Evaluation of Seed Vigor and Morphology by Varietal Difference of Sorghum (Sorghum bicolor (L.) Moench) (수수의 종자형태 및 발아세 평가)

  • This study was conducted to evaluate basic seed characteristics such as morphological characteristics and various seed vigor using 4 sorghum varieties, Nampungchal, Donganme, Hwanggeumchal and Whinchal. No difference was shown in morphological characteristics of the varieties except that the width. Whinchal has bulb-shaped seed, while the other varieties have oval-shaped seeds. The seed coat color of Whinchal is obviously different from those of the other varieties; especially, Nampungchal and Hwanggeumchal are not glossy. Germination rate were Hwanggeumchal > Nampungchal > Donganme > Whinchal in the order of higher rate in standard germination tests and cold germination test. In the ageing test, Donganme showed the lowest 15% decrease in comparison with the standard cultivar. Electric conductivity (EC) has negative correlation with standard germination tests, cold germination test and accelerated ageing test. In the percentage of emergence (PE), Hwanggeumchal was the highest 85.7% which was more than 40% to the lowest Whinchal. Mean emergence time (MET) showed significant difference between the varieties; Whinchal showed the slowest 6.0 days.

Effects of Seed-coat Softening and Prechilling on Seed Germination and Bulbil Formation of Polygonatum odoratum Druce (둥굴레 종자의 발아(發芽) 및 소주아(小珠芽) 형성(形成)에 대한 저온(低溫)과 종피연화(種皮軟化) 처리(處理)의 영향(影響))

  • Solomon's seal (Polygonatum odoratum) was traditionally propagated by its rhizomes but seed propagation has not been practiced due to their dormancy and high price. To improve the seedling production through seed pretreatments, the experiment was conducted to determine the effect of seedcoat softening and seed prechilling on seed germination and bulbil formation of the seal. The prechilling was done for 4 to 16 weeks at $3^{\circ}C$ and the softening was forced with 10% KOH for 40 minutes. Seed germination, bulbil formation and length, no. of root and root length were measured at 6 and 12 weeks after sowing. Seed germination and bulbil formation were greater at 8 to 12 week prechilling than at shorter or longer treatment when only the prechilling was enforced. Prechilling had greater effect compared to seed coat softening. In addition, the softening treatment before prechilling showed higher germination and bulbil formation compared to single treatment of them. The softening treatment prior to prechilling was better than the reverse treatment in number of root except the above two characters. Duration of prechilling treatment was able to be shortened below 8 weeks when the softening treatment was applied before prechilling.

Germination Characteristics and Daily Seed Germinating Pattern of 8 New Varieties of Perennial Ryegrass under Alternative Conditions Required by ISTA (ISTA 변온조건에서 퍼레니얼 라이그래스 신품종 8종류의 발아특성 및 일일 발아패턴)

  • Research was initiated to investigate early establislment characteristics and germination pattern of perennial ryegrass (PR, Lolium perenne L.). Eight varieties of PR were evaluated in the study. An alternative environmental condition for a PR germination test required by International Seed Testing Association (ISTA) was applied in the experiment, consisting of 8-hr light at $25^{\circ}C$ and 16-hr dark at $15^{\circ}C$ (ISTA conditions). Daily and cumulative germination pattern were measured and analyzed daily for 30 days. Significant differences were observed in germination pattern, days to the first germination, days to 50% germination, days to 75% germination, and final germination rate. Germination rates were variable among the varieties, being 66.00 to 93.75% at the end of study. There were considerable variations in early germination characteristics and germination pattern among PR varieties. The first germination in all entries tested was initiated from 3 to 5 DAS (days after seeding) under ISTA conditions, being 2 days in differences among the varieties. It was 4 to 7 DAS in days to the 50% germination, being 1 to 3 days later from the first germination and three days were variable among them. Days to the 75% germination were 5.93 to 10.50 DAS under ISTA conditions, being 4.57 days in variety differences. Pennant II and Brightstar II were the fastest varieties. The slowest ones were Revenge GLX and Brightstar SLT. Considering days to the first germination, days to 75% germination, and germination pattern etc, Pennant II and Brightstar II were regarded as excellent varieties under ISTA conditions. From tills study, information on differences in germination characteristics and pattern would be usefully applied for golf course design and construction, when established with PR.

