Identification of Seed-borne Penicillium spp. on Gramineae Crops Based on Morphological Characteristics (형태적 특성에 의한 벼과작물 종자전염 Penicillium spp.의 동정)

  • A total of 81 isolates of Penicillium were isolated from postharvest seeds of barely, Job's-tears, maize, sorghum and rice from 1997 to 2003. Based on the morphological characteristics, they were identified as P. chrysogenum, P. citrinum, P. cyclopium, P. oxalicum, P. polonicum, P. purpurogenum and P. viridicatum. P. chrysogenum was detected from Job's-tears, rice and sorghum seeds, P. citrinum from maize seeds, P. cyclopium from sorghum seeds, P. oxalicum from barely, maize, sorghum and rice seeds, P. purpurgenum from maize, rice, sorghum seeds, P. viridicatum from Job's-tears, maize and rice seeds, P. polonicum from Job's-tears, maize, rice and sorghum seeds. Among these species, P. cyclopium, P. polonicum and P. purpurogenum were first reported in Korea. Especially, about 50% of the Penicillium isolates detected from the seeds were P. polonicum. Identification of the Penicillium species using morphological characteristics was difficult especially for the species belonging to the subgenus Penicillium such as P. polonicum.

Major Morphological Characteristics of Safflower(Carthamus tinctorius L.) Accessions (홍화 수집종의 주요 형태적 특성)

  • Morphological characteristics of 16 collected safflower (Carthamus tinctorius L.)accessions were studied at Gyongbuk A.T.A. Uisong Medicinal Plant Experiment Station in 1998. As for plant height, 5 lines were tall(more than 131cm), which were mainly foreign safflower. Middle and small were 5 and 6 accessions, respectively. Number of branch per plant was 11∼13 in 2 accessions of domestic safflowers while foreign safflowers have more than 50 branches per plant. Leaf shapes were oblong(3 lines), lanceolate(10 lines) and broadly linear(3 lines). There were three types of leaf margin shape which were repand(7 lines), serrate(4 lines) and parted serrate(5 lines). Flower color was white(1 line), yellow(13 lines) and red orange(2 lines). As for flower head shape, 14 lines were conical and 2 lines were flattened. There were thorns on bract of flower head in 11 lines but in 5 lines there were not. Domestic safflower lines had seeds of conical shape. Contrastively, seed shapes of foreign saffower lines were oval.

Morphological Characteristics and Function of Hilum in Safflower Seed Germination (잇꽃 종자의 발아에서 제(Hilum)의 형태적 특성과 기능)

  • This research was carried out to clarify the germination characteristics with reference to hard seed coat in safflower. Morphologically, seed coat surface has hilum and micropyle which were evident during seed development stage. In the flower of developing seeds, the hilum area is connected with placenta of maternal tissue while the micropyle area is connected with the style of pistil. When the seeds imbibed, the hilum surface began to crack and the embryo protrudes through the hilum. To investigate the route for moisture absorption and gas exchange on the seed coat, the hilum and the micropyle were artificially sealed by paraffin. The seeds whose hilum were sealed could not germinate, which indicates that the exchange of moisture and oxygen takes place through hilum in safflower seeds. The germination was tested at $15^{\circ}C$, $20^{\circ}C$, and $25^{\circ}C$ by three substrates with different moisture conditions; top of paper method (hilum submerged in water), between-paper method, and soil seeding. The germination percentages were 31.3% at $15^{\circ}C$, 15.7% at $20^{\circ}C$ and 6.0% at $25^{\circ}C$ in the top of paper method; and 45.5% at $15^{\circ}C$, 30.0% at $20^{\circ}C$ and 14.0% at $25^{\circ}C$ in the between-paper method; and 80.0% at $15^{\circ}C$, 77.0% at $20^{\circ}C$ and 78.0% at $25^{\circ}C$ in the soil seeding, respectively. When the internal structure of hilum was investigated through SEM, it was found out consisting of vascular bundle element. In conclusion, the hilum of safflower seed was closely related with water absorption and gas exchange during initial germination process.

Morphological Variation of Foxtail Millet (Setaria italica (L.) P. Beauv.) Germplasm Collected in Korea, China and Pakistan (우리나라와 중국, 파키스탄에서 수집한 조 계통들에 대한 형태적 변이)

