강원지역 감자 역병균 Phytophthora infestans의 교배형 및 약제저항성 변화 (Changes of Mating Type Distribution and Fungicide-resistance of Phytophthora infestans Collected from Gangwon Province)

  • 강원지역의 감자포장에서 2006년과 2007년에 역병균을 분리하여, 교배형, metalaxyl과 dimethomorph 반응을 조사하였다. 각 지역에서 분리한 역병균의 교배형을 조사한 결과, 2006년 분리된 조사 균주는 155 균주 중 A1형이 58.7%, A2형이 41.3%로 확인 되었으며, 2007년 분리된 균주에서 A2 교배형은 6.7%로 발생 비율이 매우 낮았다. metalaxyl에 대한 반응성은 2006년 133균주에서 저항성 균주가 73.75%, 중도 저항성 균주가 18.8%, 감수성 균주가 7.5%로 나타났으며, 2007년 조사한 101 균주 중에서 저항성이 56.4%, 중도저항성 균이 4%, 감수성 균이 36.6%로 분리되었다. 강원도의 대부분 지역에서 A2 교배형 발생과 metalaxyl에 저항성을 갖는 균주를 확인할 수 있었는데, 이러한 결과는 병원균이 지속적으로 기주 및 환경요인에 따라 변화되고 있음을 의미한다. Dimethomorph 약제 반응 조사결과 MIC 값은 0.6미만, 06~1.0, 1.0이상인 세 개의 그룹으로 분류 되었으며, 2006년에는 $1.0\;{\mu}g/ml$이상인 균주는 56.3%이었으나 2007년에는 $1.0\;{\mu}g/ml$이상인 균주가 4%였다. $EC_{50}$ 값은 2007년에 조사한 균주들의 평균값이 $0.12\;{\mu}g/ml$로 2006년 평균값 $0.37\;{\mu}g/ml$ 보다 감소하였다. 감자 역병균들의 MIC 값과 $EC_{50}$ 값을 고려한 결과, 2007년에 조사한 병원균의 약제 반응정도가 2006년에 분리한 균주보다 감소하는 것으로 확인되었다.

흰목이버섯 대량생산을 위한 용기내 재배 최적화 연구 (Optimization of artificial cultivation of Tremella fuciformis in closed culture bottle)

  • 흰목이버섯 균주와 공생균을 수집하고 ITS 5.8S rDNA sequencing을 하여 유전자 서열을 분석하였다. Gene Bank Data homology search 결과 분리된 균의 rDNA 서열이 이 Tremella fuciformis AF042409의 rDNA 서열과 99% 일치하는 것으로 확인되었다. 그리고 함께 분리된 공생균은 같은 방법으로 Annulohhypoxylon stygium 으로 확인하였다. 분리된 T. fuciformis KG 103과 A. stygium KG 201 균주는 PD배지에서 각각 14 mm/14 days과 85 mm/14 days의 균사생육을 나타내었다. T. fuciformis KG 103 균주의 생육최적온도는 $25^{\circ}C$ (14mm/14days) 이었으며, $35^{\circ}C$ 고온과 $15^{\circ}C$이하 저온에서 균사 생장이 억제되었다. A. stygium KG 201은 흰목이버섯균과 유사한 최적온도를 나타내었다. T. fuciformis KG 103 균의 생육 최적 pH는 5.0이었으며, A. stygium KG 201도 pH 5.0에서 생육이 가장 왕성하였다. 흰목이버섯 종균용 최적 배지로 참나무톱밥 77.5%, 미강 20%, 석고 1.5%, 황백당 1%가 선정되었다. T. fuciformis KG 103과 A. stygium KG 201혼합 종균을 제조하고 흰목이버섯 자실체생산을 위한 병속재배 방법을 확립하였다. 콘코브(Corn cob) (77%와 52%)가 사용한 재료 중 최적의 자실체 성장률을 나타냈으며, 콘코브 함량을 줄일수록 생육이 저조하였다. 면실박과 참나무톱밥은 단독 사용시 생육이 저조하였고, 콘코브를 첨가시 수율이 증대되었다. 최적수분농도는 55%로 결정되었다.

