A Study of the Growth Characteristics of Starry Flounder Platichthys Stellatus in Accordance with the LED Wavelength (LED 파장에 따른 강도다리 Platichthys Stellatus 성장특성)

  • Currently the fish aquaculture industry of Korea is focused on the mass culture of flatfish (Paralichthys olivaceus) and and rockfish (Sebastes schlegeli) with completely controlled culture techniques. Recently, there has been considerable interest in new species development, such as the starry flounder (Platichthys stellatus). The value of starry flounder (Platichthys stellatus) as a raw fish increases with time because it is tasty, light, and bouncy. In this paper, the growth characteristics dependent on the LED wavelengths and the optimal growth conditions of the starry flounder were studied. In these experiments 4 different kinds of LED lighting, configurations were designed and prepared using red, green, blue and white, respectively. The fish aquaculture experiments were conducted over 10 weeks in four fish tanks, each installed with a different color of LED lighting. 10 starry flounders of 13 ~ 17g were placed into each tank. The effects of each color of light on the growth rate of the starry flounders were then examined. As a result, the starry flounders under the green LED lighting showed the highest growth rate, followed by the white, red, and blue LED lighting. Based on these results, a green light provides a suitable breeding environment for the starry flounder.

The Protective Effect of Water Extract of Phellodendri Cortex in Chronic Reflux Esophagitis-induced Rats (만성 역류성 식도염 동물 모델에 대한 황백(黃柏) 물 추출물의 식도 점막 보호 효과)

  • Objective : Reflux esophagitis is a disease caused by the reflux of gastric acid and inflammation due to unstable gastroesophageal sphincter. The aim of the present study was to clarify the effect of Phellodendri Cortex (PC) on chronic reflux esophagitis (CRE) in rats. Methods : The anti-oxidant activity of PC was measured by total polyphenol, total flavonoid contents, 1, 1-diphenyl-2-picrylhydrazyl (DPPH), and 2, 2'-azinobis-3-ethyl-enzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity. A CRE was established surgically in SD rats. And then CRE rats were treated with water or PC 200 mg/kg body weight for 14 days. Besides, the anti-oxidant and inflammatory protein levels were evaluated using western blotting. Results : PC reduced esophagus tissues injury. The total polyphenol (36.05 ± 0.25 mg/g) and total flavonoid (72.90 ± 0.61 mg/g) of PC showed a high content. PC strongly reduced radical scavenging activities (DPPH IC50 43.58 ± 1.54 ㎍/㎖; ABTS IC50 36.75 ± 0.35 ㎍/㎖). Moreover, reactive oxygen species (ROS) and peroxynitrite (ONOO-) levels in serum, the protein expression of inducible nitric oxide synthases (iNOS) were significantly reduced. In addition, the protein expression of NADPH oxidases related to oxidative stress were significantly reduced in PC compared to CRE control. PC effectively reduced inflammatory factors including, TNF-α, and IL-6 via NF-κBp65 inactivation through the inhibition of p-IκBα and increased anti-oxidant enzyme such as HO-1, SOD, catalase, and GPx-1/2 via Nrf2 activation. Conclusions : Taken together, these results show that PC can alleviate the esophageal mucosal ulcer though the inhibition of NF-κB inflammatory and the enhancement of Nrf2 anti-oxidant pathway.

Conservation Management Strategies of Protected Areas for Genetic Resources, Torreya nucifera Forest of Bulhoesa(Temple) in Naju (나주 불회사 비자림 산림유전자원보호구역의 보전관리방안)

  • This study investigated some of the specific ecological characteristics Torreya nucifera forest and its health conditions so that was drawn from four different management strategies, such as Types A, B, C, and D, for protected areas of forest genetic resources. Type A refers to passive management which routinely keeps the current status of forests by removing competitive trees or curing damaged ones. Type B, as active one which improves Torreya nucifera forest, includes increase of soil pH and removal of Sasa borealis, Phyllostachys bambusoides, and deciduous broad-leaved trees such as Quercus variabilis, Carpinus tschonoskii et al.. Type C is another passive one which reduces soil acidity within Chamae cyparisobtusa forest and maintains the existing forests. Type D is another active one which decreases soil acidity and simultaneously reduces problems associated with competitive and invasive plants to nurture the young trees. At last, it is important to note that the protected areas for forest genetic resources need to be entitled to categories III(natural monument) or IV(habitat/species management area) in accordance with protected area management guidelines of IUCN.

