Effects of Osmotic and Non-Osmotic Priming Methods for Enhancing Germination of Pepper Seed

  • This study was conducted to investigate the effects of osmotic priming, liquid smoke (LS), sonication and modified drum priming treatments to improve the germination speed and uniformity of pepper seeds for high quality export seeds. Seeds were treated in 0.5, 1.0, 5.0, 10% LS solution only or with 100mM $K_2SO_4$ solution for 6days at $15^{\circ}C$. Sonication treatment was performed for 5, 10, and 20 minutes at an intensity of 5.2, 10.4, 15.7, 21.0, 26.1kHz in water at $15^{\circ}C$. After sonication treatment, seeds were primed with water or 100mM $K_2SO_4$ for 4days. 40, 50 and 60% seed moisture content (SMC) of hydrated seeds were incubated for 84, 96 and 108h in a container with a relative humidity of 99% at 26rpm for a modified drum priming treatments. 0.5% LS treatment with water or 100mM $K_2SO_4$ priming showed significant effects with 71% GP (9.0days MGT) and 66% GP (7.4days MGT), respective, while untreated seeds resulted in only 61% GP and 10days MGT. The healthy seed % was increased by 24% by $K_2SO_4$ priming with LS treatment compared to untreated seeds. However, sonication treatment showed less germination or no difference compared to untreated seed, regardless of intensity and treatment time. The modified drum priming treatment (108h incubation after 50% SMC hydration) significantly improved the GP (on the $4^{th}$days after sowing), germination rate (GR), and MGT to 65%, $28%{\cdot}day^{-1}$, and 3.6days, whereas untreated seed resulted only 7% GP, $17%{\cdot}day^{-1}$ GR and 5.9days MGT. Thus, modified drum priming treatment effectively improved seed germination and is considered as an industrially promising treatment methods considering the shortening of the treatment period and environment-friendly aspects.

The Time for Collecting of Cryptomeria japonica Seeds

  • The time of seed collection is regarded as one of major concerns to obtain sound seeds. The physical and germinal aspects of Cryptomeria japonica D. Don (Taxodiaceae) seeds were analyzed to determine the optimum harvesting time in Korea. Cones were picked every 10 days from the $30^{th}$ of July to the $30^{th}$ of October in both 2005 and 2006. Seeds were collected from picked cones. Seed size and weight were not significant in two consecutive years. The 1,000-seed weight was 3.3 g for cones picked at the $18^{th}$ of August and 5.3 g for cones picked at the $30^{th}$ of September. The size of seeds was increased as the time of collection from the $18^{th}$ of August to the $30^{th}$ of September: from 19.3 mm to 21.3 mm in length and from 15.8 mm to 18.5 mm in width. Average germination rates in 2005 was 18.3% and 19.6% in 2006. The highest germination rate was 34.3% from seeds collected at the $30^{th}$ of September in 2005. In 2006, the highest germination rate was 31.7% for seeds collected at the same date as the 2005 seeds. After the end of September, germination rate was decreased in both years. The results implied that the best cone picking time for Korean C. japonica seeds is around the end of September.

The dehiscence process in Panax ginseng seeds and the stigmasterol biosynthesis pathway in terms of metabolomics

  • Background: Ginseng, officially known as Panax ginseng Meyer, has been traditionally used as a medicinal herb, particularly in Asia. Ginseng is propagated from seeds; however, seed germination is challenging, especially in its natural environment on farms. The seeds typically exhibit morphophysiological dormancy and require release from both morphological and physiological dormancy before germination. Although some studies have proposed methods for increasing seed germination rates, the underlying mechanisms of its dormancy release process remain unclear. Here, we investigated metabolic alterations during dehiscence in P. ginseng to determine their potential roles in dormancy release. Methods: We compared the ginseng seed metabolome before and after dehiscence and the ginsenoside and phytosterol compositions of the seeds in both periods in the presence of related enzymes. Results: After seed dehiscence, the sugar, amino acid, and squalene concentrations were significantly altered, phytosterols associated with the stigmasterol biosynthesis pathway were increased, while ginsenoside and brassinosteroid levels were not significantly altered. In addition, squalene epoxidase, cycloartenol synthase, 24-methylenesterol C-methyltransferase, and the stigmasterol biosynthesis pathway were activated. Conclusion: Overall, our findings suggest that morphological activities that facilitate ginseng seed growth are the primary phenomena occurring during the dehiscence process. This study improves the understanding of P. ginseng germination processes and promotes further research of its germination and cultivation.