나노 분광 센서를 이용한 씨감자 표면 표현형 분석 (Analysis on Surface of Seed Potato using Nano-Spectrometric Sensor)

  • 농산물의 품질 및 성분을 측정하는데 있어 기존의 화학적 분석 방식은 정밀도가 높으나 측정에 소요되는 시간과 비용이 많이 들어, 현장 적용하기에는 한계가 있다. 일반적으로 근적외선 분광 분석(Near Infra Red Spectroscopy, NIRS) 방법은 가공 과정에 따라 빠르게 변화되는 단백질 조성 및 수분함량 측정 등에 이용되고 있다. 분석에 소요 시간이 많이 걸리는 켈달법(Kjeldahi method)에 비해 NIR 분광 분석을 통한 보정으로 연속적인 모니터링이 가능하다. 본 연구에서 사용된 시료를 고정시키기 위한 프레임을 제작한 후 NIR센서와 광원인 LED의 각도를 고정시키고 측정 대상체인 사절된 감자 크기에 따라 시료를 고정시킬 수 있는 프레임을 반사면에 위치시켰다. 확산 반사법을 이용하여 프레임에 씨감자 시료를 고정 시킨 후 백색 LED를 이용하여 감자 표면에 빛을 반사시켜 3일 동안 12시간 마다 해당 시료들(열처리, 비누용액 침지, 생감자)의 스펙트럼을 측정하였다. 해당 시료들은 측정 기간 동안 저온상태($4^{\circ}C$)와 실온상태($20^{\circ}C$)에서 보관되었다. 실험 결과는 파장대 145nm에서 저온상태에서 보관된 생감자는 시간경과에 따른 흡광도의 결정 계수값($r^2$)은 0.98 이었다. 이는 감자가 저온에서 생감자의 상태 변화가 일어나고 있다는 것을 의미하고 파장대 145nm에서 시간에 따른 저온상태에서 보관한 감자의 상태 변화 예측이 가능함을 의미한다. 비누용액에 침지시킨 후 실온에 보관한 감자는 시간이 경과함에 따라 파장이 증가함에 따라 흡광도가 증가하였다. 이는 감자에 들어있는 Polyphenol Oxidase 함량 변화로 갈변 현상이 일어난 것을 알 수 있다. 또한 실온에서 보관한 생감자도 시간 경과에 따라 갈변 현상이 일어났지만 용액에 침지시킨 감자보다는 갈변 현상이 36시간 이후로 발견되었다. 열처리 후 실온에서 보관한 감자의 경우에는 갈변현상이 나타나지 않았다. 저온상태에서 보관한 감자시료들 모두 갈변형상이 나타나지 않았지만, 24시간이 지난 후 용액에 침지시킨 감자는 갈변 현상이 발생되었다. 생감자와 열처리한 감자는 시간 경과에 따른 갈변현상이 일어나지 않았으므로, 감자의 갈변현상은 감자의 표면 처리 방법에 국한되지 않고 온도에 영향을 더 많이 받는다는 것을 나타내고 있다. 본 연구는 향후 감자의 품질 및 성분 측정에서 간편하게 사용될 수 있는 감자의 품질 계측 기술에 기여할 것으로 판단된다.

