Antioxidative Ability of Lactic Acid Bacteria (유산균의 항산화 효과)

  • The health benefits of friendly bacteria first came to the attention of the general public in 1908, when Dr. Elie Metchnikoff, a Russian biologist, wrote The Prolongation of Life. The longevity may be, in part, due to the antioxidative ability of lactic acid bacteria. However, the antioxidative effect of lactic acid bacteria has been reported only recently. Many kinds of reactive oxygen species can be formed in the human body and in food system, oxidative stress plays a significant pathological role in human disease. Antioxidants are effective for the reduction of oxidation induced by oxygen radicals by scavenging reactive oxygen species. Various synthetic and natural antioxidants have been reported, but there are doubts about the safety and long term effects on health. Antioxidants from natural sources are likely to be found more desirable. An elevated scavenging ability of reactive oxygen species would be a good property for commercially applied lactic acid bacteria. Antioxidant supplement or food containing antioxidants would be greatly applied for the reduction of oxidative damage for human body, and lactic acid bacteria are potentiated candidates for the production of functional foods or natural antioxidant supplements.

Antioxidative and Antimicrobial Activities of Castanopsis cuspidata var. sieboldii Extracts (구실잣밤나무 추출물의 항산화 및 항균 활성)

  • This study was designed to investigate the possible utilization of Castanopsis cuspidata as a source of antiseptic agents. The leaves of C. cuspidata, extracted by 80% ethanol, were sequentially fractionated with n-hexane, dichloromethane, ethylacetate, and n-butanol. In order to effectively screen for a natural preservative agent, we first investigated the antioxidant activities such as DPPH radical scavenging capacity, superoxide radical scavenging capacity, and xanthine oxidase inhibitory activity of the C. cuspidata ethanol extracts and fraction. Using a screening system, we found that the ethylacetate fraction had the strongest antioxidant activity, which followed a dose-dependent manner. The antimicrobial activities were shown in the ethylacetate fraction of C. cuspidata. Among the five fractions, the ethylacetate fraction showed the highest antimicrobial activities against microorganisms tested, which were Bacillus sublitis, Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, Salmonella enteritidis and Salmonella typhimurium. In addition, leaf extracts of C. cuspidata could be suitable for the development of food preservatives.

Effects of herbal-aupuncture solution from vitis labrusca root on LPO, RNS and cytochrome P450 (포도근 약침액이 LPO, RNS 및 Cytochrome P450에 미치는 효과)

  • Objectives : In this study, we determinated protective effects of Vitis labrusca Root herbal-acupuncture solution (VLHA) against atherosclerosis and cardiovascular disease induced by oxidative damage and cytochrome P450 (CYP) induction. Methods : Antioxidant capacity of VLHA were evaluated by the ${\beta}-carotene/linoleic$ acid emulsion system and FeCl2-ascorbic acid stimulted lipid peroxidation in rat liver homogenate, respectively. Scavenging effects of VLHA on ONOO- and NO were measured by DHR 123, DAF-2 assay. Furthermore, we measured inhibitory activity of VLHA on CYP 2E1 in rat liver microsome using by Aniline assay. Results : VLHA exhibited potent inhibition rate(74%) on FeCl2-ascorbic acid induced lipid peroxidation, and revealed a strong scavenging effects on ONOO- and NO. In addition, VLHA showed inhibitory effect on CYP 2E1. Conclusions : The present study concludes that our results suggest that VLHA could be used as a good source in the prevention of atherosclerosis and alcoholic liver injury.

Studies on the Physicochemical Characteristics and NDMA Formation of Orostachys japonicus A. Berger (와송의 이화학적 특성 및 NDMA 생성에 관한 연구)

