등대풀(Euphorbia helioscopia) 용매 추출물의 항산화 및 항균활성 (Antioxidative and Antimicrobial Activities of Euphorbia helioscopia Extracts)

  • 본 연구는 대극과 식물인 등대풀(Euphorbia helioscopia)을 대상으로 에탄올 추출 및 용매분획하여 항산화 활성 및 항균활성을 비교 분석하였으며, 아울러 총 폴리페놀 및 플라보노이드 함량변화를 조사 비교함으로써 식품 저장성이나 안전성을 향상하기 위한 식품첨가제나 보존제로서의 개발 가능성을 알아보고자 하였다. 그 결과 에탄올 추출물 및 순차적 분획물의 항산화 활성은 처리 농도가 높아질수록 항산화 활성도 비례해서 증가되었으며, 특히 순차적 용매분획물 중 에틸아세테이트 분획물에서 항산화 활성이 가장 우수하게 나타났다. 또한 항균활성은 순차적 용매분획물 중에 에틸아세테이트 분획물이 본 실험에 사용된 7종의 균주 모두에서 가장 효율적인 항균활성을 보여 주었다. 이러한 등대풀 에탄올 추출물과 순차적 분획물별 항산화 및 항균활성 결과는 총 폴리페놀 및 플라보노이드의 함량과의 비례 상관관계를 보여주었으며, 한편 총 폴리페놀 함량은 에틸아세테이트 분획물>부탄올 분획물>디클로로메탄 분획물>에탄올 추출물>수용성 분획물>헥산 분획물 순으로, 총 플라보노이드 함량은 디클로로메탄 분획물>에틸아세테이트 분획물>에탄올 추출물>헥산 분획물>부탄올 분획물>수용성 분획물 순으로 나타났다.

삼릉추출물의 멜라닌 생성 저해 효과 (The Inhibitiory Effects of the Scirpi rhizoma on Melanogenesis)

  • 본 연구에서는 한방 천연 미백 소제를 개발하기 위하여 삼릉추출물의 미백 효과에 대하여 연구하였다. 삼릉추출물은 프리 라디칼 소거활성(DPPH)에서 $IC_{50}=638{\mu}g/mL$, superoxide radicals 소거활성은 $IC_{50}=21.7{\mu}g/mL$을 나타냈다. 삼릉추출물의 B16 melanoma 세포에서의 멜라닌 생합성 저해효과는 $100{\mu}g/mL 삼릉추출물을 48 h 처리한 세포에서는 멜라닌 합성량이 27% 감소되었다. 삼릉 추출물은 농도에 비례하여 B16 melanoma 세포내의 타이로시네이즈 활성을 저해하였으며, western blot을 이용하여 타이로시네이즈와 타이로시나아제 관련 단백질(TRP-1)의 발현감소를 확인하였다. 또한, RT-PCR을 이용하여 삼릉추출물이 멜라닌 생성 과정에 관련하는 유전자 발현을 조사한 결과 타이로시네이즈와 TRP-1의 mRNA발현을 억제하는 것을 확인하였다. 그러므로 삼릉추풀물은 항산화 효과와 함께 멜라닌 저해 효과가 우수하여 새로운 미백 화장품으로 응용할 수 있을 것으로 생각된다.

서리태분말을 첨가한 도토리묵의 품질특성 및 항산화성 (Quality Characteristics and Antioxidative Activites of Acorn Mook Added Seoritae Powder)

