Antimutagenicity and Cytotoxic Effect of Ethanol Extract from Korean Traditional Mackjang Added Sea Tangle (다시마 분말을 첨가한 막장 에탄을 추출물의 항돌연변이원성 및 세포독성 효과)

  • This study was carried out to investigate antimutagenic and cytotoxic effects of Korean traditional Mackjang added with sea tangle powder. The content percentage of most of minerals among general composition increased by adding sea tangle powder. By the Ames test, the antimutagenic effect of ethanol extract of Korean traditional Mackjang added with 5% sea tangle powder was strongest exceeding the control, 10%, and 15% sea tangle additions. The ethanol extract (400$\mu$g/plate) of Mackjang added with 5% sea tang1e powder in the S. typhimurium TA 100 strain showed inhibition rate of 95.0% against the mutagenesis induced by MNNG. The inhibition rate of ethanol extract (400$\mu$g/plate) of Mackjang added with 5% sea tang1e powder in the S. typhimurium TA98 and TA100 strains was 81.4% and 88.8%, respectively, against the mutagenesis induced by 4NQO. Under the same conditions, the suppression against B($\alpha$)P and Trp-P-1 in the TA98 and TA100 strains were 85.3% and 91.0%, and 96.5% and 96.5%, respectively. For the anticancer effects, an investigation was done on the cytotoxicity of Mackjang added with 5% sea tangle powder on the cell lines with human lung carcinoma (A549), human hepatocellular carcinoma (HepG2), and human gastric carcinoma (KATOIII). The cytotoxicities were inhibited with increasing the extract concentration. The treatment of 1.0 mg/mL Mackjang added with 5% sea tang1e powder showed relatively strong cytotoxicity of 61.2%, 61.8% and 66.8% against A549, KATOIII, and HepG2, respectively.

Effect of Supplementation of Dietary Sea Tangle on the Renal Oxidative Stress in Diabetic Rats (식이 다시마의 섭취가 당뇨 쥐 신장의 산화적 스트레스 및 당뇨성 병변에 미치는 영향)

  • Diabetic nephropathy has been increasing, although blood glucose and blood pressure can be controlled by angiotensin converting enzyme(ACE) or advanced glycosylation end products(AGE) inhibitors in the diabetic patients. We investigated the effect of dietary supplementation of sea tangle on the blood glucose, and pathological scoring of diabetic kidneys in the streptozotocin(STZ) induced diabetic rats. Male Sprague-Dawley rats were divided into normal rats fed control diet and diabetic rats fed control diet or control diet supplemented with powder or oater extract of sea tangle. Diabetes was induced by a single injection of STZ(60mg/kg, ip) in citrate buffer. The animals were fed the experimental diet and water for 13 weeks. Dietary supplementation of sea tangle decreased blood glucose in the diabetic rats. However, dietary supplementation of sea tangle did not affect the antioxidant enzyme activities, MDA content and pathology of diabetic kidneys. These results indicate that decreased blood glucose by sea tangle could not delay the progression of diabetic kidney disease.

Effects of γ-Aminobutyric Acid (GABA)-Enriched Sea Tangle Laminaria japonica Extract on Lipopolysaccharide-Induced Inflammation in Mouse Macrophage (RAW 264.7) Cells

  • ${\gamma}$-Aminobutyric acid-enriched sea tangle extract was obtained from the fermentation of Lactobacillus brevis BJ-20. The fermented sea tangle extract (FST) was separated into three fractions by molecular weight: FST I (greater than 10 kDa), FST II (1-10 kDa), and FST III (less than 1 kDa). The anti-inflammatory characteristics of the FST fractions were investigated by measuring the production of nitric oxide (NO) and the expression levels of inducible nitric oxide synthase (iNOS) in lipopolysaccharide (LPS)-induced mouse macrophage (RAW 264.7) cells. Both NO production and iNOS expression levels were significantly inhibited by FST treatments in a dose-dependent manner. FST III was the most effective inhibitor of processes. This demonstrates that the effect of FST on LPS-induced inflammation might be closely correlated with the inhibition of inflammatory cytokine expression.