위생교육에 따른 선식 제조기구와 작업장의 위생상태변화 및 일부 선식제품의 안정성과 영양적 평가 (Nutritional Evaluation, Stability of Cereals and Sanitation Status of Processing Utensils and Environments Based on Hygiene Education)

  • This study was carried out to investigate the effect of hygiene education on the microbiological changes of processing utensils and the environmental in the manufacture of cereals and to evaluate the Cd, Pb contents and nutrient compositions of 11 cereals. The result of microbiological evaluation was that fungi, coliforms and staphylococcus species were detected in employees, on equipment, utensils and environments in the first inspection. Fungi were detected in most of the cereals, staphylococcus species were detected in soybeans, perilla seeds and sea tangle How, and bacillus cereus was detected in sorghum and black sesame seeds. The water content of rice, barley, glutinous rice, brown rice and carrot flour in packaged products, and in carrot flour, angelica keiskei, carrot, sea mustard and potato in bulk products was in excess of 8.0%. The Pb content of cereals was 0.14-0.51 mg/kg and Cd was not found. The acid value of Job s tears flour and black sesame seed flour was higher than 5.0 mg/g oil. Cereals were manufactured from 41 different cereals and grains, legumes, seeds and nuts, vegetables, potatoes, seaweeds, fruits, glucose and salt. The average content of cereals and grains, legumes, and seeds and nuts in cereals was 75.75%, 16.19% and 4.93%, respectively. The mean nutrient content per 100 g of cereals was calories 365.8kcal, protein 13.3 g, fats 5.9 g, carbohydrates 63.4 g, Ca 91.8 mg, p 269.9 mg, Fe 3.15 mg, Na 76.2 g, K 421.8 mg, Zn 2.33 mg, Vit. A 12.5 R.E., Vit. B$_1$0.23 mg, Vit. $B_2$ 0.16 mg, Vit. $B_6$ 0.46 mg, Niacin 3.5 mg, Vit. C 1.36 mg, folic acid 62.3 $\mu$g and Vit. E 1.24 mg. When nutrients value of 48 g of cereals and 200 $m\ell$l of milk was compared to 1/3 of the RDA, the values were below than 60% of 1/3 of the RDA except Na, K, Vitamin $B_1$ and C contents. The average carbohydrate : protein : fat ratio of energy intake was 54.27 : 17.45 : 28.28, respectively. Therefore, a training program consisting of the education of the staff in surveillance and standard operating procedures, the elimination of dangerous procedures, sanitation checklist, the implementation of Preparation methods and standard recipes for cereals is required.

중부지방 적합 자운영 (Astragalus sinicus L.) 형질 특성 및 유전적 연관성 분석에 관한 연구 (Study for Morphological and Genetic Characteristics of Chinese Milk Vetch (Astragalus sinicus L.) to Select Suitable Line in Central Area of Korea)

  • 본 연구는 중북부지방 적응성 자운영 선발 및 수집 계통간 형태적, 유전적 근연관계를 분석한 것으로서 총 수집된 16개 자운영 계통 중 파주종은 엽각이 적고, 지하부의 발육이 좋아 형태적 형질이 월동에 적합한 초형이었다. 또한 구례 수집종은 개화기가 타 수집종에 비해 일주일 가량 빨랐다. 유전적 근연관계를 검정하기 위해 8개 primer 조합으로 AFLP를 실시한 결과 총 579개의 밴드를 얻었으며, 이 중 polymorphism을 갖는 밴드는 61.54%인 336개 밴드였다. AFLP 데이터를 이용해 군집분석을 실시한 결과 계통 간 유사도 지수는 0.826~0.939 사이의 값을 가지며, 구례와 파주 수집종은 타 수집종과 유전적 유사도에서 비교적 큰 차이를 보였다.

멸종 위기에 처한 Bulbophyllum auricomum Lindl. orchid의 시험관 내 종자 발아를 통한 구근 절편체의 식물 재생 및 캘러스 유도에 대한 천연 추출물 및 식물 성장 조절제(PGR)의 효과 (Effects of Different Natural Extracts and Plant Growth Regulators on Plant Regeneration and Callus Induction from Pseudobulbs Explants through in vitro Seed Germination of Endangered Orchid Bulbophyllum auricomum Lindl.)