  • 서리태분말의 첨가량(0%, 2%, 4%, 6%)을 달리하여 도토리묵을 제조하여 품질특성 및 항산화성을 실험하였다. 서리테 분말의 첨가량을 달리하여 도토리묵을 만들어 수분함량, 색도, 조직감, 항산화성, 관능평가 검사를 한 결과는 다음과 같다. 서리태 분말과 도토리분말의 수분함량은 각각 7.48%, 4.36% 였으며, 서리태분말의 첨가량을 달리하여 도토리묵을 만들어 수분함량을 분석한 결과 70.83%~81.77% 였다. 서리태분말의 첨가량이 증가할 수록 명도인 L 값은 감소하는 경향을 보였고, 적색도를 나타내는 a값은 첨가량이 증가할수록 7.29~7.79로 증가하였으며, 황색도를 나타내는 b값은 서리태 분말의 첨가량이 증가할수록 감소하였다. 조직감을 측정한 결과 서리태분말의 첨가량이 증가할수록 도토리묵의 경도(hardness), 부착성(Adhesiveness), 씹힘성(chewiness)은 유의적으로 감소하였고(p<0.05), 응집성(cohesiveness)과 씹힘성(chewiness)도 감소하는 것으로 나타났다. 항산화성 결과 서리태분말을 첨가(2%~6%)한 도토리묵의 총페놀함량은 유의적으로 증가하였으며(p<0.05), 총플라보노이드 함량, DPPH free radical 소거능, ABTS 양이온 라디칼 소거활성은 감소하는 것을 알 수 있었다. 우리는 이상의 결과 전통음식인 묵을 만들 때 항산화 기능을 갖춘 서리태 분말을 첨가하여 도토리묵을 만들 때 기초자료로 활용하는데 도움이 될 수 있을 것이라 생각한다.

Antioxidant Activity and Inhibition of MMP-9 by Isorhamnetin and Quercetin 3-O-$\beta$-D-Glucopyranosides Isolated from Salicornia herbacea in HT1080 Cells

  • Two flavonoids, isorhamnetin 3-O-$\beta$-D-glucopyranoside (1) and quercetin 3-O-$\beta$-D-glucopyranoside (2), from slander glasswort (Salicornia herbacea, Korean name hamcho) were isolated. Antioxidative and matrix metalloproteinase-9(MMP-9) inhibitory effects of these compounds were investigated in HT 1080 cell lines. These compounds suppressed the electron spin resonance (ESR) signal intensity on generation of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical in a free-cellular system. Their scavenging effects on generation of intercellular reactive oxygen species (ROS) also exhibited similar trends with DPPH radical in the free cellular system. Also, a control group combined only with Fe(II)-$H_{2}O_2$ resulted in DNA apoptosis by oxidative stress, whereas treatments with these compounds suppressed radical-mediated DNA damage. Intracellular glutathione (GSH) levels were slightly increased in the presence of compound 1 and 2. Moreover, these compounds led to the reduction of the expression levels of MMP-9 without cytotoxic influence. These results suggest that these compounds have a potential as a valuable natural antioxidant and MMP inhibitor related to oxidative stress. Therefore, these compounds not only can be developed as a candidate for a therapeutic potential but also a source for use as ingredients of health foods or functional foods to prevent metastasis involving MMP-9, closely related to ROS.

양파 열수추출물의 항산화능 및 인공소화후의 항돌연변이 효과 (Antioxidant and Antimutagenic Activities of Hot Water Extract from White and Yellow Onions after Simulated Gastric Digestion)

  • 양파 열수 추출물의 항산화능 및 인공소화 전과후의 항돌연변이 효과를 BHT와 ascorbic acid와 비교하여 조사하였다. 총 페놀 및 플라보노이드의 함량은 노란색 양파 추출물이 횐색 양파 추출물보다 많았으며, 양파 추출물의 과산화수소 소거능은 농도가 증가함에 따라 비례적으로 증가하였다. $\beta-carotene-linoleate\;system$에서 항산화능과 reducing power는 증가되었으나, BHT와 ascorbic acid보다는 낮았다. 양파열수 추출물의 인공소화 후에 IQ와 MNNG로 처리된 Salmonella typhimurium TA80과 TA100에 대하여 항돌연변이 효과가 나타났으며, 노란색 양파가 횐색 양파보다 높게 나타났다. 결론적으로 양파 추출물을 소화시키면 항산화 및 항돌연변이의 효과는 양파의 페놀과 플라보노이드의 성분 변화에 의한 것으로 판단된다.

