The Effects of Adipose Derived Stem Cells on Neurogenic Differentiation and Induction of Nerve Regeneration (인체 지방조직에서 유래한 줄기세포의 신경세포 분화능 및 신경재생 유도효과)

  • Using adipose derived stem cells(ASCs), neurogenic differentiation was induced in a mono layered culture medium containing neuronal induction agents. Cells differentiated to the neuronal cells were observed with a inverted microscope and immunofluorecent study. We made a 15 mm long defect in the sciatic nerve of 14 rats and connected a silicone tube to the defect. Then, we mixed neuronal progenitor cells differentiated from ASCs with collagen gel and grafted them to a group of rats(experimental group) and grafted only collagen gel into another group(control group). In 4 and 8 weeks after the graft, histological observation was made. According to the result, the number and diameter of myelinated axons were significantly increased in the experimental group. In addition, the nerve conduction velocity was improved more in the experimental group and neovascularity also increased. Moreover, reaction with S100 and p75 was observed in regenerated nerves in the experimental group, suggesting that the grafted cells were differentiated into supportive cells such as Schwann's cells. In conclusion, this research proved that ASCs can multiply and differentiate into neuronal cells. If they are grafted into nerve defects, the grafted cells are differ entiated into supportive cells such as Schwann's cells and thus contribute to nerve regeneration. Accordingly, the use of adipose tissue obtained easily without the limitation of donor site can be greatly helpful in treating peripheral nerve defects.

Effect of Gyehyuldeung Treatments in Peripheral Nerve Regeneration of Rat (계혈등(鷄血藤)이 Rat의 말초신경 재생에 미치는 효과)

  • Objective : Gyehyuldeung (GHD) has been widely used in oriental medicine for the treatments of cardiovascular and neurological disorders. Thus, its potential facilitatory activity on axonal regeneration was investigated in the rats. Methods: Sprague-Dawley rats were given crush injury at the sciatic nerve and the changes of axon growth after nerve injury on each nerve injury model were investigated with anti-NF-200 antibody, DiI, GAP-43 protein and Cdc2 protein Results : GHD-mediated enhancement of axonal regeneration after crush injury was measured in both qualitative and quantitative ways by immunofluorescence staining with anti-NF-200 antibody and retrograde tracing of fluorescence dye DiI. GAP-43 protein levels were elevated by GHD treatments in the distal injured sciatic nerve and DRG sensory neurons. The neurite outgrowth of DRG sensory neurons was facilitated by GHD treatment when co-cultured with Schwann cells and astrocytes prepared from injured sciatic nerves and injured spinal cord tissues, respectively. It was observed that Cdc2 protein was up-regulated in co-cultured Schwann cells or astrocytes and Cdc2 protein signals were co-localized to a certain extent with those of phospho-vimentin protein. Conclusions : These results suggest that GHD may play a facilitatory role in axonal regeneration by acting on the injured axons and adjacent non-neuronal cells. The current findings may be useful for the development of therapeutic targets through more specific explorations on molecular interactions between herbal components and endogenous factors.

  • Poly(lactic-co-glycolic acid) (PLGA) is a biodegradable synthetic polymer with acceptable mechanical strength and well-controlled degradation rate. Also, it can be easily fabricated into many shapes. Silk fibroin contains powerful bioactive molecules. We fabricated natural/synthetic hybrid films using 0, 10, 20, 40 and 80 wt% of silk fibroin. Schwann cells (SCs) were seeded on PLGA/silk fibroin hybrid films and confirmed the effects of adhesion and proliferation on SCs according to the content of silk fibroin. In this study, we confirmed PLGA/silk fibroin hybrid film containing 40% and 80% of silk fibroin interrupted adhesion and proliferation of SCs. Films containing 10% and 20% of silk, however, provided suitable environment for growth and proliferation of SCs. These results suggest that silk fibroin provides suitables surface to neural cells and its proper content provides proper culture conditions to improve cell adhesion and proliferation.

