Growth and Rotting Characteristics of Lentinus edodes Isolates in Korea and Their Adaptability to Bag-Culture Using Oak Sawdust (우리나라 표고의 생장(生長), 부후특성(腐朽特性) 및 톱밥배양(培養)에 관(關)한 연구(硏究))

  • Studies were made to find out the mycelial growth and rotting characteristics of 91 Lentinus edodes isolates collected or hybridized in Korea and abroad, and their adaptability to bag-culture. The results are as follows. 1. There were little differences in mycelial growth at PDA and oak (Quercus mongolica) sawdust-based medium, and rotting among high-, mid- and low temperature types of L. edodes isolates. But, the fruit yield of high-temperature type was much higher than those of the other two types. The fruit yields of the isolates in the same strain group were apparently different. 2. From the sawdust-based culture of 91 isolates, FRI 221, FRI 208 and FRI 169 were selected as excellent strains in yield and Quality, showing fruiting of 157g, 152g and 119g per 1kg-medium, respectively. 3. Attempts to culture in various media with different substrates resulted in almost same fruit yields, and the yield was proportional to medium weights as 2.5kg-medium showed 2.5 times more yield than 1kg-medium. Fruit yields were different according to incubation periods and the period of little more than 100days was best. 4. When the correlation analyses among mycelial growth, rotting ability, yield and fruit-shape normality were made with the 91 isolates, mycelial growth at sawdust-based medium showed highly positive correlation with that at PDA, while fruit yield was negatively correlated with mycelial growth in PDA and sawdust-based medium.

Effects of Simulated Acid Rain on Growth and Physiological Characteristics of Ginkgo biloba L. Seedlings and on Chemical Properties of the Tested Soil -III.   • One-year-old seedlings of Ginkgo biloba were treated with various simulated acid rains(pH 2.0, pH 3.0, pH 9.0 and pH 5.0) to examine the effects of simulated acid rain on the chemical properties of the tested soil. The seedlings were grown in a pot($4500cm^3$)containing one of three different soils(nursery soil, mixed soil and sandy soil). Simulated acid rain was made by diluting sulfuric and nitric acid solution($H^1SO^4$ : $HNO^3$ =3 : 1, V/V) with tap water and tap water(pH 6.4), and treated by 5mm each time for three minutes during the growing seasons(April to October 1985). Acid rain treatments were done three times per week to potted seedlings by spraying the solutions. The chemical properties of potting media were compared among three soil types as well as among the various pH levels. The results obtained in this study were as follows : 1. Exchangeable calcium and magnesium contents and base saturation of the soil decreased with decreasing pH levels of acid rain, and their decreasing rates were as follows : sandy soil was the highest, followed by mixed and nursery soils, However, exchangeable aluminum content rather increased as the pH levels decreased. 2. Available phosphate in the soil decreased as the pH levels of acid rain decreased. Its content increased in nursery soil, compared with those before acid gain treatment, Gut decreased in mined and sandy soils. 3. Soil sulfate and nitrate contents increased remarkably as the pH levels decreased, and the only significant difference in the sulfate was found among the pH levels. Soil sulfate content was the highest in nursery soil, followed by mixed and sandy soils.

Selection of Filamentous Cyanobacteria and Optimization of Culture Condition for Recycling Waste Nutrient Solution (폐양액 활용을 위한 Filamentous Cyanobacteria의 선발 및 최적배양)

  • The discharge of waste nutrient solution from greenhouse to natural ecosystem leads to the accumulation of excess nutrients that results in contamination or eutrophication. There is a need to recycle the waste nutrient solution in order to prevent the environmental hazards. The amount and kind of nutrients in waste nutrient solution might be enough to grow photosynthetic microorganisms. Hence in the present study, we examined the growth and mass cultivation of cyanobacteria in the waste nutrient solution with an objective of removing N and P and concomitantly, its mass cultivation. Four photosynthetic filamentous cyanobacteria (Anabaena HA101, HA701 and Nostoc HN601, HN701) isolated from composts and soils of the Chungnam province were used as culture strains. Among the isolates, Nostoc HN601 performed faster growth rate and higher N and P uptake in the BG-II ($NO_3{^-}$) medium when compared to those of other cyanobacterial strains. Finally, the selected isolate was tested under optimum conditions (airflow at the rate of $1L\;min^{-1}$. in 15 L reactor, initial pH 8) in waste nutrient solution from tomato hydroponic in green house condition. Results showed to remove 100% phosphate from the waste nutrient solution in the tomato hydroponics recorded over a period of 7 days. The growth rate of Nostoc HN601 was $16mg\;Chl-a\;L^{-1}$ in the waste nutrient solution from tomato hydroponics with optimum condition, whereas growth rate of Nostoc HN601 was only $9.8mg\;Chl-a\;L^{-1}$ in BG-11 media. Nitrogen fixing capacity of Nostoc HN601 was $20.9nmol\;C_2H_4\;mg^{-1}\;Chl-a\;h^{-1}$ in N-free BG-11. The total nitrogen and total phosphate concentration of Nostoc HN601 were 63.3 mg N gram dry weight $(GDW)^{-1}$ and $19.1mg\;P\;GDW^{-1}$ respectively. Collectively, cyanobacterial mass production using waste nutrient solution under green house condition might be suitable for recycling and cleaning of waste nutrient solution from hydroponic culture system. Biomass of cyanobacteria, cultivated in waste nutrient solution, could be used as biofertilizer.