  • Bulbophyllum auricomum Lindl.은 희귀 난초이며 매력적인 향기가 지닌 꽃이 피어난다. 본 연구는 미세 증식을 위한 조직 배양 방법을 조사하였다. 인공 자가 수분으로부터 파생된 캡슐은 MS 기본 배지에서 최상의 종자 발아를 위해 얻어진다. 식물 성장 조절제(1.0 mg/L의 BAP 및 2.0 mg/L의 NAA)는 원괴체 유래 유사구근 절편체의 캘러스 유도에 영향을 받았다. 캘러스 계대배양을 위해 10가지 처리 배지에서 다른 천연물 추출물을 테스트하였다. 이 중 150ml/L 코코넛 워터를 첨가한 MS 배지는 일반적으로 1개월 및 2개월에 생중량(1.75 ± 0.08) 및 (3.01 ± 0.20) 캘러스 증식 및 PLB 유도에 효과가 있었고, 그 다음으로 바나나 30g/L와 감자 추출물 20g/L의 MS 조합이 뒤를 이었다. 식물 생장 조절제와 천연 추출물을 함께 첨가한 MS배지와 천연 추출물만 첨가한 MS 배지를 비교 연구한 결과, 천연 추출물(코코넛 워터 150ml/L)과 식물 생장 조절제(BAP 2.0 mg/L 및 NAA 1.0 mg/L)를 혼합하여 MS 배지에 첨가한 결과 배양 1개월 및 2개월 후에 각각 가장 높은 새싹 재생수로 (3.37 ± 0.17) 및 (6.41 ± 0.68)을 얻었다.

이비인후, 안, 치아 및 피부증상의 민간요법에 관한 고찰 (Study on the Herb Remedies of ENT, Eyes, Teeth and Skin Problems)

