Chemical Composition and Size of Floating and Sunken Eggs of Olive Flounder Paralichthys olivaceus

  • Olive flounder (Paralichthys olivaceus) is one of the most commercially important species of farmed marine fish in Korea. Although techniques for rearing the larva of this species are improving, production costs are also increasing due to microbial influences and genetic degradation caused by successive culturing. Since the demand for healthy seed, which requires eggs of good quality of, is high, we examined the effects of nutrient composition on the size of P. olivaceus eggs. We analyzed floating (live) and sunken (dead) eggs of P. olivaceus from five different hatcheries for their size and amino and fatty acid composition. The sizes of eggs and oil globules from floating vs. sunken eggs were significantly different at p<0.05. No significant relationships were observed, however, among larval length, hatching percentage, and egg and oil globule size. The dry weight and amino acid levels of floating eggs were greater than those of sunken eggs (p<0.05), but no difference in fatty acid content was observed.

Antimycotic Activity of Allium Sativum Against Beauveria Bassiana, Pathogenic Fungus of White Muscardine Disease in Silkworm, Bombyx mori L. (Lepidoptera: Bombycidae)

  • White Muscardine is the most common fungal disease of silkworm, Bombyx mori L. caused by the pathogenic fungus, Beauveria bassiana (Balsamo) Vuillemin. In the present investigation, an attempt has been made to screen locally available medicinal/ weed plants against Beauveria bassiana. Among the plant extracts (PE) tested, 5% aqueous crude extract of the bulb of Allium sativum (Garlic) has been found to be most effective against Beauveria bassiana. The radial growth of Beauveria bassiana in vitro was inhibited to the tune of 54.9% in aqueous extract and 54.4% in ethanolic extract of Allium sativum and correspondingly mycelial dry weight gave rise to 110.7 mg and 108.7 mg against 201.7 mg in control 15 days post treatment. Similarly, silkworm larvae topically inoculated with the Beauveria bassiana conidia ($1.8{\times}10^6/ml$) registered survival up to 53.0% against 0.0% in control after treatment with aqueous extract of Allium sativum. Simultaneously, as a preventive measure, silkworm larvae were put to rear in conidia contaminated seat paper instantly treated with aqueous extract of Allium sativum that also increased survival up to 61.0% against 4.6% in control. It is also observed that the plant extract is absolutely innocuous to silkworm.

Changes in Contents and Composition of Dietary Fiber during Buckwheat Germination (메밀 발아 중 식이섬유 함량과 조성의 변화)

  • To provide the efficient application scheme of buckwheat dietary fiber and basic information of seed germination, buckwheat(Fagopyrum esculentum Moench) was germinated at 10$^{\circ}C$ for 7 days and the contents and composition of the total dietary fiber(TDF), insoluble dietary fiber(IDF), soluble dietary fiber(SDF) wire examined at 24 hour intervals. The TDF content in ungerminated seeds was 24.86o on dry weight basis. It decreased for the 1st day of germination, but gradually increased for 7 days afterwords. The contents of IDF and SDF in ungerminated seeds were 22.05, 1.42% respectively. The IDF and SDF contents decreased in the initial stage of germination, but then gradually increased. The composition of the IDF and SDF in the TDF during the germination period showed different tendencies. The IDF decreased with germination time until 5 days and then increased. The SDV increased until 5 days and then decreased gradually. The TDF contents obtained by AOAC method were generally higher than those obtained by Prosky method. The TDF contents obtained by the two method, however, were very closely correlated (r=0.9966, p< 0.01) The IDF(X1) and SDF(X2) showed the significant regression equation(p<0.01) with the root length(Y). The equation was Y: -12.6681+0.5089${\times}$ 1 $\div$ 0.6022Xa and R2 was 0.968.

The Effects of Artificial Light Sources on Lettuce Seedling Vigor and Growth

  • The aim of this study was to investigate the effects of artificial light sources on the germination and seedling vigor of lettuce, aiming to identify the optimal artificial light source for producing high-quality seedlings. Lettuce cultivar of 'Tomalin' and 'Seonpunggold' exhibited the highest seed emergence in the Metal halide(MH) lamp and High-pressure sodium(HPS) lamp treatment group, while the emergence rate parameter, T50, was the fastest in the HPS lamp treatment group. Both cultivars showed good growth characteristics such as number of leaves, root length, and stem diameter under RGB-LED, and their seedling vigor was excellent as well. The plant height was smallest in the Red + Green + Blue LED treatment, but the leaves were round and thick, resulting in higher biomass and dry weight. Single light sources of Red LED and Blue LED led to reduced growth compared with that under the mixed light treatments. Chlorophyll content in lettuce varied with the type of artificial light, with both cultivars exhibiting the highest chlorophyll content in the Red + Green + Blue LED treatment. The most suitable artificial light for lettuce seedling growth was the Red + Green + Blue LED treatment.