A Study on the Frequency of Occurrence of the Aortic Dissection using CT (CT 검사에서 대동맥박리(aortic dissection)의 발생빈도에 관한 고찰)

  • Purpose: Aortic Dissection is very dangerous, prognostic disease, which the bloodstream flow out of the true lumen of the aorta by the bursting of aortic intima resulting in a rapid dissociation of inner and outer layer from the media. It is difficult to diagnose aortic dissection clinically by normal X-ray. This study was to investigate the occurrence frequency by age and number of patients who are identified to be aortic dissection by CT (Computed Tomography) scan. Materials and methods: We investigated the trend of yearly fluctuation, gender, age, and department of clinical research of the 112 patients who conducted CT scan in C- University Hospital for two years from January 2005 to December 2006. The MIP and SSD which reconstructed CT image and the VRT image were obtained for the accurate observation. The result was investigated by comparing normal X-ray and CT scan. Results and Conclusion: 1. The yearly check of 112 patients conducted CT scan showed 37 people (41.9%) in 2005, and it was increased to 65 (58.1%) in 2006 by 1.4 times. 2. The gender distribution of patients given a CT scan showed 45 males (40.1%), and female 67 (59.9 %). The aortic dissection patients were 9 (20%) out of 45 males, 21 (31.3%) out of 67 females and women were 1.6 times more than men. Women are also 1.5 times more than men in the number of examinee. 3. The age distribution of patient's who conducted CT scan revealed that there was no patient under 30 years old while 88.3% of all patients were through 41 to 80 years old. The higher the age was, the higher the occurrence of aortic dissection was. The difference in the occurrence frequency of age was statistically significant (p<0.01). 4. The departments that requested CT scan were the emergency department 46 (41.1%), circulatory internal medicine 37 (33.0%), chest surgery 13 (11.6%), and others 6 (14.3%). The combined ratio of emergency medicine and circulatory internal medicine was 74.1% of all. The results show that the aortic dissection is a very dangerous disease whose patients visit mainly via the emergency room. 5. The aortic dissection patients had normal X-ray readings in 22 (73.3%) out of 30, and only 8 (26.7 percent) are abnormal in the X-ray diagnosis. Therefore, the CT scan needs to be enforced in order to assess accurately the disease of aortic dissection.

Hormonal Effect on the Callus Induction from Perennial Weeds (다년생잡초(多年生雜草)로부터 Callus 유도(誘導)와 생장조절제(生長調節劑)의 영향(影響))

  • This experiment was conducted to evaluate effect of various hormones on callus induction, and on plantlet formation on various media, and to detect of Londax [Methyl 2-[[[[[(4,6-dimethoxy pyrimidin-2-yl) amino] carbonyl] amono] sulfonyl) methyl] benzoate] and Basta[Ammonium-(3-amino-3-carboxy-propyl)-methyl phosphinate] on callus growth and reaction of succinate dehydrogenase in callus against TTC, using various species such as Eleocharis kuroguwai, Cyperus serotinus, Oryza sativa (samgangbyeo) and Echinochloa crusgalli P. Beauv. var. caudata Kitagawa. The optimal levels of 2,4-D in MS medium seems to be different among species tested, 2.0 ppm for rice and E. crusgalli, 1.0 ppm for Eleocharis kuroguwai, and 4.0 ppm for C. serotinus derived callus from shoot-tip. In case of combination of 2,4-D with BA, 1.0 plus 0.3 ppm appeared the most appropriate level to induce callus from rice and E. kuroguwai, and I.0 plus 0.1 ppm for C. serotinus and E. crusgalli. When 2,4-D treated with TIBA, 1.0 plus 0.5 ppm appeared the most appropriate rates to induce callus derived from seeds of rice, E. crusgalli, seeds of C. serotinus and E. kuroguwai, 1.0 plus 0.3 ppm for shoot-tip of C. serotinus. Positive reaction of succinate dehydrogenase against TTC was observed regardless of calli and herbicides tested, indicating that they all are alive, and these herbicides were not able to kill the calli tested within the short period of time 20 hrs treatment. Regardless of plant species used, the rate of plantlet formation from callus was very low. However, some plantlet formed from E. crusgalli at 0.8 ppm of 2,4-D plus 8.0 ppm of kinetin, and from E. kuroguwai at 1.6 ppm of 2,4-D plus 16.0 ppm of kinetin, showing effectiveness of 2,4-D with kinetin mixture treatment. No callus was induced from C. serotinus treated with Basta from $10^{-6}M$ to $10^{-3}M$. In general, rice was the least susceptible to Basta among plant species tested, followed by E. crusgalli, and E. kuroguwai. In Londax treatment, rice showed the least inhibition rate in callus growth. Callus was induced from rice even at $10^{-3}M$ of Londax. However, $10^{-3}M$ of Londax completely inhibited callus induction from the test species. Rice showed most tolerant to both herbicides, indicating the existence of different responses among plant species.

