Yeasts Diversity of Wild Flowers in Mountains of Korea and Their Physiological Functionalities (한국의 야생화에서 분리한 효모의 종 다양성과 생리기능성)

  • This study focused on isolation of wild yeasts from natural flowers and elucidation of yeast diversity. Wild yeasts were isolated from various flowers collected from mountains on the islands including Jejudo, Ulleungdo, Yokjido, and Seonyudo as well as inlands including Gyejoksan, Oseosan, Beakamsan, and Deogyusan in Korea. Isolated yeasts were identified by comparison of nucleotide sequences for polymerrase chain reaction-amplified D1/D2 region of 26S rDNA or internal transcribed spacer 1 and 2 including 5.8S rDNA using BLAST. 289 strains belonging to 134 yeast species were isolated. Cryptococcus genus strains were the most frequently isolated species among the identified yeasts. Metschnikowia reukaufii was also frequently isolated. Twenty three species including Cryptococcus aureus were overlapped between those of mountains on islands and inland. Physiological functionalities such as antioxidant activity, xanthine oxidase inhibitory activity, and tyrosinase inhibitory activity for the 289 identified yeast strains were investigated using their supernatant and cell-free extracts. The supernatants of Candida sp. 78-J-2 and Metschnikowia reukaufii SY44-6 showed antioxidant activity of 22.5%, and anti-gout xanthine oxidase inhibitory activity of 49.6%, respectively.

Changes in Chemical Property of Soil Affected by Termites (Reticulitermes speratus kyushuensis Morimoto) in Korea (국내 흰개미(Reticulitermes speratus kyushuensis Morimoto)에 의한 토양의 화학적 특성 변화)

  • Termites (Isoptera) are classified into approximately 3,106 species. In Korea, only one species has been identified, which is Reticulitermes speratus kyushuensis Morimoto. The termite, a social insect, is known to play an important role in nutrient cycling of the ecosystem, although some species of termites are well-known pests attacking wooden structures or any plant materials. However, there is a lack of research about termites in Korea, including aspects such the taxonomy, physiology, and ecology of termites. This study was carried out to provide valuable basic data on the ecological role of termites in an ecosystem in Korea for the future studies. For the experiments, soil and termite samples were randomly collected from Mt. Hwajang located in Jikdong-ri, Eonyang-eup, Ulju-gun, Korea between October 5 and 30, 2015. Analysis results showed that there were no significant differences in soil chemical properties between the soil samples just after air-drying and one year elapsed without any treatment. The treated soil with termites showed significantly higher than the soil without termite treatment. Chemical properties of total nitrogen, organic matter, available phosphate, pH, Calcium(Ca), Potassium(K) and Magnesium(Mg) in soil treated with termites were $1.11{\pm}0.3gkg^{-1}$, $43.3{\pm}12.4gkg^{-1}$, $27.4{\pm}2.9mgkg^{-1}$, $4.56{\pm}0.2$, $0.82{\pm}0.2cmol_ckg^{-1}$, $3.18{\pm}1.4cmol_ckg^{-1}$, $1.73{\pm}1.1cmol_ckg^{-1}$, respectively. The values of soil property of without termite treatment were $0.56{\pm}0.1gkg^{-1}$, $30.5{\pm}3.1gkg^{-1}$, $24.0{\pm}4.7 mgkg^{-1}$, $4.09{\pm}0.1$, $0.71{\pm}0.2cmol_ckg^{-1}$, $2.88{\pm}1.5cmol_ckg^{-1}$, $1.30{\pm}0.7cmol_ckg^{-1}$, respectively. These results suggest that inhabitation of termites could improve soil chemical properties in an ecosystem.