Anti-hyperlipidemia and Anti-arteriosclerosis Effects of Laminaria japonica in Sprague-Dawley Rats

  • The anti-hyperlipidemic effects of dietary supplementation with sea tangle Laminaria japonica were investigated using an animal model in which normal rats were fed either sea tangle, sea tangle ethanol extract (EE-ST) and sea tangle extracted residue (ER-ST). Total lipid and triglyceride levels in the serum were significantly (P < 0.05) reduced in rats fed ER-ST at a dose of 200 mg/kg body weight when compared to hyperlipidemic control rats. Significant decreases in serum total cholesterol and low density lipoprotein-cholesterol levels also occurred in rats fed ER-ST at 200 mg/kg body weight. In addition, the atherosclerosis index and superoxide dismutase in blood lipids were significantly (P < 0.05) lowered in rats fed ER-ST at 200 mg/kg body weight as compared to control rats. In conclusion, sea tangle and ER-ST exhibited beneficial anti-hyperlipidemic and anti-arteriosclerosis effects.

Optimization of Hot Water Extraction Conditions of Wando Sea Tangle (Laminaria japonica) for Development of Natural Salt Enhancer (천연 염미증강제 개발을 위한 완도산 다시마의 열수 추출 조건 최적화 및 염미증강 효능 평가)

  • In recent decades, health concerns related to sodium intake have caused an increased demand for salt or sodium-reduced foods. Umami substance can enhance taste sensitivity to NaCl and may offer a unique approach to replace and reduce the sodium content in foods. In this study, hot water extraction conditions of Wando sea tangle with high umami taste were investigated. Wando sea tangle harvested in June was selected for hot water extraction based on its free amino acids composition. The quality properties of sea tangle extract were investigated at various extraction temperatures ($60^{\circ}C$, $80^{\circ}C$, and $100^{\circ}C$) and times (1 h, 2 h, and 3 h). Sea tangle extracts at the extraction temperature of $100^{\circ}C$ contained the highest soluble solids (35.47%~36.93%), and crude protein (3.75%~4.00%). Viscosities of sea tangle extracts decreased with increasing extraction temperature. Umami amino acids (glutamic acid and aspartic acid) and sensory characteristics were best at extraction conditions of $100^{\circ}C$ for 2 h. Saltiness enhancement of sea tangle extract powder was determined. Saltiness intensities of NaCl solution after adding 1% sea tangle extract powder were enhanced (1.84~4.25-fold). At the same saltiness intensity, sodium contents of NaCl solution with 1% sea tangle extract powder were 12.24~24.33% lower than that of NaCl solution. These results suggest that it is possible to reduce sodium in foods with sea tangle extract as a natural salt enhancer without lowering overall taste intensity.

Effect of Viscous Materials Removal from Sea Tangle Extracts on Volatile Flavor Constituents (점성물질 제거가 다시마 추출액의 휘발성 향미성분에 미치는 영향)

  • Volatile flavor compounds of a sea tangle powder and two kinds of extract were analyzed by GC/MS. Extract I was prepared by boiling for 2 hours and centrifugation, while extract II by a sequential procedure of enzymatic hydrolysis, boiling in 1.5% NaCl solution. centrifugation and ultrafiltration to remove viscous materials. Fifty six volatile compounds from the dried sea tangle powder and the extracts were identified. The GC profiles of the extract II were different from those of the dried powder and the extract 1, indicating most volatile compounds were lost during removing viscous materials. Particularly those compounds in the initial and later parts of the GC profiles were significantly decreased and some of the compounds such as fatty acids. 3,5-nonadien-2-ol and 1-penton-3-ol were not detected.

Processing of Vinegar Using the Sea Tangle (Laminaria Japonica) Extract (다시마 추출액을 이용한 식초 제조)

  • Optimum processing conditions of vinegar using esa tangle extracts were investigated. Sea tangle vineagr was prepared by adding 3% and 5% of glucose, 6% and 10% of ethanol to sea tangle extracts and inoculating acetic acid bacteria. After fermentation for 30 days at 3$0^{\circ}C$, pH was similar to 3.22, 3.21, 3.25 and 3.28 days it was 4.75%, 4.77%, 5.61% and 5.87% respectively. Contents or reducing sugar was 0.70%, 0.70%, 0.88% and 0.89% in initial time of fermentation but it was rapidly decreased by 10 days of fermentation to 0.20%, 0.19%, 0.22%, and 0.21%, respectively. Ethanol contents was 5.88%, 9.77%, 5.75% and 9.68% in initial time of fermentation but it was rapidly decreased by 15 days of fermentation to 1.05%, 1.62%, 0.45% and 1.23%, respectively and it was addition of 5% glucose and 6% ethanol and fermentation for 20 days at 3$0^{\circ}C$. Quality for sea tangle vinegar manufactured using optimum condition were as follows respectively; pH 3.25, acidity 5.38%, total sugar 1.72%, iodine 1,537.2 ppm.