A Study on Dental Health Awareness of High School Students (고교생들의 구강보건의식 행태에 관한 연구 -경기 이천시 소재 고교-)

  • In this research, dental health awareness of 3rd graders of high schools in Icheon-city was investigated in order to use the result of this research as the basic data for school dental health program, education direction, and educational purpose. The research was done from Sep. 23. to Oct. 5 and the followings are the results of survey. (1) 85.0% of the students brush their teeth once or twice a day and more female students brush their teeth 3 times a day than male students. (2) 77% of the students had experienced dental caries and 38.0% of the students have periodontal disease, which has lower percentage than cavity. (3) Most of the students can not go to the dentists because they do not have time and 84.0% of the students have been treated in dental clinics so this tells that most of the students had experienced dental treatment. (4) 46.8% of the students had interests in dental health and male students showed more interests than female students and the difference was statistically significant(p<0.05). (5) Most of the students hear the importance and information of dental health from mass media such as TV and radio or from people around. The research shows that most of the students do not get periodic dental checkups. (6) 80.8% of the students had never taught dental health education for improvement of dental health and 76.2% of the students think that the dental health education is needed. There was no difference according to the gender. (7) Most of the students think that the dental health education is needed to prevent cavity and gingiva disease and it shows that more students tend to attend the dental health education. (8) 69.3% of the students think that brushing your teeth is important to maintain good dental health and there was no difference according to the gender. Most of the students weren't aware that periodic dental checkups, right brushing, and using fluoric toothpaste is effective for cavity protection. (9) After looking into the knowledge of the students on dental health, it shows that the knowledge is low and there is no difference according to the gender.

Effect of Cinnamomum camphora Leaf Fractions on Insulin Action (3T3-L1 지방세포에서 녹나무 잎 추출분획물이 인슐린작용에 미치는 효과)

  • In the present study, we screened candidates for enhancing insulin action and secretion from Cinnamomum camphora (CC) fractions, in 3T3-L1 adipocytes and Min6 cells by investigating insulin- stimulated glucose uptake and glucose-stimulated insulin secretion, respectively. CC were extracted by $70\%$ ethanol followed by XAD-4 column chromatography with serial mixture solvents of methanol and water, and the fractional extractions were utilized for determining insulin action and secretion, and $\alpha$-glucoamylase suppressing activity, A significant insulin-stimulated glucose uptake was observed in 3T3-L1 adipocytes, giving 0.5 or $5{\mu}g/mL$ of $40\%\;and\;60\%$ methanol fractions plus 0.2 nM insulin, compared to the treatment of DMSO plus 0.2 nM insulin. The treatments of $40\%\;and\;60\%$ methanol fractions plus 0.2 nM insulin reached the glucose uptake of 10 nM insulin treatment. The $40\%$ methanol fraction increased triglyceride accumulation by stimulating differentiation and triglyceride synthesis similar to pioglitazone, PPAR-$\gamma$ agonist. No inhibition of $\alpha$-glucoamylase activity of CC fractions was observed. They did not modulate the insulin secretion capacity In either low or high glucose media. These results suggest that $40\%$ methanol fraction contains a potential insulin sensitizer to have a similar function of PPAR-$\gamma$ agonist. Crude CC extract may improve glucose utilization by enhancing insulin-stimulated glucose uptake without elevating glucose stimulated insulin secretion.