  • To understand the morphological differentiation of the 26 accessions of Foxtail Millet collected in Korea (15 accessions), China (7 accessions) and Pakistan (4 accessions), we analyzed 9 morphological characteristics such as plant height, panicle length, leaf number, tiller number, heading time, seed weight and panicle color etc. Most accessions of foxtail millet collected in Korea showed late heading time, tall plant height and long conical panicles. While foxtail millet accessions of Pakistan showed early heading time, short plant height and short conical panicles. In case of Chinese accessions, some accessions of them showed similar characteristics with Korean accessions, and the other showed similar characteristics to Pakistan accessions. In ANOVA analysis, most of quantitative characteristics such as plant height, leaf number, internode number and heading time showed significant differences among foxtail millet accessions collected from Korea, China and Pakistan. Principal component analyses clearly discriminate foxtail millet accessions of Korea from those of China and Pakistan. In PCA analysis, most of quantitative characters such as panicle length, leaf number and internode number greatly contributed in positive direction, whereas several quantitative characters such as tiller number, seed weight and panicle color contributed in negative direction on the first axis. Thus, these morphological characteristics could be used to classify the foxtail millet accessions collected in Korea, China and Pakistan. The present results could expand our understanding of the morphological variation in foxtail millet accessions from Korea, China and Pakistan, and also could be useful for foxtail millet germplasm preservation.

Morphological Variation and Genetic Relationship among Populations of the Shortnecked Clam Ruditapes philippinarum Collected from Different Habitats

  • The characteristics of the populations of shortnecked clam (Ruditapes philipinarum) originated from three different seed-production sites, Hadong, Kochang and Ulsan along the coast of Korea, were analysed by the morphological differences and the random amplified polymorphic DNA (RAPD) profiles. The morphology of the shell and survival rate for each population were also investigated 13-months after transplantation to a farming site in Shinsung. The morphology of the populations from Hadong and Kochang showed significant differences (P<0.05), but one year after transplantation to Shinsung, the morphology of these three populations was no longer significantly different (P>0.05). The template DNA for RAPD was efficiently extracted from the digestive diverticula of the clams. Up to 13 of amplified fragments were detected using arbitrary primers. Within the species of R. philipinarum, the genetic similarities ranged from 0.196 to 0.259. The populations from Hadong and Ulsan showed the highest similarity. The survival rates of the populations from Hadong $(69.4\%)$ and Ulsan $(63.8\%)$ were higher than that from Kochang $(41.7\%)$ 13-months after transplantation. From the RAPD analysis, it could be used as one of the primary criterion in determining which shellfish populations among various seed-production sites tend to be genetically similar and more adaptable and transplantable to a farming site.

Taxonomic Study of genus Sedum and Phedimus (Crassulaceae) in Korea Based on External Morphology (외부형태 형질에 근거한 한국산 돌나물과내 돌나물속과 기린초속의 분류학적 고찰)

  • The genus Sedum comprises about 400 taxa with worldwide distribution, and about 20 taxa of the genus Phedimus distributed from Asia to Europe. According to some taxonomists, Phedimus and Sedum are two separate genera of the family Crassulaceae; however, in Korea Phedimus has been treated as a subgenus Aizoon within the genus Sedum. This ambiguous taxonomic treatment of the genus Phedimus needs further confirmation. In the study, we investigated morphological characteristics such as the whole plant height, leaf morphology, and seed coat features of these two genera. We believe that these characters are very important to distinguish Phedimus and Sedum. The result indicated that the seed surface pattern was the most useful diagnostic character. The Sedum had a hexagonal pattern with a single nipple protrusion, whereas the Phedimus had a rectangular pattern with papilla on both ends. The results of this study strongly support the separation of Phedimus from Sedum and thus Phedimus need to be treated as an independent genus.

Characteristics and Genetic Segregation of a Rolled Leaf Mutant in Rice

  • Leaf structure is one of the important agronomic traits. A rolled leaf mutant was induced from an ethyl methane sulfonate (EMS)-treated japonica rice, 'Koshihikari'. The rolled leaf mutant showed phenotypes of reduced leaf width and leaf rolling. In addition, several abnormal morphological characteristics were observed, including dwarfism, defected panicle, delayed germination, and lower seed-setting. Microscopic analysis revealed that the number of small veins was decreased and the sizes of adaxial bulliform cells were reduced in the mutant leaves. The genetic study with two $F_2$ populations from the crosses of the rolled leaf mutant with 'Koshihikari' and Milyang23 suggested that the mutant phenotype might be controlled by a single dominant gene.