벼 밀양 23호$\times$기호벼의 재조합 자식계통에서 수발아성,건열 저항성 및 종자수명간 상관관계 (Relationships among Viviporous Germination, Dry-heat Tolerance and Seed Longevity in Milyang 23/Gihobyeo RILs)

  • 밀양 23호/기호벼 유래 164 RILs($F_{17}$)을 이용하여, 종자의 휴면과 관련이 있는 수발아 저항성, 건열 저항성 및 종자수명을 검정하고, 이들 상호간 상관관계를 조사하였다. 출수 40일째에 밀양23호와 기호벼의 수발아율은 0%, 7%이었고, M/G RILs은 0$\sim$50.9%의 범위를 보였으며, 평균 3.2%였다. $90^{\circ}C$, 24시간 건열처리후 밀양23호와 기호벼는 99.2%, 37.6%의 발아율을 보였고, $T_{50}$은 2.7일, 12.9일로 유의한 차이를 보였으며, M/G RILs의 평균 발아율은 72.4%였다. 상온저장 한 종자수명은 밀양23호는 수확후 3.5년까지도 80% 이상의 종자 발아율을 유지하였으나, 기호벼는 18개월 후부터 급격히 발아력이 감소하여(30%이하), 42개월 후 발아력을 완전히 상실하였다. M/G RILs의 종자활력은 수확후 42개월까지 71계통이 90%이상의 발아율을 보였다. 수발아 저항성, 건열 저항성 및 종자수명은 밀양 23호와 기호벼 간에 뚜렷한 품종간 차이를 보였으며, M/G RILs은 양친의 중간값에 가까운 평균값을 보였다. 건열저항성과 종자수명 간에는 고도로 유의한 정의 상관관계를 보였고, 수발아와 종자수명, 수발아와 건열저항성 간에는 부의 상관관계가 성립하였다.

콩나물 생육기간 중 사포닌 함량의 변화 (Changes of Soyasaponin Contents in Soybean Sprouts)

  • 콩 재배 및 이용확대를 위한 기초자료를 얻기 위해 발아과정 중 기능성 성분인 soyasaponin의 함량 변화를 조사하였다. 1. Soyasaponin I과 soyasaponin II의 함량은 품종에 따라 차이가 있었으나 발아과정 중 떡잎부위에서 함량이 10배 이상 증가하였고, soyasaponin I은 배축에서도 종자 당 이 $140{\sim}360{\mu}g$ 수준의 함량을 나타내었다. 2. Soyasaponin III 및 soyasaponin V의 함량은 발아를 통해 증가하는 경향이었고, soyasaponin V의 경우 배축에서 그 함량이 급격히 증가하는 것으로 나타났다.

  • E. colona 종자(種子)의 휴면성(休眠性) 및 광(光)과 발아관여기체(發芽關與氣體)에 대한 발아(發芽) 반응성(反應性)을 조사(調査)하였다. E. colona는 휴면상성(休眠賞醒)을 위해 후숙기간(後熟期間)을 요(要)하지 않하였다. 종피(種皮)를 통(通)한 수분(水分) 이동(移動)은 용이(容易)하였다. 수화(水和) 및 건조(乾燥)의 반복처리(反復處理)는 종자(種子)의 발아력(發芽力)을 감소(減少)시켰다. 24 시간(時間)의 종자(種子) 침윤(浸潤)으로 수분함량(水分含量)은 21%까지 증가(增加)되었으나, 실온(室溫)에서 13 시간(時間) 후(後)에는 원상태(原狀態)로 회복(回復)되었다. 종피(種皮)의 제거(除去)는 암조건하(暗條件下)에서 발아율(發芽率)을 현저히 증가(增加)시켰으며, 광(光)은 완전종자(完全種子)의 발아(發芽)를 촉진(促進)시켰다. 일광도(日光度) 51.9$Wm^{-2}$ 이하(以下)의 조건하(條件下)에서의 발아(發芽)는 현저하게 감소(減少)되었다. 광조사(光照射) 1일(日) 1시간(時間) 처리(處理)는 1일(日) 2 시간(時間) 이상(以上) 처리(處理)에 비하여 현저하게 낮은 발아율(發芽率)을 보였다. 청색광(靑色光) 조사(照射) 종자(種子)는 적색광(赤色光) 조사(照射)에 비하여 발아(發芽)가 지연(遲延)되었으며, 발아율(發芽率)도 낮았다. 광존재하(光存在下)에 외생적(外生的)으로 처리(處理)한 에칠렌 및 탄산가스는 발아(發芽)를 촉진(促進)시켰으며, 이 두 기체(氣體)의 동시처리(同時處理)는 공력효과(共力效果)를 나타내었다. 그러나 암조건하(暗條件下)에서는 이 두 기체(氣體)의 작용력(作用力)이 나타나지 않하였다.