Detection of Pseudomonas tolaasii Causing Brown Blotch Disease of Mushroom with Species-specific DNA Probe (종 특이 DNA probe를 이용한 버섯 세균성 갈반병 병원균(Pseudomonas tolaasii)의 검출)

  • This study was carried out to develop the molecular marker for the detection of Pseudomonas tolaasii, a causative agent of bacterial brown blotch disease of oyster mushroom (Pleurotus ostreatus). When several primers designed from repetitive sequences and pectin lyase genes of bacteria were used to produce DNA polymorphism from different Pseudomonas spp. isolated from edible mushrooms, PEU1 primer derived from pectin lyase gene produced polymorphic bands differentiating P. tolaasii strains from other Pseudomonas species. Two bands, 1.0kb and 0.4kb, found commonly in 6 isolates of P. tolaasii were cloned into pGEM-T vector which were designated as pPTOP1 and pPTOP2, respectively, to use as probe. The 0.4 kb insert of pPTOP2 hybridized to only 6 isolates of P. tolaasii, but did not to the other Pseudomonas species. As few as $1.5{\times}10^3$ colony forming unit (cfu) of P. tolaasii could be detected by dot blot hybridization with the cloned 0.4kb DNA in pPTOP2.

  • An objective index for quality estimation of anchovy sauce, our traditional seafood, has been required because inadequate standard causes dispute about quality estimation. Qualities of anchovy sauce made by traditional method and commercial anchor sauce products were compared by investigating their proximate compositions, total nitrogen contents, amino acid contents, development and level of a specific peptide on SDS-polyacrylamde gel electrophoresis (SDS-PAGE) and isoelectric point (pI) of the specific peptide. Also, pIs of IEF were attemped to identify fish species of salt-fermented sauces. The 55,600 and 46,900 dalton of peptide band were identified in all experimental samples. Especially, the area of 55,600 dalton was closely correlated with total nitrogen contents. Also, specific pIs in accordance with fish species were identified. The results suggest that correlation between area of 55,600 dalton and total nitrogen contents are used of index for quality estimation, and specific pIs are used of fish species identification.

The flora of land plants (bryophytes and vascular plants) in Wangpicheon Ecosystem and Landscape Conservation Area (ELCA), Korea (왕피천 생태경관보전지역의 육상(비관속 선태식물 및 관속식물) 식물상)

  • This study presents the flora of nonvascular plants (bryophytes) and vascular plants within the Wangpicheon Ecosystem and Landscape Conservation Area in Korea. The time of the survey was divided into four seasons. The fieldwork was conducted for a total of 18 days from November of 2016 to October of 2017. The land plants in the Wangpicheon conservation area were identified as 758 taxa with 164 families, 455 genera, 698 species, 10 subspecies, 47 varieties and 3 forms. Among them, bryophytes amounted to 122 taxa and vascular plants numbered 636 taxa. Native plant amounted to 693 taxa. In total, there are 113 floristic target species in Korea, including one level V taxon, in this case Swertia wilfordii; six level IV taxa; 31 level III taxa; 38 level II taxa; and 37 level I taxa. There are nine endemic plants, including Sillaphyton (=Peucedanum) podagraria, which is the endemic genus; two rare plants; fifteen northern plants; and one southern plant as target plants adaptable to climate change. Also, there are forty-six naturalized plants and five ecosystem-disturbance organisms. The results of this study suggest several species that should be preserved and can serve as basic information with which to establish conservation and management plans for the Wangpicheon conservation area.

Ichthyotoxic Cochlodinium polykrikoides red tides offshore in the South Sea, Korea in 2014: III. Metazooplankton and their grazing impacts on red-tide organisms and heterotrophic protists

  • Cochlodinium polykrikoides red tides have caused great economic losses in the aquaculture industry in many countries. To investigate the roles of metazooplankton in red tide dynamics of C. polykrikoides in the South Sea of Korea, the abundance of metazooplankton was measured at 60 stations over 1- or 2-week intervals from May to November 2014. In addition, the grazing impacts of dominant metazooplankton on red tide species and their potential heterotrophic protistan grazers were estimated by combining field data on the abundance of red tide species, heterotrophic protist grazers, and dominant metazooplankton with data obtained from the literature concerning ingestion rates of the grazers on red tide species and heterotrophic protists. The mean abundance of total metazooplankton at each sampling time during the study was 297-1,119 individuals $m^{-3}$. The abundance of total metazooplankton was significantly positively correlated with that of phototrophic dinoflagellates (p < 0.01), but it was not significantly correlated with water temperature, salinity, and the abundance of diatoms, euglenophytes, cryptophytes, heterotrophic dinoflagellates, tintinnid ciliates, and naked ciliates (p > 0.1). Thus, dinoflagellate red tides may support high abundance of total metazooplankton. Copepods dominated metazooplankton assemblages at all sampling times except from Jul 11 to Aug 6 when cladocerans and hydrozoans dominated. The calculated maximum grazing coefficients attributable to calanoid copepods on C. polykrikoides and Prorocentrum spp. were 0.018 and $0.029d^{-1}$, respectively. Therefore, calanoid copepods may not control populations of C. polykrikoides or Prorocentrum spp. Furthermore, the maximum grazing coefficients attributable to calanoid copepods on the heterotrophic dinoflagellates Polykrikos spp. and Gyrodinium spp., which were grazers on C. polykrikoides and Prorocentrum spp., respectively, were 0.008 and $0.047d^{-1}$, respectively. Therefore, calanoid copepods may not reduce grazing impact by these heterotrophic dinoflagellate grazers on populations of the red tide dinoflagellates.

