Terminal-restriction fragment length polymorphism (T-RFLP) analysis has been optimized by using in vitro model community composed of genomic DNAs of known bacterial strains and has been applied to assess the bacterial community structure in marine sediments. The specific fluorescence-labeled terminal restriction fragments (T-RFs) between 39 and 839 base long specifying each strain were precisely measured for known bacterial strains. The addition of a co-solvent (dimethylsulfoxide or glycerol) into PCR reactions has reduced differential PCR amplification. Comparative bacterial community structure was investigated for pristine and polluted sediments. A complex T-RFLP pattern showing complex bacterial community structure was obtained in the pristine sediment, whereas simple T-RFLP pattern (low bacterial diversity) was shown in polluted sediments where caged aquaculture has been conducted for several years. The results of T-RFLP analysis were compared with that of cloning and sequencing 16S rDNA clones from the same sediments. Sequence analysis of 16S rDNA clones (72) of the pristine sediment revealed a diverse collection of lineages, largely of the class Proteobacteria ($6\%$ alpha subdivision, $46\%$ gamma subdivision, $13\%$ delta subdivision, and $3\%$ epsilon subdivision), Nitrospina $(8\%)$, high G+C gram positive $(8\%)$, Verrucomicrobia $(7\%)$, and Planctomycetes $(6\%)$. In the contaminated sediments, 17 $(59\%)$ of the 16S rDNA clones (29) were related to Campylobacter and symbiont of Rimicaris exoculata belonging to epsilon subdivision of Proteobacteria. The results obtained indicated that T-RFLP analysis is a rapid and precise technique for comparative bacterial community analysis.
Variation in the microbial biomass and community structure found in sediment of heavily polluted bays and the adjacent unpolluted areas were examined using phospholipid fatty acid analysis. Total microbial biomass and microbial community structure were responding to environmental determinants, sediment grain size, depth of sediment, and pollution due to petroleum hydrocarbons. The marker fatty acids of microeukaryotes and prokaryotes - aerobic, anaerobic, and sulfate-reducing bacteria - were detected in sediments of the areas studied. Analysis of the fatty acid profiles revealed wide variations in the community structure in sediments, depending on the extent of pollution, sediment depth, and sediment grain size. The abundance of specific bacterial fatty acids points to the dominance of prokaryotic organisms, whose composition differed among the stations. Fatty acid distributions in sediments suggest the high contribution of aerobic bacteria. Sediments of polluted sites were significantly enriched with anaerobic bacteria in comparison with clean areas. The contribution of this bacterial group increased with the depth of sediments. Anaerobic bacteria were predominantly present in muddy sediments, as evidenced from the fatty acid profiles. Relatively high concentrations of marker fatty acids of sulfate-reducing bacteria were associated with organic pollution in this site. Specific fatty acids of microeukaryotes were more abundant in surface sediments than in deeper sediment layers. Among the microeukaryotes, diatoms were an important component. Significant amounts of bacterial biomass, the predominance of bacterial biomarker fatty acids with abundance of anaerobic and sulfate-reducing bacteria are indicative of a prokaryotic consortium responsive to organic pollution.
Analysis of one sinkhole, the Dodgeville sinkhole, developed in Ordovician dolostones in the Driftless Area of Wisconsin in the Upper Midwest'd Driftless Area reveals homogenous clayey sediment fills reflecting a range of dissolutional processes during the Quaternary or Pre-Quaternary. Granulometric analysis, graphical moments statistics, carbonate minerals, ana sand grain lithology were used to differentiate sinkhole sediment sources and modes of accumulation. Sediments in the dolostone sinkholes developed by dissolution. Sediments contain two major types of sediments : residual redish clay( autogenic sediments) and aeolian silt (allogenic sediments). The massive clay is generated from the weathered dolostone bedrocks as a in situ materials. The loessial silt is mostly derived from transportation of the surrounding surface materials, with some evidences of penetrated deposition. Unlike the collapsed sandstone sinkholes (Oh et al., 1993), dolostone sinkholes reveal homogenous, autogenic clay materials, and a geochemical composition indicative of in situ autogenic karstification. Dolostone sinkhole si1ts (26.9%) and sands (34.9%) are derived from weathered Plattevi1le-Galena dolostones, and contain high carbonate(37.5%), chert (57.2%) and lead ore (3%). Graphical moments statistics for sorting, skewness, and kurtosis indicate that sand grains from dolostones were derived entirely from local bedrock by in situ dissolution. Upper sinkhole sediments are pedagogically very young as carbonate is unleashed. Materials of the sinkhole sediment are definitely inherited from internal dolostones by dissolution and weathering, because not only a granulomatric comparison of dolostone and sandstone sediments demonstrates that they have heterogeneous paticle size distributions, but also 1ithologic analyses displays they differ completely.
This study was carried out geochemically and mineralogically to define how Kunsan shore sediments are related to their terrestrial source rocks in the region of Keum River Basin, western Korea. As a whole the chemical composition for major elements, trace elements and rare earth elements analysis from shore sediments and river bed sediments doesn't show the big difference, and especially rare earth elements chondrite normalized patterns are almost same. Heavy minerals of shore sediments are identified as hornblende, epidote, ilmenite, garnet, hematite, magnetite, sphene and rutile. Compared with Taean Area of Seo et al. (1998) and Byeonsan Area of Kwon et al. (1999), Kunsan shore sediments of this study area were origined mostly from Keum River Basin.
