Induction of insulin receptor substrate-2 expression by Fc fusion to exendin-4 overexpressed in E. coli: a potential long-acting glucagon-like peptide-1 mimetic

  • Exendin-4 (Ex-4), a peptide secreted from the salivary glands of the Gila monster lizard, can increase pancreatic $\beta$-cell growth and insulin secretion by activating glucagon-like peptide-1 receptor. In this study, we expressed a fusion protein consisting of exendin-4 and the human immunoglobulin heavy chain (Ex-4/IgG-Fc) in E. coli and explored its potential therapeutic use for the treatment of insulin-resistant type 2 diabetes. Here, we show that the Ex-4/IgG-Fc fusion protein induces expression of insulin receptor substrate-2 in rat insulinoma INS-1 cells. Our findings therefore suggest that Ex-4/IgG-Fc overexpressed in E. coli could be used as a potential, long-acting glucagon-like peptide-1 mimetic.

Study on Relationship between a TiO2 Photo-Electrode Fabrication Conditions and Efficiency of a Dye-sensitized Solar Cells (산화티탄 광전극 제작조건과 염료감응 태양전지 효율과의 상관관계 연구)

  • In this work, the effect of addition of acetylacetone on microstructure and quality of nanoporous $TiO_2$ photo-electrodes was studied in dye-sensitized solar cells (DSCs) and structure and electrical properties of fabricated cells were investigated. From the results, the DSCs fabricated with acetyl acetone showed highest photovoltaic performances. This behavior may be attributed to paste agglomeration decrease and interconnection and bonding improvement between $TiO_2$ particles. Furthermore, the most favorable dye absorption time results to be 10h: exceeding this time a decrease in efficiency is observed despite the increasing amount of dye absorption. The $TiO_2$ photo-electrode prepared under the conditions of acetylacetone ratio of 15% and dye absorption time of 10hr showed the better photovoltaic performance ($J_{sc}=12.48mA/cm^2$, $V_{oc}=0.69V$, ff=0.68, ${\eta}=5.86%$).

  • Short ($\tau$=40 $mutextrm{s}$) and high-voltage ($U_{sub}$=2~8 kV) negative substrate bias pulses were used to assist pulsed magnetron sputtering DLC films deposition. Space- and time-resolved probe measurements of the plasma characteristics have been performed. It was shown that in case of high-voltage substrate bias spatial non-uniformity of the magnetron discharge plasma density greatly affected DLC deposition process. By Raman spectroscopy it was found that maximum percentage of s $p^3$-bonded carbon atoms (40 ~ 50%) in the coating was attained at energy $E_{c}$ ~700 eV per deposited carbon atom. Despite rather low diamond-like phase content these coatings are characterized by good adhesion due to ion mixing promoted by high acceleration voltage. Short duration of the bias pulses is also important to prevent electric breakdowns of insulating DLC film during its growth.wth.

  • 다양한 라이프스타일에 따른 유비쿼터스 환경에서 디자인 요소와 감성공학을 결합시키는 상호작용 시스템이 요구되고 있으며 많은 연구가 진행되어 왔다. IT융합기술을 이용하여 감성 디자인을 제공하는 것은 제품 서비스 전략의 중요한 요소이다. 본 논문에서는 상황 기반의 감성 분석을 이용한 자전거 디자인 추천 방법론을 제안하였다. 제안된 방법은 자신의 감성에 부합하는 자전거 디자인을 제공함으로써 이를 얻기 위한 시간과 비용을 줄여주고, 원하는 디자인 스타일에 적용하도록 한다. 감성에 따른 자전거 디자인을 추천하기 위해 협력적 필터링을 사용하여 개인화 서비스를 제공한다. 이를 사용자 인터페이스로 구축하여 논리적 타당성과 유효성을 검증하기 위해 실험적인 적용을 시도하고자 한다.

  • The development of an exclusive 8-axis multi-tasking machine to finish multiple cutting processes by a single piece of power equipment for securing the high-precision machining and high productivity of the series of shafts (a core part of the automotive powertrain that delivers engine power) is needed. The rigidity of the structure must be improved and the weight of the structure must be reduced to develop a multi-tasking machine with high precision and high productivity. In this paper, we perform a static structural analysis of the initial design of the multi-tasking machines and compare the results of the multi-tasking machines improved by the reinforced design and the results of the initial one. According to the results of the structural analysis, the rigidity of the reinforced machine was increased and the overall weight was decreased. Therefore, the productivity was increased.

Changes in Epistemological Beliefs in Chemistry Following Completion of Advanced Chemistry in Science High School Students

  • We studied the effects of science high school students on the change of epistemological beliefs in chemistry and the academic achievement of chemistry by completing 'advanced chemistry'. For seven months from July 2023 to January 2024, 80 first-year students at G Science High School in Gyeongnam were surveyed and analyzed for epistemological beliefs about chemistry before and after classes in advanced chemistry. Chemistry academic achievement was classified by 'upper' and 'lower' levels based on the end-of-semester grades of 'advanced chemistry' in the second semester of the first year and analyzed with the SPSS 28 program. After completing advanced chemistry, the epistemological belief in chemistry increased in the proportion of favorable responses. After completing advanced chemistry, the proportion of favorable responses increased in detailed factors such as 'effort', 'math link', 'outcome', 'reality link', and 'concepts', while the 'visualization' factor decreased. Although completing 'advanced chemistry' positively changed students' epistemological beliefs about chemistry, visual expression showed little contribution to understanding chemical concepts. Based on the above results, we will have to focus on the design of instructors' teaching-learning, such as learner-centered inquiry experiments, creative visual expressions, etc., for successful chemistry teaching-learning.

How Many Korean Science High-school Students Find the Same Scientific Problem as Kepler Found in Optics and Physiology? (얼마나 많은 과학고등학교 학생들이 케플러가 광학과 생리학에서 발견한 과학적 문제를 발견하는가?)

  • The aims of this study are to investigate how Kepler found a scientific problem for the retinal image theory and to investigate how the science high-school students respond when the same situation is applied to them. And their results was compared with general high-school students' results. Kepler found the scientific problem in the eye vision through the critical analysis of contemporary theories of vision, based on his relevant knowledge of optics. When we applied the same situation to the Korean science high school and general high-school students, only a few of science high-school students found the scientific problem as same as Kepler's finding. From the results, it is suggested that in development of creativity teaching material, the situations like Kepler's problem finding need to be included in the programs.