Development of natural Seasoning Based on Mushroom (버섯 천연조미료 제조에 관한 연구)

  • This study was conducted to provide basic data for the development of natural seasoning. The quality chatacteristics of water exacts from Pleurotus ostreatus and Lentinus edodes were determind. Also, savory taste, fragrance, color and overall preference was evaluated. The preference test on the seasonings prepared from Pleurotus ostreatus and Lentinus edodes with the addition of sea tangle was also performed. The crude protein content of dried, Lentinus edodes and Pleurotur ostreatus was 20.4% and 30.4% respectively. The total organic acid contents were higher in Plourotus ostreatus than in Lentinus edodes extracts. The total free amino acid adn total nucleotide contents were higher in Pleurotus ostreatus than in Lentinus edodes extracts. Three nucleotide, adenosine monophosphate(AMP) , guanosine monophosphate(GMP), xanthine monophosphate(XMP) were found in mushroom seasonings while inosine monophosphate(IMP) was not detected. Pleurotus ostreatus extract showed higher score in savory taste and overall acceptability than Lentinus edodes extract, Pleurotur ostreatus seasoning added with 1% sea tangle and Lentinus edodes seasoning added with 1.5% sea tangle were rated as high in overall quality In a sensory test of foods cooked with mushroom seasoning. It was shown that panels in their twenties well used to the taste of monosodium glutamate while the people in their thirties preferred the specific flavor of the mushroom seasoning.

Fermented sea tangle (Laminaria japonica Aresch) Accelerates Osteoblast Differentiation in murine osteoblastic MC3T3-E1 Cells (MC3T3-E1 골아세포에서 발효 다시마 추출물에 의한 조골세포 분화의 촉진)

  • The Laminaria japonica Aresch (Sea tangle) belongs to the brown algae and has a long history as a food material in Asia, including Korea. Recent studies have found that the fermented Sea tangle extract (FST) inhibited the differentiation of osteoclasts and protected osteoblasts from oxidative damage. This study aims to explore the possibility that FST can induce the differentiation of osteoblasts and identify the responsible mechanism. According to our results, FST induced differentiation into osteogenic cells in the presence of osteoblastic MC3T3-E1 cells under non-toxic conditions.. This finding was confirmed by phalloidin staining, increased alkaline phosphatase activity, and calcium deposition. Additionally, it was found that this process was achieved by increasing the expression of key factors involved in osteoblast differentiation, such as runt-related transcription factor-2, osterix, β-catenin, and bone morphogenetic protein-2. Moreover, FST increased autophagy, which may contribute to the maintenance of the bone formation homeostasis, and is associated with the activation of the phosphatidylinositol 3-kinase/Akt and mitogen-activated protein kinase signaling pathways. Although further research about the bioactive substances contained in FST and the tests of their efficacy are required, the results of this study indicate that FST has incredible applicability as a functional material for maintaining the bone homeostasis.

Effect of Extraction Temperature on Some Quality of Sea Tangle Extract (다시마 추출액의 특성에 미치는 추출온도의 영향)

  • Aqueous extraction of sea tangle at the temperature range of 60~10$0^{\circ}C$ was studied for temperature effects on soluble solids and protein yields, amino nitrogen, turbidity and viscosity of extracts. The solids and protein yield were increased as the temperature increased and most of solids and protein were extracted during 1 hour. The supernatant ratio after centrifugation showed significantly low for the extraction at 6$0^{\circ}C$. More temperature effects were found on turbidity and viscosity than yields. A significantly higher total amino nitrogen contents was obtained from higher temperature at initial stage of extraction and then the differences of them became to be narrow as the extraction prolonged further. The low values of 24.1% solids and 13.5% protein yields after 2 hours of extraction at 10$0^{\circ}C$ indicated that most of solids in sea tangle are insouble.