  • The purpose of this study was to analyze the changes in physicochemical characteristics and NDMA formation of Orostachys japonicus A. Berger according to harvest times. The results showed moisture, carbohydrate, crude fiber and crude protein contents of <$48.5{\pm}1.4%$, $38.2{\pm}1.2%$, $l5.8{\pm}0.4%$ and $13.2{\pm}0.6%$ respectively. The total mineral content was 4,817.1 mg/100 g, where Ca was highest($2,577.0{\pm}3.2$ mg/100 g), followed by K, Mg, Na, P and Fe. The composition amino acid of O. japonicus A. Berger showed high levels of glutamic acid and aspartic acid. And among the five free sugars detected, galactose and glucose were most abundant at $32.2{\pm}0.02$ mg/100 g and $25.3{\pm}0.1O$ mg/100 g, respectively. Upon comparing O. japonicus A. Berger extracts that came from plants harvested during $August{\sim}October$, total phenolic compound($2,703.l{\pm}5.2l{\sim}2,428.0{\pm}3.52$ mg/100 g) and flavonoid($634.2{\pm}2.33{\sim}1,324.6{\pm}1.87$ mg/100 g) contents were higher in the methanol extract than in the water extract. Also, within a reaction system, nitrite scavenging ability and NDMA inhibition were most effective at pH 2.5, and increased in proportion to the extract concentration. Finally, the methanol extract of O. japonicus A Berger harvested during $August{\sim}October$ had the highest nitrite scavenging and NDMA inhibition effects.

  • Antiinflammatory and antioxidative effects of mulberry fruit were investigated by using bioassay screening system. The extract of muberry fruits showed inhibitory effect in phospholipase $A_2$ activity and cyclooxygenase II activity. Among nine varieties of mulberry tree, the antiinflammatory activities of the Shingwangppong, Ficus, Cheongilppong and Keomseolppong were higher than that of the other varieties. Also, antioxidative activity of mulberry fruit was examined by DPPH free radical scavenging method. The radical scavenging activity of the mulberry fruit decreased as following order : Shingwangppong > Keomseolppong > tetraploid Ficus> diploid Ficus.

  • Objective : The objective of this study was to investigate the antioxidative effects of Carthami Flos extract. Methods : Total antioxidant status was examined by total antioxidant capacity(TAC) and total antioxidant response(TAR) against potent free radical reactions. The effect of Carthami Flos extract was examined far details of total phenolic content concentration at which 1,1-dipheny1-2-picrylhydrazyl(DPPH) radical scavenging activity was inhibited, the inhibitory effect on lipid peroxidation, and the effect on reactive oxygen species(ROS) generation. Results : TAC of Carthami Flos extract at the concentration of 5 mg/ml was 1.84 mM Trolox equivalent. 2. TAR of Carthami Flos extract, on the other hand, couldn't be determined due to interference from unidentified compounds. 3. Total phenolic content of Carthami Flos extract at the concentration of 5 mg/ml was 2.01 mM gallic acid equivalent. 4. Concentration of Carthami Flos extract at which DPPH radical scavenging activity was inhibited by 50% was 6.43 mg/ml as compared to 100% by Pyrogallol solution as a reference. 5. The inhibitory effect of the extract on lipid peroxidation was examined using rat liver mitochondria induced by FeS04/ascorbic acid. Carthami Flos extract at the concentration of 10 ms/ml slightly but significantly decreased TBARS concentration. The extract continued to prevent lipid peroxidation in a dose-dependent manner. 6. The effect of Carthami Flos extract on reactive oxygen species(ROS) generation was examined using a cell-free system induced by hydrogen peroxide/FeS04. Addition of 1 mg/ml of Carthami Flos extract significantly reduced dichlorofluorescein(DCF) fluorescence. Carthami Flos extract caused concentration-dependent attenuation of the increase in DCF fluorescence, indicating that the ektract significantly prevented ROS generation in vitro. Conclusion: : Antioxidant efffcts of Carffami ffor extract seem to be due, at least in part, to the prevention offree radical-induced oxidation, fellowed by inhibition of lipid peroxidation.

The effect of Mori Folium extract was examined by measuring total phenolic content, concentration at which 1,1-dipheny1-2-picrylhydrazyl(DPPH) radical scavenging activity was inhibited, inhibitory effect on lipid peroxidation, and the effect on reactive oxygen species(ROS) generation. Results : 1. TAC and TAR of Mori Folium extract at the concentration of 5 mg/ml were 1.61 and 1.24 mM Trolox equivalents, respectively. 2. Total phenolic content of Mori Folium extract at the concentration of 5 mg/Ml was 1.70 mM gallic acid equivalent. 3. Concentration of Mori Folium extract at which DPPH radical scavenging activity was inhibited by 50% was 2.29 m9/m4 as compared to 100% by Pyrogallol solution as a reference. 4. The inhibitory effect of the extract on lipid peroxidation was examined using rat liver mitochondria induced by FeSO$_4$/ascorbic acid. Mori Folium extract at the concentration of 10 mg/ml significantly decreased thiobarbituric acid reactive substances(TBARS) concentration. The extract prevented lipid peroxidation in a dose-dependent 5. Scavenging Effects of Ginkgo biloba Extract on Paraquat Induced Toxicity (Paraquat 유도독성에 대한 Ginkgo biloba Extract의 독성경감효과(I))