Changes in the Physicochemical Characteristics and Triglyceride Molecular Species of Corn oil during Hydrogenation (수소첨가에 따른 옥수수유의 트리글리세리드 분자종 및 이화학적 특성의 변화)

  • Changes in the physicochemical characteristics and triglyceride molecular species of corn oil under the following condition of hydrogenation; temperature $180^{\circ}C,\;H_{2}$, pressure $2.0{\pm}0.3bar$, the amount of Ni catalyst 0.048%(Ni/oil by wt.) and agitation speed 300 rpm. The rate of hydrogenation, expressed as the reduction rate of the iodine value with respect to time, is first order and high (K>0.01). When the reduction rate of the iodine value was 39.9%, hydrogenation time was 30 min, 18:1 was highest(77.06%), thereafter that was decreased and 18:0 increased. In the triglyceride composition, OLL, LLL were reduced markedly in 10 min, thereafter reduced slightly. And PLO, PLL, OLO were eliminated in first 30 min. On the other hand, POO, PLS(CN52) and OOO, SLO(CN54) were increased sharply, and then that showed little change. The melting point(MP) of hydrogenated corn oil were $27.8^{\circ}C\;and\;44.1^{\circ}C$ after 20 min and 60 min, respectively. Trans isomer content increased to 46.8% during 40 mins of hydrogenation and then decreased insignificantly. The solid fat content were linearly increased with hydrogenation time. Accordingly, it is confirmed that this condition of hydrogenation was selective, preferential elimination of polyunsaturated fatty acid went stepwise and trans isomer was formed promotedly. These results suggest that fat modification techniques can be used for practical application.

Effects of Seaweeds on Matrix Metalloproteinases Derived from Normal Human Dermal Fibroblasts and Human Fibrosarcoma Cells (사람피부섬유아세포 및 섬유아육종세포로부터 유래된 기질금속단백질효소에 대한 해조류의 효능)

  • In recent years novel potential pharmocological candidates have been looked for in animal, seaweed, sponge, fungi and marine bacteria resources. In this study, matrix metalloproteinases (MMPs) that play an important role in metastasis, arthritis, chronic inflammation and wrinkle formation were used as target enzymes to screen therapeutic agents. The inhibitory effects of several marine algae including green algae (5 species), red algae (18 species) and brown algae (4 species) methanolic extracts on MMPs were investigated in human dermal fibroblasts and human fibrosarcoma cell line (HT1080 cells) using gelatin zymography. In human dermal fibroblasts, the inhibition of MMP-2 was observed in Laurencia okamurae, Polysiphonia japonica, Grateloupia lanceolate and Sinkoraena lancifolia of red algae. In contrast, MMP-2 activation was enhanced in Enteromorpha compressa and E. linza of green algae, and Peltaronia bighamiae and Sargassum thunbergii of brown algae. In human fibrosarcoma cells, MMP-9 activation was decreased in the presence of S. thunbergii of brown algae, Polysiphonia japonica in red algae and E. compressa and E. linza of green algae. The interesting finding is that E. compressa and E. linza of green algae, and S. thunbergii of brown algae exhibited a positive effect on MMP-2 in normal cells, but a negative effect on MMP-9 in cancer cell lines. These results suggest that E. compressa and E. linza of green algae, and S. thunbergii of brown algae contain potential therapeutic ingredients for cancer treatment.