Total Sugars, $\alpha$-amylase Activity, and Germination after Priming of Normal and Aged Rice Seeds

  • Osmoconditioning and hardening effects for the seed germination of normal and naturally aged rice seeds were studied through analyzing the total sugars and $\alpha$-amylase activity. The normal seeds which used to have high germination rate accelerated germination with the osmoconditioning at the suboptimal temperature of 17$^{\circ}C$. On the other hand, the aged seeds did not affect germination rate at $25^{\circ}C$, while increased germination rate and accelerated germination a little at 17$^{\circ}C$, Hardening of aged seeds increased germination rate by 10-15% compared with control seeds at both 17 and $25^{\circ}C$ and accelerated germination. Total sugar content and $\alpha$-amylase activity of normal seeds were higher than did aged seeds. The aged seeds with treatment of osmoconditioning and hardening increased total sugar content and $\alpha$--amylase activity, but hardening was more effective than osmoconditioning. The $\alpha$--amylase activity was positively correlated with the total sugar content and germination rate.

생물학적 검정법을 이용한 소규모 수계내 수질 오염물질의 환경독성 평가 -상추씨 발아시험과 Microtox 시험 비교- (Evaluation of Environmental Toxicities for Priority Water Pollutants in a Small Watershed by Bioassays - Comparision between Lettuce Seed Germination Test and Microtox Bioassay -)

  • Environmental toxicities of priority water pollutants were evaluated by two selected bioassays, Lettuce seed germination/elongation test and Microtox acute toxicity test. Toxic chemicals (heavy metals, polycyclic aromatic hydrocarbons, and phenolic compounds) inhibited the germination rate and root elongation of Lettuce seed, as well as the bioluminescence of Microtox bacteria. When test biota were exposed to target chemicals, the sensitivity of Lettuce bioassay was relatively lower than that of Microtox bioassay. However, Lettuce bioassay may be a good candidate for prescreening the environmental toxicities of priority water pollutants, since the testing method with Lettuce seed was relatively easier and more economic than with Microtox bacteria. Toxicity tests were conducted to compare the validity and sensitivity of both bioassays for sediment from a small stream passed through urban area as well as leachate from a municipal solid waste landfill. From experimental results, we found that Lettuce test and Microtox test are compensated each other as a battery of bioassay for evaluating the environmental toxicities of field samples obtained from a small stream contaminated by pollutants.

Effect of Sta-Green on Leaf and Stem Production of Angelica acutiloba

  • This study was conducted to develop effective production system in greenhouse for leaves and stems of Angelica acutiloba by fertilizing of Sta-Green in pots. The results obtained are summarized as follows. Germination rate of Angelica acutiloba seeds collected in 2003 was $13\%$, while germination rate of seeds Collected in 2004 was above $91\%$. Seed germination rate and plant biomass of Angelica acutiloba collected in 2004 were higher than seed gathering in 2003. Especially, plant growth and yield of Angelica acutiloba grown in pot(The pots was filled with soil mixtures of Sta-Green and Peat Moss mixed with 45:55 ratio.) was the highest. These results indicate that leaf and stem production of Angelica acutiloba can be improved by fertilizing of Sta-Green in pots and optimizing seed collecting time in greenhouse.

땅콩 종실의 크기와 자엽절단정도가 발아 및 수량에 미치는 영향 (Effects of Seed Size and Cotyledon Removal on Germination and Yields in Peanuts)

  • 땅콩품종에 대한 입중의 변이를 조사하였으며, 동일품종에서의 종자크기에 따른 발아력, 생육 및 수량을 비교하는 한편 자엽을 인위적으로 절단, 조절했을 때의 생육과 수량에 대한 반응을 검토하였다. 1. 입중의 변이는 소립품종보다 대립종에서 크게 나타났으며, 2. Cold test에 의한 발아시험결과 종자가 작을수록 발아율이 낮았으며 포장출아율도 떨어졌다. 3. 종자크기에 따른 후기생육의 차이는 없었으나 소립품종의 경우는 종자가 큰 것을 파종했을 때 수량이 많았다. 4. 자엽의 일부를 절단하였을 때는 개화성기의 생육이 정상종자보다 저조했으나 협비대기이후에는 차이가 없었다. 5. 소립품종에서는 자엽절단이 수량을 감소시키나대립종에서는 정상립과 차이가 없었다.