The Effect of Different Concentration of Glyphosate on the Growth of Coconut Seedlings

  • Coconut (Cocos nucifera L) is one of the predominant plantation crops in Sri Lankan economy which is known to have existed for over thousands of years. During the past decades coconut production had been reduced by a significant quantity. The usage of poor quality planting materials is a major reason for the low coconut production. Thus much attention needs to be paid in coconut nurseries. Weed management is a critical management practice in the nursery. Though glyphosate application is becoming popular in nurseries it can affect weeds as well as coconut seedlings growth. Therefore the effects of glyphosate were evaluated by determining the growth of shoot and root of coconut seedlings. Poly bag nursery was prepared and three treatments were used. Treatments were no glyphosate and manual weeding ($T_1$), application of glyphosate 1.08 ai kg $ha^{-1}$ at 2 monthly interval ($T_2$) and application of glyphosate 1.44 ai kg $ha^{-1}$ at 2 monthly interval ($T_3$). Application of glyphosate at early stage of seedling growth had a no significant effect on growth parameters tested. However, the concentrations of glyphosate negatively affected numbers, volumes and dry weights of secondary, tertiary and quaternary roots at the latter stage of seedling growth. The leaf area and the height of seedling were significantly reduced by the highest concentration of glyphosate. Among the growth parameters tested, seedling girth and shoot dry weight were not affected by the application of glyphosate. These results revealed that the usage of glyphosate at both concentrations negatively affected root growth of coconut seedlings. Based on these results, the both concentration levels of glyphosate should be applied to coconut nurseries before sprouting the seed nuts.

Mitigation Effects of Foliar-Applied Hydrogen Peroxide on Drought Stress in Sorghum bicolor (과산화수소 엽면 처리에 의한 수수에서 한발 스트레스 완화 효과)

  • Global climatic change and increasing climatic instability threaten crop productivity. Due to climatic change, drought stress is occurring more frequently in crop fields. In this study, we investigated the effect of treatment with hydrogen peroxide (H2O2) before leaf development on the growth and yield of sorghum for minimizing the damage of crops to drought. To assess the effect of H2O2 on the growth of sorghum plant, 10 mM H2O2 was used to treat sorghum leaves at the 3-leaf stage during growth in field conditions. Plant height, stem diameter, leaf length, and leaf width were increased by 7.6%, 9.6%, 8.3% and 11.5%, respectively. SPAD value, chlorophyll fluorescence (Fv/Fm), photosynthetic rate, stomatal conductance, and transpiration rate were increased by 3.0%, 4.9%, 26.0%, 23.4% and 12.7%, respectively. The amount of H2O2 in the leaf tissue of sorghum plant treated with 10 mM H2O2 was 0.7% of the applied amount after 1 hour. The level increased to approximately 1.0% after 6 hours. The highest antioxidant activity measured by the Oxygen Radical Absorbance Capacity assay was 847.3 µmol·g-1 at 6 hour after treatment. However, in the well-watered condition, the concentration of H2O2 in the plant treated by the foliar application of H2O2 was 227.8 µmol·g-1 higher than that of the untreated control. H2O2 treatment improved all the yield components and yield-related factors. Panicle length, plant dry weight, panicle weight, seed weight per plant, seed weight per unit area, and thousand seed weight were increased by 8.8%, 18.0%, 24.4%, 24.7%, 29.9% and 7.1%, respectively. Proteomic analysis showed that H2O2 treatment in sorghum increased the tolerance to drought stress and maintained growth and yield by ameliorating oxidative stress.