  • The intention of this study is to apprehend the con. tents and methods of herb remedies that are commonly conducted when there are health-problem cases of ENT, eyes, teeth and skin. Methods of this study are divided into two stages : 1) For a period of six months from December 1994 to June 1995, some 40 persons who are believed to be well versed with herb remedies have been randomly chosen, and we made a survey on herb remedies by symptoms: and 2) we have endeavored to make their grounds evident through the studies on literatures with the focus on the basic data collected. Their results are as follows: 1) When one feels a pain in ears, such herb remedies are employed as pouring the vapor into ears, which is made by steaming Alaska pollack, or as applying or wiping with the juices of radish or the ginkgo, or' alum. Applying the radish juice is effective for sterilization and fever removal: and applying the ginkgo juice is effective for cleanliness. But, plastering alum, sesame oil or castor oil, or pouring the vapor of Alaska pollack into ears are perhaps effective but do not have any pharmacologic grounds. 2) When one bleeds at the nose, such kinds of herb remedies are applied as stimulating nose or head with cold water, pressing nose or ridge of nose, or filling up nares with mashed mugworts. In addition, they have utilized garlic or leeks. Such methods as stimulating with cold water or just pressing nose and ridge of nose is based on reasonable grounds, i.e. vasoconstriction and vascular compression ; and applying mashed garlic on the sole of foot is good for the circulation of Qui ; and the use of mugworts and leeks is based upon the pharmacological function of hemostasis. 3) When one feels a sore throat such kinds of herb remedies are employed as gargling or rinsing throat with brine, drinking hot gruel or water, or drinking the juice of mugwort, radish, ginger or Chinese quince. Gargling with brine or drinking the juice of mugwort, radish or ginger is based upon the pharmacological function of pain alleviation, fever removal, and detoxication. 4) When a boil is formed in mouth, such herb remedies are applied as spreading honey, brine or alum water, and taking gall nut, Chinese matrimony vine, lotus root, etc, for drugs. Spreading honey, brine or water that is made by infusing gallnut, Chinese matrimony vine, lotus root is based upon such functions as hematosis, astriction, antibacterial, and antiphlogistic, Alum, eggplant and licorice are said to be effective, but their pharmacological effects have no grounds. 5) When one has conjunctivitis such herb remedies are commonly applied as irrigation with brine and dropping breast milk in eyes. Moreover, such other drugs are used as plantain. shepherd's purse, and purslane, etc. The use of brine, breast milk, plantain, shepherd's purse and purslane is based upon such functions as sterilization, antiphlogistic, disinfection and pain relieving. Eriocaulon sieboldianum, bean stem, bean pod and narcissus leaves are said to be effective, but their pharmacological action have no basis. When one has a stye, such herb remedies are applied as extracting eyelashes, stimulating by a massage of middle finger, third finger or big toe, as well as sear ing with a heated bamboo comb that is fine-toothed. Other than these, plantain and nightshade's nuts are used as drugs for it. Extracting eyelashes corresponds with exclusing suppurative node and draining the stye of pus ; and the use of plantain is based upon disinfection: and nightshade's nuts are said to be effective, however, their pharmacological action has no grounds. 6) For a treatment of toothache, such herb remedies are commonly employed as rinsing mouth with brine and holding cold water or gasoline in the mouth ; and as the drugs that are believed to be effective have been Welsh onion, ginger and castor-oil, plant, etc. The use of Welsh onion is based upon pain killing, antiinflammatory actions, and the use of ginger is based upon detoxication and disinfection ; and seeds of castor-oil plants are said to be effective, but they have no pharmacological basis. 7) When one has hives, such herb remedies are commonly applied as rubbing burned straw in affected parts, exposing to its smoke, rubbing with salt, sweeping down with a broom, and spreading and drinking boiled water of trifoliate orange. The use of cassia tora seeds, walnut, aloe and radish is said to be effective. The use of cassia tor a seeds has the functions of intestinal order, anti-paralysis, etc. The use of walnut has resulted in an increase of blood by invigorating spirits ; and the use of aloe is based upon disinfection, antibiotic, anti-salt, antihistamine and detoxication action. But, the effects of radish juice and straw's smoke have no pharmacological grounds. 8) When one gets a boil, such herb remedies are commonly used as applying a plaster, paste of flour mixed with yolk, soy sauce or honey, as well as spreading pounded elm tree. Other remedies that have been said to be effective are ; heating with mugwort, brine, wild rocambole, aloe, onion, squid's bone, etc. The use of mugwort is based upon pain killing, astringent antiinflammatory and tranquility. Wild rocambole is based upon the generation and maintenance functions of cell-joining textures ; elm tree upon antiphlogistic ; aloe upon fever removal and antiphlogistic ; onion on pain killing, fever removal, antiphlogistic and tranquility ; squid's bone on astriction: and brine or vinegar on sterilization. Pine resin and gardenia seed are said to be effective, but they have no pharmacological basis. 9) When one cuts his skin, such herb remedies are commonly employed as spreading mugwort's juice or squid's bone powder, or pressing the wounds. In addition, kalopanax, onion and fine soil are employed. The use of mugwort, kalopanax and squid's bone is based upon such functions as hemostasis, sedation, pain killing, antibacterial ; and fine soil is said to be effective, but it has no pharmacological basis. 10) When one suffers from whitlow, such herb remedies are commonly utilized as heating with boiled soy sauce, spreading soybean paste, or dipping into eggs, etc. Other drugs that have been employed are onion root, brine, eggplant, potato, loach, etc. The use of onion is based upon pain killing and antiphlogistic functions ; and that of brine upon antiphlogistic function. The use of soy sauce or soybean paste, fomentation, eggplant, potato and loach is said to be effective, but it has no pharmacblogic ground. 11) For the treatment of frostbite, such herb remedies are commonly used as dipping the affected part into frozen soybean sack, using boiled water of eggplant stem, garlic caulis, onion, hot pepper, caulis. Onion is based upon antiphlogistic and tranquility actions garlic upon disintection, metabolic exacerbation, tonic and aphrodisiac actions and the use of eggplant and hot pepper is based upon help blood circulation, dissolution and excretion of waste matters in vein. 12) For the treatment of burn, such herb remedies or drugs are commonly used as cleansing with Korean gin, spreading eggs, cleansing with cold water and soap water ; and as brine, cactus, moss, soybean paste, oil, etc. The cleansing with Korean gin, cold water, soap water, brine, vinegar is based upon cleaning and sterilizing functions ; and the use of cucumber is based upon nu. trition provision, and strengthening of resisting power by adjustment of metabolism. The use of potato, cactus, moss, oil and eggs is said to be effective, but their phamacological functions are not clarified. In view of the above results, we can realize that the drugs that have been employed in herb remedies are quite diverse. However, in regard to majority of herb remedies that have been employed by symptoms, the pharmacological functions of their drugs have not been clarified, and they are merely known as effective. Furthermore, they have not been recorded in the literature as yet ; and we confirm that there have been many herb remedies that were executed without the proper knowlege of their effects. It is now our view that the results of this survey may be utilized for consulting data in regard to the use of herb remedies.