Antioxidant Activities and Phenolic Compounds Composition of Extracts from Mulberry (Morus alba L.) Fruit

  • The objective of this research was to evaluate the ability of water and ethanol extracts from mulberry fruit (Morus alba L.) to influence the inhibitory activity of angiotensin converting enzyme (ACE) and xanthine oxidase(XOase). The total phenol contents and sixteen phenolic compounds were investigated in water and ethanol extracts. In order to understand the factors responsible for the potent antioxidant and antihypertensive ability of mulberry, it has been evaluated for anti-oxidative activity using Fenton's reagent/ethyl linoleate system and for free radical scavenging activity using the 1,1-diphenyl-2-picryl hydrazyl free radical generating system. The total phenol contents and total of phenolic compounds in ethanol extract showed higher levels than water extract in mulberry fruit six phenolic compounds (chlorogenic acid, narigin, syringic acid, quercetin, naringenin, kampferol) has a higher individual phenolic compound content in the 60% ethanol extraction than 80% ethanol extract. The inhibitory activity on angiotensin converting enzyme (ACE) were highest in 80% ethanol extract (9.0%). Also, activity of xanthine oxidase(XOase) inhibition appeared highest in 80% ethanol extracts and correlated well with the total phenolic content, which was modulated by the concentration of individual phenolic compounds. This result revealed, that strong biological activity was caused by specific phenol compound contents. Utilization of water and ethanol extracts from mulberry fruit are expected to be good candidate for development into source of free radical scavengers and anti-hypertentive activity

상추잎의 Paraquat 내성에 미치는 Nitric oxide의 영향 (Effect of Nitric Oxide on Paraquat-Tolerance in Lettuce Leaves)

  • The protective effect of nitric oxide (NO) on the antioxidant system under paraquat(PQ) stress was investigated in leaves of 8-week-old lettuce (Lactuca sativa L.) plants. PQ stress caused a decrease of leaf growth including leaf length, width and weight. Application of NO donor, sodium nitroprusside (SNP), significantly alleviated PQ stress induced growth suppression. SNP permitted the survival of more green leaf tissue preventing chlorophyll content reduction and of higher quantum yield for photosystem II than in non-treated controls under PQ exposure, suggesting that NO has protective effect on chloroplast membrane in lettuce leaves. Flavonoids and anthocyanin were significantly accumulated in the leaves upon PQ exposure. However, the rapid increase of these compounds was alleviated in the SNP treated leaves. PQ treatment resulted in lipid peroxidation and induced accumulation of hydrogen peroxide ($H_2O_2$) in the leaves, while SNP prevented PQ induced increase in malondialdehyde (MDA) and $H_2O_2$. These results demonstrate that SNP serves as an antioxidant agent able to scavenge $H_2O_2$ to protect plant cells from oxidative damage. The activities of two antioxidant enzymes that scavenge reactive oxygen species, superoxide dismutase (SOD) and catalase (CAT) in lettuce leaves in the presence of NO donor under PQ stress were higher than those under PQ stress alone. Application of 2-(4-carboxyphenyl)-4, 4, 5, 5-tetramethylimidazoline-1-oxyl-3-oxide (PTIO), a specific NO scavenger, to the lettuce leaves arrested SNP mediated protective effect on leaf growth, photosynthetic pigment and antioxidant systems. However, PTIO had little effect on lettuce leaves under PQ stress compared with that of PQ stress alone. The obtained data suggest that the damage caused by PQ stress is in part due to increased generation of active oxygen by maintaining increased antioxidant enzyme activities and SNP protects plants from oxidative stress. From these results it is suggested that NO might act as a signal in activating active oxygen scavenging system that protects plants from oxidative damage induced by PQ stress and thus confer PQ tolerance.

RI-CNG 엔진에서 연료 분사시기에 따른 연소특성에 관한 연구 (Study on Combustion Characteristics with Fuel Injection Timing in a RI-CNG Engine)

  • The RI gasoline engine haying a sub-chamber had a high cycle variation due to the difficulty of the residual gas scavenge in the sub-chamber. To solve this problem and improve the combustion performance of RI engine, we devised a method to inject directly CNG fuel into the sub-chamber. A DI diesel engine of single cylinder was converted into a RI-CNG engine and an electronic control unit for the engine was manufactured. In this study, the combustion characteristics of the RI-CNG engine were investigated with the injection timings and air excess ratios at the load conditions of 50% throttle open rate and 1700rpm. As the results from this study, the RI-CNG engine worked reliably under the condition of the ignitable lean limit of $\lambda=1.7$ by showing the $COV_{imep}$ below about 5%. And the highest thermal efficiency could be obtained in the injection timing that produced the high imep and the low $COV_{imep}$ at the same time. The CO emission concentration indicated very low values and the THC and $NO_x$ showed an opposite pattern. With a view to improving the thermal efficiency and reducing the harmful emissions, the proper control region of the ignition timing and the mixture ratio were nearly ATDC $20^{\circ}\sim50^{\circ}$ and $\lambda=1.4$ respectively.