Comparison of Germination Characteristics and Daily Seed Germination Pattern in 15 New Cultivars of Kentucky Bluegrass under Alternative and Natural Conditions at Room Temperature (자연실온 및 변온환경에서 15종류의 켄터키 블루그래스 신품종의 발아특성 및 일일발아패턴 비교)

  • Research was initiated to investigate early establishment characteristics and germination pattern of Kentucky bluegrass (KB, Poa pratensis L.) under natural room temperature and to analyze differences between alternative and natural conditions for a practical application. Fifteen cultivars of KB were evaluated in the study. Daily and cumulative germination pattern were measured and analyzed on a daily basis. Differences were observed in germination percentage, germination pattern, days to the first germination and days to 75% germination. Germination percentage was variable with cultivars at the end of study. It was 75.75 to 90.25% under natural condition. There were considerable variations in early germination characteristics and germination pattern among KB cultivars according to germination conditions. The first germination in all entries tested was initiated between 6 and 9 DAS (days after seeding) under ISTA (International Seed Testing Association) condition. But it was observed between 9 and 12 DAS under natural condition, being 3 to 5 days later as compared with ISTA condition. On the first date of a germinating process, the germination rate was 0.25% to 25.00% under ISTA condition, while 0.50% to 13.25% under natural condition. Days to the 75% germination were 15.08 to 28.80 DAS under ISTA condition and 17.78 to 28.75 DAS under natural condition. Midnight II, Excursion and Midnight were the fastest cultivars under ISTA condition, while Odyssey the fastest one under natural condition. Regardless of germination condition, the slowest cultivar was Voyager II, being over 4 weeks to 75% germination percentage. For the most cultivars, days to 75% germination under natural room temperature condition were 1 to 8 days slower than under ISTA condition. From this study information on differences in germination characteristics and pattern among cultivars would be usefully applied for KB establishment such as golf course construction. Prior to on-site seeding, a comprehensive germination test is practically recommended due to a possible decline in germination capability during domestic distribution and storage after import.

Changes of Germination Rate of Pulses Seed Germplasm after Long-term Conservation

  • The seeds of soybean (Glycine max), adzuki bean (Vigna angularis), mung bean (Vigna radiata), and kidney bean (Phaseolus vulgaris L.) were examined the germination rate after 10 years of long-term storage ($-18^{\circ}C$) conservation. For soybean seeds, 2,313 accessions were examined and germination rate of 1,082 accessions was decreased with below 15% of initial germination rate. For 227 accessions of soybean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 589 accessions was increased and showed no change for 415 accessions after 10 years of long-term storage. For adzuki bean seeds, 2,058 accessions were examined and germination rate of 739 accessions was decreased with below 15% of initial germination rate. For 63 accessions of adzuki bean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 535 accessions was increased and showed no change for 721 accessions after 10 years of long-term storage. For mung bean seeds, 438 accessions were examined and germination rate of 139 accessions was decreased with below 15% of initial germination rate. For 5 accessions of mung bean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 155 accessions was increased and showed no change for 139 accessions after 10 years of long-term storage. For kdney bean seeds, 366 accessions were examined and germination rate of 7 accessions was decreased with below 15% of initial germination rate. For 65 accessions of kidney bean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 201 accessions was increased and showed no change for 93 accessions after 10 years of long-term storage.

Characteristics of Seed and Germination of Rhododendron mucronulatum by Collection Dates and Germination Temperatures (채취 시기와 발아 온도에 따른 진달래의 종자 및 발아 특성)

  • This study was carried out in order to secure basic data of seedling mass propagation technique of Rhododendron mucronulatum which is the native tree species of Korea by surveying the characteristics of its fruit and seed. The fruits were collected at Mt. Goryeo in Ganghwa-gun on different dates in 2013; August 26th, September 5th, September 12th, October 4th. The seed germination test was carried out at 5, 10, 15, 20, 25 and $30^{\circ}C$. Moisture content of the fruit was highest (54.5%) in the fruit collected on September 5th. Number of the seeds in a fruit was 91.3 to 116.3, regardless of the collection date. Seed length was highest ($1947.4{\mu}m$) in the seeds collected on October 4th and seed width was highest ($727.3{\mu}m$) in the seeds collected on September 12th. Germination rate of the seeds was highest at $25^{\circ}C$ regardless of the seed collection date, which showed the highest value(27.3%) in the seeds collected September 12th. Meanwhile, the seeds were not germinated not at all at 5, 10 and $30^{\circ}C$. $T_{50}$ and mean germination time of the seeds got shorter at the higher temperature. Germination uniformity got lower when the collection date got later. Germination speed of the seeds was fastest at $25^{\circ}C$. According to the results of this study, it seems that the appropriate time to collect fruit and seed is between September 12th and October 4th, and the appropriate temperature for the seed germination is $25^{\circ}C$.

