Monographic Study of the Endemic Plants in Korea IX. Taxonomy and interspecific relationships of the genus Melandrium (한국 고유식물의 종속지 IX. 장구채속식물의 분류와 종간유연관계)

  • To clarity the species of genus Melandrium (Caryophyllaceae) in Korea, the taxonomic characters were described and interspecific relationships were discussed to the extemal morphology with light and scanning electron microscopy. The position and size of appendages on petal, the shape and venation pattern of leaf, and the external morphology of seed and pollen were approved as the good characters to classify genus Melandrium in Korea. As the result, genus Melandrium in Korea could be arranged as 6 species, 1 variety, 1 forma. M. umbellatum was exclusively distributed in north Korea, and its taxonomic treatment was reserved.

  • The mathematical implementation for inverse airfoil design of wind turbines is presented using vortex panel method based on assumptions of the two-dimensional incompressible potential flow. The vortex panel method employs linear distribution of the vortex strength to obtain the well converged solution. Stream function is adopted to get the basic formula for the inverse airfoil design, and a symmetric seed airfoil is given for initial data of the iteration approach. The final airfoil shape has been compared with the original airfoil shape for validation of the mathematical procedure.

  • Surface enhanced Raman Scattering (SERS) has attracted attention because the technique enables detection of various chemicals, even down to single molecular scale. Among the diverse candidates for SERS substrates, Au nanoparticles are considered promising due to their fine optical properties, chemical stability and ease of surface modification. Therefore, the fabrication and optical characterization of gold particles on solid supports is highly desirable. Such structures have potential as SERS substrates because the localized surface plasmon resonance of gold nanoparticles is very sensitive to combined molecules and environments. In addition, it is well-known that the properties of Au nanoparticles are strongly dependent on their shape. In this work, arrays of shape-controlled Au nanoparticles were fabricated to exploit their enhanced and reproducible optical properties. First, shape-controlled Au nanoparticles were prepared via seed mediated solution-phase synthesis, including spheres, octahedra, and rhombic dodecahedra. Then, these shape-controlled Au nanoparticles were arranged on a PDMS substrate, which was nanopatterned using soft lithography of poly styrene particles. The Au nanoparticles were selectively located in a pattern of hexagonal spheres. In addition, the shape-controlled Au nanoparticles were arranged in various sizes of PDMS nanopatterns, which can be easily controlled by manipulating the size of polystyrene particles. Finally, the optical properties of the fabricated Au nanoparticle arrays were characterized by measuring surface enhanced Raman spectra with 4-nitrobenezenethiol.

Improvement of cultural efficiency using DNA markers in anther and seed culture of rice (DNA marker를 이용한 벼의 조직배양 효율개선)

  • The purpose of this study was to improve the culturability of 'IR 36', a indica type rice cultivar using DNA marker associated with the ability of plant regeneration in anther and seed culture. The culturability of 6 rice cultivars and 2 indica/japonica lines ('MGRI 036', 'MGRI 079') were investigated in anther and seed culture. The culturability of 3 japonica rice cultivars were much higher than tongil and indica rice cultivars, and 'MGRI 036' and 'MGRI 079' has high culturability with 20% regenerability, also. 34 $BC_2F_4$ 4 lines were selected by marker screening using RZ400 among 90 $BC_2F_4$ lines derived from a cross between 'MGRI 079' and 'IR 36'. The frequency of callus formation of 10 $BC_2F_4$ lines were higher than 'IR 36' in anther culture among selected 34 $BC_2F_4$ lines. The ability of plant regeneration of 10 lines were higher than 'IR 36' in the seed culture among selected 34 $BC_2F_4$ lines. A promising line, $BC_2F_4$-28, was selected to have better culturability in the anther and seed culture among selected 34 $BC_2F_4$ lines. The heading date and grain shape of the $BC_2F_4$-28 was similar to 'IR 36'. Using the RZ400 DNA marker associated with the culturability will be useful method for improving of indica rice culticvar's culturability in rice breeding program.

Several Factors Affecting Seed Germination of Hydrangea petiolaris Siebold & Zucc. (등수국의 종자발아에 미치는 몇 가지 요인)

  • This study was carried out to establish seed propagation method of Hydrangea petiolaris Siebold & Zucc., one of the specific and protected plant species for the floral region. Seed size ranged $1.36{\times}0.84mm$, weight of thousand seeds was $2.3{\pm}0.02mg$. Observation of seed morphology characters showed wrinkled surface and elliptical shape, indicating amber color and fine seeds. Moisture contents of seeds, increased rapidly by water-soaking treatment, recording maximum moisture contents (31.9%) after 48 hours. Percent germination of seeds was higher under the light than dark condition. Seed germination was the best at $25^{\circ}C$ under the light condition (78.0%) among temperature and light conditions treated. Percent germination, germination energy and $T_{50}$ was greatly improved by soaking in $GA_3$ solutions for 48 hours. Especially, $100{\sim}500mg{\cdot}L^{-1}$ $GA_3$ treatment resulted in the highest germination rate as 90.0% and above.