Study on Morphological Characteristics of Maiden silvergrass(Miscanthus sinensis) Cultivars (참억새(Miscanthus sinensis) 재배종의 형태적 특성에 대한 고찰)

  • 참억새(Miscanthus sienesis)는 화본목 벼과 억새속에 속한 다년생 단자엽식물로 동아시아와 오세아니아 동쪽 해역에 자생하고 있으며, 국내에서는 관상가치가 높아 정원용이나 조경용 소재로 이용되고 있는 대표적인 산림식물이다. 억새속(Miscanthus)은 물억새(M. sachariflorus), 참억새(M. sinensis), 억새아재비(M. longiberbis), 장억새(M. changii)로 분류하고 있으며, 이는 종을 구분하는 화기구조가 작고, 변이가 다양하기 때문이다. 그 중 참억새는 이러한 변이를 이용하여 다양한 초장과 엽 무늬를 가진 재배품종이 많이 개발되었으며, 내한성 또한 강해 정원용 관상식물로서의 가치가 높이 평가되고 있다(Lewandowsik et al, 1964). 지금까지 참억새 재배품종은 엽 무늬를 중심으로 개발되어 품종간 높은 유사성을 가지고 있어 구별하는데 어려움이 있었다. 본 연구에서는 새로운 참억새 품종개발시 기존품종과의 차이를 구별할 수 있는 기준을 제시하고자 기존 참억새 재배품종들에 대해 형태적 특성을 조사 분석하여 고찰해 보았다. 조사결과 가로무늬를 가진 'Strictus'와 'Zebrinus' 품종은 형태적으로 매우 유사하였으나 전체적인 모습에서 'Zebrinus'가 'Strictus' 품종에 비해 잎의 끝부분이 늘어지는 정도가 커 처진 특성을 나타내었다. 또한, 세로무늬를 가진 'Cosmopolitan'과 'Carbaret' 품종의 경우 초장 및 엽신의 길이가 유사하였으나 엽 무늬 위치에 차이가 있는 것으로 나타났다. 엽 폭이 좁은 세엽종인 'Morning Light'와 'Little Kitten' 품종은 형태적 차이는 없지만 가장자리 엽 무늬 유무로 구분이 가능하였다. 반면에 'Adagio'와 'Little Kitten, 품종은 형태적으로 매우 유사하여 좀 더 세밀한 조사가 필요할 것으로 보인다. 앞으로 더 많은 참억새 재배품종의 형태적 특성을 조사한 자료를 이용한다면 신품종 개발시 기존 재배품종과의 차이를 비교하는데 효율적으로 이용될 것으로 기대된다.

Evaluation of Horticultural Characteristics on Water Dropwort (Oenanthe stolonifera DC.) Genetic Resources for Various Utilization

  • Water dropwort (Oenanthe stolonifera DC.) is a perennial herbaceous plant that grows wild throughout Korea. As of 2020, 24,819 tons (51.7% of open field, 48.3% of facility) are produced in 1,278 ha (59.9% of open field, 40.1% of facility) nationwide. Water dropwort, which is rich in nutrients such as vitamins and iron, is mainly cultivated by vegetative propagation method using local traditional species, however, seed propagation and breeding of cultivars are insufficient so far. Since securing, propagating and continuous characterization of various genetic resources are required to breed new cultivars, this study was conducted to compare the main characteristics of domestic genetic resources and to improve their utilization. Growth characteristics such as plant height, fresh weight, plant type, and flowering date were investigated for the 89 varieties of genetic resources owned by Jeollabuk-do Agricultural Research and Extension Services after vegetative propagation in the individual pots. Also, the morphological image information of leaves and flowers was constructed. Genetic resources were collected nationwide and originated in 30 regions including Hwaseong, Siheung and Jeju. Their plant types could be classified into straight, intermediate and creeping types according to their morphological characteristics, and at this time, the number of intermediate types accounted for the largest proportion. Flowering was carried out under high-temperature and long-day conditions in summer. According to the flowering date, they could be classified into early, middle and late flowering varieties, and at this time, the middle flowering varieties occupied the largest proportion. As a result of the investigation of vegetative growth characteristics, varieties with long plant height, heavy fresh weight and thick stem thickness were identified. Along with this result, it is thought that classification and selection of genetic resources for various purposes will be possible through additional investigations such as analysis of components and antioxidant activity. Moreover, it is judged that such results can be used as basic data for breeding new water dropwort cultivars in the future.

Phylogenetic Relationship of Araliaceae in Korea by Seed Morphological Characteristics (종자 외부형태학적 특성에 의한 한국산 두릅나무과(Araliaceae) 식물의 종간 유연관계)

  • The purpose of this study is to describe and compare the morphological characteristics of seeds of 12 taxa of Araliaceae, and to evaluate their possible use for taxonomic considerations. For light microscope observations and measurements, the seeds were observed using Image Analyzing System. The observations were made on twenty randomly selected seeds of each species. Obtained data were statistically processed using analysis of variance. Principal Components Analysis indicated four groups of characters, the genera Aralia, Eleutherococcus, Panax and the others, which explained 65.47% of the total variation. As a results of Cluster Analysis using the eleven variables, 12 species of Araliaceae were also discriminated into four groups. Eleutherococcus senticosus and E. gracilistylus were closely related, which is well supported by the results from recent molecular studies. Also, the genera Dendropanax and Eleutherococcus were closely related in terms of seed characters.