"감자에 대한 항생제(抗生劑) 피마리신의 통계적(統計的) 효과(效果) 분석(分析)" (INVESTIGATION OF THE EFFECT OF AN ANTIBIOTIC "P" ON POTATOES)

  • An antibiotic 'P', which is one of the products of the Gist Brocades N. V. is being tested by its research department as fungicide on seed-potatoes. For this testing they designed experiments, with two control groups, one competitor's product, eight formulations of the antibiotic to be tested in different concentrations and one mercury treatment which can not be used in practice. The treated potatoes were planted in three different regions, where bifferent conditions prevail. After several months the harvested potatoes are divided in groups according to their diameter, potato illness is analysed and counted. These data were summarised in percentage and given to us for Analysis. We approached and analysed the data by following methods: a. Computation of the mean and standard deviation of the percenage of good results in each size group and treatment. b. Computation of the experimental errors by substraction of each treatment mean from observed data. c. Description of the frequency table, plotting of a histogram and a normal curve on same graph to check normality. d. Test of normality paper and chi-sqeare test to check the goodness of fit to a normal curve. e. Test for homogeneity of variance in each treatment with the Cochran's test and Hartley's test. f. Analysis of Variance for testing the means by one way classifications. g. Drawing of graphs with upper and lower confidence limits to show the effect of different treatments. h. T-test and F-test to two Control mean and variance for making one control of Dunnett's test. i. Dunnett's Test and calculations for numerical comarision of different treatments wth one control. In region R, where the potatoes were planted, it was this year very dry and rather bad conditions to grow potatoes prevailed during the experimental period. The results of this investigation show us that treatment No.2, 3 and 4 are significantly different from other treatments and control groups (none treated, just like natural state). Treatment no.2 is the useless mercury formulation. So only No. 3 and 4, which have high concentrations of antibiotic 'P', gave a good effect to the potatoes. As well as the competitors product, middle and low concentrated formulations are not significantly different from control gro-ups of every size. In region w, where the potatoes got the same treatments as in region R, prevailed better weather conditions and was enough water obtainable from the lake. The results in this region showed that treatment No. 2, 3, 4, and 5 are Significantly different from other treatments and the control groups. Again No.2 is the mercury treatmentin this investigation. Not only high concentrated formulation of antibiotic 'P', but also the competitor's poroduct gave good results. But, the effect of 'P', was better than the competitors porduct. In region G, where the potatoes got the same treatments as in the regions R and w. and the climate conditions were equal to region R, the results showed that most of the treatments are not significantly different from the control groups. Only treatment no. 3 was a little bit different from the others. but not Significantly different. It seems to us that the difference between the results in the three regions was caused by certain conditions like, the nature of the soil the degres of moisture and hours of sunshine, but we are not sure of that. As a conclusion, we can say that antibiotic 'P' has a good effect on potatoes, but in most investigations a rather high concentration of 'P' was required in formulations.