Comparative seed morphology of the tribe Alsineae (Caryophyllaceae) in Korea and its taxonomic implications (한국산 Alsineae (석죽과) 종자의 형태비교 및 분류학적 유용성)

  • The seed morphology of 25 taxa (8 genera) of Alsineae (Alsinoideae, Caryophyllaceae) in Korea was examined using stereo (SM) and scanning electron microscopy (SEM) in order to evaluate their diagnostic values and taxonomic implications. Circular, comma-shaped, ellipsoid, ovate, pyriform, reniform, and sub-circular seed shapes were identified. The seeds of the examined taxa range from 0.28 to 4.10 mm in length and from 0.33 to 3.10 mm in width; the smallest one was found in Sagina japonica, while the largest was in Honkenya peploides var. major. A hilar appendage was found in Cerastium, Honkenya, Moehringia, and strophioles were only observed in the genus Moehringia. Based on the examination of epidermal cells of the seed surface, four micromorphological types can be distinguished, viz., the colliculate-papillate (Col-Pap), colliculate-columellate (Col-Cll), colliculate-simple (Col-Sim), and smooth (Sm) types. Descriptions of the testa shape, margin, and the degree of elevation of the anticlinal cell walls, and the outer periclinal cell walls are summarized for the tribe. Taxonomic implications of the seed micromorphological characters were also discussed, in comparison with an earlier taxonomic system.

Comparison of color and major components of hempseed oils extracted with pressuring and extruding methods (압착식, 압출식 착유 대마 종실유의 색깔과 주요성분 비교)

  • Hemp [Cannabis sativa L.] has been cultivated as a fiber crop for long history, but it was a good oil crop because its seed contain plenty of lipid which is high ratio of unsaturated fatty acid. Hemp seed oil was extracted with a extruding method in many countries including Europe. The color of oil extracted with extruding method is dark green which could be difficult to attract consumer's interest in Korea because of insufficient information about hemp seed oil. This study was conducted to know correct information about hemp seed oils which were extracted with pressuring and extruding methods. In extruding method, seeds were crushed during the extracting process and discharged oil cake in shape of thin ribbon, but maintained seed shape although the seed were slightly flatted in pressuring method. Oil yield were higher in the extruding method in comparison with pressuring method. The oil have lower degree of lightness but higher degree of greenness and yellowness in the extruding method in comparison with pressuring method because of higher content of chlorophyll A, B and carotenoid. In fatty acid composition, the ratio of palmitic acid, stearic acid, oleic acid and ${\gamma}$-linolenic acid were higher but linoleic acid and ${\alpha}$-linolenic acid were lower in the extruding method in comparison with pressuring method. The content of total tocopherol and ${\gamma}$-tocopherol were lower in the extruding method in comparison with pressuring method.

Phylogenetic study of East-Asia Astragalus L. based on morphological characters (형태형질에 기초한 동아시아산 황기속 식물의 계통분류)

  • In order to find useful taxonomic morphological characters, 17 taxa from East-Asia Astragalus and Oxytropis anertii were investigated. In this study, seed coat structure, fruit surface, and fruit anatomical structures were selected to classify the species in Astragalus. As a results, useful morphological and anatomical characters in East-Asia Astragalus were life cycle, leaflet shape, raceme, petal color, seed color, pod shape, absent or present of hairs in pod surface, bent degree of dorsal suture, and thickness of endocarp. Using medicinal sources in China, A. floridus, A. membranaceus, A. membranaceus var. mongholicus and A. tongolensis were formed same group. However, these were not completely consensus in the morphological characters. The other hand, A. koraiensis and A. sikokianus, which have controversial taxonomic issue, have clearly no differences morphological characters each other. Therefore, A. koraiensis, A. sikokianus, and A. bhotanensis should be conducted a comparative study on the DNA sequences and molecular cytology in order to clarify the taxonomic treatments in the near future.

Microstructures and Mechanical Properties of SiC Containing $SiC_{platelet}$ Seeds of Various Size (여러 가지 크기의 $SiC_{platelet}$ Seed를 함유한 SiC 세라믹스의 미세구조 및 기계적 특성)

  • Liquid phase sintered silicon carbides were obtained by sintering of $\alpha$-SiC and $\beta$-SiC powders as starting materials at 2173K and 2273K respectively. The SiCplatelet seeds of different sizes were obtained by a repeated ball milling and sedimentation. Their mean size (d50) were 2.217 ${\mu}{\textrm}{m}$ 13.67 ${\mu}{\textrm}{m}$, 22.17${\mu}{\textrm}{m}$ respectively 6wt%Al2O3-4 wt% Y2O3 was used as the sintering additives for the liquid phase sintering. The two silicon carbides had a bimodal microstructure consisting of small matrix grains and large platelike grains when the SiCplatelet seeds were added. In the case of the $\beta$-SiC the appreciable phase transformation occurred as sintering temperature increased from 2173K to 2273K and resulted in matrix shape change from equiaxed into platelike grains. In contrast there was no shape change for the $\alpha$-SiC. The size of large grains in the $\alpha$-SiC of large grains in the $\alpha$-SiC was larger than that of the large grains in the $\beta$-SiC These results suggested that the growth of the $\alpha$-SiCplatelet in the $\alpha$-SiC matrix was more favored than that of the $\alpha$-SiCplatelet in the $\beta$-SiC matix. The three point flexural strength decreased as the added seed size increased. Fracture toughness values of samples sintered at 2273K were higher than those of samples sintered at 2173K.

Two-step Boundary Extraction Algorithm with Model (모델 정보를 이용한 2단계 윤곽선 추출 기법)

  • We propose an algorithm for extracting the boundary of a desired object with shape information obtained from sample images. Considering global shape obtained from sample images and edge orientation as well as edge magnitude, the Proposed method composed of two steps finds the boundary of an object. The first step is the approximate segmentation that extracts a rough boundary with a probability map and an edge map. And the second step is the detailed segmentation for finding more accurate boundary based on the SEEL (seed-point extraction and edge linking) algorithm. The experiment results using IR images show robustness to low-quality image and better performance than conventional segmentation methods.