Analysis was done on the particle size distribution of sediments flown into Saemangeum from the Mankyung and Dongjin River. The organic pollutants and heavy metal existing in the sediments were analyzed, which was further used to study the properties of pollution in the sediments according to the particle size distribution. Conclusions shown below were made from these analyses. The particle size distribution showed a big difference between the upriver areas of Mankyung and Dongjin River. Particles under $75{\mu}m$ showed to be around 85% at Dongjin River, while it showed to be around 70% at Mankyung River. This kind of distribution in particle size concluded in greatly affecting the contamination density of the sediments. From the analysis done on the soil type of sediments, deposition in Mankyung River categorized into Silty loam and Sandy loam, where Silty loam covered most of area and deposition in Dongjin River categorized into Sand, Loamy sand, Silty loam, Sandy loam. Considering the weight ratio, the density of contamination of the sediments by particle size at Dongjin and Mankyung River has been analyzed to show that organic pollutants and heavy metals occupy more than 70% of the whole contamination in the range under the particle size of $75{\mu}m$.
determine the distribution of heavy metals in estuarine sediments, forty surface sediment samples were collected from the Youngsan estuary, and analysed for their contents of Fe, Mn, Co, Cr, Cu, Ni, Zn and Pb. Contents of most analysed metals were comparable to those in the near shore environment around Korean peninsula, and little anthropogenic effect was identified. However, there was a contrasting behavior among the metals with respect to their relationship with the grain-size of sediments. Contents of Fe, Co, Cr and Ni demonstrated a fairly linear relationship with the grain-size of sediments whereas those of Mn, Cu, Zn and Pb did not. This different behavior In these metals was Interpreted as being the result of the additional inputs. The R-mode factor analysis also confirmed the different behavior of Cu, Zn, Pb and Mn from that of Fe, Co, Cr and Ni.
order to investigate physical characteristics and element concentrations of sediments, coastal bottom sediments were collected at 20 stations in the vicinity of Youngkwang Nuclear Power Plant. After air drying of samples in the laboratory. article size distribution was examined by Master sizer (X-350F), radio-activity by HPGe ${\gamma}$-spectrphotometer, and element concentrations by ICP-AES and AAS. According to particle size analysis , sediments are mainly composed of silt fraction weith 23% of sand, 65% of silt and 12% of clay on average. Most sediments are derived from muddy environment that silt dominates with the characteristics of 5.3${\varsigma}$ mean particle size, poorly sorted, very fine skewed and lepto-kurtic. Only two sediments are well sorted with sandy silt owing to wind, winnowing action, tide and current andits complex reactions. Element concentrations in the coastal bottom sediments are relatively high at finer sediment and show significant relationship with grain size. Index of geoaccumulation by heavy metals at every sampling station is classified as practically unpolluted. The radioactivities of the sediments were measured for 15 isotope elements, and 2 elements of K-40 and Cs-137 were detected in most sediments. The K-40 is the natural nuclide and the artificial nuclide of Cs-137 was thought to be derived from the fallout of past nuclear weapon test. The results of correlation coefficient between grain size and radioactivity shows that the activity of Cs-137 significantly increases in finer grain.

In this study, we tried to determine the origin of fine-grained sediments in Southeastern Yellow Sea Mud patch (SEYSM) using principal component analysis coupled with semi-quantitative X-ray diffraction analysis for 4 major clay minerals. We used 51 marine surface sediments from SEYSM and 33 surface sediments of rivers flowing into the Yellow Sea. We made bioplot diagram using R program with principal component 1 and component 2 because the two components might contain about 98% of all data. The content of each clay mineral in the south and north regions of SEYSM are almost similar. In the biplot, SEYSM sediments distribute close to Korean rivers sediments than Huanghe and Changjiang sediments. Based on these results, we suggest that SEYSM is originated from the Korean rivers sediments. The higher accumulation rate in the SEYSM compared to the sediment discharge from neighboring Korean rivers can be explained by erosion and reworking of surface sediments in this area. The principal component analysis can be used for the provenance research of marine sediments around the Korean Peninsula.
In this study, it was analyzed the soil components and elution characteristics of heavy metal ions in sediment samples to examine the effect of sediments embedded in Andong and Imha reservoirs on water quality. Major elements of sediments were shown to be Al, Si, K, and Fe by EDS analysis and major soil components of the collected 6 different sediments were illite (I), kaolinite (Ka), quartz (Q) and feldspar (F). And especially quartz took up a considerable part of sediments by XRD analysis. The total concentrations of Zn, As, Cr, Cu and Pb in sediments of Andong reservoir were relatively higher than those of Imha reservoir. The elution property of heavy metal from sediments had various aspects according to pH. Among the heavy metals in Andong reservoir sediments, As and Zn were significantly eluted at pH 6 compared with the other heavy metals. In the case of the adsorption tests using the sediments, the adsorptive capacities of Zn, Cd and Cu were very weak, on the while those of Pb and Cr were high.
A Q-mode factor analysis for 115 surficial bottom sediments on the continental shelf off the southeast coast of Korean Peninsula(the Korea Strait) has been carried out to determine the depositional environments. The first four factors are taken as the "principal" factors, and the results are considered to represent the depositional history and sedimentary processes in relation to the Holocene sea-level changes. The fact implies that the sediments are grouped as outer-shelf relict sand sediments, inner-shelf modern muddy sediments, and finally the palimpsest sediments that are mainly distributed within and around the Korea Trough.