Antioxidant Effect of Berberine and its Phenolic Derivatives Against Human Fibrosarcoma Cells

  • Berberine (B1), isolated from stems of Coscinium fenestratum (Goetgh.) Colebr, was used as a principle structure to synthesize three phenolic derivatives: berberrubine (B2) with a single phenolic group, berberrubine chloride (B3) as a chloride counter ion derivative, and 2,3,9,10-tetra-hydroxyberberine chloride (B4) with four phenolic groups, to investigate their direct and indirect antioxidant activities. For DPPH assay, compounds B4, B3, and B2 showed good direct antioxidant activity ($IC_{50}$ values=$10.7{\pm}1.76$, $55.2{\pm}2.24$, and $87.4{\pm}6.65{\mu}M$, respectively) whereas the $IC_{50}$ value of berberine was higher than $500{\mu}M$. Moreover, compound B4 exhibited a better DPPH scavenging activity than BHT as a standard antioxidant ($IC_{50}=72.7{\pm}7.22{\mu}M$) due to the ortho position of hydroxyl groups and its capacity to undergo intramolecular hydrogen bonding. For cytotoxicity assay against human fibrosarcoma cells (HT1080) using MTT reagent, the sequence of $IC_{50}$ value at 7-day treatment stated that B1 < B4 < B2 ($0.44{\pm}0.03$, $2.88{\pm}0.23$, and $6.05{\pm}0.64{\mu}M$, respectively). Berberine derivatives, B2 and B4, showed approximately the same level of CAT expression and significant up-regulation of SOD expression in a dose-dependent manner compared to berberine treatment for 7-day exposure using reverse transcription-polymerase chain reaction (RT-PCR) assays. Our findings show a better direct-antioxidant activity of the derivatives containing phenolic groups than berberine in a cell-free system. For cell-based system, berberine was able to exert better cytotoxic activity than its derivatives. Berberine derivatives containing a single and four phenolic groups showed improved up-regulation of SOD gene expression. Cytotoxic action might not be the main effect of berberine derivatives. Other pharmacological targets of these derivatives should be further investigated to confirm the medical benefit of phenolic groups introduced into the berberine molecule.

병포장 갓김치의 항산화 효과에 대한 연구 (Antioxidation Activities of Bottled Mustard Leaf Kimchi during Fermentation)

  • Mustard Leaf Kimchi (MLK) is a traditional fermented Korean vegetable food. This study was conducted to investigate the effects of part vacuum treatment on MLK packed in a glass bottle during fermentation. There have been a few previous studies that examined the chemical and microbial changes during MLK fermentation. However, the major object of this study was to investigate the antioxidative activities of vacuum treated MLK. In this study, the antioxidative activity of vacuum treated mustard leaf Kimchi (VM) and control mustard leaf Kimchi (CM) were examined. VM and CM were fermented at $5^{\circ}C$ for 8 weeks. A model system was designed to evaluate the antioxidative activity of crude chlorophylls and carotenoids (CCC) extracts from Mustard leaf Kimchi. The oxidative reaction of the linoleic acid mixture system at $30^{\circ}C$ in the dark was quantified determining the peroxide value and conjugated dienoic acid content. The effect of the CCC extracts on lipid peroxidation in a rat liver homogenate was examined. Formation of lipid peroxides was estimated by the TBA value, and the CCC extracts were found to inhibit the TBA value. Chlorophyll a and b, and carotenoids, Which are the major components in the CCC extracts of Kimchi were isolated on a DEAE-sepharose CL-6B and Sepharose CL-6B column and TLC. The effects of chlorophyll a and b, caroteins on linoleic acid autoxidation were measured by determining the peroxide value. In addition, their effects on free radical scavenging were investigated by DPPH. In this assay, chlorophyll a showed the greatest antioxidative activity followed by chlorophyll b, and carotenoids. MLK contains a sufficient content of chlorophyll a and b, and carotenoid which have strong antioxidative activities.