Germination Characteristics and Maturity by Production Time of Chamaecrista nomame, Lespedeza cuneata and Lespedeza bicolor Seed in Fabaceae Plant (콩과식물 차풀, 비수리, 싸리 종자의 채종시기별 성숙도 및 발아특성)

  • This text was experimented and investigated the optimum production time and germination characteristic of Fabaceae plant seed that collect in Korean Chungnam. Optimum time of seed production was early-mid October of Chamaecrista nomame, early November of Lespedeza cuneata and early-mid November of Lespedeza bicolor. Epiphytic amount of seed full ripe was the most times in early October of Chamaecrista nomame, early November of Lespedeza cuneata and Lespedeza bicolor, respectively. Seed rate of maturity was the highest times in early November of Chamaecrista nomame, mid November of Lespedeza cuneata and Lespedeza bicolor, respectively. Germination rate by seed collection time was the highest times in early October of Chamaecrista nomame (99.8%), mid November of Lespedeza cuneata (68.3%) and early November of Lespedeza bicolor (24.3%). Germination days by seed production time was the shortest times in mid October of Chamaecrista nomame, mid November of Lespedeza cuneata and Lespedeza bicolor, respectively. Optimum temperature of germination was $15{\sim}35^{\circ}C$ in Chamaecrista nomame, $25{\sim}30^{\circ}C$ in Lespedeza cuneata, and Lespedeza bicolor was difficult judgment of germination optimum temperature because of low germinating rate.

Bioassessment of Nanoparticle Toxicity based on Seed Germination and Germination Index of Various Seeds (다양한 씨앗의 발아 및 발아지수에 근거한 나노입자 생물학적 독성평가)

  • This study investigated the effects of six metal oxide nanoparticles (NPs: CuO, NiO, TiO2, Fe2O3, Co3O4, ZnO) on seed germination and germination index (G.I) for five types of seeds: Brassica napus L., Malva verticillata L., Brassica olercea L., Brassica campestris L., Daucus carota L. NPs of CuO, ZnO, NiO show significant toxicity impacts on seed activities [CuO (6-27 mg/L), ZnO (16-86 mg/L), NiO (48-112 mg/L)], while no significant effects were observed at > 1000 mg/L of TiO2, Fe2O3, Co3O4. Tested five types of seed showed different sensitivities on seed germination and root activity, especially on NPs of CuO, ZnO, NiO. Malva verticillata L. seed was highly sensitive to toxic metal oxide NPs and showed following EC50s : CuO 5.5 mg/L, ZnO 16.4 mg/L, NiO 53.4 mg/L. Mostly following order of toxicity was observed, CuO > ZnO > NiO > Fe2O3 ≈ Co3O4 ≈ TiO2, where slightly different toxicity order was observed for carrot, showing CuO > NiO ≈ ZnO > Fe2O3 ≈ Co3O4 ≈ TiO2.

Germination Behaviors and Seed Longevities of Three Ulmus Species in Korea (한국산(韓國産) 느릅나무속 3수종(樹種)의 종자저장성(種子貯藏性) 및 발아특성(發芽特性))

  • This study was conducted to investigate temperature effect on seed germination behaviors and seed viability changes by drying periods of three Ulmus species (Ulmus davidiana var. japonica (Rehder) Nakai, U. pumila L., and U. parvifolia Jacq.) distributed in Korea. Statistical analysis showed that temperature had a significant effect on mean germination time in three Ulmus species, but its effect on germination percentage was only shown in U. parvifolia (p<0.01). U. davidiana var. japonica and U. pumila germinated relatively consistent in all temperature conditions, especially the latter showed high germination behaviors in low temperatures (10-15$^{\circ}C$). U. parvifolia germinated well in high temperature (25$^{\circ}C$), while this species rarely germinated in low temperature conditions (10-20$^{\circ}C$). The results confirm that seeds of U. pumila and U. parvifolia are orthodox seeds which can be stored under 4-8% moisture condition, and U. davidiana var. japonica seed is recalcitrant seed which is difficult to store for a long time. In conclusion, these differences of the germination behaviors for three Ulmus species seem to be adaptive regimes of their natural habitats, respectively.

Effect of Calcium Cyanamide Soil Fumigation on Sterilization of Rhizoctonia solani, Pythium sp., Soil Microbes and Plant Seed (석회질소 토양훈증의 라이족토니아 소라니, 피시움, 토양미생물과 씨앗의 사멸효과)

  • The effect of calcium cyanamide (China-made) soil fumigation on the growth of the ginseng pathogen Rhizoctonia, Pythium), soil microbes, and seed germination of lettuce and radish was investigated. At twice the recommended level (2S0-ppm $CaCN_2$), the growth of Rhizoctonia and Pythium, and the seed germination, were not inhibited. Rather, the effective level was 10,000 ppm. The powder form was more effective than the granular form in inhibiting pathogen growth and seed germination. The lettuce seed was also more sensitive than the radish seed. Calcium cyanamide appearedto decrease the fungi population and to increase Actinomycetes in the soil.