Waterlogging Effects on Nitrogen Accumulation and $N_2$ Fixation of Supernodulating Soybean Mutants

  • Soybean is sensitive to waterlogging stress, leading to reduce their growth and yield significantly. The objective of this study was to characterize the relative sensitivities of biomass accumulation and specific nodule activity under waterlogging stress between supernoduating mutants, 'SS2-2' and 'Sakukei 4' and their wild-type soybeans, 'Sinpaldalkong 2' and 'Enrei', respectively. Flooding treatment was performed to soybean plants grown in a pot by waterlogging for 15 days from the beginning bloom(R1) stage under natural light. The nodule number and weight were considerably decreased by waterlogging stress. The bleeding sap rate of waterlogging soybean plants was decreased by 78-80% in supernodulating mutants and 65-74% in their wild types compared to control plants. The relative ureide-N content was also decreased by waterlogging and the reduction was high in supernodulating mutants. This may cause the massive reduction of shoot and root dry weight and leaf area in waterlogged soybean plants. There was a varietal difference in response to the waterlogging stress. During the waterlogging, supernodulating mutants maintained higher spad value than their wild types. Particularly, the difference between soybean varieties was clear in low rank leaves from the top. Also, supernodulating mutants showed a weak waterlogging tolerance than their wild types. Under waterlogging conditions, massive nodules were considerably destroyed and specific nodule activity after waterlogging may not be recovered when compared to their wild-type soybeans. Supernodulating mutants showed lower seed yield than their wild types in waterlogging conditions.

Dehisced Seed Germination and Seedling Growth Affected by Chilling Period in Eleutherococcus senticosus Maxim. (가시오갈피 개갑종자의 저온처리에 의한 발아 묘목의 생육특성)

  • This experiment was carried out to study the optimal chilling period for breaking the physiological dormancy of dehisced E. senticosus seeds and to investigate the critical seeding date in Kangwondo. Storage at $5^{\circ}C$ for 85 days was most effective in breaking dormancy of dehisced seeds, but didn't germinated synchronously. Only 28.1% of undehisced seeds germinated after 125 days of low temperature storage. For improving seedling survival rate in the field, 50% shading was more effective than 30% shading. Delayed seeding date resulted in decreased seedling fresh weight, dry weight, leaf area and root length, but seeding date has no significant affecte on stem length.

Studies on Major Agronomic Characteristics of Korean Artemisia annua L. (한국산 개똥쑥의 작물학적 특성)

  • In order to development medicinal Artemisia herbs of high quality, Korean A. annua L. were investigated with its ecological and morphological characteristics, and basic statistical data of agronomic characteristics. This species, which is annual herb, is mainly distributed to marginal land, riverside, roadside, grassland. Ecological niche is low species in competition of the others. Its pollination is basically anemogamous, but is frequency pollinated by insects. This species is characterized as tap root, 1~3 pinnate compound leaf of ovate or narrowly ovate, stem is green, erect and solitary. Inflorescence is paniculate, receptacle is not hair. Capitulum, consist of ray floret and disk floret, is subglobose shape. Additionally, this species could be easily discriminated from related Artemisia herbs by the capitulum size. Ray floret is female, disk floret is bisexual. Flowering season is from August to October. Seed is achene unattached hair. Stem length ranged from 179 cm to 225 cm, and stem diameter and number of branch were $17.14\;{\pm}\;1.68\;mm$, $2.43\;{\pm}\;0.51\;mm$, respectively. Length and width of leaf were $14.5\;{\pm}\;0.5\;cm$, $15.0\;{\pm}\;1.0\;cm$, and leaf number of main stem were $48.06\;{\pm}\;10.57\;cm$, respectively. Fresh weight of aerial parts and root were $364.7\;{\pm}\;14.1\;g$, $32.6\;{\pm}\;5.1\;g$, and its dry weight were $136.6\;{\pm}\;10.0\;g$, $14.9\;{\pm}\;2.34\;g$, respectively.

Growth Characteristics and Phenol Compounds Analysis of Collected Perilla frutescens Resources From China and Japan (중국과 일본 들깨 수집 자원의 생육 특성 및 페놀 성분 분석)

  • This study was conducted to find out the variation in agronomic trait and chemical composition in the collected Perilla frutescens from China and Japan. From the results of growth investigation, the maximum number if branches was 26.7ea in Japan 134 line, followed by 25 nodes number in China 119 line. Among the different lines investigated, maximum number of panicle number (108.8) were observed in China 114 line. 1000 seed weight was maximum (4.12 g) in China 118 line. Flowering time of different collected lines varied significantly with average value of 175.5 days and the average line required for maturation of seedlings was 205.1 days. Plant height was the highest (248.9 cm) in China 107 line. Highest number of total picking leaves was 965ea, and the average picked period was 54 days. The major phenol compounds contained in Perilla frutescens showed wide variation for Syringic acid, Benzoic acid, Naringin, o-Coumaric acid, Myricetin, Naringenin and Hesperetin. Japan 139 line showed the highest level of total phenol contents ($8254.0{\mu}g/g$, dry weight).