Six unrecorded species from the family Lejeuneaceae (Marchantiophyta) in Korea (작은귀이끼과(Lejeuneaceae)의 한반도 미기록 태류식물)

  • While preparing a floristic study of Korean hepatics, we discovered the six unrecorded species of Lejeuneaceae from Mt. Seolak, Mt. Gaya, Geojedo, Wando and Gageodo. Archilejeunea kiushiana, Drepanolejeunea angustifolia, Lejeunea aquatica, Lejeunea otiana, Leucolejeunea japonica, and Spruceanthus semirepandus, are reported here. A. kiushiana was characterized by unlobed underleaves, homogeneous oil-bodies, and four to six perianth-keels. D. angustifolia is similar to D. ternatensis, but it differs in the dorsal margin of the leaf-lobe, which is entire or denticulate, spines never present, and leaf cells smooth or with minute dorsal bulging. L. aquatica is similar to L. japonica in that it has a wider and similar shape (wider than long) of its underleaves and has homogeneous oil-bodies. The former, however, has minute leaf-lobules (5-15 cells), 1/10 to 1/5 the length of the leaf-lobe, versus leaf-lobules 1/4 to 1/3 the length of the leaf-lobe. L. otiana is similar to L. kodamae in that it has a large and acute first tooth of the leaf-lobule but differs in having plain leaf-lobules obliquely quadrate and an indistinct second tooth. Drepanolejeunea (Spruce) Schiffn. is widely distributed in tropical and subtropical regions (Mizutani, 1961). L. japonica is characterized by entire underleaves; one large, coarsely segmented oil-body per leaf cell; and a large second tooth of the leaf lobule, two to six cells long, and two to three cells wide at the base. S. semirepandus is widely distributed in tropical and subtropical regions in Asia. This species is characterized by small homogeneous and numerous oil-bodies, up to 10 smooth perianthkeels, and leaf-lobe acute at the apex.

Mixotrophy in the newly described dinoflagellate Ansanella granifera: feeding mechanism, prey species, and effect of prey concentration

  • Mixotrophic protists play diverse roles in marine food webs as predators and prey. Thus, exploring mixotrophy in phototrophic protists has emerged as a critical step in understanding marine food webs and cycling of materials in marine ecosystem. To investigate the feeding of newly described mixotrophic dinoflagellate Ansanella granifera, we explored the feeding mechanism and the different types of species that A. granifera was able to feed on. In addition, we measured the growth and ingestion rates of A. granifera feeding on the prasinophyte Pyramimonas sp., the only algal prey, as a function of prey concentration. A. granifera was able to feed on heterotrophic bacteria and the cyanobacterium Synechococcus sp. However, among the 12 species of algal prey offered, A. granifera ingested only Pyramimonas sp. A. granifera ingested the algal prey cell by engulfment. With increasing mean prey concentration, the growth rate of A. granifera feeding on Pyramimonas sp. increased rapidly, but became saturated at a concentration of $434ngCmL^{-1}$ (10,845 cells $mL^{-1}$). The maximum specific growth rate (i.e., mixotrophic growth) of A. granifera feeding on Pyramimonas sp. was $1.426d^{-1}$, at $20^{\circ}C$ under a 14 : 10 h light-dark cycle of $20{\mu}Em^{-2}s^{-1}$, while the growth rate (i.e., phototrophic growth) under similar light conditions without added prey was $0.391d^{-1}$. With increasing mean prey concentration, the ingestion rate of A. granifera feeding on Pyramimonas sp. increased rapidly, but slightly at the concentrations ${\geq}306ngCmL^{-1}$ (7,649 cells $mL^{-1}$). The maximum ingestion rate of A. granifera feeding on Pyramimonas sp. was 0.97 ng C $predator^{-1}d^{-1}$ (24.3 cells $grazer^{-1}d^{-1}$). The calculated grazing coefficients for A. granifera feeding on co-occurring Pyramimonas sp. were up to $2.78d^{-1}$. The results of the present study suggest that A. granifera can sometimes have a considerable grazing impact on the population of Pyramimonas spp.
