Microorganisms associated with seed-rhizome rot of gingers preserved in three underground storage caves were identified with respect to rot types. Rot patterns were grouped into 4 different types : yellow soft rot, brown rot, localized ring rot, and water-soaked rot. Water-soaked rot was highest in frequency with 40% and ring rot the least with 14%. Causal pathogens differed with rot type, yellow soft rot by Erwinia carotovora and Pseudomonas aeruginosa, brown rot by Fusarium solani and Pseudomonas aeruginosa, localized ring rot by F. solani, and water-soaked rot by Pythium spinosum and P. ultimum. Pythium myriotylum, the causal pathogen of ginger rhizome rot which occurs severely in fields was rarely detected from storage seed-rhizomes suggesting its minor involvement with storage rot. Pathogenic Pythium isolates were frequently obtained from both rhizome surface and inner tissues of rotten rhizomes. Detection frequency of Pythium isolates in inner tissues decreased as increasing distance from rhizome surface. In wound-inoculation tests, above pathogens caused a varying degree of rot on healthy rhizomes at 15$^{\circ}C$, 2$0^{\circ}C$ and 3$0^{\circ}C$ with increasing severity at higher temperatures.

Antioxidant Activity of Radish Seed Oil and the Quality and Storage Characteristics of Pork Patties with Added Radish Seed Oil

  • This study investigated the antioxidant activity of radish seed oil (RSO) and its effects on the quality and storage characteristics of pork patties. To assess the antioxidant capacity of RSO, this study analyzed fatty acid composition, peroxide value (PV), and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity. Pork patties were manufactured with the addition of RSO-0.4%, 0.8%, 1.6%, and 2.4%-and measured in terms of proximate composition, pH, water holding capacity (WHC), cooking loss (CL), color, texture profile analysis, and a sensory evaluation. Total microbial count (TMC), volatile basic nitrogen (VBN), thiobarbituric acid reactive substances (TBARS), and PV were measured at 1, 3, and 7 days of refrigerated storage. The DPPH radical scavenging activity of RSO was found to be 75.46%. In the cases of WHC and CL, there was no significant differences observed between RSO0.4%, RSO0.8%, and positive control (PC; p>0.05). Meanwhile, RSO2.4% showed significantly lower hardness, springiness, gumminess, and chewiness than PC (p<0.05), and these values tended to decrease with the addition of increasing RSO. In terms of storage characteristics, with an increase in the amount of RSO added, TMC, VBN, TBARS, and PV all decreased; among the treatment groups, RSO2.4% showed the lowest values. In conclusion, RSO exhibits antioxidant activity, but when added in large amounts, it negatively affects the quality characteristics of patties while positively impacting their storage properties, thus necessitating a balanced consideration of both outcomes. Therefore, adding 1.6% RSO is considered to be the most appropriate level for formulations to be used in practice.

Studies on Seed Germination Characteristics and Patterns of Protein Expression of Lithospermum erythrorhizon by Plant Growth Regulators and Seed Primings (생장조절제와 프라이밍 처리에 의한 지치종자의 발아특성과 단백질 발현 양상에 관한 연구)

  • This study was conducted to investigate the quality of seeds, the germination rates and the days required for germination, to examine the patterns of protein expressions during the germination and to improve the techniques of managing and storing seeds and viability of the seeds of Lithospermum erythrorhizon Sieb. et Zucc. After collecting and harvesting seeds, they were classified to white and brown colors of seed coat through testing their seed size, weight, and quality. The germination rates, the days required for germination, and the protein expressions were examined with different colors of seed coats, storing temperatures and durations by treating the different plant growth regulators and primings. One hundred seed weight of white color was heavier about 1.17 g than those of brown one about 0.81 g. The germination rates in white color of seed coat was higher, 3.05 ~ 5.75%, than those in brown one. Its rates were decreased with getting longer in storage durations. There was no big differences on germination rates between storage temperatures. The plant growth regulator of $GA_3$ and Kinetin was affected to improve the seed germination. $GA_3$ increased the seed germination clearly at 25 ppm level, while kinetin increased it gradually from 25 to 100 ppm levels. In germination by seed primings, PEG6000 made higher germination rate with increasing their levels, whereas $KNO_3$ increased the germination until 100 mM level and then decreased it with 200 mM unlike PEG6000. The protein expressed during the seed germination were appeared more and clearer bands in the seed after germination, especially 20 ~ 30 kDa, compared to those in the seed before germination. These results showing more and clearer bands were positively related to the germination rates which were different by seed colors, storage temperatures and durations, and plant growth regulators and primings.