Effects of Cytokinin and Auxin on Organ Formation in Leaf Scale Tissue of Allium Sativum L. (Cytokinin과 Auxin이 Allium Sativum L.의 인엽배양(鱗葉培養)에서 기관(器官)의 분화(分化)에 미치는 영향(影響))

  • In order to determine the effects of cytokinin and auxin on organ formation from tissue of garlic cloves, leaf blades and basal tissues contained meristem of garlic (Allium sativum L.) cloves harvested in 1979 (old cloves) and 1980 (new cloves) were explanted on a MS medium contained various levels of BA ($N^6$-benzyl amino purine), NAA (naphthalene acetic acid), and 2, 4-D (2, 4-dichlorophenoxyacetic acid). And some of the new cloves were explanted on a media contained BA and NAA after chilling treatment at $4^{\circ}C$ for 10, 20, 30 and 40 days. 1. In a culture of leaf blades of old cloves, shoots were differentiated on a medium supplemented with 2mg/l of BA and NAA. 2. Callus was grown as a quite straw-coloured globular mass on a medium contained 0.2 or 2mg/l 2.4-D. 3. As subcultures of globular calli, shoots and roots were differentiated on a medium contained 2mg/l BA and 0.5 or 1 mg/l NAA, whereas no shoots was shown on a conterol. 4. Shoots were differentiated in a culture of leaf blades of new cloves, but they were not in an old cloves in control, and better effect was shown on a medium contained 2mg/l BA and 1mg/l NAA. However shoots were no differentiated from leaf blades chilled at $4^{\circ}C$ for 30 or 40 days at the same condition. 5. Large numbers of adventitious shoots could be obtained from basal region of garlic cultured on a medium contained 1mg/l BA and 4mg/l NAA, or 2mg/l BA and 2mg/l NAA.

Histological studies on in vitro Propagation of Pulsatilla koreana Nakai (할미꽃 기내증식(器內增殖)에 관(關)한 조직학적(組織學的) 연구(硏究))

  • This study was carried out to investigate the optimal condition for multiple propagation through leaf tissue culture and to apply anther culture techniques to Pulsatilla koreana Nakai breeding. Leaf and anther of Pulsatilla koreana Nakai were cultured on MS, MT, LS and $B_5$ media supplemented with several growth regulators and nitrogen sources under various conditions. For callus induction and differentiation from the Pulsatilla koreana leaf segments were more effective in the combination of zeatin and auxin than auxin alone. The color of the callus was green when treated with IBA alone. Shoot differentiation was more effective when treated with zeatin than auxin alone, especially the best hormoal combination for shoot differentiation was zeatin 1.0mg/l +NAA 0.1mg/l, while 2,4-D inhibited shoot differentiation. The appeared rate of S pollen was 35% in vivo, while that of S pollen by low temperature$(4^{\circ}C)$ pretreatment for 4 days was increased by 53% and the optimum culture time for callus induction from anther was uni-nucleate stage. $B_5$ basal medium supplemented with NAA 0.5mg/l and zeatin 1 mg/l was the most effective on callus formation and the best results of plant regeneration were obtained from combination of NAA 0.5mg/l and zeatin 0.5mg/l in anther culture. $NH_{4}NO_3$ as more effectives as the nitrogen source than $KNO_3$ and the combination with zeatin 2.0mg /L was the best effective. The best combination for plant regeneration in callus induced from anther was $NH_{4}NO_3$ 1650mg/l + $KNO_3$ 3800mg/l + zeatin 2.0mg/l. Ploidy level of anther-derived plants appeared 28% haploid, 47% diploid and the others were triploid, tetraploid and mixploid. In compare with E.S.T, M.D.H and P.X banding patterns were distinguished among callus, haploid and diploid plants in electrophoresis.

Investigation of some harmful bacteria in commercial Kimchi (시판김치 중 유해세균의 조사)

  • Three different commercial Kimchi industry-made packaged, department store-made and ordinary market-made were analyzed for their pH, acidity and microbiological characteristics including certain harmful bacteria on selective media during a storage-fermentation period of 7 days at $2^{\circ}C\;or\;20^{\circ}C$ The pH of all the Kimchi samples wasdecreased from 5.85 to 3.82 and their total acidities increased from 0.2 to 1.18 as the fermentation continued during the storage-fermentation. E. coli and Salmonella sp. of $0\~1101$ cfu/mL were found in the industry-made Kimchi just after purchase time. As the storage-fermentation proceeded, the viable numbers of these bacteria had been reduced in all Kimchi samples tested and no bacteria were detected after 5 days at $2^{\circ}C$ and 2 days at $20^{\circ}C$, respectively. On the other hand, a range of $7.9102\~2.9103$ cfu/mL of Staphylococcus sp. was detected in the department store-made and ordinary market-made Kimchi samples at the purchase time, which was higher than that of the industry-made Kimchi, and this range wasn't reduced during storage-fermentation. The viable number of yeasts in the market-made Kimchi was 2.1103 cfu/mL. These results suggest that some commercial Kimchis were contaminated by some harmful bacteria and that a portion of these bacteria remained alive in the Kimchi, even with high acidity during the edible period.