  • Subcellular localization of a protein containing nuclear localization signals (NLS) has been well studied in many organisms ranging from invertebrates to vertebrates. However, no systematic analysis of NLS-containing proteins available from Mollusks has been reported. Here, we describe in silico screening of NLS-containing proteins using the mollusks database that contains 22,138 amino acids. To screen putative proteins with NLS-motif, we used both predict NLS and perl script. As a result, we have found 266 proteins containing NLS sequences which are about 1.2% out of the entire proteins. On the basis of KOG (The eukaryotic orthologous groups) analysis, we can't predict the precise functions of the NLS-containing proteins. However, we found out that these proteins belong to several types of proteins such as chromatin structure and dynamics, translation, ribosomal structure, biogenesis, and signal transduction mechanism. In addition, we have analysed these sequences based on the classes of mollusks. We could not find many from the species that are the main subjects of phylogenetic studies. In contrast, we noticed that cephalopods has the highest number of NLS-containing proteins. Thus, we have constructed mollusks NLS database and added these information and data to the mollusks database by constructing web interface. Taken together, these information will be very useful for those who are or will be studying NLS-containing proteins from mollusks.

Construction of web-based Database for Haliotis SNP (웹기반 전복류 (Haliotis) SNP 데이터베이스 구축)

  • The Web-based the genus Haliotis SNP database was constructed on the basis of Intel Server Platform ZSS130 dual Xeon 3.2 GHz cpu and Linux-based (Cent OS) operating system. Haliotis related sequences (2,830 nucleotide sequences, 9,102 EST sequences) were downloaded through NCBI taxonomy browser. In order to eliminate vector sequences, we conducted vector masking step using cross match software with vector sequence database. In addition, poly-A tails were removed using Trimmest software from EMBOSS package. The processed sequences were clustered and assembled by TGICL package (TIGR tools) equipped with CAP3 software. A web-based interface (Haliotis SNP Database, http://www.haliotis.or.kr) was developed to enable optimal use of the clustered assemblies. The Clustering Res. menu shows the contig sequences from the clustering, the alignment results and sequences from each cluster. And also we can compare any sequences with Haliotis related sequences in BLAST menu. The search menu is equipped with its own search engine so that it is possible to search all of the information in the database using the name of a gene, accession number and/or species name. Taken together, the Web-based SNP database for Haliotis will be valuable to develop SNPs of Haliotis in the future.

Flow Rate·Water Quality Characteristics of Tributaries and a Grouping Method for Tributary Management in Nakdong River (낙동강 지류·지천의 유량·수질 특성 및 하천관리를 위한 등급화 방안 연구)

  • In this study, the major 38 tributaries in Nakdong River were monitored for flow rate and water quality in order to understand the characteristics of the watershed and to find improvement plan. The flow rate and water quality for each target tributary were evaluated based on the monitoring data in 2013~2014 using a statistical package SPSS-22.0. In addition, the tributary grouping method was conducted using a $BOD_5$ concentration/flowrate and TP concentration/flowrate monitoring data. The average values of $BOD_5$, $COD_{Mn}$, TP and TOC concentrations in Gumicheon, Gyeonghocheon, Jincheoncheon, Gisegokcheon, Yonghacheon and Yonghocheon located at Nakdong Waegwan and Nakdong Goryung watershed were high and in the grade of III or IV (5~8 mg/L). The Pearson correlation coefficients of TOC with $BOD_5$, $COD_{Mn}$, and TP were greater (r=0.8, p<0.01) than those of the other water quality parameters (12 species). The tributaries with high values of water quality parameters ($BOD_5$ > 3.0 mg/L, TP > 0.1 mg/L) and flowrate (Q > $0.1m^3/sec$) were selected for improving water quality according to the stream grouping method. Five tributaries (Gumicheon, Gisegokcheon, Yonghacheon, Yeongsancheon, Mijeoncheon and Yonghocheon) were classified as Group I, which require polices and plans for water quality improvement.

  • The objective of this study was carried out in order to evaluate the antioxidative activities of n-hexane fractions from medicinal herbs. Certain parts from 6 species of edible and medicinal herbs were extracted with methanol. Radical scavenging activity of the n-hexane fractions against DPPH and ABTS radicals were observed to be high in Psoralea corylifolia, and Scutellsria barbata; it also significantly increased (p<0.05). For a comparison of the effect of various concentration of fractions, as the concentration of treatment increased, the antioxidative effects on radical scavenging activity also oncreased; yet it was not significant. The antioxidative effect of medicinal plants was dependent on the concentration but was not significant. Therefore, these results indicate that edible wild plants, such as Psoralea corylifolia, and Scutellsria barbata may be useful as potential antioxidant sources for improving the human antioxidant defense system against disease.