  • These experiment were conducted to evaluate the environmentally acceptable polymers, and 10 polymers were used in these study, and to investigate conductivity, germination percentage, water uptake of polymeric coating soybean seed. The conductivity of polymeric coating seed is higher than that of none coating seed and the highest conductivity was obtained with waterlock coating seed among the 10 polymer coating seed. As the soaking time was long, the conductivity was increased. The conductivity of large seed was higher than that of small seed, and that of long period storage seed was higher than that of short period storage seed. The effects of seed coating polymers on uptake water were various, and daran 8600 inhibited uptake water of low quality seed. The waterlock, captan, klucel and sacrust was rised germination percentage, and daran 8600 was declined germination percentage, and the effect of coating polymers on germination percentage of low quality seed was higher than that of high quality seed.

Change of Seed Dormancy at Different Storage Conditions in Rice (벼 저장조건에 따른 종실의 휴면성 변화)

  • This study was conducted to investigate change of seed dormancy during storage in rice. The result of germination rate observed at different storage duration after harvest indicated that two Japonica rice varieties, Sangpungbyeo and Hwaseongbyeo, tow sare type local varieties, Jakwangdo and Mongeunsare, and a Tongil type rice, Samgangbyeo, showed no dormancy in rice seed harvested at 40 days after heading, However, weak dormancy was found in matured rice seed of Sobaekbyeo and Odaebyeo, and strong seed dormancy was found in Jungwonbyeo, IR-20 and IR-42. Rice seed stored for 30 days after harvest at 2$0^{\circ}C$ constant temperature or 25/15$^{\circ}C$ of day /night temperature fluctuation showed higher germination rate above 90% in all varieties tested. But germination rate of seeds stored at 4$^{\circ}C$ for 30 to 180 days showed varietal difference, higher in rice varieties having weak seed dormancy and lower in strong dormancy varieties. Days to 80% germination and average days for germination were shortened by storage for 30 to 180 days after harvest at 2$0^{\circ}C$, 4$^{\circ}C$ with RH 40% or day /night 25/15$^{\circ}C$ temperature condition in all rice varieties tested, but the degree of acceleration was lower at the condition of 4$^{\circ}C$ with RH 40%.

Effects of Alkaline Ionic Water and Grapefruit Seed Extract Added Immersion Solutions on Storage Characteristics of Mulberry Leaf Soybean Curd (침지액에 알칼리성 이온수 및 자몽종자추출물 첨가가 뽕잎두부의 저장성에 미치는 효과)

  • This study was conducted to analyze the acidity and turbidity changes of immersion solutions as well as changes in aerobic bacteria, E. coli, anaerobic bacteria, yeast and mold counts of mulberry leaf soybean curds during storages at $4^{\circ}C$ and $25^{\circ}C$ in different immersion solutions such as distilled water, grapefruit seed extract (300 ppm) and alkaline ionic water. The acidities of immersion solutions of distilled water, grapefruit seed extract and alkaline ionic water after 18 days of storage at $4^{\circ}C$ were 0.021, 0.008 and 0.002%, respectively. After 5 days of storage at $25^{\circ}C$ were 0.042, 0.029 and 0.009%, respectively. The turbidities of the above mentioned immersion solutions were 0.50, 0.29 and 0.21 after 18 days of storage at $4^{\circ}C$ and 0.38, 0.34 and 0.27, respectively, after 5 days of storage at $25^{\circ}C$. The acidity and turbidity changes of immersion solutions were sensitive to storage temperatures. The aerobic bacteria count of mulberry leaf soybean curds after 18 days of storage at $4^{\circ}C$ was still below $10^7\;CFU/g$, the beginning point of soybean curd putrefaction; in contrast, this value was reached within one day in distilled water at $25^{\circ}C$ and between 2 and 3 days in alkaline ionic water. Grapefruit seed extract and alkaline ionic water had a better preservative effect at $4^{\circ}C$ than at $25^{\circ}C$ storage temperature.