  • Schwannoma(neurilemmoma) is a benign neoplasm that originates from the Schwann cells. Schwannoma most commonly arises in the soft tissue of the head and neck. Intraoral lesions are uncommon, however, and intraosseous schwannoma are even rare. The most common site of occurrence for these unusual lesions is the mandible. This article documents a case of an intraosseous schwannoma located in the mandibular ascending ramus. The lesion was surgically removed and no radiographic evidence of recurrence was observed after 1 year.

  • A schwannoma is a benign neurogenic tumor derived from Schwann cells. A rare case of a large painful schwannoma in the foot with metatarsal deformity was presented. Due to suspicion of malignancy, amputation had been recommended previously. We report on a rare case of a large forefoot schwannoma causing pain and paresthesia of the forefoot.

  • Granular cell tumor (GCT) is a rare, benign neoplasm of Schwann cell origin. GCT is composed of cells with eosinophilic granular cytoplasm. GCT presents as a solitary painless nodule. Because of their subtle clinical presentation, GCTs are often misdiagnosed. This report of a 47-year-old woman with an auricular GCT serves to highlight that complete excision and histopathological evaluation should be attempted even in apparently benign cases, to ensure complete cure.

Effects of Bogijetong-Tang on Diabetic-peripheral Neuropathy Induced by Streptozotocin in the Mouse (보기제통탕(補氣除痛湯)이 Streptozotocin으로 유발된 당뇨병성 말초신경병증 동물 모델에 미치는 영향)

  • Objectives: Previous reports have shown that Bogijetong-Tang (BJT) is effective in peripheral neuropathy induced by taxol and crush injury. In this study, we researched the effects of BJT on diabetic neuropathy induced by STZ in the mouse. Methods: We performed both in vitro and in vivo experiments to verify the effects of BJT on diabetic neuropathy induced by STZ in mice. Changes in axonal recovery were observed with immunofluorescence staining using NF-200, Hoechst33258, $S100{\beta}$, caspase 3 and anti-cdc2. Proliferation and degeneration of Schwann cells were investigated by immunofluorescence staining and western blot analyses. Results: BJT showed considerable effects on neurite outgrowth and axonal regeneration in diabetic neuropathy. BJT contributed to the creation of NF-200, GAP-43, Cdc2, phospho-vimentin, ${\beta}1$, active ${\beta}1$, ${\beta}3$ integrin, phospho-Erk1/2 protein. Conclusions: Through this study, we found that BJT is effective for enhanced axonal regeneration via dynamic regulation of regeneration-associated proteins. Therefore, BJT had a pharmaceutical property enhancing recovery of peripheral nerves induced by diabetic neuropathy and could be a candidate for drug development after more research.

  • The group of tumors now generally termed Neurilemmoma or Schwanno ma was first described by Verocay, in 1908, and in 1910, he termed them "neurinoma". In 1935 Stout proposed the term neurilemmoma, believing that they arise from nerve sheath or Schwann's cells. Neurilemmoma is an encapsulated, solitary tumor arising in any nerves with Schwann's cell sheath. They may occur at any age and have no preponderance of sex and developing site, however, usually occur in the head and neck area in 25% of cases and have not shown metastasis and are radioresistant. Authors report 5 cases of neurilemmoma surgically surgically removed under the local and general anesthesia at ENT department of SNUH during the last 5 years, with review of the literatures.

  • Granular Cell Tumors(GCT) were originally described as myoblastic myomas. This tumor is believed to originate from Schwann cells based on subsequent scientific investigations. Although it usually appears in the head and neck, it can also appear in other organs as well. Endobronchial granular cell tumors are rather rare and should be differentiated from other common endobronchial diseases such as bronchogenic carcinoma and endobronchial tuberculosis, especially. A case of a patient with an extremely rare condition of endobronchial granular cell tumor concurrent with malignant mediastinal tumor is reported.