The Genetic Approach on Analyzing the Habitat Characteristics of Fairy Pitta Pitta Nympha Inhabiting Jeju Island, the Korean Peninsula and Taiwan (제주도, 한반도 및 대만 내 팔색조의 생태적 서식특성 분석을 위한 유전적 접근)

  • A Fairy Pitta is a bird known to breed only in mainland China, Taiwan, Japan and Korea and is listed as Vulnerable in the IUCN Red List. We carried out a DNA analysis to contribute to conserve the genetic diversity of Fairy Pitta. 32 samples were collected at Jeju Island, the Korean Peninsula and Taiwan from 2004 to 2013 and DNA was extracted from them and several sequences were amplified-it through PCR. And then we performed the population genetic analysis. We found there was a transversion between nucleotide sequences at CO1 gene, while there was no changes at Cyt-b gene. And we confirmed the polymorphism from two genes was caused from genetic drift not from selection. Through this analysis, the group within the Peninsula was found bigger than other two groups based on the analysis of CO1 gene, and the group from Taiwan was found bigger than other two groups through the analysis of Cyt-b gene. The population genetic structure of mitochondria gene of three group was showing CO1 gene had 5 haplotypes and Cyt-b gene had 6 haplotypes. Haplotype 2 in CO1 gene was found in three group and many individuals of samples had this haplotype. Like CO1 gene, haplotype 2 in Cyt-b gene was found in three group and was included in plenty of individuals. Other haplotypes were not overlaped and broke off among the three groups. To prevent from the extinction of Fairy Pitta and to obtain the genetic diversity, we need to compare with other regional group such as Japan, China and perform additional research in the non-breeding area.

Variation of Fatty Acid Composition and Content in Domestic and Imported Solar-Salt by GC-MS (국산과 수입산 천일염의 지방산 분석)

  • BACKGROUND: Inorganic component is made up largely of salt, because the criteria are difficult to split into domestic and imported ingredients, organic examined the fatty acid composition is to see a possible use as a marker using gas chromatography-mass spectrometry (GC-MS). The 14 domestic samples and 8 imported samples were collected from China, India and other countries were investigated on item of 37 fatty acids. METHODS AND RESULTS: The major components were six species, myristic acid ($C_{14:0}$), palmitic acid ($C_{16:0}$), palmitoleic acid ($C_{16:1}$), stearic acid ($C_{18:0}$), oleic acid ($C_{18:2,n9}$), linoleic acid ($C_{18:2,n6}$) among detected twenty fatty acids including 9 unknowns. The content of palmitic acid were highest as 25.2 to 50.8% of total fatty acids contents. Domestic has seen the most amount of salt in the Taepyeong salts $250.8{\mu}g$/100g, imports $135.2{\mu}g$/100g salt in Chinese Weifang salts showed the lowest content. CONCLUSION(s): The total fatty acid content has seen the most amount of salt in 352.3 ug/100g Christmas island salts showed the lowest content of $164.0{\mu}g$/100g, as a result it is difficult to distinguish the domestic salts and imported by the composition of fatty acid.

Nobiletin attenuates neurotoxic mitochondrial calcium overload through K+ influx and ∆Ψm across mitochondrial inner membrane

  • Mitochondrial calcium overload is a crucial event in determining the fate of neuronal cell survival and death, implicated in pathogenesis of neurodegenerative diseases. One of the driving forces of calcium influx into mitochondria is mitochondria membrane potential (${\Delta}{\psi}_m$). Therefore, pharmacological manipulation of ${\Delta}{\psi}_m$ can be a promising strategy to prevent neuronal cell death against brain insults. Based on these issues, we investigated here whether nobiletin, a Citrus polymethoxylated flavone, prevents neurotoxic neuronal calcium overload and cell death via regulating basal ${\Delta}{\psi}_m$ against neuronal insult in primary cortical neurons and pure brain mitochondria isolated from rat cortices. Results demonstrated that nobiletin treatment significantly increased cell viability against glutamate toxicity ($100{\mu}M$, 20 min) in primary cortical neurons. Real-time imaging-based fluorometry data reveal that nobiletin evokes partial mitochondrial depolarization in these neurons. Nobiletin markedly attenuated mitochondrial calcium overload and reactive oxygen species (ROS) generation in glutamate ($100{\mu}M$)-stimulated cortical neurons and isolated pure mitochondria exposed to high concentration of $Ca^{2+}$ ($5{\mu}M$). Nobiletin-induced partial mitochondrial depolarization in intact neurons was confirmed in isolated brain mitochondria using a fluorescence microplate reader. Nobiletin effects on basal ${\Delta}{\psi}_m$ were completely abolished in $K^+-free$ medium on pure isolated mitochondria. Taken together, results demonstrate that $K^+$ influx into mitochondria is critically involved in partial mitochondrial depolarization-related neuroprotective effect of nobiletin. Nobiletin-induced mitochondrial $K^+$ influx is probably mediated, at least in part, by activation of mitochondrial $K^+$ channels. However, further detailed studies should be conducted to determine exact molecular targets of nobiletin in mitochondria.