  • Reactive oxygen species (ROS) are highly reactive molecules due to their unpaired electron. They have been suspected as one of the major tissue damage inducers in biological metabolic systems. Antioxidant enzymes, such as catalase and superoxide dismutase, could not repair all the oxidative damages resulting from those excessive toxic ROS. It is, therefore, urgent to develop effective antioxidants to relieve from the oxidatire damages. In this study antioxidative effects were investigated by using two flavonoids such as quercetin and naringenin and a flavonoid-rich extract, Ginkgo biloba extract in combination with paraquat that is known as a strong generator of oxygen radicals. The results are summeringed as follows: 1. To assess radical scavenging ability reduction concentrations (IC$_{50}$) of 1,1-diphenyl-2-picrylhydrazine (DPPH) within 15 minutes were measured. The values of the IC$_{50}$ of quercetin and Ginkgo biloba extract were 15.4 $\mu$M and 13.2$\mu$g/ml, respectively. Their radical removing activities showed concentration-dependent manners. 2. In the hydrogen peroxide assay by using PMS-NADH system, quercetin, naringenin and Ginkgo biloba extract led to removing hydrogen peroxide in concentrationdependent manner whose removing abilities at 100$\mu$M or 100 $\mu$g/ml were 75.6, 25.8 and 26.0%, respectively. 3. In the hydrogen peroxide-induced rat blood hemolysis assay all three compounds led to similar effects whose hemolysis inhibition ratios at 100$\mu$M or 100$\mu$g/ml were 68.0, 5.14 and 55.8%, respectively. 4. In the xanthinee oxidase assay by measuring degree of NADH oxidation in the presence of hypoxanthine and xanthinee oxidase, both quercetin and Ginkgo biloba extract showed excellent activities showing 42.8 and 24.2% inhibiting xanthine oxidase activity at 100$\mu$M or 100$\mu$g/ml concentrations, respectively.

Antioxidant Flavone Glycosides from the Root of Pteroxygonum giraldii

  • Two new flavone glycosides, giraldiin A and B, together with three known compounds, annulatin, myricetin 3-O-$\alpha$- L-rhamnopyranoside and gallic acid, were isolated from the ethanol extract of the root of Pteroxygonum giraldii Damm. et Diels. The structures of giraldiin A and B are designated as 3'-($\alpha$-L-arabinopyranosyloxy)-4',5,5',7- tetrahydroxy-3-methoxyflavone and 4'-($\beta$-D-glucopyranosyloxy)-5,5',7-trihydroxy-2',3-dimethoxyflavone, respectively, on the basis of detailed spectroscopic analyses. The free radical scavenging activity of giraldiin A was evaluated by decolouring spectrophotometry of pentamethine cyanine dye (Cy5) with $Fe^{2+}-H_2O_2$ Fenton radical generating system. The results indicated the hydroxyl free radical scavenging activity of giraldiin A (E$D_{50}$ = 23.7 nmol/mL) is higher than that of some known antioxidants such as rutin, puerarin, daidzein and 2,6-di-tertbutyl-4-methylphenol.

Effect of High Speed Drying on Antioxidant Properties of Enzymatic Digests from Citrus By-products and Their Protective Effect on DNA Damage Induced by $H_2O_2$

  • The aim of this study was to evaluate the antioxidant activities of enzymatic digests from citrus by-products (CBPs) prepared by high speed drying (HSD). HSD needs a short time (60 min) for drying and can be used in a commercial scale. Enzymatic digests were prepared from the CBPs using 6 enzymes such as aminoglucosidase (AMG), celluclast, pectinex, termamyl, ultraflo, and viscozyme. Antioxidant activities of AMG digest from CBPs were evaluated by different in vitro models such as 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl, alkyl, $H_2O_2$ scavenging, metal chelating, lipid peroxidation, and comet assays, and exhibited strong activities. The antioxidant compounds were detected by an high performance liquid chromatography (HPLC) coupled on-line to an 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid ($ABTS^+$) scavenging detection system, and found that hesperidin was the key compound. Therefore, the results obtained in this study suggest that HSD is an effective method to transform wet CBPs into dried form, and CBPs are potential source of natural antioxidant.
